Bluetongue virus proteins derived from baculovirus expression vectors have been administered in different combinations to sheep, a vertebrate host susceptible to bluetongue virus, and the neutralizing antibody responses were measured. Vaccinated sheep were subsequently challenged, and the indices of clinical reaction were calculated. The results indicated that the outer capsid protein VP2 alone in doses of greater than 50 micrograms per sheep elicited protection. A dose of ca. 50 micrograms of VP2 protected some but not all sheep. However, when used in combination with ca. 20 micrograms of the other outer capsid protein, VP5, 50-micrograms quantities of VP2 not only protected all the vaccinated sheep but also elicited a higher neutralizing-antibody response. The addition of viral core proteins VP1, VP3, VP6, and VP7, the nonstructural proteins NS1, NS2, and NS3, and the outer capsid proteins VP2 and VP5 did not enhance this neutralizing-antibody response.

A postal survey of the techniques being used for the treatment and control of footrot in sheep flocks between November 1999 and October 2000 was conducted in England and Wales in November 2000. Of the 392 questionnaires circulated, 251 (64 per cent) were returned, and 209 of these were usable. Negative binomial regression analysis indicated that the isolation of bought-in sheep, and the separation and individual treatment of diseased sheep with parenteral antibiotics, foot trimming and topical foot sprays were associated with a significantly lower prevalence of footrot in a flock. In contrast, ewe flocks which were routinely foot trimmed more than once a year had a significantly higher prevalence of footrot. No evidence was found that footbathing a flock reduced the level of footrot, except on the 14 per cent of farms where the penning and race facilities for footbathing were reported by the farmer to be excellent. Vaccination had no significant beneficial effect on the level of footrot in a flock

The purpose of the computer simulation was to use experimentally measured parameters of placental water transfer to compute conceptual water acquisition during the last third of ovine pregnancy and to evaluate the possible role of each of these parameters in the pathophysiology of polyhydramnios. Total conceptual water at birth was almost insensitive to the value of the placental filtration coefficient. It was more sensitive to fetal and maternal placental blood flows, the concentrations of actively transported or metabolically produced solutes in fetal plasma (bicarbonate, fructose, alpha-amino acids, urea, lactate), and the hydrostatic pressure difference across the placental barrier. It was quite sensitive to the NaCl reflection coefficient and permeability. We conclude that the actively transported or produced solutes in fetal plasma constitute a primary driving force. The opposing diffusion gradient of NaCl, and to a much lesser extent that of glucose, are essential to restrain the process. The causes of polyhydramnios in this species are, in order of probability, an increase in the placental diffusion permeability of NaCl, a decrease in the placental reflection coefficient for NaCl, or an increase in the concentration of an osmotically effective solute in fetal plasma, by active transport or metabolic production.
