[Possible participation of cyclic AMP in regulating acid hydrolase activity in muscle tissue in avitaminosis E and denervation].

In the muscles of denervated and vitamin E-deficient rabbits the level of 3', 5'-cyclic AMP proved to decrease with a simultaneous increase in the activity of cAMP phosphodiesterase. In vivo experiments showed that at the concentration of 10(-4) cAMP was capable of retarding the release of acid phosphatase from the lysosome-rich fraction obtained from the muscles of E-deficient rabbits. Thus, in muscular dystrophy elevation of acid hydrolase activity in the skeletal muscle was due to leakage of the enzymes from the lysosomes as a result of decreased lysosome membrane stability because of decreased cAMP level.

[Structural changes in sarcolemma with E-avitaminosis dystrophy].

Sorption properties of sarcolemma preparations isolated from skeletal muscles of normal and E-avitaminous rabbits were studied relative to organic ions. Analysis of isotherms of sarcolemma equilibrium binding of neutral red cations and turquoise direct lightfast "K" anions made it possible to determine the number of positively and negetively charged sorption centres, which fix the mentioned dyes. With E-avitaminous muscular dystrophy the number of the centres increases considerably. A larger number of the positively charged centres fixing the surquoise dye are found both in the control and in case of dystrophy. The calcium ions prevent the neutral red sorption and intensify the turquoise direct sorption. In the sarcolemma preparations isolated from the muscles of the E-avitaminous rabbits the content of calcium ions is almost twice as high and the number of sulphydryl groups is 30-40% less as compare to the normal level. The data presented evidence for structural changes in sarcolemma with E-avitaminous muscular dystrophy.

[Effect of alpha- and gamma-tocopherols on the cytochrome P-450 system in the liver of rats with E-avitaminosis. Effectiveness of liposomal forms of gamma-tocopherol].

The effects of low doses of intraperitoneally injected alpha- and gamma-tocopherols dispersed in isotonic solutions of NaCl and of liposomal gamma-tocopherol on endogenous lipid peroxidation and the cytochrome P-450 system of vitamin E-deficient rats were compared. It was found that all tocopherol preparations under study caused a twofold decrease of the lipid Schiff base concentration in the liver in comparison with tocopherol-deficient rats. However, the increase in cytochrome P-450 and b5 levels and the high functional activity of the microsomal oxidation system in the benz(a)pyrene hydroxylation reaction was induced only after liposomal gamma-tocopherol injection. It was concluded that the efficiency of the liposomal form of gamma-tocopherol can be conditioned both by preventing gamma-tocopherol oxidation in lecithin vesicles during its transport and/or by an increased liposomal gamma-tocopherol uptake by the liver from the circulating blood.

[Non-antioxidative mechanism of cytochrome P-450 stabilization by alpha-tocopherol: the effectiveness in avitaminosis E].

The efficiency of alpha-tocopherol as a 7-etoxycumarine deethylase protector in rat liver microsomes damaged by phospholipase A2 at various levels of vitamin E was studied. No selective damage of cytochrome P-450 isoforms possessing a catalytic activity towards 7-etoxycumarine under vitamin E deficiency was observed. Phospholipase A2 decreased the deethylase activity of cytochrome P-450, the efficiency of the damaging action being dependent on vitamin E content in the liver. Exogenous alpha-tocopherol exerts an antiphospholipase effect and protects 7-etoxycumarine deethylase; the protective action is inversely proportional to vitamin E level in the liver. Under normal conditions the damaging effect of phospholipase A2 on cytochrome P-450 is mainly provided for by lysophospholipids, while under vitamin E deficiency both lysophospholipids and free fatty acids exert a damaging action. A possible mechanism of the stabilizing effect of alpha-tocopherol may consist in the interaction of the chromanol nucleus in the vitamin E molecyule both with lysophospholipids and with free fatty acids.

[Glycosaminoglycans of the aorta in avitaminosis].

It has been shown that pregnancy specific beta 1-globulin (SBG) is found in the rat blood serum on the 6--7th day of pregnancy. SBG reaches the maximum by the end of pregnancy and is not detected already at the 3d--4th day after delivery. The placenta is the site of the SBG snythesis. Other organs of the pregnant rat are not able to incorporate radioactive aminoacids into the protein in vitro.
