Rab3D and actin reveal distinct lamellar body subpopulations in alveolar epithelial type II cells.

Rab3D is a small GTP-binding protein associated with secretory vesicles in various exocrine and endocrine cells, where it has been implicated in regulated exocytosis. Data obtained previously in pancreas have suggested that rab3D is involved in the coating of secretory granules with filamentous actin. In the present study we employed Western blot analysis, immunofluorescence, and immunoelectron microscopy to examine the distribution of rab3D in rat lung. Rab3D immunoreactivity was detected in bronchiolar Clara cells and alveolar epithelial type II (AET-II) cells. In both cell types, rab3D displayed preferential localization to secretory vesicles that were identified using specific antibodies against Clara Cell Secretory Protein and p180 lamellar body protein, respectively. Interestingly, rab3D was associated with only 24% of the lamellar bodies in AET-II cells. Rab3D-positive lamellar bodies were typically in close proximity of the apical plasma membrane, where exocytosis occurs. Another subpopulation of lamellar bodies, constituting only 2%, was not only rab3D-positive but could also be labeled with the filamentous-actin probe phalloidin. A third subpopulation, constituting 9%, displayed actin coating without rab3D staining. We propose that these three lamellar body subpopulations represent consecutive intermediates along the regulated exocytotic pathway, implying that rab3D release and actin coating are intimately linked processes.

[ENT emergencies due to foreign bodies at a tertiary hospital].

A study of the patients who, through a year, came to the Emergencies Department showing foreign bodies in ENT-area has been accomplished. Among ENT urgencies foreign bodies constitute the most frequent cases. Age, sex, date, time of admission, sitting, kind of body and treatment are taken in consideration. The intruders were most commonly found in the pharyngeal area. A striking correspondence between age and location is underlined.

Evidence for the presence of substance P in cat nasal receptor afferents.

The afferents of the nasal receptors responsible for many upper airway protective reflexes are carried in the ethmoidal branch of the ophthalmic division of the trigeminal nerve. Previous electrophysiological studies indicate that a significant number of ethmoidal afferents respond to noxious stimuli applied to the nose. The objective of the present study was to identify ethmoidal nerve cell bodies within the trigeminal ganglion which demonstrated the presence of the neurotransmitter substance P (SP). SP is believed to be involved in the relay of nociceptive information. A double-labelling technique was employed and involved tracing the afferents to their cell bodies using horseradish peroxidase (HRP) and subsequent identification of SP-immunoreactivity within HRP-filled cells using monoclonal antibody immunohistochemistry. SP-immunoreactive cell bodies constituted 43 per cent-50 per cent of the total number of labelled ethmoidal cell bodies within the trigeminal ganglion. Although ethmoidal cell bodies were much smaller than the overall population of trigeminal ganglion cells, the size of SP-immunoreactive ethmoidal cell bodies was not significantly different from that of ethmoidal cell bodies not exhibiting SP-immunoreactivity.

Energy metabolism in the brain, adaptation towards starvation.

It is hypothesized that the brain's incapacity to utilize energy sources other than glucose and ketone bodies, constitutes a mechanism of protecting it from self degradation, at the expense of the rest of the body, during periods of starvation. This protection is based upon the brain's inability to catabolize substances of which it is built, for energy production.

Islet amyloid polypeptide (amylin) is expressed in sensory neurons.

Islet amyloid polypeptide (IAPP) or amylin is a hormone candidate predominantly expressed in insulin cells. A role for IAPP in the regulation of glucose homeostasis and the development of non-insulin-dependent diabetes mellitus has been proposed. IAPP is structurally related to the sensory neuropeptide calcitonin gene-related peptide. In the present study, using in situ hybridization, immunocytochemistry, and immunochemistry, the expression of IAPP in sensory neurons in the rat was investigated. IAPP was expressed in a population of small- to medium-sized nerve cell bodies in dorsal root ganglia from all levels and in the jugular-nodose and trigeminal ganglion; IAPP-expressing nerve cell bodies constituted a subpopulation of those expressing calcitonin gene-related peptide. In addition, IAPP-like immunoreactivity occurred in nerve cell bodies storing substance P and pituitary adenylate cyclase-activating polypeptide. IAPP-immunoreactive nerve fibers were encountered in the dorsal horns of the spinal cord, and to a lesser extent in peripheral tissues receiving sensory innervation; IAPP-immunoreactive fibers constituted a subpopulation of those containing calcitonin gene-related peptide and/or substance P. The immunochemical determinations demonstrated a low level of IAPP-like immunoreactivity in the dorsal root ganglia and spinal cord, which chromatographically coeluted with authentic rat IAPP. We conclude that IAPP is expressed in sensory neurons, thus being a novel sensory neuropeptide candidate for which a physiological role remains to be identified.

The parenchymal cell mass in normal human parathyroid glands.

The parenchymal cell mass in parathyroid glands from 368 autopsy cases without apparent parathyroid or kidney disease was investigated. By measuring the glandular density with a density gradient technique and the glandular weight, the parenchymal to fat cell ratio and the weight of the parenchymal cell mass could be determined. The mean parathyroid parenchymal cell weight was calculated to be 22 mg and the maximal normal parenchymal cell weight 39 mg. The glandular weight was evidently dependent on the body constitution, as the fat tissue content of the glands was related to the amount of fat tissue in the body as a whole. The parenchymal cell weight was much less variable. Also, the amount of fat tissue in the parathyroid glands was positively correlated to the size of glands. Thus, the amount of fat cells within parathyroid glands seems less adequate as a histological criterion and the parenchymal cell weight should be of greater value in the histological diagnosis that the weight of the whole gland. The age-and-sex-related variations of the parenchymal cell mass were slight and therefore a value of 39 mg may be considered an appropriate upper limit of the weight of this mass in parathyroid glands of adult persons.

[Orbital foreign bodies -- diagnostics, therapy and management].

BACKGROUND: Orbital foreign bodies constitute an interdisciplinary challenge, diagnosis being especially difficult because of inconspicuous entrance orifices. Indication for the removal of an orbital foreign body has always to be decided upon individually taking into account the benefits and risks resulting from an operation. The latter is particularly indicated if there are acute and chronic functional restrictions or inflammatory reactions either present or to be expected. The objective of our study was to produce systematic guidelines concerning the therapeutic procedure of dealing with orbital foreign bodies with a view to a benefit-risk assessment. PATIENTS AND METHODS: Over the last ten years ten patients with an orbital foreign body were treated in our Department, four of them requiring acute attention. CTs were taken of all patients. Only in one case additional MRTs were produced. Eight out of ten patients underwent operation, of which seven resulted in a complete removal of the foreign body. In the case of two patients we decided against an operation after interdisciplinary consultation. RESULTS: In our view the imaging--providing diagnostics by means of computer tomography is the most promising method for determining a foreign body in the orbit. Decision for an operative removal of a foreign body was invariably taken interdisciplinary in cooperation with an ophthalmologist. Surgical approach was chosen in accordance with the localisation of the foreign bodies. None of the patients suffered from a loss of vision or a restriction of functions as a result of the operation. CONCLUSIONS: In the indication the foreign material is of essential importance. Orbital foreign bodies that have been present in the tissue for a longer time can be allowed to stay, unless loss of functions are progressive. Loss of vision always being a consequence of the initial trauma.

Substantia nigra Marinesco bodies are associated with decreased striatal expression of dopaminergic markers.

Marinesco bodies are nuclear inclusions found in pigmented neurons of the substantia nigra and locus ceruleus of humans and monkeys. It has long been known that the frequency of these inclusions increases with advancing age, but no pathologic associations have ever been established. We quantified Marinesco body frequency in human autopsy subjects, classified as young normal controls, elderly controls, dementia with Lewy bodies (DLB), Alzheimer disease (AD), and Parkinson disease (PD). Elderly controls, AD cases, and DLB cases had significantly increased Marinesco body frequencies relative to young controls and DLB cases had significantly increased frequencies relative to elderly controls, while PD cases did not differ from young controls; cases with AD did not differ from elderly controls. Lewy body-containing neurons had significantly higher Marinesco body frequencies than non-Lewy body-containing neurons. Marinesco body frequency in elderly control cases correlated significantly, in inverse fashion, with striatal concentrations of the dopaminergic neuron markers dopamine transporter and tyrosine hydroxylase. These statistical associations suggest that Marinesco bodies constitute or mark a pathologic process that may be related to, or contribute to, age-related motor dysfunction and/or Lewy body disorders. Further studies are needed to ascertain the molecular basis of Marinesco body formation; preliminary studies indicate that proteasome dysfunction can lead to similar abnormalities in cultured cells.

Respiratory responses to electrical stimulation of the cervical sympathetic nerves in decerebrate, unanaesthetized cats.

1. The effects of respiration of electrical stimulation of the peripheral cut end of the cervical sympathetic nerve were studied in seventeen decerebrate, unanaesthetized cats.2. The tidal volume increased and the end-tidal CO(2) fell within 2-3 breaths after the onset of stimulation in fourteen cats.3. In three experiments tidal volume and respiratory rate fell during stimulation and increased above the control when the stimulus was interrupted.4. All respiratory responses to sympathetic stimulation were abolished after cutting the carotid sinus nerves and are attributed to activation of chemo- and baroreceptor afferents.5. With the carotid sinus nerves intact, respiratory responses could be eliminated completely only by cutting the post-ganglionic sympathetics that pass directly to the carotid body and the post-ganglionic sympathetics that join the glossopharyngeal nerves to course with the sinus nerves to the carotid bifurcation.6. Pentobarbitone (10 mg/kg) or chloralose (40 mg/kg) given intravenously depressed spontaneous ventilation and responses to sympathetic nerve stimulation.7. Stimulation changed blood pressure slightly in fourteen experiments; in ten it fell and in four it increased 10 mm Hg or less. After pentobarbitone or chloralose, stimulation elicited a pronounced pressor response.8. The results of the study indicate that activation of sympathetic pathways to the carotid body constitutes an effective stimulus to ventilation.

The relation between bone mineral density, insulin-like growth factor I, lipoprotein (a), body composition, and muscle strength in adolescent males.

Osteoporosis is the most common metabolic bone disease. A low peak bone mass is regarded a risk factor for osteoporosis. Heredity, physical activity, and nutrition are regarded important measures for the observed variance in peak bone mass. Lp(a) lipoprotein is a well-known risk factor for atherosclerosis. Serum insulin-like growth factor I (IGF-I) has been found to be increased in males with early cardiovascular disease. In this study, we evaluated the association between bone mass, body constitution, muscle strength, Lp(a), and IGF-I in 47 Caucasian male adolescents (mean age, 16.9 yr). Bone mineral density (BMD) and body composition were measured by dual x-ray absorptiometry, muscle strength of thigh using an isokinetic dynamometer, IGF-I by RIA, and Lp(a) by enzyme-linked immunosorbent assay. IGF-I was only associated with Lp(a) (r = 0.38, P < 0.01). Lp(a) was related to total body (r = 0.40, P < 0.01), skull (r = 0.45, P < 0.01), and femoral neck BMD (r = 0.44, P < 0.01). Lp(a) was also related to fat mass (r = 0.34, P < 0.05) and muscle strength (r = 0.30-0.42, P < 0.05). After multiple regression and principal component (PC) analysis, the so-called PC body size (weight, fat mass, lean body mass, and muscle strength) was the most significant predictor of BMD (beta = 0.28-0.51, P < 0.05-0.01), followed by the so-called PC physical activity (beta = 0.28-0.38, P < 0.05-0.01, weight-bearing locations). However, the PC analysis confirmed that Lp(a) was an independent predictor of total body, skull, and femoral neck BMD (beta = 0.33-0.36, P < 0.01). The present investigation confirms that BMD, body size, and muscle strength are closely related and that the level of physical activity is a major determinant of BMD. However, the positive relation of Lp(a), a major risk factor for cardiovascular disease, to BMD has not previously been described. The importance of this observation has to be further investigated.

Subdivisions of the medial geniculate body in the tree shrew (Tupaia glis).

The medial geniculate body of the tree shrew has 3 major divisions which can be identified on the basis of cytoarchitecture. Each of these major divisions can be subdivided further. The present paper describes these divisions and compares them in terms of their patterns of afferent and efferent connections. In particular, anterograde degeneration and autoradiographic techniques are used to demonstrate that the dorsal and medial divisions receive projections from the deep layers of the superior colliculus and from the tegmentum. Results are also presented which indicate that the dorsal division projects to a non-primary cortical area adjacent to primary auditory cortex. Evidence is discussed which suggests that large sectors of what we have called the medial geniculate body constitute subdivisions of the auditory thalamus.

Lipid bodies: intracellular sites for eicosanoid formation.

Lipid bodies, therefore, represent specialized intracellular domains that form rapidly in response to agents that activate protein kinase C. These structures contain eicosanoid-precursor arachidonate esterified in specific phospholipids. Arachidonate-releasing phospholipases probably act at lipid bodies, and an eicosanoid-forming enzyme, PGH synthase (cyclooxygenase), definitely localizes to lipid bodies. In addition, the heightened presence of lipid bodies in cells likely to be producing eicosanoids as part of inflammatory reactions indicates that lipid bodies are dynamic, specialized intracellular domains with roles pertinent to the metabolic transformation of arachidonate into paracrine mediators of inflammation. With their prominence in cells in association with inflammation, lipid bodies constitute specialized sites at which eicosanoid formation could occur for the heightened generation of eosinophil eicosanoid mediators of inflammation. This compartmentalization of eicosanoid formation at lipid bodies would provide a nonmembrane pool of arachidonate whose metabolic utilization could occur without perturbation of membranes if membranes were the sole stores of substrate fatty acid used for quantities of eicosanoids synthesized as paracrine mediators of inflammation. Moreover, lipid bodies would serve as sites at which the coordinated and regulated enzymatic events involved in arachidonate mobilization and oxidative metabolism could occur.

Cell cycle-dependent association of PML bodies with sites of active transcription in nuclei of mammalian cells.

Promyelocytic leukemia (PML) nuclear bodies constitute one class of intranuclear domains that may be directly involved in the expression of specific genes. Here we have analyzed the spatial relationship between PML bodies and sites of transcriptional activity by indirect immunofluorescence and confocal microscopy during the cell cycle. In unsynchronized mammalian cells approx 30% of PML bodies are spatially associated with transcription sites. These sites contain hyperphosphorylated RNA polymerase II, indicating active mRNA transcription tightly associated with the PML body. In G1 phase of the cell cycle more than 70% of PML bodies contain active transcription foci. A similarly high degree of colocalization (approx 80%) between PML bodies and sites of active transcription was also observed when the cells were exposed to interferon-gamma. We also show that the hypophosphorylated form of RNA polymerase II and the transcriptional coactivator CBP colocalize within PML bodies predominantly in G1. Our observations suggest that PML bodies may be recruited to nuclear sites of induced or up-regulated mRNA transcription where it may serve as a scaffold for factors involved in expression of specific genes.

[Mutation in the beta3-adrenergic receptor gene (Trp64Arg) does not influence insulin resistence, energy metabolism, fat distribution and lipid spectrum in young people. Pilot study].

UNLABELLED: A missence mutation Trp64Arg in the beta3-adrenergic receptor gene is associated with obesity, insulin resistance, a lower metabolic rate and the earlier onset of NIDDM but the published results are controversial. We investigated the effect of this mutation on insulin resistance (euglycemic hyperinsulinemic clamp), on fat mass and fat distribution (anthropometry, bioimpedance, CT) and resting metabolic rate (indirect calorimetry), lipid spectrum and other metabolic disturbances in Czech juveniles recruited from juvenile hypertensives (H, n = 68) and controls (C, n = 81). The frequency of this mutation (determined by digestion of 210 bp PCR product with MvaI) was double in H than in C (14.7%, vs. 7.4%) and the carriers of Arg64 allele had sig. higher fasting glucose (H: p = 0.002. C: p = 0.025). Four Trp64/Arg64 and six Trp64/Trp64 men (age 23 +/- 4.2, vs. 22.5 +/- 1.9 y, BMI 26 +/- 5.5, vs. 22.9 +/- 5.1 kg/m2) took part in a detailed pilot study. But no signif. differences (Horn's method) in fasting glucose (4.6 +/- 0.6, vs. 4.9 +/- 0.4 mmol/l), in parameters of insulin resistance (M-value150-180 min. 9.1 +/- 1.1, vs. 8.9 +/- 1.5 mg glucose/kg.min(-1)), resting metabolic rate/lean body mass (RMR/kg LBM: 78.6 +/- 4.6, vs. 85.6 +/- 23.2 kJ/kg), lipid spectrum and other screened parameters were found. The lowest resting metabolic rate (RMR/kg LBM 55.4; 62.6 kJ/kg) was found in brothers (both C, Trp64/Trp64) who highly differ in body constitution (BMI 19.0 resp. 32.4 kg/m2). We suppose that in this case the energy metabolism is probably determined by other genetic loci and does not correlate with body fat mass. CONCLUSION: Our pilot study does not confirm the influence of Trp64Arg mutation in heterozygous carriers on insulin resistance, energy metabolism and lipid spectrum.

[Mind, body and the Freudian concept of the psychic].

The purpose of this paper is twofold. In the first place, I intend to set up a general framework for the classification of standard philosophical theories concerning the so-called mind-body problem. Starting with a description of some relevant features of the pretheoretical distinction between the body and the mind. I follow the received distinction between dualistic theories (namely, theories maintaining that mind and body constitute two different ontological realms), monistic theories (that is, theories asserting that, ontologically speaking, only mind or matter exists), and attributive theories (namely, theories positing that mind and body are attributes of a neutral staff). I analyse several versions of each type, paying due attention to interactionism and to the so-called identity theory. In the second place I rise a crucial question concerning Freud's views about the nature of the psychical: was Freud a dualist, a monist or and attributionist? Is it possible to assign Freud a neat position on the matter? I argue that Freud's views of the psychical amount to a kind of monism of a materialistic or physicalistic type. As to the version of monism ascribable to Freud, I maintain that it is impossible to produce a precise and simple answer. He seems to fluctuate between a plain reductionism, a sort of identity theory and, even, some version of epiphenomenism.

[The effect of inbreeding on the manifestations of some physiological traits in the Kladrub Black Horse strain].

Inbreeding was used in the regeneration process in the breeding of the Kladrub Black Horse. Inbreeding rate is realized in practice, as related to the manifestation of the selected characteristics of this unique horse population. The evaluation of the effect of inbreeding on conception rate and fertility did not demonstrate any differences between the group of inbred mares and non-inbred mares. The study was conducted in mares used for breeding for 10 years. The results provide evidence that inbreeding causes no depression in this characteristics in the Kladrub Black Horse which would otherwise be the first physiological function in the reproduction process to be altered as a result of inbreeding. The evaluation of the effect of inbreeding on growth rate by comparing it with the standard growth curves which were characterized by the exponential function y = A (1 -- e-k(t-to)) did not indicate any statistically significant differences in growth dynamics between foals with a zero Fx value and inbred foals. This study was based on the evaluation of growth rate in the basic body traits and weight. It is inferred that inbreeding does not reduce growth dynamics of different body measurements and weight. Similarly, the evaluation of the body dimensions and weight of inbred and non-inbred adults showed no significant differences. Similar results were obtained from the evaluation of the relation between inbreeding and body constitution, character, and temperament. All the correlation coefficients are very low, almost equal to zero.

Protein bodies of mung bean cotyledons as autophagic organelles.

We present evidence that protein bodies constitute the principal lytic compartment in storage parenchyma cells of mung bean cotyledons and propose that they play a role in cellular autophagy. We developed a method to isolate protein bodies by incubating tissue slices with cell wall-degrading enzymes and fractionating the cellular organelles on a Ficoll gradient. About 75-80% of the protein bodies present in the protoplasts were recovered intact in a band at the 5/25% Ficoll interface. This band contained a similar proportion of the cellular alpha-mannosidase, N-acetyl-beta-glucosaminidase, ribonuclease, acid phosphatase, phosphodiesterase, and phospholipase D. beta-Amylase was present in the cells but not in the protein bodies. Ultrastructural observations showed that on the 3rd day of seedling growth protein bodies contain small vesicles (0.3-1.0 mum) with a cytoplasmic content (ribosomes, membrane vesicles, mitochondria). Later in seedling growth these vesicles appeared empty. We believe that these are autophagic vesicles resulting from invaginations of the protein body membrane and that their cytoplasmic contents are digested by the acid hydrolases present in the protein bodies.
