Bioelectrical impedance analysis (BIA) was used to assess body composition in rats fed on either standard laboratory diet or on high-fat diet designed to induce obesity. Bioelectrical impedance analysis predictions of total body water and thus fat-free mass (FFM) for the group mean values were generally within 5% of the measured values by tritiated water (3H2O) dilution. The limits of agreement for the procedure were, however, large, approximately +/- 25%, limiting the applicability of the technique for measurement of body composition in individual animals.
Hindered Diels-Alder adducts have been prepared from 3,6-dibromophencyclone, 2, with cis-1,4-diacetoxy-2-butene, 3; cis-2-butene-1,4-diol, 4; and N-benzylmaleimide, 5. The adduct from the parent phencyclone, 1, with N-benzylmaleimide was prepared for comparison. One- and two-dimensional (1D and 2D) proton and carbon-13 NMR studies (at 7.05 tesla, ambient temperatures), including high-resolution COSY45 and HETCOR (XHCORR) chemical shift correlation spectra, were performed, allowing extensive rigorous assignments for protons and protonated carbons. Substantial anisotropic shielding was seen for the ortho protons of the N-benzyl group in the adducts of 5 with 1 or 2, with these aryl protons resonating at 6.25 ppm (CDCl3) for each adduct. The unsubstituted bridgehead phenyls of all four adducts showed slow exchange limit (SEL) 1H and 13C spectra. Greater shift dispersions for the bridge-head phenyl protons in the adducts from 5 relative to those from 3 or 4 suggested the role of the imide carbonyls for anisotropic contributions or for influences on adduct geometry. Ab initio geometry optimizations were performed at the Hartree-Fock level with the 6-31G* basis set (or the LACVP* basis set for the bromine-containing compounds) for each of the adducts. For the two adducts from benzylmaleimide, separate minima were located corresponding to conformers in which the benzyl group was directed into the adduct cavity (syn) or out of the adduct cavity (anti). Calculated energies and geometric parameters for the adducts are presented, and these suggested a significantly different structure for the dibromo diacetate adduct, in terms of general symmetry and bridgehead phenyl geometries, compared to the other adducts.
The water solubility (Sw) in the temperature range from 283 to 308 K and 1-octanol/water partition coefficient (Kow) for the nonionic form of brominated phenols (BPhs) of varying bromine content (4-bromophenol, 2,4-dibromophenol, 2,4,6-tribromophenol, and pentabromophenol) were measured by the generator-column or shake-flask method and by the high-performance liquid chromatography method, respectively. The temperature dependence of Sw, namely enthalpy of solution (deltaHsolution), was calculated using the van't Hoff Equation. To determine the activity coefficients in water (gammaw) of BPhs from the Sw data, enthalpy of fusion (deltaHfusion) and melting point (Tm) were measured by a differential scanning calorimetry method. The gammaw was used for evaluating data consistency as well as for predicting other physico-chemical properties, such as the Henry's law constant (Hw). The present work systematically reports, to our knowledge for the first time, the experimental values of Sw, Kow, deltaHsolution, deltaHfusion, Tm, and gammaw for BPhs with one to five bromines, and it discusses the effects of bromine content on these physico-chemical properties. Finally, the overall physico-chemical characteristics of BPhs are compared with those of polychlorinated phenols, polycyclic aromatic hydrocarbons, and brominated benzenes with the same solubility level as BPhs.
Persistent organic pollutants (POPs), such as polychlorinated biphenyls (PCBs), some organochlorinated pesticides (OCPs), polybrominated diphenyl ethers (PBDEs), and hexabromocyclododecane (HBCD), were analyzed in yolk sacs of kittiwake (Rissa tridactyla) hatchlings from Kongsfjorden at Svalbard (Norwegian Arctic; 79 degrees N) and from Runde, an island on the coast of Norway (62 degrees N). Retinol (vitamin A), retinyl palmitate, and alpha-tocopherol (vitamin E) were measured in plasma and liver of the hatchlings to examine whether these vitamins were associated with the POPs. Higher levels of PCBs and OCPs were found in hatchlings from Kongsfjorden than in hatchlings from Runde. However, HBCD levels were significantly lower in hatchlings from Kongsfjorden compared to those in hatchlings from Runde. No differences between the two populations of kittiwake hatchlings were found regarding levels of PBDEs. In comparison with seabirds from other European waters and the Canadian Arctic, the kittiwakes seemed to have relatively high levels of PBDEs and HBCD. No effects on morphological variables or vitamin levels by the POPs were revealed, but multivariate regression indicated that liver tocopherol levels may be influenced by POPs.
There are few histochemical techniques for staining rubber in the cells of guayule. With regard to specificity, clarity and retention of the stain we found staining with oil red O or iodine-potassium iodide for bright field, and a combination of oil red O and dansyl chloride for epifluorescence microscopy to be the best methods. The plausible mechanisms of staining are discussed.
A series of thioxanthenes was tested for cytotoxicity to isolated rat hepatocytes, as measured by the loss of an intracellular enzyme (GOT) to the surrounding medium. The relative order of potency was found to be: SKF 10812 greater than cis- = trans-chlorprothixene = N-756A greater than cis- = trans-flupenthixol greater than xanthiol greater than methixene = cis- = trans-clopenthixol = N-716 greater than N-710. The presence of an unsaturated exocyclic bond increased the apparent toxicity as did the presence of a substituent (trifluoromethyl, chlorine, bromine) at the two position of the tricyclic nucleus. The trifluoromethyl substituted thioxanthenes were three to four times more potent than their halogenated analogs, but there were no differences in potency among the halogenated (chlorine or bromine) thioxanthenes. Compounds which had a dimethylaminopropylidene side chain were five to six times stronger in causing enzyme leakage than were their analog which had a hydroxyethylpiperazinylpropylidene side chain. Although only the cis-isomers of each compound are highly active neuroleptics, both isomers were equipotent at causing the efflux of GOT from rat hepatocytes.
The subcooled liquid vapor pressures (P0(L),25S) and aqueous solubilities (Sw,25s) were determined and Henry's law constants (H25s) were estimated for a number of brominated flame retardants (BFRs) at 25 degrees C. The established methods of the gas chromatography-retention time and generator column techniques were used to experimentally determine P0(L),25 and Sw,25 for hexabromobenzene and a series of brominated diphenyl ether (BDE) congeners. The H25 was estimated as the ratio of P0(L)25 to the subcooled liquid aqueous solubility. Values of PL0(L),25 obtained ranged from 0.000000282 Pa (BDE-190) to 0.259 Pa (BDE-3); Sw,25 ranged from 0.00000087 g/L (BDE-153 and BDE-154) to 0.00013 g/L (BDE-15); and H25 ranged from 0.0074 Pa m3/mol (BDE-183) to 21 Pa m3/mol (BDE-15). An increase in the bromine content of polybrominated diphenyl ether congeners resulted in significant decreases Of P0(L),25, Sw25, and H25. A simple four-compartment equilibrium distribution model suggested that the majority of BFRs being released into the environment would reside in soil and sediment and have localized distributions. The model also suggested that lower brominated congeners tend to be somewhat more mobile. Degradative debromination reactions that yield these congeners would mobilize them environmentally, and ultimately affect the fate and distribution of BFRs.
