Does androgen excess contribute to the cardiovascular risk profile in postmenopausal women with type 2 diabetes?

The purpose of this study was to determine if postmenopausal women with type 2 diabetes have clinical and biochemical evidence of androgen excess as a potential contributor to an increase in risk for coronary heart disease when compared with women without diabetes. Fasting glucose, insulin, lipids, sex hormone-binding globulin (SHBG), and sex steroids (from pooled samples) (total testosterone and free testosterone [non-SHBG-T], androstenedione [A-dione], total estrogens) were measured at baseline in 16 postmenopausal women with type 2 diabetes treated with diet or a sulfonylurea and 17 age-matched controls. Measurements of glucose, insulin, and sex steroids were repeated at hourly intervals for 3 hours after oral glucose administration. Hirsutism scores and insulin sensitivity (homeotasis model assessment [HOMA] insulin [SI]) were obtained. Women with type 2 diabetes were more hyperglycemic, hyperinsulinemic, and insulin-resistant (HOMA SI, 46.7 +/- 7.0 vs 12.9 +/- 2.0, P < .001), and had higher total to high-density lipoprotein cholesterol (TC/HDL) ratios, lower SHBG (20.8 +/- 3.5 vs 59.3 +/- 14.4 nmol/L, P < .05), higher non-SHBG-T (0.225 +/- 0.025 vs 0.135 +/- 0.021 nmol/L, P < .05), and higher hirsutism scores (1.1 +/- 0.3 vs 0.3 +/- 0.2, P = .004) than those without diabetes. No changes in sex steroids occurred after the oral glucose challenge. HOMA SI and area under the curve for glucose correlated significantly with SHBG (r = -0.42), non-SHBG-T (r = 0.40), and TC/HDL (r = 0.41) (all P < .05) in the combined groups. Postmenopausal women with type 2 diabetes have both clinical and biochemical evidence of androgen excess that may contribute to more adverse cardiovascular risk profiles.

Characterization of the surface organization of nanostructured hybrid organic-inorganic materials by time-of-flight secondary ion mass spectrometry

Time-of-flight secondary ion mass spectrometry (TOF-SIMS) has been used to analyse the surface composition of organic-inorganic hybrid solids obtained by a sol-gel process. Gels of type O(1.5)Si-R-SiO(1. 5), obtained from bis-silylated precursors (R'O)(3)-R-Si(OR')(3) (R' = Me, Et and R = (-CH(2))(n)-, n = 1, 2, 6, 10, 12;--CH=CH-; (-CH(2))(3)NH(CH(2))(3)-; 1, 1'-ferrocenyl; (CH(2))(n)-Ph-(CH(2))(n)- with Ph = 1,4-phenylene and n = 0, 1, 2; Ph = 1,3,5-phenyl and n = 0) were analysed. The results were highly dependent on the nature of the organic group. When the organic group was small or 'rigid', the main peaks detected corresponded to SiOH and SiOR' residual groups. Fragment ions from the organic group were poorly detected in this case. When the organic group was larger and more 'flexible', characteristic mass fragment ions were detected at higher relative intensities, indicative of a different organization of the organic units in the solid. TOF-SIMS clearly showed the differences between the xerogels derived from mono- and bis-silylated organic precursors : the organic group is present at the surface of mono-silylated xerogels, whereas for bis-silylated ones, the organization is dependent on the length and the flexibility of the organic units. These TOF-SIMS results are in agreement with other features already reported. Copyright 1999 John Wiley & Sons, Ltd.

[Internal osteosynthesis after unstable pelvic ring fracture in a 3-year-old child].

A 3-year-old child was trapped under the heavy load of a forklift truck and sustained an unstable pelvic ring fracture (Tile type C) with complete SI disruption, disruption of the public symphysis and external rotation injury of the contralateral SI joint. An immediate internal fixation was performed, exposing the SI joint and the public symphysis simultaneously. For stabilization an H-plate was used for anterior plate fixation of the SI joint, while the public symphysis was stabilized by screws and cerclage wires. After one revision of the symphysis the clinical course was uneventful with anatomical healing of the pelvic ring. The implants were removed after 4 months. Clinical and radiological follow-up after 12 months showed no signs of maldevelopment of the pelvic ring.

Morphology of layer V pyramidal neurons in the cat somatosensory cortex: an intracellular HRP study.

Pyramidal tract (PT) or corticopontine neurons of the cat somatosensory cortex (SI) were identified with antidromic activation on stimulation of the bulbar pyramid or pontine nuclei (PN) and stained intracellularly with HRP after examining the electrophysiological properties. Comparison of the conduction velocity of the stem axons and the soma-dendritic morphology revealed that in the cat SI, there exists two types of layer V pyramidal neurons, i.e. one has smooth apical dendrites with larger soma (51.6 +/- 9.5 x 22.7 +/- 2.8 micron) and the other has richly spinous apical dendrites with smaller soma (34.0 +/- 8.8 x 15.3 +/- 3.3 micron). The former group responded antidromically at latencies shorter than 1 ms by PT stimulation or 1.5 ms by PN stimulation, respectively. These values were consistent with the borderline latencies between two similar groups of layer V pyramidal neurons in the motor (fast and slow PTNs) and parietal (aspiny and spiny layer V corticopontine neurons) cortices in the cat.

Development of a standardized analysis strategy for basic drugs, using ion-pair extraction and high-performance liquid chromatography - II. Selection of preferred HPLC-systems.

A test set of 100 basic drugs has been chromatographed on 16 preselected HPLC-systems using four different types of stationary phase (Si-, NH2-, CN- and C18-). A numerical treatment of the chromatographic data, based on the discriminating power concept, results in the selection of two preferred HPLC systems for basic drugs. both using the CN-bonded phase. The preferred eluents are n-heptane-dichloromethane-acetonitrile-propylamine (25:50:25:0.1) and acetonitrile-water-propylamine (90:10: 0.01). The two preferred HPLC systems are adopted in a standardized analysis strategy.

Identification of membrane-anchoring domains of RLIP76 using deletion mutant analyses.

RLIP76 (RALBP1) is a multifunctional transporter involved in signaling and transmembrane movement of solute allocrites, which include glutathione conjugates and several natural product antineoplastic agents [Awasthi, S., et al. (2000) Biochemistry 39, 9327-9334; (2001) Biochemistry 40, 4159-4168]. Our previous studies suggested that the membrane-anchoring domain resides in the N-terminus of RLIP76, despite the lack of identifiable membrane-spanning domains. Amino acid sequence analysis indicated that this region of RLIP76 contains sequences that are similar to those of vector peptides. We, therefore, have studied the effect of a series of deletion mutant proteins on hydrophobicity and transport activity. RLIP76 or one of its derived deletion mutants was expressed in Escherichia coli, and bacteria were lysed and extracted in buffer without or with the nonionic detergent polidocanol. The ratio of RLIP76 in the detergent/aqueous extracts was found to be 2.5 for the wild-type protein, but decreased to 0.7 in the mutant in which amino acids 154-219 were deleted. Deletion of only one segment of this region (amino acids 171-185) alone resulted in a significant decrease in this ratio to 1.0. For the mutants with deletions within the region from amino acid 154 to 219, loss of hydrophobicity correlated with less incorporation of mutants into artificial liposomes, and decreased transport activity toward doxorubicin and dinitrophenyl-S-glutathione. In contrast, deletion of one of the two ATP-binding sites (at amino acids 65-80 or 415-448) or both sites did not affect hydrophobicity but reduced or abrogated transport activity. NSCLC (H358) stably transfected with del171-185 and del154-219 showed that loss of these regions results in a decrease in the extent of membrane association of RLIP76. Confocal laser immunohistochemistry colocalized amino acids 171-185 with her2/neu on the cell surface. Depletion of wild-type RLIP76 using si-RNA directed to this region in cells transfected with del171-185 resulted in the loss of cell surface expression. These finding demonstrate that amino acids 171-185 constitute a cell surface epitope which is necessary for optimal transport of anthracycline and glutathione conjugates by RLIP76, and that this peptide could be a novel target for antineoplastic therapy.

Potential for hostility and faking-good in high-hostile men.

We investigated if Type A Structured Interview (SI)-assessed hostility (PoHo) is subject to voluntary underreporting, or faking-good, by hostile subjects. Sixteen men who previously scored above the clinical cutoff of 3 on SI hostility were randomly assigned to an experimental (N = 8) or control (N = 8) group. Both groups underwent a second videotaped SI, prior to which experimental subjects were asked to present themselves in a favorable light. Repeated-measures ANOVA revealed that experimental subjects were able successfully to decrease their global PoHo scores when instructed to do so (38% scored below the clinical cutoff score). The SI-derived hostility score is vulnerable to faking-good, and attention should be paid to both the motivational set of subjects and the instructions given prior to the SI.

Associations of blood pressure with self-report measures of anger and hostility among black and white men and women.

This study examined associations between blood pressure (BP) and dispositional variables pertaining to anger and hostility. Black and White 25- to 44-year old male and female normotensives and unmedicated mild to moderate hypertensives completed four reliable self-report scales--the Cook-Medley Hostility (Ho) Scale, the Trait Anger subscale of the State-Trait Anger Scale (STAS-T), and the Cognitive Anger and Somatic Anger subscales of the Cognitive-Somatic Anger Scale--plus the Framingham Anger Scale and the Harburg Anger Scale. They also engaged in three laboratory tasks--Type A Structured Interview (SI), a video game, and a cold pressor task--that elicit cardiovascular reactivity. Ambulatory BP readings at home and at work were also obtained from most subjects. Blacks had significantly higher Ho and lower STAS-T scores than did Whites. Women reported higher levels of somatic anger than did men. White women showed significant positive correlations between STAS-T and systolic BP (SBP) and diastolic BP (DBP) both at rest in the laboratory and during the SI. Black women revealed significant positive relationships between STAS-T and SBP and DBP at rest in the laboratory and at work as well as with DBP during the cold pressor test. For Black men, cognitive anger and DBP at rest were positively related. In contrast, White men revealed significant negative correlations between Ho scores and SBP at rest and during the video game; these men also showed significant negative relationships between somatic anger and SBP and DBP reactivity during the cold pressor test. Women, but not men, showed significant positive relationships between all four anger measures and ambulatory BP at work. Whereas main effects relating anger and cardiovascular measures were not apparent as a function of race, Blacks demonstrated significantly greater SBP and DBP reactivity than Whites during the cold pressor test, with the converse occurring during the SI. Men demonstrated significantly greater DBP reactivity than women during the video game. The present findings indicate that self-reports on anger/hostility measures and cardiovascular responses to behavioral tasks differ as a function of race but that relationships between anger and BP regulation need to take into account possible race-sex interactions and selection of anger/hostility measures.

Pollen performance before and during the autotrophic-heterotrophic transition of pollen tube growth.

For species with bicellular pollen, the attrition of pollen tubes is often greatest where the style narrows at the transition between stigmatic tissue and the transmitting tissue of the style. In this region, the tubes switch from predominantly autotrophic to predominantly heterotrophic growth, the generative cell divides, the first callose plugs are produced, and, in species with RNase-type self-incompatibility (SI), incompatible tubes are arrested. We review the literature and present new findings concerning the genetic, environmental and stylar influences on the performance of pollen before and during the autotrophic-heterotrophic transition of pollen tube growth. We found that the ability of the paternal sporophyte to provision its pollen during development significantly influences pollen performance during the autotrophic growth phase. Consequently, under conditions of pollen competition, pollen selection during the autotrophic phase is acting on the phenotype of the paternal sporophyte. In a field experiment, using Cucurbita pepo, we found broad-sense heritable variation for herbivore-pathogen resistance, and that the most resistant families produced larger and better performing pollen when the paternal sporophytes were not protected by insecticides, indicating that selection during the autotrophic phase can act on traits that are not expressed by the microgametophyte. In a study of a weedy SI species, Solanum carolinense, we found that the ability of the styles to arrest self-pollen tubes at the autotrophic-heterotrophic transition changes with floral age and the presence of developing fruits. These findings have important implications for selection at the level of the microgametophyte and the evolution of mating systems of plants.

Propagation of surface plasmon polaritons on semiconductor gratings.

We present time-domain measurements of terahertz surface plasmon polaritons (SPPs) propagating on gratings structured on silicon surfaces. Using single-cycle pulses of terahertz radiation to excite SPPs in a broad frequency range, we observe that the efficient SPPs scattering on the semiconductor periodic structure introduces significant dispersion and modifies the SPPs propagation. A stop gap, or a frequency range where SPPs are Bragg reflected, is formed by the structure. This gap depends strongly on the Si doping density and type. The resonant scattering at the edge of the gap reduces the group velocity by more than a factor of 2. The measurements show a good agreement with our numerical calculations based on the reduced Rayleigh equation.

Structural derivation and crystal chemistry of apatites.

The crystal structures of the [A(1)(2)][A(2)(3)](BO(4))(3)X apatites and the related compounds [A(1)(2)][A(2)(3)](BO(5))(3)X and [A(1)(2)][A(2)(3)](BO(3))(3)X are collated and reviewed. The structural aristotype for this family is Mn(5)Si(3) (D8(8) type, P6(3)/mcm symmetry), whose cation array approximates that of all derivatives and from which related structures arise through the systematic insertion of anions into tetrahedral, triangular or linear interstices. The construction of a hierarchy of space-groups leads to three apatite families whose high-symmetry members are P6(3)/m, Cmcm and P6(3)cm. Alternatively, systematic crystallographic changes in apatite solid-solution series may be practically described as deviations from regular anion nets, with particular focus on the O(1)-A(1)-O(2) twist angle phi projected on (001) of the A(1)O(6) metaprism. For apatites that contain the same A cation, it is shown that phi decreases linearly as a function of increasing average ionic radius of the formula unit. Large deviations from this simple relationship may indicate departures from P6(3)/m symmetry or cation ordering. The inclusion of A(1)O(6) metaprisms in structure drawings is useful for comparing apatites and condensed-apatites such as Sr(5)(BO(3))(3)Br. The most common symmetry for the 74 chemically distinct [A(1)(2)][A(2)(3)](BO(4))(3)X apatites that were surveyed was P6(3)/m (57%), with progressively more complex chemistries adopting P6(3) (21%), P3; (9%), P6 (4.3%), P2(1)/m (4.3%) and P2(1) (4.3%). In chemically complex apatites, charge balance is usually maintained through charge-coupled cation substitutions, or through appropriate mixing of monovalent and divalent X anions or X-site vacancies. More rarely, charge compensation is achieved through insertion/removal of oxygen to produce BO(5) square pyramidal units (as in ReO(5)) or BO(3) triangular coordination (as in AsO(3)). Polysomatism arises through the ordered filling of [001] BO(4) tetrahedral strings to generate the apatite-nasonite family of structures.

Microscopy and quantitative morphology of aluminum silicate nanoparticles grown on organic templates.

Biomimetic synthesis of ceramic materials is increasing in popularity because it offers many advantages. In this work, aluminum silicate nanoparticles were obtained on a self-assembled organic multilayer at near room temperature (30 < or = T < or = 50 degrees C) and at atmospheric pressure. Morphological and microanalytical characterization was carried out by means of transmission electron microscopy and subsequent image analysis. The roles of some process parameters such as template type, reactant concentration, [Al]:[Si] molar ratio, number of initiation steps (IS) of mineralization, and reaction time (rt) were assessed by comparing images, diffraction patterns, and EDX spectra. Generally, the Si-rich phase exhibited higher crystallinity, whereas the Al-rich phase was mostly amorphous. Crystal structure resulted with rt > or = 4 days for template-grown materials. Images of materials obtained at T = 50 degrees C, rt = 3 days, and 1 < or = IS < or = 4 were further analyzed by "spectrum enhancement," an algorithm based on the Fourier transform. Morphological indicators were extracted from suitably processed power spectral densities, a correlation matrix was formed, and multivariate statistics was carried out. Visual differences in nanoaggregate morphology were quantitatively translated. Materials were ranked by the spatial uniformity of nanoparticle distribution: the most uniform aggregates were those grown on templates by IS > or = 2. Univariate statistics validated the conclusion: the particles of those same materials had a narrower size distribution and sharper edges. This last property has been ascribed to crystalline structure, independently demonstrated by diffraction patterns.

Study of the mechanism underlying the difference in motility between the large and small intestine: the "single" and "multiple" pacemaker theory.

The common movement of the small intestine (SI) is peristalsis and of the large intestine (LI) are giant migrating contractions. The mechanism underlying the difference in the type of motility between the SI and LI is yet to be elucidated; the current communication investigated this point. 7 pigs (4 male) were anesthetized, abdomen opened and a balloon-ended catheter was introduced into the right colon through ileotomy. Balloon was filled in increments of 10 ml of saline and the EMG activity of the colonic longitudinal and circular muscle layers was recorded before and after myotomy performed between 2 of the 3 electrodes fixed to the colon. The balloon was then withdrawn, located in the terminal ileum and distended in increments of 2 ml; the ileal EMG activity was registered before and after myotomy done between 2 of the 3 electrodes applied to the ileum. The LI showed slow waves or pacesetter potentials (PPs) and action potentials (APs) which had the same frequency, amplitude and conduction velocity from the 3 electrodes of the same animal. The waves were recorded from the longitudinal and not the circular muscle coat. Upon LI distension, the electric activity increased and was recorded also from the circular muscle. At 40-50 ml distension, the balloon was dispelled to the transverse colon. Electric activity from SI was similar to that of the LI, but was not the same from the 3 electrodes; it diminished aborally. It increased with increasing balloon distension until, at 8-10 ml distension, the balloon moved slowly aborally. Electric waves were recorded proximally but not distally to colonic myotomy, and the balloon moved up to the cut. In the SI, waves were recorded both proximally and distally to the ileal myotomy, and the balloon moved across the cut. The fact that the colonic electric waves displayed the same variables from the 3 electrodes and that they were not recorded distally to the colonic myotomy, would suggest the presence of a "single" colonic pacemaker, probably situated in the cecum. This is in contrast to the hypothesis of the "multiplicity" of the small intestinal pacemakers, which is based on the fact that the electric activity diminished as the waves propagated aborally and on the existence of slow waves distal to the ileal myotomy. The concept of "single" and "multiple" pacemakers explaining the difference in the motility of small from that of large gut needs further studies.

Responses from area 3b of somatosensory cortex to textured surfaces during active touch in primate.

(1) The purpose of this experiment was to characterize the responses of neurons in somatosensory cortex while the hand was actively moved (stroked) across a textured surface. Surfaces consisted of horizontal gratings that varied by spatial period or ridge-groove ratio (roughness). Surfaces were attached to rectangular blocks. TOP and BOTTOM halves of each block could contain surfaces of different roughness. (2) Velocity and force of the stroke were behaviorally constrained within certain limits and continuously measured and recorded during the stroke. (3) Response samples for each neuron were obtained for repeated presentations of each surface. Statistical analyses consisted of analysis of variance and t tests across surfaces on the data of each neuron, and summary statistics on groups of neurons with similar response characteristics. The interaction effects of behavioral variables (velocity and force) were examined and found not to be significant. (4) The sample mainly consisted of rapidly adapting neurons in area 3b of somatosensory area I (SI). Three main response types were found: (a) GRADED cells showed a monotonic increase in firing rate to increasingly rougher surfaces. This effect was seen in one-third of cells studied and is consistent with other reports. These cells seem to code roughness in the magnitude of their response. (b) In some cells, response to a BOTTOM surface depended on the roughness of the preceding TOP surface. This is analogous to contrast in the visual system. These CONTRAST cells are a novel finding in the somatosensory system. (c) Some cells only responded to surfaces that were completely smooth. These "OFF"-response-type cells were seen in proximity to other cells that responded in a reciprocal fashion to surfaces with ridges, but not to smooth surfaces. SMOOTH cells did not respond to punctate or passively applied stimuli, and therefore could not be classified by adaptation of the responses. (5) An increase in firing rate as spatial period (roughness) increases (with a constant ratio of ridge to groove) seems contrary to vibratory models of texture perception. As spatial period increases, temporal frequency decreases, and thus "tuned" cells should show a decreased response rate. Yet GRADED cells showed an increased response. In addition, response varied on surfaces with different groove size, where spatial period, and thus temporal period, was constant. This suggests that in rapidly adapting neurons, at least for these simple surfaces, texture is coded by the magnitude of the firing rates rather than by its temporal fidelity.(ABSTRACT TRUNCATED AT 400 WORDS)

Development and testing of a new hostility scale.

Evidence suggests that assessment of global type A behaviour as a coronary risk factor may not be the most fruitful strategy because some components, particularly hostility, may be more harmful than others. Measurement of hostility has been primarily through two types of instrumentation: the Type A Structured Interview (SI) and the Cook-Medley (Ho) scale. Deficiencies in the measurement of hostility have resulted from a lack of conceptual clarity because definitions of hostility have been blurred by overlap with the concepts of anger and aggression. The main objective of this study was to develop and test an instrument that focuses hostility on attitude rather than emotion or behaviour. Psychometric properties of the Hostile Attitude Scale (HAS) were Cronbach's alpha, 0.80, and correlation of 0.56 with the Ho scale and 0.43 with the SI. In a clinical investigation that used the HAS, SI and Ho scale, all measures were significantly correlated with angiogram outcome, although the strongest relationship was with the HAS. Hostile attitude alone (HAS) accounted for 6% of explained variance in angiogram outcome based on regression analysis. The HAS is offered as a short, easily administered measure of hostility, with particular emphasis on cognitive aspects of the construct.

Identification of self-incompatibility (S-) locus linked pollen cDNA markers in Petunia inflata.

Solanaceous type self-incompatibility (SI) is controlled by a single polymorphic locus, termed the S-locus. The only gene at the S-locus that has been characterized thus far is the S-RNase gene, which controls pistil function, but not pollen function, in SI interactions between pistil and pollen. One approach to identifying additional genes (including the pollen S-gene, which controls pollen function in SI) at the S-locus and to study the structural organization of the S-locus is chromosome walking from the S-RNase gene. However, the presence of highly repetitive sequences in its flanking regions has made this approach difficult so far. Here, we used RNA differential display to identify pollen cDNAs of Petunia inflata, a self-incompatible solanaceous species, which exhibited restriction fragment length polymorphism (RFLP) for at least one of the three S-haplotypes (S1, S2, and S3) examined. We found that the genes corresponding to 10 groups of pollen cDNAs are genetically tightly linked to the S-RNase gene. These cDNA markers will expedite the mapping and cloning of the chromosomal region of the Solanaceae S-locus by providing multiple starting points.

Hostility and facial expression in young men and women: is social regulation more important than negative affect?

Behavioral differences may clarify the link between hostility and health. This study examined facial expression. Seventy-two low- and high-hostile undergraduates underwent the Type A Structured Interview (SI) and a test of social anxiety. Facial behavior was measured with the Facial Action Coding System. Low-hostile participants displayed non-Duchenne smiles more frequently than high-hostile participants during the SI. There were no group differences in the expression of disgust. The results identify differences in the nonverbal behavior of hostile people. Restricted use of non-Duchenne smiles may reflect limited use of appeasement, contributing to uncomfortable interpersonal relations and limited social support. The findings are consistent with a behavioral ecology perspective and suggest that social regulation may be as important as negative affect in determining the consequences of hostility.

[Use of micturition cystourethrography in evaluation of urinary incontinence in women].

Miction cystourethrography (MCU) is an investigation of the conditions of suspension of the bladder. Assessment of the conditions of suspension has been employed by som urologists for 1) diagnosis of genuine stress incontinence (SI), 2) choice of operative procedure for SI and 3) assessment of the surgical result after operation for SI. It is generally agreed that MCU, as a routine method, does not contribute any important diagnostic information on account of the very low predictive values. Whether MCU can contribute information of significance in the choice of operative procedure for SI is still controversial. Recent investigations suggest that the suprapubic approach is more effective than vaginal procedures and that this should be preferred for SI, regardless of the type of possible suspension defect and the presence of slight to moderate cystocele. This reduces the significance of a meticulous radiological diagnosis. Whether large cystoceles should be operated upon vaginally and/or suprapubically is not yet elucidated but these are not indications for MCU as they can be diagnosed by gynaecological examination. Postoperative MCU has revealed poor correlation between cure and relief of a possible suspension defect. As MCU is a resource-demanding examination which may be difficult to interpret, it cannot be recommended as a routine in assessment of urinary incontinence in women.
