[The isolation and identification of PAF inhibitors from Piper wallichii (Miq.) Hand-Mazz and P. hancei Maxim].

Platelet activating factor (PAF) is a highly potent endogenous phospholipid mediator, involved in various inflammatory and cardiovascular disorders. As part of a research program dealing with PAF inhibitors isolated from Piper plant species, we have isolated kadsurenone (I), denudatin B (II), and N-isobutyl-deca-trans-2-trans-4-dienamide (III) from Piped wallichii (Miq.) Hand-Mazz. and P. hancei Maxim. In a continuing search for potential PAF inhibitor from plants, using PAF induced platelet aggregation as a guide, a new neolignan named hancinone D (IV) was isolated from P. hancei maxim. By X-ray analysis it was identified as a racemate. The X-ray analysis led to a revision of the previously made structure assignment of hancinone C. Another new neolignan named wallichinine (V), which was identified as an analogue of (IV), along with the known compounds hancinone C (VI), galgravin (VII), dihydropiperlonguminine (VIII) and crotepoxide (IX) were isolated from P. wallichii (Miq.) Hand-Mazz. The structure determination was based upon spectroscopic analysis. All of the compounds were for the first time obtained from both plants. In the test of platelet aggregation caused by PAF, I, II, V, VI, VII showed inhibitory activity, whereas III, IV, VII, IX showed no activity.

[Studies on the chemical constituents of Piper macropodum C.DC].

Piper macropodum is used in Chinese folk medicine for the treatment of rheumatism, toothache, epilepsy and stomach ache. From the ethanolic extract of the viny stem of this plant five components were isolated, a new compound and four known compounds. On the basis of spectroscopic analysis (IR, UV, MS, 1HNMR and 13CNMR) the structure of this new compound was elucidated as (2E,4E)-N-isobutyl-2,4-icosadienamide (I). The other four compounds were identified as piperine (II), piperyline (III), daucosterol (IV) and beta-sitosterol (V). All of them were isolated from this species for the first time.

Chemical Phenomena↗

Effect of Piper betle L. and its extracts on the growth and aflatoxin production by Aspergillus parasiticus.

Ground powder of the leaf and fruit of Piper betle L., a tropical spice plant grown in Southeast Asia, was prepared and extracted by chloroform, ethanol and water with one solvent only or with 3 solvents in sequence. The betel powder and various extracts were added to YES broth to determine their effects on the growth and aflatoxin production by Aspergillus parasiticus. Results showed that betel leaf powder exhibited higher antimycotic activity than fruit. One half percent of ground leaf powder completely inhibited the growth and aflatoxin production by A. parasiticus. Among the solvent extracts, chloroform and ethanol extracts of betel leaf prepared from a single solvent extraction showed more antimycotic activity. The ethanol extract of betel leaf at the level of 450 micrograms/ml would eliminate A. parasiticus growth and aflatoxin production. The antimycotic activity of this ethanol extract was most pronounced at pH 4.

[Studies on some derivatives of crotepoxide--an antitumor constituent of Piper futokadzura].

Through reactions of deacetylation and partial deacetylation with selected opening of epoxide ring, seven derivatives of crotepoxide (futoxide), a known antitumor constituent from Piper futokudzura, were prepared. Preliminary pharmacological tests showed that individual derivative possess significant inhibitory activity at 2 x 10(-4) mol/L to the PAF-induced platelet aggregation, while crotepoxide has no effect even at fivefold of this concentration.

[Anti-platelet activating factor constituents, 2,5-diaryltetrahydrofuran type lignans, from Piper futokadsura Sied. et Zucc].

Besides the known components kadsurenone, futoquinol and futoxide, three anti-platelet activating factor principles have been found from the stem part of Piper futokadsura (Piperaceae), the Chinese drug haifengteng. They were separated and identified as galgravin, galbelgin and veraguensin on the basis of HPLC and spectral analysis. The natural existence of these three anti-PAF constituents in haifengteng is reported for the first time.

[PAF antagonistic benzofuran neolignans from Piper kadsura].

In a continuing search for PAF antagonists, five benzofuran neolignans have been isolated from the aerial part of Piper kadsura (Choisy) Ohwi, a Chinese traditional drug used for the treatment of inflammation and rheumatic conditions. The structure determination was based upon spectroscopic analysis. Two of the neolignans were found to have new structures and were named as (-)-denudatin B (the enantiomer of denudatin B, II) and kadsurenin M (7S,8S-3,4,3'-trimethoxy-7'-oxo-nor-8',9'-7.O. 4',8,5'-neolignan, V). The known compounds kadsurenon (I), (-)-acuminatin(III) and (+)-licarin A(IV) were also obtained from the same source. (-)-Denudatin B (II) showed potent PAF antagonistic activity in 3H-PAF receptor binding assay.

[Studies on PAF antagonistic bicyclo(3,2,1) octanoid neolignans from Piper kadsura].

As a continuation of our previous report dealing with the structures of novel neolignans from Piper kadsura (Choisy) Ohwi, two other new bicyclo(3,2,1) octanoid neolignans, named kadsurenin K and kadsurenin L, were isolated. On the basis of spectral analysis and chemical derivatization, their structures were established as 7R,8R,3'R,5'R-delta 8'-3,5'-dimethoxy-4-hydroxy-2',3',4',5'-tetrahydro-2', 4'-oxo-7.3',8.5'-neolignan and 7R, 8R, 3'S, 4'R, 5'R-delta 8'-3,4,5'- trimethoxy-4'-acetoxy-2',3',4',5'-tetrahydro-2'-oxo-7.3', 8.5'-neolignan respectively. Kadsurenin B and kadsurenin C were known compounds. In 3H-PAF receptor binding assay, kadsurenin B, C, K and L demonstrated significant PAF-antagonistic activity.

Effect of orally administered betel leaf (Piper betle Linn.) on digestive enzymes of pancreas and intestinal mucosa and on bile production in rats.

The influence of two varieties of betel leaf (Piper betle Linn.) namely, the pungent Mysore and non-pungent Ambadi, was examined on digestive enzymes of pancreas and intestinal mucosa and on bile secretion in experimental rats. The betel leaves were administered orally at two doses which were either comparable to human consumption level or 5 times this. The results indicated that while these betel leaves do not influence bile secretion and composition, they have a significant stimulatory influence on pancreatic lipase activity. Besides, the Ambadi variety of betel leaf has a positive stimulatory influence on intestinal digestive enzymes, especially lipase, amylase and disaccharidases. A slight lowering in the activity of these intestinal enzymes was seen when Mysore variety of betel leaf was administered, and this variety also had a negative effect on pancreatic amylase. Further, both the betel leaf varieties have shown decreasing influence on pancreatic trypsin and chymotrypsin activities.

Organic amendments and control of foot rot of Piper betle caused by Phytophthora parasitica var. piperina.

Experiments have been performed to control foot rot of Piper betle by organic amendments and antagonists in soil. Soil amended with corn straw gave best control. Corn straw supplemented with nitrogen (NH4NO3) gave still better control. The effect of different antagonists growing on corn-straw and til (Sesamum indicum) oil cake amendments showed that Trichoderma viride gave best control. Aspergillus terreus failed to control the disease although it showed great antifungal activity under controlled condition in Petri dishes.

Piper methysticum (kava kava).

Piper methysticum (kava kava) is a plant native to the Pacific Island region, and has been used ceremonial for thousands of years. The active ingredients are a group of substances know as kava lactones (AKA kava pyrones). Four lactones in kava have been found to have significant analgesic and anesthetic effects via non-opiate pathways. Kava's most popular application is as a natural anxiolytic, comparing favorably in several studies to a number prescription medications, including benzodiazepines. CNS effects seem to be mediated by several mechanisms. Studies have been conflicting regarding its GABA-receptor-binding capacity, although this has been found to occur in some studies. In vitro kava has been found to block norepinephrine uptake. It also has some anti-convulsant capabilities, which appear to be mediated by Na+ channel receptor sites. The therapeutic dosage is in the range of 50-70 mg kava lactones three times daily. The most common side effect, usually seen only with long-term, heavy usage of the herb, is a scaly skin rash called "kava dermopathy." It has also been know to potentiate other medications such as barbiturates and Xanax.

Aflatoxin B1-induced micronuclei and cell cycle alterations in lung and bone marrow cells and their modulation by Piper argyrophyllum extract.

Aflatoxin B, (AFB1) is a clastogen that causes cellular damage by covalent modification of nucleic acids. In this investigation, male rats were injected i.p. with AFB1 (8 mg/kg b.w.) in DMSO and the same dose of AFB1 was also administered intratracheally (i.t.) to the animals separately. The animals were killed after 26 h of the carcinogen treatment, femur bone was removed, and bone marrow cells were isolated and stained with Mayer's hematoxylin and eosin. Micronuclei (Mn) were scored by using light microscopy. Bronchoalveolar lavage (BAL) was prepared from rats administered AFB1 i.t. A part of BAL was fixed with 70% ethanol, stained with the fluorochrome DAPI, and analysed for cell cycle variations; the other part of the lavage was used for making slides to record Mn with a fluorescent microscope. A significantly greater proportion of lung cells were found to enter cell cycle with extended S-phase due to AFB1 treatment. Mn were induced in polychromatic erythrocytes (PCE) as compared to normochromatic erythrocytes (NCE) in the bone marrow of AFB1-treated rats, where there was nearly a three-fold increase in the number of Mn of bone marrow cells. The administration of AFB1 resulted in a two-fold rise in the Mn in the lung cells. The effect of BSO, DEM, and PB, the modulators of AFB1 metabolism, was studied on AFB1-induced Mn formation. A significant increase in the Mn score in PCEs of BSO- and DEM-treated rats was noted, while a slight reduction in the Mn score was noted in the case of PB-treated rats. The administration of the methanol extract of the leaves of Piper argyrophyllum (taken up in DMSO) to rats for a week exhibited normalising effect on AFB1-induced Mn in bone marrow cells. These observations record the induction of Mn in lung cells due to AFB1 for the first time. We propose the utility of AFB1-induced Mn as a model for screening plant extracts as inhibitors of genotoxicity. Prevention of genotoxic changes described above by phytochemicals is being pursued in our Laboratories.

Efficacy of piperine, an alkaloidal constituent from Piper nigrum on erythrocyte antioxidant status in high fat diet and antithyroid drug induced hyperlipidemic rats.

The main aim of this study was to investigate the effect of piperine on erythrocyte antioxidant status in high fat diet (HFD) and antithyroid drug induced hyperlipidemic rats. Male Wistar rats were divided into eight groups. The first four groups were fed a control diet and in addition were given respectively 1% carboxymethyl cellulose (CMC); 10 mg/kg body weight carbimazole (CM); 10 mg CM + 40 mg/kg body weight piperine and 10 mg CM + 2 mg/kg body weight atorvastatin (ATV). A similar pattern was followed for the next four groups except that they were all fed HFD instead of the control diet. Erythrocyte osmotic fragility, total cholesterol, phospholipids, lipid peroxidation products, enzymic and non-enzymic antioxidant status were studied in all experimental groups. Significantly increased osmotic fragility, total cholesterol/phospholipid ratio, thiobarbituric acid reactive substances and lipid hydroperoxides were observed in the plasma and erythrocytes of HFD fed and CM treated rats compared to the control. Superoxide dismutase, catalase, glutathione peroxidase, vitamin E and reduced glutathione in erythrocytes and vitamin C in the plasma were also significantly lowered in HFD fed, antithyroid drug treated rats compared to control animals. Concurrent piperine supplementation along with HFD and antithyroid drug administration normalized erythrocyte osmotic fragility, reduced lipid peroxidation, and improved the enzymic and non-enzymic antioxidant status compared to those rats that did not receive piperine. Thus, our results indicate that piperine supplementation markedly protects erythrocytes from oxidative stress by improving the antioxidant status in HFD fed antithyroid drug treated rats.

Antioxidant activity of prenylated hydroquinone and benzoic acid derivatives from Piper crassinervium Kunth.

Growing evidence suggests that RNOS (reactive nitrogen and oxygen species) are involved in the damage of biomolecules, contributing to the aetiology of several human diseases. Thus, the demand for antioxidants has stimulated the search for new compounds with potential use in this field. The in vitro antioxidant potential of prenylated hydroquinones and prenylated 4-hydroxy-benzoic acids from fruits of P. crassinervium was evaluated in terms of their capacity to suppress both DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and chemiluminescence produced from luminol, using 2,2'-azo-bis(2-amidinopropane) (ABAP) as a peroxyl radical source. The inhibition of lipid peroxidation was assessed using liposomes from phosphatidylcholine as a membrane model. The prenylated hydroquinones had higher antioxidant activity than the benzoic acids and, among the hydroquinones, the E isomer was more efficient than the Z isomer.
