Comparative immunological recognition of proteins from New Guinea "C" dengue virus type 2 prototype and from a dengue virus type 2 strain isolated in the State of Rio de Janeiro, Brazil.

The protein profiles of the New Guinea "C" dengue virus type 2 (DENV-2)prototype and those of a Brazilian DENV-2 isolated in the State of Rio de Janeiro in 1995 were compared. SDS-PAGE analysis showed that the virus from Rio de Janeiro expresses NS5 (93.0 kDa), NS3 (66.8 kDa) E (62.4 kDa) and NS1 (41.2 kDa) proteins differently from the New Guinea "C" virus. The immunoblot revealed specificity and antigenicity for the NS3 protein from DENV-2 Rio de Janeiro mainly in primary infections, convalescent cases, and in secondary infections in both cases and only antigenicity for E and NS1 proteins for both viruses in primary and secondary infections.

A model of the transmission of dengue fever with an evaluation of the impact of ultra-low volume (ULV) insecticide applications on dengue epidemics.

We have developed a deterministic susceptible, exposed, infectious, resistant or removed (SEIR) model of dengue fever transmission that enables us to explore the behavior of an epidemic, and to experiment with vector control practices. Populations of both host and vector are divided into compartments representing disease status (susceptible, exposed, infectious, and, for humans, resistant), and the flow between compartments is described by differential equations. Examination of the equilibrium points leads to a formulation of the basic reproduction rate (Z0) of the disease. With a base set of parameters, Z0 = 1.9 and the model realistically reproduces epidemic transmission in an immunologically naive population. Control of adult mosquitoes by ultra-low volume (ULV) aerosols is simulated by an abrupt decrease in vector densities, followed by gradual recovery of the vector population. The model indicates that ULV has little impact on disease incidence, even when multiple applications are made, although the peak of the epidemic may be delayed. Decreasing the carrying capacity of the environment for mosquitoes, and thus the basic reproduction rate of the disease, by source reduction or other means, is more effective in reducing transmission.

Dengue vaccines: preparing to roll back dengue.

Dengue is an emerging viral pathogen of growing global importance, infecting up to 100 million people annually. Although there have been many efforts over the last six decades to produce a vaccine to combat this infection, few have been able to meet the challenges posed by the unusual interplay between this virus and its human host. However, recent progress in molecular-based vaccine strategies, as well as a renewed commitment by the World Health Organisation to co-ordinate global efforts on vaccine development, finally provides hope that control of this serious disease may be at hand.

Genetic variations and relationship among dengue virus type 3 strains isolated from patients with mild or severe form of dengue disease in Indonesia and Thailand.

Sequence analysis was conducted on structural and non-structural genes of 7 strains of dengue virus type-3 (DENV-3 virus) isolated in Indonesia and Thailand in the year 1973, 1994, and 1998 from patients with different clinical manifestations. In general, sequence similarity among isolates was greater than 93%, indicating that the mutation rate of DENV-3 circulating in this region was not more than 7% in the last 3 decades and suggesting that sequences that may responsible for viral architectures and/or biological function were strictly conserved. Mutations unique to viral strains associated with specific clinical manifestations were not found. Alignment of PrM/M and E nucleic acid sequences followed by parsimony analysis of sequences obtained in this study and published elsewhere allowed generation of phylogenetic trees, demonstrating that DENV-3 strains isolated in Indonesia in 1998 belonged to a separate cluster (subtype 2) from those isolated between 1973-1985 (subtype 1).
