Differences in the effectiveness of EDTA to induce SCEs and chromosomal aberrations in CHO and Allium cepa chromosomes.

The chelating agent EDTA was able to produce chromosome aberrations (CA) in CHO cells when it was administered simultaneously with BrdUrd (2 x 10(-5) M), without any concomitant effect on the yield of sister chromatid exchanges (SCEs). Root meristematic cells of Allium cepa did not show any type of CA when they were treated with different doses of EDTA (with or without BrdUrd 10(-4) M) while the SCE frequency was increased in a dose-dependent fashion. These effects of EDTA have not been previously reported. It is suggested that deprivation of divalent cations (Ca(+)+/Mg(+)+) probably play an important role in DNA replication and repair processes.

Antidiabetic and hypolipidemic effects of S-methyl cysteine sulfoxide isolated from Allium cepa Linn.

S-methyl cysteine sulphoxide (SMCS), a sulphur containing amino acid isolated from onion (Allium cepa Linn) showed antidiabetic and antihyperlipidemic effects. Oral administration of SMCS daily at a dose of 200 mg/kg body weight for a period of 45 days to alloxan diabetic rats controlled significantly their blood glucose and lipids in serum and tissues and altered the activities of liver hexokinase, glucose 6-phosphatase and HMG CoA reductase towards normal. The above effects of SMCS were comparable to those of glibenclamide and insulin.

[Nucleotide sequence and organization of tRNA-Gly (UCC), tRNA-Arg (UCU), and alpha-subunits of CF1ATPase in chloroplast DNA of Allium porrum].

The nucleotide sequence of 1029 bp BamHI-fragment of leek chloroplast DNA (Allium porrum, fam. Liliaceae) has been determined. The fragment contains the 3'-terminal part of the tRNA-Gly (UCC) gene, the tRNA-Arg (UCU) gene and the 3'-terminal domain of the CF1ATPase alpha-subunit gene. The gene arrangement and the nucleotide sequence of this fragment are similar to those of the tobacco chloroplast DNA but differ significantly from that of other monocots in which the region containing these genes underwent extensive recombination.

Antiplatelet activity in onion (Allium cepa) is sulfur dependent.

Plants of the genus Allium such as onion and garlic are often consumed as a source of compounds which inhibit human platelet activity, with the goal of decreasing vascular disease. Antiplatelet activity in these plants is determined in part by native concentrations of organosulfur compounds. Evaluation of four onion genotypes grown in a field study at four US locations in 1994 demonstrated onions with mild flavor and low sulfur content exhibited significantly lower antiplatelet activity than those containing high levels of sulfur. Antiplatelet activity was significantly positively correlated with genotypically determined bulb sulfur content and dissolved solids, indicating these latter factors are good predictors of antiplatelet strength. These data demonstrate antiplatelet activity is genotype dependent and correlated with bulb sulfur content. Genotype and bulb sulfur content should be taken into account in studies assessing onion antiplatelet effects.

Competence for nuclear replication and the NOR-chromosomes of Allium cepa L.

Autotetraploid (4n = 32) cells were induced in Allium cepa L. root meristems by successively treating with a multipolarizing agent in anaphase (carbetamide) and an inhibitor of cell plate formation in telophase (caffeine). This treatment produced cells with their 32 chromosomes distributed in more than two nuclei. During G1, one of the nuclei in the resulting trinucleate cells had a DNA content which was equivalent to an average of 16 chromosomes, while the other 16 chromosomes were randomly distributed in two aneuploid nuclei. In the set of 16 chromosomes forming the onion diploid complement, there are 4 NOR-chromosomes and 5 chromosomes carrying DNA domains providing a nucleus with the competence to replicate, as previously shown. Expected probabilities derived from the different possible models for cosegregation of both kinds of chromosomes in the two aneuploid nuclei of the trinucleate cells were estimated by a computer simulation. These expected values were compared with the recorded frequencies of aneuploid nuclei which were able to organize a nucleolus and to respond to inducers of replication. The present data are compatible with the existence of sequences providing a nucleus with the competence to replicate in three out of the four NOR-bearing chromosomes, as well as in two other chromosomes of this diploid complement. The scarcity of chromosomes bearing early origins able to initiate replication in a nucleus is a striking feature of this huge onion genome.

[On the effect of Chinese lacquer upon the cell division of root tip of Allium cepa].

Chinese Lacquer, as a fine coating, has been studied and applied for thousands years. The allergic reaction in Chinese Lacquer on the human skin has also been known early. The reaction of Chinese Lacquer on mitosis of cell in plant meristem have not been reported yet and was carefully studied in this paper. The result showed that Chinese Lacquer induced severe abnormality of mitotic division in Allium cepa root tips. This was more obvious in the anaphase and telophase, especially in the former phase laggard chromosomes, chromosome bridges, acentric fragments and polypolar distribution could be seen frequently. A lot of polynuclear bodies were observed in the telophase. Therefore, we think that the Chinese Lacquer can be used as a plant cell mutagen, and suggest geneticists and physiologists to do more researches on the effects of Chinese Lacquer at the genetic variation, metabolism etc.

Mitotic effects of the aqueous leaf extract of Cymbopogon citratus in Allium cepa root tips.

Aqueous extracts of the lemon grass, Cymbopogon citratus, were used to clear the malaria parasite in infected mice, although they died some days later. Allium cepa roots grown in aqueous extracts from 3, 6, 12 and 20 g of chopped leaves for 1, 3, and 6 h, showed some mitotic abnormalities including c-mitotic and mitodepressive effects. The abnormalities were not peculiar to any concentration or duration of extract treatment. The highest frequency of affected cells was 0.75% in the treatment with the 20 g concentration, but the 3 h treatment group had the greatest variety of effects. The mitodepressive effect of the extract increased significantly with concentration and time, and persisted even after 24 h in tap water. The chromosomal effects of the extract occur at a very low frequency but the mitodepressive effects may have implications for man.

Allium sativum (garlic) treatment for murine transitional cell carcinoma.

BACKGROUND: Currently, immunotherapy with Bacillus Calmette-Guerin (BCG) is the most effective treatment for superficial bladder carcinoma, but treatment-related toxicity may limit its use in some patients. Alternative treatments are needed for patients who fail to respond to BCG immunotherapy. Allium sativum (AS), or garlic, is known to have a broad range of biologic activities, including immune stimulation and reported antitumor activity. For these reasons, the authors conducted a series of experiments designed to explore the possible therapeutic effects of AS in the MBT2 murine bladder carcinoma model. METHODS: C3H/HeN mice were randomized prior to initiation of each experimental protocol. Mice received 1 x 10(3) MBT2 cells in 0.1 mL RPMI-1640, administered subcutaneously into the right thigh, on Day 0 of the experiment. AS was injected at the site of tumor transplantation on Day 1 and at 2- to 7-day intervals up to Day 28. To evaluate the effects of oral AS in this model, treatment was initiated 30 days prior to tumor inoculation and continued for 30 days after tumor inoculation. Animals in all experiments were followed for tumor incidence, tumor growth, and survival. RESULTS: In the initial experiments, subcutaneous AS significantly reduced tumor volume compared with the saline control (P < 0.05). Unfortunately, treatment-related death was also observed, requiring reduction in the total dose of AS. Animals that received 5 weekly immunizations of AS (5 mg, 5 mg, 1 mg, 1 mg, and 1 mg; cumulative dose = 13 mg) had significantly reduced tumor incidence, tumor growth, and increased survival when compared with animals that received the saline control. No treatment-related deaths were observed with this treatment schedule. To determine whether systemic AS administration might be effective, orally administered AS was tested at doses of 5 mg, 50 mg, and 500 mg per 100 mL of drinking water. Mice that received 50 mg oral AS had significant reductions in tumor volume (P < 0.05) when compared with animals that received the saline control, and mice that received 500 mg oral AS had significant reductions in both tumor volume and mortality (P < 0.05). CONCLUSIONS: The significant antitumor efficacy of subcutaneous and oral AS warrants further investigation and suggests that AS may provide a new and effective form of therapy for transitional cell carcinoma of the bladder.

Allium sativum (garlic) suppresses leukocyte inflammatory cytokine production in vitro: potential therapeutic use in the treatment of inflammatory bowel disease.

BACKGROUND: Cytokines involved in inflammatory bowel disease (IBD) direct a predominantly cell-mediated T- helper-1 (Th1) immune response. The nonspecific anti-inflammatory treatment being used in the management of patients with IBD has not changed much since the 1970s and new therapeutic agents are keenly sought. Several compounds isolated from Allium sativum (garlic) modulate leukocyte cell proliferation and cytokine production. METHODS: To investigate the possible therapeutic effects of garlic in the treatment of patients with IBD, whole blood and peripheral blood mononuclear cells (PBMCs) were stimulated in the presence of various concentrations of garlic extract and the effect on leukocyte cytokine production was determined in vitro using multiparameter flow cytometry. RESULTS: Monocyte interleukin (IL)-12 production was inhibited significantly in the presence of low concentrations of garlic extract (>or=0.1 microg/ml total protein). Monocyte IL-10 production increased significantly and monocyte tumor necrosis factor-alpha (TNF-alpha), IL-1alpha, IL-6, IL-8, T-cell interferon-gamma (IFN-gamma), IL-2, and TNF-alpha decreased significantly in the presence of >or=10 microg/ml garlic extract. Twenty to fifty percent of the immunomodulatory activity of garlic extract on cytokine production was acid labile. The inhibitory activity of methylprednisolone, a commonly used anti-inflammatory in IBD, with garlic on leukocyte cytokine production was additive. CONCLUSIONS: By inhibiting Th1 and inflammatory cytokines while upregulating IL-10 production, treatment with garlic extract may help to resolve inflammation associated with IBD. An in vivo animal model study needs to be undertaken to determine the significance of these in vitro findings.

Isolation of allicepin, a novel antifungal peptide from onion (Allium cepa) bulbs.

From the bulbs of the onion Allium cepa, a novel antifungal peptide distinct from the antimicrobial peptide previously reported from onion seeds was isolated. The antifungal peptide, designated allicepin, was purified with a procedure that involved aqueous extraction, ion exchange chromatography on DEAE-cellulose, affinity chromatography on Affi-gel blue gel and FPLC-gel filtration on Superdex 75. Allicepin was unadsorbed on DEAE-cellulose and adsorbed on Affi-gel blue gel. The molecular weight of allicepin was estimated to be 10 K by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel filtration on Superdex 75. Allicepin exerted an inhibitory activity on mycelial growth in several fungal species including Botrytis cinerea, Fusarium oxysporum, Mycosphaerella arachidicola and Physalospora piricola.

The protective effect of Allium sativum L. clove aqueous and methanolic extracts against hypoxia-induced lethality in mice.

The antihypoxic activity of Allium sativum clove (garlic) aqueous and methanolic extracts was studied in mice. The extracts of garlic showed that the antihypoxic effect was dose-dependent. The minimum effective doses of aqueous and methanolic extracts were 0.2 g/kg and 5.12 g/kg, respectively. Phenytoin, 50 mg/kg, and R-phenylisopropyladenosine (R-PIA), 1.6 mg/kg (R-PIA) as positive controls increased survival time up to 52.5 +/- 2.9 min and 120.5 +/- 6 min, respectively, compared to normal saline (34.73 +/- 0.71 min). The high doses of aqueous (16.9 g/kg) and methanolic (12.8 g/kg) extracts increased survival time up to 73.17 +/- 4.9 and 68.41 +/- 3.7, respectively. These results indicated that the extracts of A. sativum cloves have a protective effect against hypoxia-induced lethality in mice.

Prevention of bone loss by oil extract of garlic (Allium sativum Linn.) in an ovariectomized rat model of osteoporosis.

The effects of oil extract of garlic (Allium sativum Linn.) on different primary and secondary osteoporotic marker changes were tested in an ovariectomized rat model of osteoporosis. Experiments were performed on three different rat models: sham-operated control, ovariectomized and ovariectomized supplemented with garlic oil. In ovariectomized group, there has been a significant increase in different relative organ weights compared to sham-operated control, while the uterine weight was found to be decreased. Supplementation with oil extract of garlic could effectively reverse these changes. Also low bone densities that developed in the ovariectomized group were significantly recovered in the garlic oil supplemented group. In our study, the development of high rate of bone turnover and osteoporosis in the ovariectomized animals were confirmed by significant alteration of serum alkaline phosphatase activity, serum tartrate resistant acid phosphatase activity, urinary excretion of calcium, phosphate, hydroxyproline and urinary calcium to creatinine ratio, when compared with the sham-operated control group. Garlic oil extract supplementation, apart from its unique influence in lowering blood cholesterol, could also prevent ovariectomy-induced rise in all the above-mentioned marker changes. The results of this study emphasize that oil extract of garlic possibly has a positive role in suppressing ovariectomy-induced bone resorption.

Effect of cadmium on the nucleoli of meristematic cells of onion Allium cepa L: an ultrastructural study.

This study of the effect of cadmium on nucleolar ultrastructure was carried out with meristematic cell populations of Allium cepa L. Meristems, grown at 25 degrees C, were treated with 10 ppm cadmium chloride. Conventional and silver staining techniques were carried out, and the ultrastructure was analyzed using electron microscopy. Observation showed alterations in the nucleoli of the cells that had been treated with cadmium and this effect varied according to the time of exposure to the metal. After 4h of treatment, nucleolar segregation was observed in interphase, probably because of the effect of cadmium on the synthesis of ribosomal RNA precursors. A decrease in the fibrillar to granular component ratio also occurred in the cells exposed to Cd2+ for 8 h. Some changes were observed in the G1 cells; their chromatin still remained very condensed, and prenucleolus bodies remained scattered within the nucleus. At the same time, there was a large amount of interchromatin granules. These changes produced by cadmium resembled those produced during inhibition of RNA synthesis. The fibrillar bodies, another morphologic feature, resulting from a blocked transcription, were also evidenced. All these observations suggest that one of the ways that cadmium exercises its toxicity is by altering the biosynthesis of the preribosomal RNA precursor.

Allergic contact dermatitis to garlic (Allium sativum L.). Identification of the allergens: the role of mono-, di-, and trisulfides present in garlic. A comparative study in man and animal (guinea-pig).

Garlic (Allium sativum L.) water- and ethanol-soluble extracts were prepared and purified by column chromatography. They were tested on garlic-sensitive patients and showed that the allergenic fraction was well located in a few column chromatography fractions. Guinea-pigs were sensitized with garlic water-soluble extracts and tested (open epicutaneous tests) with several fractions. The presence of diallyldisulfide was detected in the sensitizing chromatographic fractions. Guinea-pigs were successfully sensitized to this product and cross-reacted to garlic; animals sensitized to garlic extracts cross-reacted to diallyldisulfide. Both groups reacted to allicin, an oxidized derivative of diallyldisulfide present in garlic. Garlic-sensitive patients showed positive tests to diallyldisulfide, allylpropyldisulfide, allylmercaptan and allicin.

Ultrastructural localization of DNA and RNA in Allium porrum interphase cells by means of nuclease-gold complexes.

Nucleic acids have been localized in Allium porrum interphase meristematic cells by means of labelling with nuclease-gold complexes, a technique which provides high resolution and improved specificity. DNase-gold labelling was observed over dense chromatin and to a lesser extent over dispersed chromatin. Nucleolar labelling was restricted to the dense fibrillar component, very few particles being located over the fibrillar centres. Labelling by the RNase-gold complex was present over both the cytoplasm and the nucleoplasm. Cytoplasm labelling was intense over the rough endoplasmic reticulum but absent over vacuoles. In the nucleoplasm many gold particles were located at the border between the condensed and the dispersed chromatin. Nucleolar labelling was intense over the granular zones but many gold particles were also seen over the dense fibrillar component. Fibrillar centres showed, however, no labelling with the RNase-gold complex. These results are consistent with previous autoradiographic and cytochemical observations carried out on the same plant material.
