Effect on microorganisms of volatile compounds released from germinating seeds.

Volatile compounds evolved from germinating seeds of slash pine, bean, cabbage, corn, cucumber, and pea were evaluated for their ability to support growth of microorganisms in liquid mineral salts media lacking a carbon source. Growth of eight bacteria was measured turbidimetrically and of six fungi as dry weight of mycelium. Volatiles caused increased growth of Pseudomonas fluorescens, Bacillus cereus, Erwinia carotovora, Agrobacterium tumefaciens, A. radiobacter, Rhizobium japonicum, Mucor mucedo, Fusarium oxysporum f. conglutinans, Trichoderma viride, and Penicillium vermiculatum but not of Sarcina lutea, Serratia marcescens, Chaetomium globosum, or Schizophyllum commune. Spores of Trichoderma viride showed higher germination in the presence of volatiles. Effects on growth were apparent only during the first 3 or 4 days after planting the seeds. Killed or dried seeds had no effect. The volatiles did not support microbial growth in the absence of nitrogen nor did they supply growth factors. Passing volatiles through KMnO4 or hydrazone reduced growth of the bacteria, indicating that oxidizable organic compounds, primarily aldehydes, were the active components. The volatiles were not absorbed by sterile soil, clay minerals, or water, but they were absorbed by non-steril soil and activated charcoal.

Volatilization of mercury by an iron oxidation enzyme system in a highly mercury-resistant Acidithiobacillus ferrooxidans strain MON-1.

A highly mercury-resistant strain Acidithiobacillus ferrooxidans MON-1, was isolated from a culture of a moderately mercury-resistant strain, A. ferrooxidans SUG 2-2 (previously described as Thiobacillus ferrooxidans SUG 2-2), by successive cultivation and isolation of the latter strain in a Fe2+ medium with increased amounts of Hg2+ from 6 microM to 20 microM. The original stain SUG 2-2 grew in a Fe2+ medium containing 6 microM Hg2+ with a lag time of 22 days, but could not grow in a Fe2+ medium containing 10 microM Hg2+. In contrast, strain MON-1 could grow in a Fe2+ medium containing 20 microM Hg2+ with a lag time of 2 days and the ability of strain MON-1 to grow rapidly in a Fe2+ medium containing 20 microM Hg2+ was maintained stably after the strain was cultured many times in a Fe2+ medium without Hg2+. A similar level of NADPH-dependent mercury reductase activity was observed in cell extracts from strains SUG 2-2 and MON-1. By contrast, the amounts of mercury volatilized for 3 h from the reaction mixture containing 7 microM Hg2+ using a Fe(2+)-dependent mercury volatilization enzyme system were 5.6 nmol for SUG 2-2 and 67.5 nmol for MON-1, respectively, indicating that a marked increase of Fe(2+)-dependent mercury volatilization activity conferred on strain MON-1 the ability to grow rapidly in a Fe2+ medium containing 20 microM Hg2+. Iron oxidizing activities, 2,3,5,6-tetramethyl-p-phenylenediamine (TMPD) oxidizing activities and cytochrome c oxidase activities of strains SUG 2-2 and MON-1 were 26.3 and 41.9 microl O2 uptake/mg/min, 15.6 and 25.0 microl O2 uptake/mg/min, and 2.1 and 6.1 mU/mg, respectively. These results indicate that among components of the iron oxidation enzyme system, especially cytochrome c oxidase activity, increased by the acquisition of further mercury resistance in strain MON-1. Mercury volatilized by the Fe(2+)-dependent mercury volatilization enzyme system of strain MON-1 was strongly inhibited by 1.0 mM sodium cyanide, but was not by 50 nM rotenone, 5 microM 2-n-heptyl-4-hydroxy-quinoline-N-oxide (HQNO), 0.5 microM antimycin A, or 0.5 microM myxothiazol, indicating that cytochrome c oxidase plays a crucial role in mercury volatilization of strain MON-1 in the presence of Fe2+.

Nonhost angiosperm volatiles and verbenone disrupt response of western pine beetle, Dendroctonus brevicomis (Coleoptera: Scolytidae), to attractant-baited traps.

Nonhost angiosperm volatiles and verbenone were tested for their ability to disrupt the response of western pine beetle, Dendroctonus brevicomis LeConte (Coleoptera: Scolytidae), to attractant-baited multiple funnel traps. Verbenone significantly reduced attraction; however, no difference was observed between 4 and 50 mg/24-h release rates. Combinations of six bark volatiles (benzyl alcohol, benzaldehyde, trans-conophthorin, guaiacol, nonanal, and salicylaldehyde), three green leaf volatiles [(E)-2-hexenal, (E)-2-hexen-1-ol, and (Z)-2-hexen-1-ol], and the nine compounds combined did not significantly reduce D. brevicomis response to attractant-baited traps. However, a significant effect was observed when the bark and green leaf volatiles were combined with verbenone. The nine nonhost angiosperm volatiles (NAVs) significantly augmented the effect of both release rates of verbenone, reducing trap catches to levels significantly below that of either release rate of verbenone alone. trans-Conophthorin, a compound reported to have behavioral activity in a number of other scolytids, was not critical to the efficacy of our NAV blend. Our results suggest that the addition of nonhost angiosperm volatiles to verbenone could be important for developing successful semiochemical-based management techniques for D. brevicomis.

[Analysis of residual volatiles in recycled polyethylene terephthalate].

The residual volatiles in recycled polyethylene terephthalate (PET) were analyzed using headspace/GC/MS. Recycled PET samples were made from PET bottles used for beverages, alcohol and soy sauce, and they were recycled in physical recycling plants, chemical recycling plants and superclean-like recycling trials. The physically recycled PET flakes contained small amounts of volatiles such as ethanol, limonene, 2-methyl-1,3-dioxolane, acetone, octanal and nonanal. Most of them originated from foods packed in bottles, and only 2-methyl-1,3-dioxolane was derived from polymer impurities. In contrast, the superclean-like or chemically recycled PET contained no detectable volatiles, like new PET pellets. The PET sheets shaped from physically recycled PET had no detectable volatiles. Not only the chemically and superclean-like recycled PET, but also the physically recycled PET contained no hazardous volatiles. It was concluded that there is no safety concern about volatiles in recycled PET, for the present use.

[Effects of Rhizoma kaempferiae volatile oil on tumor growth and cell cycle of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice].

OBJECTIVE: To evaluate the effects of Rhizoma kaempferiae volatile oil on tumor growth and cell cycle of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice. METHODS: One hundred and five nude mice orthotopically transplanted with MKN-45 human gastric cancer cells were randomly divided into seven groups: untreated group, normal saline-treated group, dissolvant-treated group, cyclophosphamide (CTX)-treated group and high-, medium-, and low-dose Rhizoma kaempferiae volatile oil-treated groups. Corresponding interventions were implemented in each group except the untreated group. The antitumor effects in vivo were evaluated. Cell cycle distribution and apoptosis of MKN-45 human gastric cancer cells were determined by using flow cytometry (FCM). The ultrastructure of MKN-45 gastric cancer cells was observed by a transmission electron microscope. RESULTS: In the high-, medium-, and low-dose Rhizoma kaempferiae volatile oil-treated groups, the growth inhibition rates of gastric cancer were 57.2%, 28.0% and 5.0% respectively, and the gastric cancer cells were arrested at G(0)/G(1) phase. This antitumor effect was dose-dependent. The apoptotic cells occurred more frequently in the high-dose Rhizoma kaempferiae volatile oil-treated group and the CTX-treated group than those in the medium- and low-dose Rhizoma kaempferiae volatile oil-treated groups. CONCLUSION: The Rhizoma kaempferiae volatile oil is an effective composition for growth inhibition of gastric cancer, and its mechanism may be related to regulating the cell cycle and inducing apoptosis.

[Retention of selenium volatility using lime in coal combustion].

For understanding the volatility of selenium, the effect of the contents of exchangeable cations of coal on it, and the retention of selenium using CaO in coal combustion, the sequential chemistry extraction, the fixed bed and circulating fluidized bed (CFB) combustion, X-ray diffraction (XRD) and atomic fluorescence spectrometry (AFS) were undertaken. The results showed that the volatility of selenium was more than 97% in coal combustion at 815 degrees C, and the volatility of selenium was affected by the content of exchangeable cations of coal in low-middle temperature. It was identified that lime can restrain the volatility of selenium. In fixed bed combustion of coal, the retention rates of selenium volatility were between 11.6% and 50.7% using lime. In circulating fluidized bed combustion of coal, partitioning of selenium changed very much in ash of different size fraction between without lime and with lime. Comparing with combustion without lime, the content of selenium in ash from chimney was less than fourth times and that in leaching water from chimney decreased by two orders of magnitude using lime. Retention of selenium volatility using lime is so effective in coal combustion, especially in CFB combustion of coal.

Application of membrane-coupled anaerobic volatile fatty acids fermentor for dissolved organics recovery from coagulated raw sludge.

To investigate the treatment performance of membrane-coupled anaerobic volatile fatty acids fermentor system, the effects of operational parameters for volatile fatty acids production were evaluated through experiments and a mathematical model. The volatile fatty acids recovery ratio was largely affected by the change of hydraulic retention time, reaching its maximum value at 12 hrs. Over the range of hydraulic retention time 8 to 96 hrs, the volatile fatty acids recovery ratio decreased with the increase of hydraulic retention time above 12 hrs, while the ratio of mineralization and gasification increased. Hydraulic retention time and membrane filtration ratio should be maintained less than 1 day and above 0.9, respectively, to attain over 40% of organic materials recovery ratio at 10 days of solids retention time. When the hydrolysis rate constant was 0.01 hr-1, the organic loading rate should be maintained at above 1.0 (kgC/m3/day) to attain over 45% of volatile fatty acids recovery ratio. Based on experimental and simulated results, membrane-coupled anaerobic volatile fatty acids fermentor system was thought to be effective for dissolved organics recovery from coagulated sewage sludge.

[Pharmacological comparison between volatile oil and water extract].

OBJECTIVE: To compare the pharmacological differences between volatile oil and water extract in eight aromatic drugs for resolving dampness. METHOD: Volatile oil and water extract of eight commonly used aromatic drugs for resolving dampness were ig administered to rats respectively, and the effects of volatile oil and water extract on the secretion of gastric juice, volume of gastric mucous blood flow and activity of SOD in gastric mucous membrane were studied. RESULT: It was shown that the effects of aromatic volatile oil in eight drugs on the indexes mentioned above were generally equal or similar to those of water extract, and it was obvious that the volatile oil in some high oil content drugs such as Amonum cardamomum L(Baidoukou), Alpinia katsumadai Hagata (Caodoukou), Amomum villosum Lour (Saren), and Amomum tsao-ko Crevost et Lem (Caogou) had better effects on the secretion of gastric juice and volume of gastric mucous blood flow that those of water extract. CONCLUSION: It has been pharmacologically proved that in using aromatic drugs for resolving dampness, the loss of aromatic (volatile) components should be avoided.

Volatile compounds in virgin olive oil: occurrence and their relationship with the quality.

The stimulation of the human sensory receptors by volatile compounds present in virgin olive oils gives rise to the sensory attributes that describe its delicate and fragrant aroma. The composition of the volatile compounds and their biogenesis is briefly illustrated. Analytical methodologies for evaluating the volatile fraction and the sensory properties of virgin olive oils are elucidated. Compounds responsible for typical flavours are examined and the influence of the main factors on the composition of volatile compounds is discussed. The origin of off-flavours are also described and the consequent changes of volatile composition and of sensory characteristics are analysed. The relationships between volatile compounds and sensory attributes are discussed.

[Mitigation effect of several controlled-release N fertilizers on ammonia volatilization and related affecting factors].

By using static absorption and soil column leaching methods, this paper studied the behaviors of several controlled-release N fertilizers in soil under laboratory conditions. The results showed that under the application rate of 450 mg x kg(-1), total ammonia volatilization from three controlled-release fertilizers decreased by 49.7%, 28.0% and 71.2%, respectively, in comparing with common urea. When the application rate was 600 mg x kg(-1), total ammonia volatilization decreased by 34.6%, 12.3%, 69.9%, respectively. Controlled-release fertilizers could markedly reduce total ammonia volatilization from soil and decrease environment pollution via fertilization. The results also indicated that total ammonia volatilization correlated significantly with soil urease activity, pH value and N leaching rate. The correlation coefficient between total ammonia volatilization and accumulated N leaching rate was 0.9533, and that between total ammonia volatilization and soil urease activity and pH value was 0.9533 and 0.9908, respectively.

[Comparing analysis of components in volatile oils of nutmeg and prepared nutmeg by GC-MS].

OBJECTIVE: To compare components in volatile oils of nutmeg and prepared nutmeg. METHOD: Volatile oil from nutmeg and prepared nutmeg were extracted by vapor distillation. The chemical components in two kinds of volatile oils were determined and indentified by GC-MS. RESULT: The change in quantity and quality of components in volatile oils were observed after processing. 13 new components occurred and 4 components disappeared in volatile oils after processing. The contents of methyleugenol and methylisoeugenol that are active ingredients were increased. The contents of myristicin and safrol that are toxic ingredients in volatile oils were decreased. CONCLUSION: The processing method of nutmeg by soaking with water and roasting with bran is scientific.

How rainfall, relative humidity and temperature influence volatile emissions from apple trees in situ.

Headspace volatiles from apple-bearing twigs were collected in the field with a Radiello sampler during three different diurnal periods over the complete fruit growing season. Analyses by thermal desorption-GC-MS identified a total of 62 compounds in changing quantities, including the terpenoids alpha-pinene, camphene, beta-pinene, limonene, beta-caryophyllene and (E,E)-alpha-farnesene, the aldehydes (E)-2-hexenal, benzaldehyde and nonanal, and the alcohol (Z)-3-hexen-1-ol. The variations in emission of these plant odours were statistically related to temperature, humidity and rainfall in the field. Remarkably, rainfall had a significant positive influence on changes in volatile release during all three diurnal periods, and further factors of significance were temperature and relative humidity around noon, relative humidity in the late afternoon, and temperature and relative humidity during the night. Rainfall was associated consistently with an increase in the late afternoon in terpene and aldehyde volatiles with a known repellent effect on the codling moth, one of the key pests of apple fruit. During the summer of 2003, a season characterized by below-average rainfall, some postulated effects of drought on trees were tested by establishing correlations with rainfall. Emissions of the wood terpenes alpha-pinene, beta-pinene and limonene were negatively correlated with rainfall. Another monoterpene, camphene, was only detected in this summer but not in the previous years, and its emissions were negatively correlated with rainfall, further supporting the theory that drought can result in higher formation of secondary metabolites. Finally, the two green leaf volatiles (E)-2-hexenal and (Z)-3-hexen-1-ol were negatively correlated with rainfall, coinciding well with the expectation that water deficit stress increases activity of lipoxygenase. To our knowledge, this work represents the first empirical study concerning the influence of abiotic factors on volatile emissions from apple trees in situ.

Characterization of volatile sulphur production by pathogenic and non-pathogenic strains of oral Bacteroides.

Marked differences were observed in intermediate sulphur metabolism between non-pathogenic strains of Bacteroides melaninogenicus var melaninogenicus (CP-) and pathogenic Bacteroides melaninogenicus asaccharolyticus (CP+). The CP+ strains, which produced collagenase and protease and caused formation of abscesses when injected subcutaneously into groins of guinea pigs, produced copious amounts of volatile sulphur compounds (VSC) which consisted predominantly of CH3SH and (CH3S)2. Hydrogen sulphide occurred in considerably lesser amounts. CP+ cultures yielded 8-fold more total volatile S, 15-fold more CH3SH and 260-fold more (CH3S)2 during 24 h of incubation in trypticase-yeast extract medium. Whereas H2S accounted for 60 per cent of the total volatile S content of the head-space of CP- cultures, it represented only 8 per cent of the volatile S in CP + systems. Although the CP-organisms did not grow as well as CP +, the differences in concentration of VSC may be only partly related to the disparity in growth rates. When the VSC concentrations were calculated on the basis of equivalent optical density of 1.0, the CP + strains still produced over 3-fold more total volatile S, 6-fold more CH3SH and 100-fold more (CH3S)2. A similar allowance for growth rate suggests that CP-strains may possess a greater potential to produce H2S. Both groups metabolized S-containing amino acids and serine, resulting in appreciable increases in H2S production by CP-. However, the two groups appeared to metabolize the carbon moiety of cystine an cysteine by different pathways. The addition of glucose to the medium depressed total volatile S production by both CP+ and CP-strains, attributable mostly to lower H2S levels. Whereas the omission of yeast extract and charcoal treatment of trypticase did not adversely effect the activity of CP+, it further markedly reduced the capacity of CP-cultures to produce VSC. These results suggest that VSC analysis offers a convenient means of assessing strain differences and pathogenic potential of B. melaninogenicus.

Distribution of moderately volatile trace elements in fine-grained chondrule rims in the unequilibrated CO3 chondrite, ALH A77307.

The concentrations of Ni, Cu, Zn, Ga, Ge, and Se in five, fine-grained chondrule rims in the highly unequilibrated CO3 chondrite ALH A77307 (3.0) have been determined for the first time by synchrotron X-ray fluorescence (SXRF) microprobe at Brookhaven National Laboratory. These elements are especially useful for tracing the role of condensation and evaporation processes which occurred at moderate temperatures in the solar nebula. Understanding the distribution of moderately volatile elements between matrix and chondrules is extremely important for evaluating the different models for the volatile depletions in chondritic meteorites. The data show that the trace element chemistry of rims on different chondrules is remarkably similar, consistent with data obtained for the major and minor elements by electron microprobe. These results support the idea that rims are not genetically related to individual chondrules, but all sampled the same reservoir of homogeneously mixed dust. Of the trace elements analyzed, Zn and Ga show depletions relative to CI chondrite values, but in comparison with bulk CO chondrites all the elements are enriched by approximately 1.5 to 3.5 x CO. The abundance patterns for moderately volatile elements in ALH A77307 chondrule rims closely mimic those observed in the bulk chondrite, indicating that matrix is the major reservoir for these elements. The close matching of the patterns for the volatile depleted bulk chondrite and enriched matrix is especially striking for Na, which is anomalously depleted in ALH A77307 in comparison with average CO chondrite abundances. The depletion in Na is probably attributable to the effects of leaching in Antarctica. With the exception of Na, the volatile elements show a relatively smooth decrease in abundance as a function of condensation temperature, indicating that their behavior is largely controlled by their volatility.

A competitive effect of androgen signaling on male mouse attraction to volatile female mouse odors.

Olfaction plays an important role in animal communication. We hypothesized that males recognize the attractive volatile odors attributed to female reproductive ability. We measured the period during which a male mouse spent sniffing volatile odors from a sham-operated female mouse or an ovariectomized mouse without visual or tactile contact. Intact male mice spent more time sniffing volatile odors from proestrous, estrous or metestrous females than from ovariectomized females. There was no difference in castrated male mice. To investigate the involvement of sexual hormone in this behavior, castrated male mice were treated with 17 alpha-estradiol (E), dihydrotestosterone (DHT), or both. E-treatment did not affect sniffing behavior. Regardless of the estrous stages, DHT-treated castrated males spent less time sniffing the volatile odors from sham-operated than from ovariectomized female mice. Both E- and DHT-treated castrated males spent less time sniffing the volatile odors from proestrous or estrous females than from ovariectomized females. These results suggest that neither androgen nor estrogen is sufficient for reproducing male attraction to volatile female mouse odors, and that androgen signaling has a competitive effect against the attraction.

An assessment of the cerebroprotective potential of volatile anaesthetics using two independent methods in an in vitro model of cerebral ischaemia.

Previous studies using a rat brain slice model of cerebral 'ischaemia' (hypoxia and hypoglycaemia) have suggested that volatile anaesthetics may have cerebroprotective potential. In this study, we tested the cerebroprotective profile of four volatile anaesthetics in this model by two independent means: voltammetric measurement of 'ischaemia'-induced dopamine (DA) release and post-'ischaemic' tissue staining with 2,3,5-triphenyltetrazolium chloride (TTC). 'Ischaemia' caused a characteristic pattern of DA release. Halothane, isoflurane and enflurane did not affect the time from onset of 'ischaemia' to the initiation of DA release. However, all three volatile agents significantly increased (P<0.01, P<0.05, P<0.001, respectively) the time taken for 'ischaemia'-induced DA release to reach maximum and reduced the rate of DA release. Enflurane, unlike halothane or isoflurane, reduced the maximal extracellular DA concentration induced by 'ischaemia' (P<0.01). The effects of sevoflurane were inconsistent. At the higher concentrations used, the volatile anaesthetics frequently changed the character of DA release from monophasic to biphasic, an effect only previously seen in this model with Na(+) channel blockers. 'Ischaemia' also diminished the subsequent level of tissue staining with TTC. When the effects of the volatile agents were analysed by TTC staining, only enflurane showed any cerebroprotective effects and these were limited to the striatum (P<0.01). High concentrations of halothane, isoflurane and enflurane appeared to have some 'toxic' effects, reducing TTC staining in control slices. In summary, we do not find any consistent evidence that volatile anaesthetics are cerebroprotective in this model.

Genetic diversity of Ocimum gratissimum L. based on volatile oil constituents, flavonoids and RAPD markers.

Morphological, chemical and genetic differences of 12 tree basil (Ocimum gratissimum L.) accessions were studied to determine whether volatile oils and flavonoids can be used as taxonomical markers and to examine the relationship between RAPDs to these chemical markers. Eugenol, thymol, and geraniol were the major volatile oil constituents found in Ocimum gratissimum. Xantomicrol and cirsimaritin were the major external flavones. The accessions morphologically described as O. gratissimum var. gratissimum contained eugenol as the major volatile oil constituent, and cirsimaritin as the major flavone. Ocimum gratissimum var. macrophyllum accessions contained thymol as the major volatile oil constituent, and xantomicrol as the major flavone. A distinct essential oil and flavone chemotype (producing geraniol and a mixture of the flavones cirsimaritin, isothymusin, xanthomicrol, and luteolin) was found in an accession genetically more distant from the other two groups when analyzed by molecular markers. The accessions could be divided based on volatile oil constituents into six groups: (1) thymol: alpha-copaene (ot24, ot25, ot26, and ot28); (2) eugenol:spathulenol (ot17, ot63, and ot52); (3) thymol:p-cymene (ot65); (4) eugenol:gamma-muurolene (ot27 and ot29); (5) eugenol:thymol: spathulenol (ot85); and (6) geraniol (ot84). Cluster analysis of RAPD markers showed that there are three groups that are distinct genetically and highly correlated (r=0.814) to volatile oil constituents.

Influence of assimilable nitrogen on volatile acidity production by Saccharomyces cerevisiae during high sugar fermentation.

We analyzed the variability of volatile acidity and glycerol production by Saccharomyces cerevisiae on a large sample of high sugar musts. The production of volatile acidity was inversely correlated with the maximum cell population and the assimilable nitrogen concentration. The higher the nitrogen concentration, the less volatile acidity was produced. An approach to minimize volatile acidity production during high sugar fermentations by adjustment of assimilable nitrogen in musts was investigated in terms of both quantity and addition time. It was found that the optimal nitrogen concentration in the must is 190 mgN.l(-1). The best moment for nitrogen addition was at the beginning of fermentation. Addition at the end of the growth phase had less effect on volatile acidity reduction. We suggest that by stimulating cell growth, nitrogen addition provides NADH in the redox-equilibrating process, which in turn reduces volatile acidity formation.
