Developmental studies of the lethal gene Bld in the mouse. I. Post-implantation development of the lethal homozygote.

In matings of Bld/+ x Bld/+ mice a characteristic type of abnormal embryo is found on days 6 and 7 after impregnation which dies at 8 days and accounts for about 25% of all living embryos. These embryos are regarded as the fethal Bld/Bld homozygotes. Before 6 days the embryos appear slightly retarded. Entodermal cells invade the yolk sac and the trophectoderm does not fuse to the uterine decidua. Late on day 6 the yolk sac is filled with a cap of unorganized cells of entodermal origin, surrounded by both a thick membrane of non-living material, corresponding to Reichert's membrane, and external to this a continuous layer of trophectoderm: there is still almost no contact with uterine tissue. At 7 days 10 h amniotic folds are formed. Mesoderm appears occasionally but is not always present; when it appears it does not grow out from its place of origin. Entodermal cells, particularly in the proximal part of the extra-embryonic region, become polyploid. At the same time, the trophectoderm makes contact with the uterine decidua and gives rise to primary giant cells. Twelve hours later, the embryonic cells begin to degenerate, first in the region of the amniotic fold and the mesoderm. The embryo dies shortly afterwards. It is suggested that the primary target tissues affected by the Bld/Bld constitution are trophectoderm and entoderm. Many but not all of the abnormal features appearing later can by ascribed to insufficient nutrition of the embryo, due to failure of attachment of the trophectoderm and the filling of the yolk sac with entodermal cells.

[Effect of prostaglandin F2-alpha on the course of pregnancy and 17-beta-estradiol concentration in blood plasma of mice].

The effect of prostaglandin F2 alpha (at a dose of 2 mg/kg, per each stage of development) on the mouse preimplantation development and 17 beta-estradiol concentration in the blood plasma was studied. Prostaglandin F2 alpha caused inhibition of mitoses in the embryo and decreased the percentage of embryos liberated from zona pellucida. Meanwhile blood plasma showed diminution of 17 beta-estradiol level as a result of administering prostaglandin F2 alpha. In physiological conditions a significant rise in 17 beta-estradiol level was recorded at the stage of blastocyst liberated from zona pellucida.

[Effect of bee venom and its low- and high-molecular fractions on embryogenesis in rats].

The authors injected 3 groups of pregnant rats with 1/5 of LD50 of whole bee venom and its low and high weight molecular fractions daily between the 6th and 14th day of gestation. The embryos of the treated animals, aged 20 days, showed a reduction of the weight and dimunution of the activity of lactate dehydrogenase and isocitrate dehydrogenase in their liver homogenate. There were autolisis and dead rats in the uterus of the treated animals (embryotoxic effect). There were no teratogenic alterations. The low weight molecular fraction of the bee venom revealed more manifested embryotoxic effect.

[The effect of repeated applications of carbutamide on the embryonic development of the rat (author's transl)].

The present paper will check the effects of repeated applications of the oral "antidiabeticum" Carbutamide (substitute of the "sulfonylurea") on the embryonic development of wistar rats. The agent was given the pregnant animal by the means of a throat sonde at the 5th, 6th and 7th day post coitum suspended in Tween 80 in a dosage of 800 mg/kg bodyweight. In order to value the intrauterine development the following parameters were used: mean implantation rate, resorptions rate, bodyweight of fetuses, statemend of animalies by an accurate inspection of the body surface shape, of the skeleton and the inner organs. The rates of implantation do not differ from that of the control group. The rate of resorption increases dependend on the beginning of the treatment. The later the treatment begins the higher the resorptions rate. The body weight of surviving fetuses is diminished strongly. Damage of the skeleton appears as the presence of 14th ribs at one or each side or as ossifications at the 7th neck vertebra (neck rib). Also this results depend on the beginn of the treatment.

Antitumor effects of RNA isolated from murine tumors and embryos.

We describe the antitumor effects of a certain RNA(s) isolated from murine tumors and embryos. A single i.v. injected of 10 to 30 microgram of this RNA(s) induces necrosis, hemorrhages, and regression of solid tumors in the strain of origin; in pregnant mice it is embryotoxic, causing resorption of the embryos, but has no toxic effect upon the tumor-bearing or pregnant host. Its action is highly specific, and its presence in both tumors and embryos suggests that it may result from reexpression of embryonic genes in tumor cells.

[Experimental study of the teratogenicity and embryotoxicity of endodan].

An experiment was carried out on albino rats for the assessment of the effect, during gestation, of the fungicide endodan (ethylene - thiuram monosulphide) on the intrauterine and postnatal development of the progeny. A low teratogenic and embryotoxic activity was established after a single treatment with I/2 LD50 on 13 th gestation day. The preparation is with a pronounced cumulative effect in a daily administration of 1/5, 1/10 and 1/20 LD50, manifested by clinical symptomatics and lethality of a part of the pregnant animals. Via tests for the assessment of the behaviour, deviations in the functional state of the nervous system of the progeny were established, with no changes in the integral indices - viability and body weight gain during the postnatal period.

[Effect of lofenal on rat embryogenesis].

The embryotoxic and teratogenic effect of lofenal was studied at different stages of embryogenesis of albino rats. Postimplantation death of embryos and foetus anomalies are stated to depend on both the dose of the administered preparation and on the period of embryogenesis.

[Development and survival of mouse embryos in intra- and interstrain transplants].

Survival rate and weight of embryos were studied in the Balb/c, CBA//ac and C57BL/6j mice, developed from several combinations of transplantation. The weight of allogenic fetus as well as syngenic fetus which developed together with allogenic ones was detected to increase. This increased weight does not result from survival of certain genotype embryos, but seems to be due to genetical differences in the mother-embryo system.

[Lactate dehydrogenase activity of the ova and early embryos in BALB/c and C57BL/6 mice and their reciprocal hybrids].

The lactate dehydrogenase (LDH) activity in eggs and 8-cell mouse embryos of BALB/c, C57BL/6 strains and their reciprocal hybrids was studied. LDH pattern of mouse eggs and 8-cell embryos is represented by only one fast migrating isozyme LDH-1. For determining the water-soluble LDH activity water extraction of enzyme was used. For determining the total LDH activity triton X-100 extraction of enzyme was used. The bound LDH activity was estimated as a difference between total LDH and water-soluble LDH activities. The total LDH activity of eggs and 8-cell embryos for all genotypes is reliably higher than the water-soluble LDH activity. Differences between strains and hybrids are reliable. There is diversity of values of eggs and 8-cell embryos LDH activity. The diversity of values of water-soluble LDH activity is higher than that of the total LDH activity in all cases. The development from the egg to 8-cell embryo is accompanied by the decrease of the total LDH activity, the water-soluble LDH activity and the bound LDH activity for all genotypes except BALB/c. Only the water-soluble LDH activity decreases by BALB/c strain.

[Effect of ethanol on the genetic apparatus of mammalian germ cells].

A study was made of the frequency of dominant lethal mutations in germ cells at different stages of spermatogenesis, the level of chromosome aberrations in embryonic cells of male progeny treated with ethanol as well as the influence of ethanol on chromosomal apparatus of germ cells. Ethanol possesses a mutagenic activity, induces dominant lethal mutations which are more pronounced when gametes were treated 2 weeks before fertilization.
