Furanochromone radical cations: generation, characterization and interaction with DNA.

The radical cations of naturally occurring furanochromones visnagin (VI) and khellin (KH) have been generated and identified for the first time by use of laser flash photolysis and pulse radiolysis techniques. The lifetimes of VI(.+) and KH(.+) are determined as approximately 6 and approximately 35 micros under these conditions, respectively. Direct 308-nm excitation of VI in aqueous buffer at physiological pH results in monophotonic photoionization to generate VI(.+), with a quantum yield of 0.075, which is much higher than that of 8-methoxypsoralen and KH under identical conditions. Though VI(.+) is a more powerful oxidant than KH(.+), both of them react with guanosine mononucleotide (k=1.2x10(9) and 3.8x10(7) dm(3) mol(-1) s(-1), respectively) via electron transfer to give the guanine radical cation. Furthermore, selective oxidation of guanine in single and double strand DNA by VI(.+) was also observed. These novel findings suggest that electron transfer reactions involving furanochromone radical cations may be of considerable importance in furanochromone photochemotherapy.

Synthesis and vasorelaxant activity of new coumarin and furocoumarin derivatives.

We have synthesized a new series of coumarins and furocoumarins and evaluated their vasorelaxant activity in rat aorta rings pre-contracted with noradrenaline or by depolarisation with high KCl. The new furocoumarins relax smooth vascular muscle with a profile similar to that of khellin (a furochromone that directly relaxes smooth muscle) and at a greater potency, suggesting that these compounds have a potential interest for the development of new and more efficient vasodilator drugs.

A pulse radiolysis study of the complexing of furocoumarins with DNA and proteins.

The complexing of furocoumarins with DNA in aqueous solution has been investigated using the fast-reaction technique of pulse radiolysis. Association constants are given for 8-methoxypsoralen, psoralen, khellin, angelicin, 5,7-dimethoxycoumarin, 5-methoxypsoralen and 4'-aminomethyl-4,5',8-trimethylpsoralen. In addition, the complexing of some furocoumarins to the protein, bovine serum albumin, has also been studied.

Measurement of daphnoretin in plasma of freely moving rat by liquid chromatography.

Daphnoretin (7-hydroxyl-6-methoxy-3,7'-dicoumaryl ether), isolated from Wikstronemia indica C.A. Mey. (Thymelaceae), has been reported to induce rabbit platelet aggregation through protein kinase C activation and anticancer activity. In this study, we developed an automated blood sampling system coupled to a simple and sensitive HPLC system to determine plasma concentration of daphnoretin in rats. This method was applied to investigate the pharmacokinetics of daphnoretin in a freely moving rat. Separation of daphnoretin in the rat plasma was achieved using a reversed-phase C18 column (250 mm x 4.6 mm, 5 microm) with a mobile phase of methanol-10 mM NaH2PO4 (adjusted to pH 3.0 with H3PO4) (55:45, v/v), and the flow rate of 1.0 ml/min. The UV detector was set at 345 nm. The automated blood sampling system (DR-II has been applied for blood sampling in a conscious and freely moving rat. The blood samples were centrifuged at 3000 x g for 10 min and the plasma samples were then deproteinized by acetonitrile containing an internal standard (khellin 1 microg/ml). After centrifugation (8000 x g for 10 min), the aliquot of supernatant was injected into the HPLC system for analysis. The concentration-response relationship from the present method indicated linearity over a concentration range of 0.05-1.00 and 1.00-100 microg/ml. Intra- and inter-assay precision and accuracy of daphnoretin fell well within the predefined limits of acceptability (< or = 15%). After daphnoretin (500 mg/kg) was given orally, the maximum concentration was 0.17 microg/ml at the time of 5 min. The oral bioavailability was about 0.15%.

Cytotoxic activity of five compounds isolated from Colombian plants.

Cytotoxic effect of five known compounds, khellin, berberine, lupeol, scopolin, rapanone, obtained from Colombian plants, were determined against human tumor cell lines as an indicator of the potential anticancer activity of these compounds. Berberine and rapanone presented interesting cytotoxicity.

Inhibition of the mutagenicity of 2-nitrofluorene, 3-nitrofluoranthene and 1-nitropyrene by flavonoids, coumarins, quinones and other phenolic compounds.

When 56 flavonoids, 32 coumarins, five naphthoquinones, 12 anthraquinones and five structurally-related compounds were tested for their antimutagenic potencies with respect to mutagenicities induced by 2-nitrofluorene (2-NF), 3-nitrofluoranthene (3-NFA) and 1-nitropyrene (1-NP) in Salmonella typhimurium TA98 distinct structure-activity relationships were detected. First, the tetracyclic nitroarenes 3-NFA and 1-NP were in general more effectively antagonized by potent antimutagenic flavonoids and coumarins than the tricyclic 2-NF, while there were only minor differences with quinones. Secondly, antimutagenicity of natural compounds of plant origin correlated with the aglyconic nature 10 of a total of 15 glycosides were inactive, four flavonoid glycosides exerted antimutagenicity but to a distinctly lower degree than the corresponding aglycones. Thirdly, within flavonoids, coumarins and anthraquinones positive correlations were found between antimutagenic potency and the polarity of a molecule with the existence of an optimum of activity within flavonols and anthraquinones. However, polarity seemed to be unimportant within the chalcone and naphthoquinone series. Among flavonoids, the parent compounds flavone and flavanone were inactive, but all flavones and many flavonoids with phenolic hydroxyl groups exerted antimutagenicity. Antimutagenic potency reached a maximum with the presence of four hydroxyl functions-luteolin, kaempferol-though the position of hydroxyls was also a determinant of antimutagenic potency. Methylation of phenolic hydroxyl groups, however, always reduced antimutagenicity. A carbonyl group at carbon 4 was essential for antimutagenicity: two catechins and anthocyanidins each were inactive. On the other hand, ring C of the flavane nucleus was not essential for antimutagenicity: chalcones and dihydrochalcones were potent antimutagens. Among coumarins, the parent compound showed antimutagenicity against 1-NP and 3-NFA, although dihydrocoumarin, methylcoumarins and compounds with bulky substituents were inactive. Otherwise, antimutagenic activity depended on the presence of polar hydroxyl, amino or carboxyl groups at carbons 3, 4 or 7 but was diminished by interactions of hydroxyl groups vicinal to carbon 7. Again, antimutagenic potencies were reduced by alkylation or acetylation. Among furanocoumarins xanthotoxin exerted strong and bergapten moderate antimutagenicity, while psoralen (except against 3-NFA), isopimpinellin and the furanochromanones visnagin and khellin were inactive. Among anthraquinones, the principles delineated here were valid again, resulting in potent antimutagenicity of most phenolic compounds and inactivity of anthraquinone itself. Among compounds structurally related to anthraquinones, anthrone, acridone and xanthone exerted antimutagenicity, anthrone being the most potent one, while thioxanthone and 9-fluorenone were inactive. All naphthoquinones were potent antimutagens irrespective of the presence of methyl or hydroxyl functions. Plumbagin, 2-methyl-5-hydroxynaphthoquinone, however, showed exceptional antimutagenicity.

Twenty years of sodium cromoglycate treatment: a short review.

Sodium cromoglycate (Intal) was first synthesized from khellin, a naturally occurring plant chromone, by Roger Altounyan and his colleagues in 1965. It was introduced as a therapeutic agent in 1968 and marked a new era in the management of asthma. Numerous studies on the use of sodium cromoglycate in the treatment of asthma have since been published. Both short-term and long-term controlled studies indicate unequivocally that sodium cromoglycate is of significant clinical benefit in 60-70% of asthmatic children and adults. Clinical experience during the last 20 years has indicated that it should be the drug of first choice in the short-term treatment of asthma provoked by irritants, allergens and exercise, as well as in chronic asthma. Side effects are usually minor and sodium cromoglycate is the safest therapeutic agent in current use for the treatment of asthma.

Synthesis and antiarrhythmic activity of new benzofuran derivatives.

Various 5-aminobenzofuran derivatives were prepared from khellin and screened intravenously in the dog for their potential antiarrhythmic activity against ouabain-induced ventricular arrhythmia and in the Harris test. From systematic structural variations it was found that two methoxy groups in positions 4 and 7 on the benzofuran ring, a tertiary aminoethoxy side chain in position 6, and a N-methylurea group in position 5 led to the most active compounds. These were then tested orally in the Harris test in the dog. The two long-acting derivatives N-[4,7-dimethoxy-6-(2-pyrrolidinoethoxy)-5-benzofuranyl]-N'-methylurea (8j) and N-[4,7-dimethoxy-6-(2-piperidinoethoxy)-5-benzofuranyl]-N'-methylurea (8m) showed advantages when compared to quinidine and disopyramide and have been selected for further studies.

Synthesis and antidiabetic activity of some new furochromonyl-2,4-thiazolidinediones.

A new series of furochromone-2,4-thiazolidinedione derivatives (VIIa-h) was prepared by Knoevenagel reaction of substituted-2,4-thiazolidinediones (VIa-h) with Khellin-2-carboxaldehyde (IV). The prepared compounds were tested for their insulinotropic activities in INS-1 cells. Compounds VIId and VIIf (at lower concentration; 1 microgram/ml) were able to increase insulin release in the presence of 5.6 mmol/l glucose. Both these compounds (VIId and VIIf) and VIIg increased glucose uptake in NIH-3T3 cells. Thus these 3 compounds should be tested for antidiabetic effects in vivo.

The chemical investigations on the ripe fruits of Ammi visnaga (Lam.) Lamarck growing in Turkey.

Two furanochromones and one furanochromone glycoside were isolated from the fruits of Ammi visnaga (L.) Lam. They were identified as khellin, visnagin, and khellol glycoside by interpretation of spectral analyses. Quantitative determination of furanochromones in A. visnaga (L.) ripe fruits from Hatay region (Turkey) was carried out by ultraviolet spectrophotometry and gas chromatography. In addition, photochemical properties of furanochromones and chemical composition of essential oil were determined.

Quantitative assessment of epidermal melanogenesis in C3H/Tif hr/hr mice treated with topical furocoumarins and UVA radiation.

We report quantitative data on epidermal melanogenesis by established and new furocoumarins. The ears and dorsal skin of pigmented hairless mice were treated for 12 d with compounds in ethanol, at equi-optical concentrations, and exposed to subphototoxic doses of ultraviolet A. Increased pigmentation was observed with 6,4,4'-trimethylangelicin > psoralen > 8-methoxypsoralen > 5-methoxypsoralen > 4,4',5'-trimethylazapsoralen = bergamot oil. Assessment of melanocyte numbers and morphology in epidermal sheet dihydroxyphenylalanine preparations showed that 6,4,4'-trimethylangelicin was the best compound with 536 ear melanocytes/mm2 +/- 15 SEM compared with 46 +/- 4 in controls. Psoralen induced 297/mm2 +/- 33, compared with its methoxy derivatives with ranges between 200 and 240/mm2.6,4,4'-trimethylangelicin had a striking effect on dorsal skin with 462 +/- 18 melanocytes/mm2 compared to less than 80/mm2 in all other ultraviolet A treatment groups. Khellin, 5-GOP and ultraviolet A only and all non-ultraviolet A controls had no effect. Melanogenesis was associated with increased dendricity, melanocyte size, especially with 5-methoxypsoralen, and giant melanocytes were noted with some treatments. The potency of 6,4,4'-trimethylangelicin, which does not form DNA interstrand crosslinks, may be related to its high DNA binding constant. Our data may be useful in the selection of compounds to treat vitiligo.

The actions of some vasoactive polypeptides and their antagonists on the anococcygeus muscle.

1 The action of three polypeptides, bradykinin, substance P and eledoisin known to inhibit vascular smooth muscle has been examined on the anococcygeus muscle of the rat, cat and rabbit.2 In the atonic rat muscle, bradykinin and substance P had little or no effect on tone but eledoisin produced a sustained dose-related contraction which could be abolished by phentolamine (1 muM) and is, therefore, probably an indirect sympathomimetic effect. On the motor response to field stimulation of adrenergic nerves, bradykinin had no effect whereas both substance P and eledoisin reduced this response. The mechanism of action was further analysed with eledoisin by examining its effect on the response to noradrenaline. Eledoisin did not alter the dose-response curve to noradrenaline and its inhibitory action is likely, therefore, to be presynaptic.3 In the rat anococcygeus muscle in which the tone was raised by guanethidine or carbachol, bradykinin and substance P reduced this tone whereas eledoisin continued to exert a motor action. Compared with substance P the inhibitory effect of bradykinin appeared at lower concentrations (threshold 0.01 mug/ml), developed more rapidly and the size of the response was greater.4 The effect of bradykinin on the tonically contracted cat and rabbit anococcygeus muscles was examined in addition to that of the rat. In all three species bradykinin caused inhibition and the magnitude of the response was equal to the maximum effect of inhibitory nerve stimulation. None of the peptides affected the inhibitory response to nerve stimulation itself.5 The effects of three substances, hesperitin, khellin and apiin, reported in other tissues to antagonize the action of bradykinin were examined both on the inhibitory response to bradykinin and to field stimulation. None of them was able to inhibit either response, although they reduced tone when given by themselves. During these experiments it was found that ethanol antagonized the inhibitory response to field stimulation.6 The possibility that bradykinin or some related peptide might play a part in the inhibitory response to nerve stimulation in the anococcygeus is discussed.

Calcium antagonist and antiperoxidant properties of some hindered phenols.

1. The calcium antagonist and antioxidant activities of certain synthetic and natural phenols, related to BHA (2-t-butyl-4-methoxyphenol), were evaluated in rat ileal longitudinal muscle and in lipid peroxidation models respectively. 2. Compounds with a phenol or a phenol derivative moiety, with the exception of 2,2'-dihydroxy-3,-3'-di-t-butyl-5,5'-dimethoxydiphenyl (di-BHA), inhibited in a concentration-dependent manner the BaCl2-induced contraction of muscle incubated in a Ca(2+)-free medium. Calculated pIC50 (M) values ranged between 3.32 (probucol) and 4.96 [3,5-di-t-butyl-4-hydroxyanisole (di-t-BHA)], with intermediate activity shown by khellin < gossypol < quercetin < 3-t-butylanisole < BHA < nordihydroguaiaretic acid (NDGA) < 2,6-di-t-butyl-4-methylphenol (BHT) and papaverine. 3. The Ca2+ channel activator Bay K 8644 overcame the inhibition sustained by nifedipine, BHA and BHT, while only partially reversing that of papaverine. 4. BHA, BHT, nifedipine and papaverine also inhibited in a concentration-dependent fashion CaCl2 contractions of muscle depolarized by a K(+)-rich medium. This inhibition appeared to be inversely affected by the Ca(2+)-concentration used. 5. The inhibitory effects of nifedipine, papaverine, BHA and BHT were no longer present when muscle contraction was elicited in skinned fibres by 5 microM Ca2+ or 500 microM Ba2+, suggesting a plasmalemmal involvement of target sites in spasmolysis. 6. Comparative antioxidant capability was assessed in two peroxyl radical scavenging assay systems. These were based either on the oxidation of linoleic acid initiated by a heat labile azo compound or on lipid peroxidation of rat liver microsomes promoted by Fe2+ ions. Across both model systems,di-t-BHA, NDGA, BHT, di-BHA, BHA and quercetin ranked as the most potent inhibitors of lipid oxidation, with calculated pICso (M) values ranging between 7.4 and 5.7.7. Of the 32 compounds studied only 15 phenolic derivatives exhibited both antispasmogenic andantioxidant activity. Within this subgroup a linear and significant correlation was found betweenantispasmogenic activity and antioxidation. These bifunctional compounds were characterized by the presence of at least one hydroxyl group on the aromatic ring and a highly lipophilic area in the molecule.8. Di-t-BHA is proposed as a lead reference compound for future synthesis of new antioxidants combining two potentially useful properties in the prevention of tissue damage after ischaemia reperfusion injury.

Guidelines for the treatment of vitiligo.

At present, there is no universally effective drug for vitiligo therapy; there are, however, various therapeutic modalities that may be beneficial. Therapeutic regimens used to treat vitiligo include psoralens and ultraviolet A light (PUVA), topical corticosteroids, fluorouracil locally applied with skin abrasion, a variety of surgical techniques to transplant autologous melanocytes from pigmented skin to nonpigmenting areas, a new photochemotherapeutic regimen using oral khellin with UVA phototherapy, and a recently proposed treatment with oral phenylalanine in combination with UVA exposure. PUVA and topical corticosteroids are the 2 most frequently used modalities. The use of effective sunscreens with a high sun protection factor (SPF) is of help in preventing the vitiliginous areas from burning and normal skin from becoming tanned. Cosmetic camouflage is also useful to disguise the white areas of skin. Finally, depigmentation should be considered in patients with greater than 50% cutaneous involvement who fail to respond or are unwilling to undergo treatment. The selection of patients for therapy should take into consideration the patient's motivation, the psychological impact of the disease and the clinical presentation of vitiligo, and should weigh the risks and benefits of prolonged therapy.

Asthmatic drugs and competitive sport. An update.

Almost all asthmatics are prone to asthma triggered by moderate to severe exercise. Fortunately there are a number of pharmaceutical agents now available which can prevent and/or reverse exercise-induced asthma (EIA) and allow many asthmatics to participate in vigorous physical activities with minimum respiratory disadvantage. Regular exercise is an accepted part of the management of asthma and EIA can now be controlled so successfully that a number of elite sportspersons, in almost all types of sporting events, are asthmatic. This control of EIA, which is essential if asthmatics are to participate safely, requires that the patient and his/her doctor initiate a strategy to manage the disease during sport and other physical activities. In recent years the mortality and morbidity from asthma have been increasing and this has indicated the need to improve patient care. One of the most important innovations aiming to improve the control and treatment of asthma has been the recent development of the 6 point asthma management plan which is a strategy to simplify and optimise the long term management of asthma. It aims to improve the quality of life of most asthmatics and more importantly, prevent deaths due to asthma. Because antidoping controls operate in many high performance sports it is essential that the EIA management plan rely on those medications which are permitted. The list of allowable drugs is in continual flux as new ones are added and others are challenged on the grounds of possible ergogenicity. All aerosol beta 2-agonists except fenoterol, the khellin derivatives, theophylline, ipratropium bromide and the aerosol corticosteroids are currently permitted. Some nonasthmatic athletes who are aware of the improved performance of asthmatic athletes when using pre-exercise medication have been known to take antiasthma medication in the hope that it might improve their performance. Current evidence indicates, however, that the permitted medications are not ergogenic and do not give the asthmatic any advantage over the nonasthmatic athlete but merely removes the respiratory disadvantage under which he/she competes.

Epoxidation of some natural furocoumarins and furochromones using gamma-ray.

Epoxidation of imperatorin (1a) with m-chloroperbenzoic acid (mcpba) under the irradiation of gamma-ray gave a mixture of epoxide 3a and dioxofuran 4a. Whereas, alloimperatorin (1b) under the same condition obtained the epoxide 3b as a sole product. On the other hand, it was successfully epoxidized xanthotoxin (1c) with mcpba under the same condition as above. In addition, visnagin (2a) was epoxidized to give a mixture of epoxides 5a, 6a. While, khellin (2b) gave the epoxide 5b, no other products were observed.

Stimulation of furanochromone accumulation in callus cultures of Ammi visnaga L. by addition of elicitors.

In order to check the possibility of producing secondary metabolites, in vitro cultures of A. visnaga callus were established. The best growth of A. visnaga callus was obtained on Murashige and Skoog medium (MS) containing 6-benzyladenine (BA) and alpha-naphtaleneacetic acid (NAA). The study was concentrated on the induction of production of secondary metabolites by exposing callus to abiotic elicitors: benzo(1,2,3)-thiadiazole-7-carbothionic acid S-methyl ester (BION) and a suspension of silica (SiO2) and biotic elicitors: autoclaved lysates of Enterobacter sakazaki and scleroglucan. GC analysis indicated that not-elicited callus of A. visnaga grown in darkness accumulated 2 times more visnagin than the one which was grown under a 16-h photoperiod. The highest accumulation of visnagin was observed in the callus culture elicited with scleroglucan or BION. Scleroglucan induced also the accumulation of khellin in A. visnaga callus. The presented work shows that biosynthesis of pharmacologically important secondary metabolites in A. visnaga cultures could be stimulated by application of elicitors.

Antagonism of calcium-induced contractions by some non-specific spasmolytics in depolarized smooth muscles.

The effects of papaverine MgCl2, cocaine, DNP, KCN and khellin on responses of some rabbit and rat tissues to CaCl2 were studied in vitro in a depolarizing medium. Guinea pig taenia coli preparation was used for comparison. In rabbit tracheal chain and vas deferens and guinea pig taenia coli preparations all spasmolytics shifted the concentration-response curves of CaCl2 to the right without affecting the maxima or slopes. In rat tracheal chain and vas deferens preparations all spasmolytics shifted the concentration-response curves of CaCl2 to the right. Furthermore all agents (except cocaine in tracheal chain preparations) depressed the maximum responses. The slopes were unaffected in either preparations. The initial competition and subsequent noncompetition observed in certain tissues is discussed in the light of the reported poor capacity of some tissues to retain Ca++ and the absence of releasable firmly bound Ca++ (11).
