Aryl isocyanate, carbodiimide, and isocyanide prepared from carbon dioxide. A metathetical group-transfer tale involving a titanium-imide zwitterion.

Carbon dioxide can be readily converted quantitatively and under mild conditions into the aryl isocyanate and symmetrical carbodiimide via a metathetical reaction involving a zwitterionic titanium imide (nacnac)Ti=NAr(CH(3)B(C(6)F(5))(3)) (nacnac(-) = [ArNC((t)Bu)](2)CH, Ar = 2,6-(i)Pr(2)C(6)H(3)). The metathetical process to generate isocyanates allows also for facile formation of sterically demanding aryl isocyanide, by a deoxygenation route. Labeling studies using enriched (13)CO(2) are also described.

Synthesis and characterization of an isocyanate functionalized polyhedral oligosilsesquioxane and the subsequent formation of an organic-inorganic hybrid polyurethane.

Organic-inorganic hybrids are an important class of new materials that offer improved thermal and mechanical properties over normal polymers. They may be produced by either the sol-gel route or through the use of inorganic compounds possessing reactive functional groups. Polyhedral oligosilsesquioxanes (POSS) are completely defined molecules of nanoscale dimensions that may be functionalized with reactive groups suitable for the synthesis of new organic-inorganic hybrids. Here we report the synthesis and characterization of a novel POSS possessing eight isocyanate groups via the hydrosilylation of octakis(hydridodimethylsiloxy)octasilsesquioxane (Q8M8H) and m-isopropenyl-alpha,alpha'-dimethylbenzyl isocyanate (m-TMI). The suitability of this new macromer to the synthesis of a organic-hybrids has been explored by forming a new type of highly cross-linked polyurethane elastomer via reaction of the macromer with poly(ethylene glycol) using dibutyltin dilaurate catalyst.

Dynamic kinetic asymmetric cycloadditions of isocyanates to vinylaziridines.

The first examples of the use of racemic vinylaziridines in a Pd-catalyzed dynamic kinetic asymmetric transformation have been examined. Optimization studies of the Pd-catalyzed addition of vinylaziridines to isocyanates revealed that the chiral ligand between trans-1,2-diaminocyclohexane and 2-diphenylphosphino-1-naphthoic acid is superior to that involving 2-diphenylphosphino benzoic acid. Surprisingly, high ee's required the use of an acid whose pKa was about 4.7 +/- 0.1 as a cocatalyst. Both acetic acid and hydroxybenzotriazole meet this requirement. Less electrophilic isocyanates (e.g., benzyl, p-methoxyphenyl) gave higher ee's than more electrophilic ones (phenyl or benzoyl). Both N-benzyl and N-arylaziridines react well to give good yields and ee's, whereas N-tosylaziridines gave lower ee's. A 1,1-disubstituted aziridine led to the formation of a tertiary C-N bond with ee's comparable to the formation of the secondary C-N bond. The products were easily reduced almost quantitatively to the sensitive imidazolidines which can be readily hydrolyzed to the vicinal diamines. The reactivity pattern is consistent with a Curtin-Hammett situation wherein the enantiodiscriminating event is the cyclization of a rapidly equilibrating dynamic pi-allyl palladium intermediate.

Nickel-catalyzed cycloaddition of alkynes and isocyanates.

A mild and general route for preparing 2-pyridones from isocyanates and diynes is described. Ni imidazolyidene complexes were used to mediate cyclizations between both internal and terminal diynes with aryl and alkyl isocyanates. In addition, the efficacy of this protocol allows for the preparation of a fused seven-membered pyridone and for three component cyclizations.

Ruthenium-catalyzed [2 + 2 + 1] cocyclization of isocyanates, alkynes, and CO enables the rapid synthesis of polysubstituted maleimides.

Intermolecular [2 + 2 + 1] cocyclization of isocyanates, alkynes, and CO (1 atm) proceeded smoothly in the presence of a catalytic amount of Ru3(CO)12 (3.3 mol %) in mesitylene at 130 degrees C for 3 approximately 42 h to give a variety of polysubstituted maleimides in excellent yields with high selectivity. The reaction may involve an azaruthenacyclopentenone intermediate derived from oxidative cyclization of an isocyanate and an alkyne on an active ruthenium species.

Polycyclic heteroaromatics from reactions of acylbenzotriazoles with aryl isocyanates

N-Acylbenzotriazoles react with aryl isocyanates to form, depending on the type of acyl group, compounds based on five different classes of polycyclic heteroaromatics. Higher alkanoyl-, acetyl-, acetoacetyl-, aroyl-, and cinnamoylbenzotriazoles yield, respectively, derivatives of quinoline, pyrimidino[5,4-c]quinoline, benzo[b]-1,8-naphthyridine, phenanthridine, and indolo[2, 3-b]quinoline by incorporating 3, 3, 4, 2, and 2 molecules, respectively, of the isocyanate per acylbenzotriazole molecule.

Biradicals/zwitterions from thermolysis of enyne-isocyanates. Application to the synthesis of 2(1H)-pyridones, benzofuro[3,2-c]pyridin-1(2H)-ones, 2,5-dihydro-1H-pyrido[4,3-b]indol-1-ones, and related compounds.

Thermolysis of benzannulated enyne-isocyanates 13 and enyne-isocyanates 36 and 37 promoted the cycloaromatization reactions to generate in situ O,4-didehydro-2-hydroxyquinolines and O,4-didehydro-2-hydroxypyridines, respectively, as reactive intermediates. These cycloaromatized intermediates could be captured either as biradicals and/or as zwitterions depending on the nature of the substituent at the alkynyl terminus. The intermediate derived from cycloaromatization of 13a bearing a phenyl substituent could be regarded as biradical 14, which then abstracts hydrogen atoms from gamma-terpinene leading to 2(1H)-quinolinone 15. Alternatively, the same intermediate could also be regarded as zwitterion 14', which then undergoes an initial hydride abstraction from gamma-terpinene followed by protonation to produce 15. The presence of a 2-phenylethyl substituent in 13b and 37a or a 2-methylphenyl substituent in 37b also allowed the resulting intermediates to be captured intramolecularly either as biradicals or as zwitterions, producing 2(1H)-quinolinone 19, 2(1H)-pyridone 39, and benzopyranopyridine 43, respectively. On the other hand, with a 2-methoxyphenyl, a 2-(dimethylamino)phenyl, or a 3-methoxypropyl substituent, the chemical behavior of the cycloaromatized adduct could be best accounted for in terms of a zwitterionic intermediate leading to benzofuro[3,2-c]quinolin-6(5H)-one (20), 5,11-dihydro-11-methyl-6H-indolo[3,2-c]quinolin-6-one (25), benzofuro[3,2-c]pyridin-1(2H)-one 44, 2,5-dihydro-2,5-dimethyl-1H-pyrido[4,3-b]indol-1-one 46, and related compounds. Interestingly, thermolysis of 37f bearing a 2-(methoxymethyl)phenyl substituent at the alkynyl terminus produced the unexpected benzopyranopyridine 56 as the major product in a process involving the cleavage of the bond between the methoxyl oxygen and the adjacent methylene carbon. The efficiency and selectivity of the cycloaromatization reaction could also be enhanced by the introduction of 1.1 to 10 equiv of dimethylphenylsilyl chloride to the reaction mixture to capture the resulting zwitterion.

High nucleophilicity of cyclic amidocarbene toward aryl isocyanates, new approach to spiro[azetidinone-4,3'-indolinone] derivatives.

The nucleophilic addition of beta-lactam-4-ylidenes 2, a type of ambiphilic cyclic amidocarbene, to aryl isocyanates has been studied and their application in organic synthesis has been demonstrated. Thermolysis of spiro[beta-lactam-4,2'-oxadiazolines] 1 in the presence of aryl isocyanates afforded both N-lactam and O-lactam substituted spiro[azetidine-2-one-4,3'-indole-2'-one] derivatives 5 and 6 in the total yield of 65-86%. Upon hydrolysis, products 5 and 6 were converted into spiro[azetidine-2-one-4,3'-indole-2'-one] 9 that was analogous to known biologically active compounds.

Empty level structure in phenyl and benzyl isocyanates.

The energies of the lowest-lying anion states of phenyl (C6H5N=C=O) and benzyl (C6H5CH2N=C=O) isocyanates have been determined experimentally in the gas phase for the first time using electron transmission spectroscopy (ETS), and their localization properties have been evaluated using HF/6-31G, MP2/6-31G*, and B3LYP/6-31G* calculations. The lowest-lying anion state of phenyl isocyanate, mainly of benzene ring character but with some contribution also from the N=C=O pi-system, lies at significantly higher energy than that of other benzenes substituted by pi-functionals, such as benzaldehyde or styrene. The scaling with the use of suitable empirical equations of the virtual orbital energies (VOEs) for orbitals with predominantly pi*(ring) character calculated for the neutral-state molecules leads to vertical attachment energies (VAEs) which closely correspond to those determined experimentally, whereas those calculated for the predominantly pi*(CO) and pi*(NC) orbitals (3rd and 4th LUMO, respectively) are significantly different from the corresponding measured values notwithstanding the fact that the calculations reproduce the shortening of the N=C and C=O double bonds.

Ultrathin coatings from isocyanate-terminated star PEG prepolymers: layer formation and characterization.

In this study we present the preparation of thin and ultrathin coatings from six-arm star-shaped isocyanate-terminated prepolymers on amino-functionalized silicon wafers. The backbone of the stars is a statistical copolymer of ethylene oxide and propylene oxide in the ratio 80:20 (Star PEG). Film preparation by spin coating from aqueous THF resulted in a variety of film morphologies that are determined by the water content of the solvent. Water is indispensable for activation of the isocyanate-terminated stars in solution and for proper cross-linking of the coatings on the substrate. This cross-linking results in a dense network of PEG chains on the substrate linked via urea groups with a mesh size of the network that corresponds to the arm length of the stars. Layer thickness variations between 3 and 500 nm revealed a strong dependence of the contact angle with water on the layer thickness which is explained by the chemical composition of the coatings. Due to the high functionality of the star-shaped prepolymers, free amino groups remain in the films that were detected by fluorescence microscopy after reaction with 4-chloro-7-nitrobenzofurazan (NBF). To test the system for the ability to prevent unspecific interaction with proteins, adsorption of fluorescence-labeled avidin was examined with fluorescence microscopy. For layer thicknesses between 3 and 50 nm, no protein adsorption could be detected.

Cobalt-mediated macrocyclizations. Facile synthesis of 2-oxo pyridinophanes via [2 + 2 + 2] cycloaddition of alpha, omega-diynes and isocyanates.

[reaction: see text] Cobalt-mediated [2 + 2 + 2] cycloaddition of alpha,omega-diynes and isocyanates provides a direct approach to macrocyclic 2-oxopyridinophanes. This macrocyclization process, which proceeded most efficiently with aliphatic isocyanates, was conveniently performed at a moderate temperature (85 degrees C) without irradiation or syringe-pump addition.

[reaction: see text] A novel one-pot procedure for the three-component coupling reaction of isocyanates, aldehydes, and dienophiles (IAD reaction) has been developed. Condensation of isocyanates and aldehydes and subsequent Diels-Alder reactions with electron-deficient dienophiles furnishes endo-selective amino-substituted cyclohexenes in good yield.

Rhodium-catalyzed chemo-, regio-, and enantioselective [2 + 2 + 2] cycloaddition of alkynes with isocyanates.

[reaction: see text] We have developed a cationic rhodium(I)/modified-BINAP complex-catalyzed chemoselective [2 + 2 + 2] cycloaddition of alkynes with isocyanates leading to a wide range of 2-pyridones. This method was successfully applied to the chemo-, regio-, and enantioselective synthesis of axially chiral 2-pyridones from unsymmetrical alpha,omega-diynes, bearing an ortho-substituted phenyl at one terminal position, and alkyl isocyanates.

Determination of isocyanic acid in air.

A method is presented for the determination of isocyanic acid (ICA), HNCO, in air samples as a di-n-butylamine (DBA) derivative. The method is based on sampling in midget impinger flasks containing 10 ml of 0.01 mol l-1 DBA in toluene. Quantification was made using liquid chromatography (LC) and electrospray mass spectrometry (MS) monitoring positive ions. The instrumental detection limit for the LC-MS was 10 fmol of ICA-DBA. ICA was generated by thermal decomposition of urea. A standard solution containing the DBA derivatives of ICA was prepared by collecting the emitted ICA in an impinger flask containing DBA. ICA in the reference solution was characterised by LC and time-of-flight (TOF) MS and quantified by LC chemiluminescent nitrogen detection (LC-CLND). The instrumental detection limit for the LC-CLND was 1 ng of nitrogen. ICA was emitted during thermal degradation of PFU resins and polyurethane (PUR) lacquers, from car metal sheets. ICA was the most dominant isocyanate and in PUR coating up to 8% of the total weight was emitted as ICA and for PFU resins up to 14% was emitted as ICA. When air samples were collected in an iron foundry during casting in sand moulds with furan resins, concentrations of ICA in the range 50-700 micrograms m-3 were found in the working atmosphere.

Selective activation of human heat shock gene transcription by nitrosourea antitumor drugs mediated by isocyanate-induced damage and activation of heat shock transcription factor.

Treatment of cultured human tumor cells with the chloroethylnitrosourea antitumor drug 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) selectively induces transcription and protein synthesis of a subset of the human heat shock or stress-induced genes (HSP90 and HSP70) with little effect on other stress genes or on expression of the c-fos, c-myc, or beta-actin genes. The active component of BCNU and related compounds appears to be the isocyanate moiety that causes carbamoylation of proteins and nucleic acids. Transcriptional activation of the human HSP70 gene by BCNU is dependent on the heat shock element and correlates with the level of heat shock transcription factor and its binding to the heat shock element in vivo. Unlike activation by heat or heavy metals, BCNU-mediated activation is strongly dependent upon new protein synthesis. This suggests that BCNU-induced, isocyanate-mediated damage to newly synthesized protein(s) may be responsible for activation of the heat shock transcription factor and increased transcription of the HSP90 and HSP70 genes.

A new method for the generation of standard atmospheres of organo isocyanates.

A new and versatile method for the generation of standard atmospheres of several organo isocyanates based on permeation technique has been developed. Several membranes were screened for integrity, compatibility and permeability of these isocyanates. Silicone membrane was found to be the best membrane for the construction of permeation devices. Two types of permeation are described: liquid phase and gas phase. The liquid phase permeation tubes are recommended for generation of high concentrations (ppm) and gas phase permeation is preferred for generation of atmospheres with low concentrations (ppb). Several simple designs for construction of permeation devices are described. The permeation of isocynates were evaluated gravimetrically and were shown to be constant, provided the temperature and the flow rate of air over the permeation tube are maintained constant.

Respiratory variables and exposure-effect relationships in isocyanate-exposed workers.

Respiratory variables in 95 isocyanate workers and 37 control workers were compared. The exposed workers had a slightly higher frequency of cough and shortness of breath than the controls and a significantly lower frequency of family history of asthma, hay fever, and bronchitis. The isocyanate workers had slightly lower baseline lung function than the control workers but demonstrated significantly larger declines in their pulmonary function over the work shift. Both groups showed some intraday and intraweek variation in lung function. The changes in lung function over the work shift varied with different job categories, the largest changes occurring in finishing-area workers. A gradation of response was observed when exposure was categorized as nil, low, or high, but no exposure-effect relationships could be demonstrated by regression analysis of either area or personal results.
