5-Endo-dig electrophilic cyclization of 5-alkynyl-2'-deoxyuridines with N-iodosuccinimide or N-bromosuccinimide in acetone at room temperature gives 3-(2'-deoxy-beta-D-ribofuranosyl)-5-halo-2,3-dihydrofuro[2,3-d]pyrimidin-2-ones that usually precipitate from the reaction mixture (86-74%).
Hypobromite ion, BrO(-), is an effective alpha-nucleophile that reacts rapidly with activated phosphorus(V) and sulfonate esters. The parent acid rapidly oxidizes organic sulfides and aryloxide ions. At pH 10-11 BrO(-) and HOBr coexist in water and are potentially useful as decontaminants of chlorosulfide blister agents and the phosphonyl nerve agents. Bis(dialkylamide)hydrogen dibromobromates are well characterized, stable, solids which rapidly form HOBr-BrO(-) in mildly alkaline water. Reactions of 4-nitrophenyl diethyl phosphate and phosphonate, which are simulants of the phosphonofluoridate nerve agents, and of 4-nitrophenyl tosylate, with BrO(-) are rapid (t(1/2) = 60-700 s) with 0.1 M BrO(-), under conditions in which oxidations of organic sulfides are too fast to be followed by conventional methods.
Chemical reactions are known to behave differently, depending upon their local environment. While the interactions with neighboring molecules may alter both the kinetics of chemical reactions and the overall equilibrium conversion, we have performed simulations of the latter. The particular environment that we address is the vapor-liquid interface, since only a few, limited studies have explored the influence of an interface on equilibrium reaction behavior. Simple dimerization reactions are modeled, as well as more complex multicomponent reactions, using the reactive Monte Carlo (RxMC) simulation technique. We find that the conversion of a reaction can be markedly different at an interface as compared to the bulk vapor and liquid phases, and these trends are analyzed with respect to specific intermolecular interactions. In conjunction, we calculate the surface tension of the reacting fluids at the interface, which is found to have unusual scaling behavior, with respect to the system temperature.
[reaction: see text] Multifunctional palladium catalysis is utilized in the one-pot stereocontrolled synthesis of tetrasubstituted alkenes. The homogeneous palladium dihalide catalyst utilized for bromo-/chloroallylation of alkynes is then reused in situ for a subsequent Suzuki cross-coupling reaction.
[reaction: see text] Multifunctional palladium catalysis is utilized in the one-pot stereocontrolled synthesis of tetrasubstituted methyl ketones and enynes. The homogeneous palladium dihalide catalyst utilized for the bromo-/chloroallylation of alkynes is reused in situ for subsequent Wacker-Tsuji oxidation or Sonogashira cross-coupling.
[reaction: see text] An efficient copper-catalyzed amination of aryl bromides with primary alkylamines was developed that uses commercially available diethylsalicylamide as the ligand. This amination reaction can be performed at 90 degrees C in good yield. A variety of functional groups are compatible with these reaction conditions. Preliminary results show that this reaction can be carried out under solvent-free conditions with comparable yields.
Synthesis of 2'-deoxyguanosine-C8 adducts (dG-C8 adducts) with mutagenic/carcinogenic heterocyclic amines (HCAs) was achieved via the Buchwald-Hartwig arylamination reaction. By using tris(dibenzylideneacetone)dipalladium (Pd(2)dba(3)) and 9,9-dimethyl-4,5-bis(diphenylphosphino)xanthene (xantphos) with a cesium carbonate (Cs(2)CO(3)) base at a reaction temperature of 100 approximately 120 degrees C, we obtained derivatives of dG-C8 adducts with 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-6-methyldipyrido[1,2-a:3',2'-d]imidazole (Glu-P-1), 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1), 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) in 69% approximately 97% yield from the cross-coupling of an 8-bromodeoxyguanosine derivative. In the case of PhIP, it was found that dimethyl sulfoxide (DMSO) was the critical solvent for the arylamination reaction. Subsequent deprotection of the resulting dG-C8 adduct derivatives yielded authentic samples of dG-C8 adducts with HCAs. The dG-C8-PhIP adduct was further converted into a suitably protected phosphoramidite derivative for automated DNA synthesis. Synthesis of oligonucleotides wherein PhIP adducted on each G within a triple G sequence in codon 869 (TCC GGG AAC) of rat Apc genes was performed with a modification in the coupling time and deprotection procedures.
Histone acetylation is important in chromatin remodelling and gene activation. Nearly all known histone-acetyltransferase (HAT)-associated transcriptional co-activators contain bromodomains, which are approximately 110-amino-acid modules found in many chromatin-associated proteins. Despite the wide occurrence of these bromodomains, their three-dimensional structure and binding partners remain unknown. Here we report the solution structure of the bromodomain of the HAT co-activator P/CAF (p300/CBP-associated factor). The structure reveals an unusual left-handed up-and-down four-helix bundle. In addition, we show by a combination of structural and site-directed mutagenesis studies that bromodomains can interact specifically with acetylated lysine, making them the first known protein modules to do so. The nature of the recognition of acetyl-lysine by the P/CAF bromodomain is similar to that of acetyl-CoA by histone acetyltransferase. Thus, the bromodomain is functionally linked to the HAT activity of co-activators in the regulation of gene transcription.
A series of structurally characterised, monovalent metal-halide complexes incorporating neutral amidine and guanidine ligands is reported. N,N'-diphenylbenzamidine reacted with copper(I) chloride to afford the bis-ligand complex [CuCl(PhC[NPh][NHPh])2]2 (1), that exists as a chlorine bridged dimer in the solid state, with a non-symmetrical distribution of NH...Cl interactions within the 'Cu2Cl2' metallacycle. In contrast, only one equivalent of the guanidine, Me2NC[NiPr][NHiPr] (2), is coordinated in the copper(I) iodide complex [CuI(Me2NC[NiPr][NHiPr])]2 (3), which was also isolated as the dimer with bridging halide atoms. The molecular structure of the bicyclic guanidine, 1,3,4,6,7,8-hexahydro-2H-pyrimido[1,2-alpha]pyrimidine (hppH), is reported, revealing a hydrogen bridged dimer with extensive delocalisation throughout the ligand framework. Coordination of hppH to lithium chloride afforded the dimeric bis-ligand complex [LiCl(hppH)2]2 (4) in which each hppH molecule interacts with a different chlorine atom of the central 'Li2Cl2' core of the molecule via NH...Cl hydrogen bonding. In contrast the 2:1 ligand to metal complex is formed with silver(I) chloride to afford AgCl(hppH)2 (5), a unique example of a monomeric, three-coordinate silver chloride supported by nitrogen-based ligands. The series of mixed ligand complexes [CuX(hppH)(PPh3)]n (6, X = Cl, n= 1; 7, X = Br, n= 2; 8 X = I, n= 2) have also been synthesised and structurally characterised, allowing comparisons of the relative coordinating behaviour of hppH and PPh3 as neutral donors at copper(I) centres to be made.
Exposure of 1-alkynyl[p-(trifluoromethyl)phenyl](tetrafluoroborato)-lambda3-bromanes to 2-mercaptobenzimidazole or benzothiazole in dichloromethane at 0 degrees C under argon resulted in a domino Michael addition-carbene rearrangement-cyclization reaction to produce directly tricyclic heterocycles in high yields, whereas the reaction with 2-mercaptobenzoxazole afforded 1-alkynyl sulfides.

1. The oxidation of NN-dimethyl-p-phenylenediamine (DPD) by inorganic oxidants and by caeruloplasmin was studied. Some experiments were also made with NNN'N'-tetramethyl-p-phenylenediamine (TPD). 2. E(mM) (550) of the first free radical oxidation product of DPD (DPD(+)) was 9.8 and E(mM) (563) of the corresponding product of TPD (TPD(+)) was 12.5. 3. The non-enzymic decomposition of DPD(+) was studied with respect to temperature, pH, concentration and DPD/DPD(+) ratio, thus defining conditions for enzyme experiments under which DPD(+) extinction at 550mmu was proportional to enzyme activity. 4. Rates of oxidation of DPD to DPD(+) by caeruloplasmin were constant over a range of DPD concentrations. At low DPD concentrations a lag period occurred, which was eliminated by addition of DPD(+). 5. A lag period was not observed with TPD, but at low TPD concentrations the rate of TPD(+) formation was greater when TPD(+) was added. This suggests that TPD(+) may compete weakly as a substrate with TPD and may be oxidized further by the enzyme before a non-enzymic reaction with TPD to form more TPD(+). 6. With DPD sulphate or acetate or TPD sulphate as substrate, Lineweaver-Burk plots were curved. With DPD hydrochloride the chloride ion caused inhibition at higher concentrations, opposing the curvature. 7. Curved Lineweaver-Burk plots were interpreted in terms of two types of substrate binding site with different K(m) values but similar V(max.) values. 8. The apparent thermodynamic changes associated with enzyme-substrate-complex formation at the sites with higher K(m) suggest that considerable conformational change may occur on binding at these sites. 9. With substrate concentrations at which only the low-K(m) sites are involved 2mol. of DPD(+)/mol. of caeruloplasmin are formed before a steady state is established. At higher substrate concentrations up to 3.2mol. of DPD(+)/mol. of caeruloplasmin are formed at this initial stage. 10. Results are discussed in relation to caeruloplasmin structures in which (a) two valence-changing and two permanently cuprous copper atoms are more accessible than the remaining four copper atoms or (b) binding of substrate at one site hinders access of substrate to another site.
Two bifunctional reagents designed to probe the active site of pepsin and other acid proteinases are described. One of these, the bisdiazoketone 1,1-bis(diazoacetyl)-2-phenylethane inactivates pepsin at pH 5.0 much more rapidly than the corresponding monodiazoketon 1-diazoacetyl-2-phenylethane, whereas the other, the bromodiazoketone dl-1-diazoacetyl-1-bromo-2-phenylethane is less effective in this regard. The inactivation is greatly accelerated by the presence of Cu(II), and the pH dependence of the process is consistent with the interaction of the enzyme with the metal complex of the carbene derived from the reagent. The bisdiazoketone appears to react stoichiometrically with pepsin in a 1:1 ratio to form a product whose apparent molecular size is the same as that of untreated pepsin. The inactivation of pepsin by the bromodiazoketone is accompanied by the release of stoichiometric amounts of bromide ions and the formation of a major product whose apparent size is similar to that of pepsin, and a minor component larger than the untreated enzyme.
Mode of radicalcyclization (5-exovs. 6-endo) was investigated by using 6-(bromomethyl)dimethylsilyl derivatives of 1',2'-unsaturated uridine with an aim to develop a new stereoselective method for the introduction of an alpha-hydroxymethyl group at the anomeric position. Although the 2'-unsubstituted derivative underwent only 6-endo-cyclization, substrates bearing 2'-CO2Me or 2'-OBz group gave exclusively 5-exo-cyclized products with a high stereoselectivity.

Recent advances in the design of photostimulable phosphor (PSP) plates for computed radiography (CR) systems have made it possible to manufacture plates made of BaFI:Eu phosphor material in the cassette form. The image quality of this plate, six BaF(Br,I):Eu plates, and one BaFBr:Eu plate were evaluated in terms of presampling modulation transfer function (MTF), normalized Wiener spectra (WSN), and detective quantum efficiency (DQE). Compared with the best-performing BaF(Br,I):Eu plate, the BaFI:Eu plate provided DQE that was higher, at spatial frequencies of 0.5, 1.0, and 2.0 cycles/mm, by 12% (21.8 versus 19.4), 13% (18.8 versus 16.7), and 11% (12.0 versus 10.8), respectively. Since presampling MTF values of the two plates were comparable, the BaFI:Eu plate's higher DQE is attributable to total WSN conversely being lower by 17% [8.65 x 10(-5) (mm2) versus 10.38 x 10(-5) (mm2)], 17% [5.85 x 10(-5) (mm2) versus 7.03 x 10(-5) (mm2)], and 12% [2.82 x 10(-5) (mm2) versus 3.19 x 10(-5) (mm2)] at the specified frequencies, respectively, primarily due to the contribution of x-ray quantum WSN. This jibes with the high x-ray absorption provided by the 27%-higher x-ray attenuation coefficients (7.54 versus 6.07, at 60 KeV) that BaFI offers over BaF(Br(0.85),I(0.15)), a result of the high atomic number of BaFI's exclusively iodine halide content. The results were consistent with earlier studies of several of these same plates and indicate that BaFI:Eu is a promising avenue to lower image noise and higher overall CR system image quality.
