Expression in Saccharomyces cerevisiae of chimeric cytochrome P450 cDNAs constructed from cDNAs for rat cytochrome P450c and P450d.

Three chimeric cytochrome P450 cDNAs were constructed by replacing the central region, carboxy-terminal region, or both central and carboxy-terminal regions of cytochrome P450c cDNA with the corresponding regions of cytochrome P450d cDNA. These were inserted between the alcohol dehydrogenase I promoter and terminator of yeast expression vector pAAH5 to form expression plasmids pACDC2, pACCD1, and pACDD2. On introduction of each of these plasmids into Saccharomyces cerevisiae AH22 cells, chimeric cytochrome P450 proteins were expressed in AH22/pACDC2, AH22/pACCD1, and AH22/pACDD2 cells at the level of at least 10(5), 4 X 10(5) molecules per cell, respectively. The reduced CO-difference spectra showed that AH22/pACCD1 and AH22/pACDD2 cells contained 4 X 10(5) and 10(5) molecules per cell of the corresponding chimeric cytochrome P450 hemoproteins, designated as cytochrome P450ccd and cytochrome P450cdd, respectively. Cytochrome P450ccd exhibited higher monooxygenase activities toward 7-ethoxycoumarin, acetanilide, and benzo[alpha]pyrene than cytochrome P450c, although the substrate specificity of cytochrome P450ccd seemed to be the same as that of cytochrome P450c. Cytochrome P450cdd exhibited lower activities toward 7-ethoxycoumarin and benzo[alpha]pyrene, and a higher activity toward acetanilide as compared with those of cytochrome P450c and cytochrome P450ccd. Therefore, the substrate specificity of cytochrome P450cdd seemed to be the same as that of cytochrome P450d. These results suggest that the central one-third region of cytochrome P450c and cytochrome P450d is responsible for substrate-binding, and that the carboxy-terminal third of both cytochromes P450 plays an important role in electron transport.

Evidence in rat and mouse liver for temporal control of two forms of cytochrome P-450 inducible by 2,3,7,8-tetrachlorodibenzo-p-dioxin.

In the liver of perinatal rats or mice, the ratio of 2,3,7,8-tetrachlorodibenzo-p-dioxin-induced aryl hydrocarbon hydroxylase to total cytochrome P-450 content decreases, whereas the ratio of 2,3,7,8-tetrachlorodibenzo-p-dioxin-induced acetanilide 4-hydroxylase to total cytochrome P-450 content increases, between 18 or 19 days and 22 days following conception. The ontogenesis of inducible aryl hydrocarbon hydroxylase corresponds well with increases in a 56000-Mr electrophoretic band; we suggest this band represents the cytochrome P1-450 subunit. The later temporal expression of inducible acetanilide 4-hydroxylase closely parallels 2,3,7,8-tetrachlorodibenzo-p-dioxin-induced increases in size of a 54000-Mr electrophoretic band and a 2--3-nm hypsochromic shift in the Soret peak of the total microsomal reduced cytochrome P-450 . CO complex. We suggest this band represents the cytochrome P-448 subunit. Previous work from this laboratory has shown that this developmental difference is separated by several weeks in rabbit liver, as compared with several day's separation shown in this report with rat or mouse liver. The data here therefore provide evidence in the rodent for temporal control of the expression of different structural gene products regulated by the Ah locus.

Purification and characterization of a novel enzyme, arylalkyl acylamidase, from Pseudomonas putida Sc2.

A novel enzyme, arylalkyl acylamidase, which shows a strict specificity for N-acetyl arylalkylamines, but not acetanilide derivatives, was purified from the culture broth of Pseudomonas putida Sc2. The purified enzyme appeared to be homogeneous, as judged by native and SDS/PAGE. The enzyme has a molecular mass of approximately 150 kDa and consists of four identical subunits. The purified enzyme catalyzed the hydrolysis of N-acetyl-2-phenylethylamine to 2-phenylethylamine and acetic acid at the rate of 6.25 mumol.min-1.mg-1 at 30 degrees C. It also catalyzed the hydrolysis of various N-acetyl arylalkylamines containing a benzene or indole ring, and acetic acid arylalkyl esters. The enzyme did not hydrolyze acetanilide, N-acetyl aliphatic amines, N-acetyl amino acids, N-acetyl amino sugars or acylthiocholine. The apparent Km for N-acetylbenzylamine, N-acetyl-2-phenylethylamine and N-acetyl-3-phenylpropylamine are 41 mM, 0.31 mM and 1.6 mM, respectively. The purified enzyme was sensitive to thiol reagents such as Ag2SO4, HgCl2 and p-chloromercuribenzoic acid, and its activity was enhanced by divalent metal ions such as Zn2+, Mg2+ and Mn2+.

Stabilization of the hydrophilic sphere of non-ionic monomers: are all protected in a similar way?

The present study attempts to investigate whether incorporation of a methyl group as second substituent in the tertiary amido group of the two benzamide side chains of iobitridol (Xenetix) increases the stability of the hydrophilic sphere around this molecule as claimed by its manufacturer, and whether this hydrophilic sphere is unique to this particular molecule or to what extent other monomer non-ionic contrast media show this feature. Five non-ionic monomer contrast medium molecules, ioversol, iohexol, iobitridol, ioxilan and iopromide, were studied. Barriers to the rotation of acetanilide and benzamide side chains, and to the rotation of the amide bond in the benzamide chains, were calculated at a semi-empirical quantum mechanical level of theory with the Mopac/Ampac computer program. The five studied contrast medium molecules showed very similar energy barriers to the rotation of complete substituent chains (benzamide and acetanilide) around their bond with the benzene ring. The magnitude of the barriers fell in the range of values mentioned in the literature. In conclusion, introduction of a methyl group as second substituent in the tertiary amido group of the benzamide substituent chains of iobitridol does not increase the studied rotation barriers, i.e. it does not seem to stabilize the hydrophilic sphere to a greater extent compared with similar monomer non-ionic molecules. The sphere is shown to exist in all five analysed molecules. Introduction of a group with a more hydrophobic character has to be considered and eventually questioned, bearing in mind that a more hydrophilic molecule interacts to a lesser degree with body systems.

Pharmacological actions of some simple analogues of reserpine. II. Anticonvulsant effects.

Reserpine analogues obtained from strip-down parts of the parent molecule were studied for their effects on experimental convulsive seizures and amphetamine antagonism. The unsubstituted and M-methoxy substituted piperidino- and morpholino acetanilides derived from A, B and C rings or reserpine showed prominent anticonvulsant activity as against the known lowering of threshold activity by reserpine. Methoxy morpholino derivative showed prominent activity with least sedative effect. gamma-Piperidino acetanilide showed a dual effect such that at higher doses it itself produced clocin covulsions. The effects were non-specific since they unpreferentially antagonized the maximal electrochock, Metrazol and strychnine convulsions Compounds derived from C, D and E rings of reserpine did not significantly increase or decrease the seizure thresholds.

Coordinated maternal imprinting and paternal genotype determined the ultimate level of hepatic cytochrome P-450 and associated monooxygenase activities.

To understand the pre- and postnatal maternal influence on hepatic drug metabolism later in adult life, we have chosen to study the mouse hybrid (129/sv x LS) offspring derived from contrasting parental lines in a designated maternal environment. Results of these studies revealed that the following: 1) sex of the offspring was the major determinant for cytochrome P-450-specific content and for the activity of aryl hydrocarbon hydroxylase, testosterone 16 alpha-hydroxylase, acetanilide hydroxylase, and NADPH-cytochrome c reductase; 2) maternal factor(s) affected cytochrome P-450-specific content and the activities of acetanilide hydroxylase and aryl hydrocarbon hydroxylase but not of testosterone 15 alpha- and 16 alpha-hydroxylases or NADPH-cytochrome c reductase; 3) superimposed upon sex and maternal determination, paternal genotype significantly contributed to adult hepatic cytochrome P-450-specific content; 4) interactions between sex of the offspring and maternal factor(s) accounted for the development of testosterone 15 alpha- and 16 alpha-hydroxylase activities later in adult life; and 5) with the exception of testosterone 16 alpha-hydroxylase activity, other yet undefined factors, including the paternal genotype, contributed to most of the measured hepatic monooxygenase activities. The mechanism of coordinated maternal imprinting and paternal genotype in determining the levels of hepatic cytochrome P-450 and its associated monooxygenase activities is discussed.

[Anilides with analgesic and antipyretic activity].

Several trimethylcyclopentyl and trimethylcyclopentenyl acetanilides, mono- and di-substituted on the aromatic nucleus as well as the corresponding acyl derivatives from aniline it self and corresponding phenylacetanilides, were prepared and tested as analgesics and antipyretics. Several compounds exhibit considerable activity in some cases superior to that of acetanilide.

Effects of cytochrome b5 on cytochrome P-450-catalyzed reactions. Studies with manganese-substituted cytochrome b5.

The effects of cytochrome b5 with manganese-protoporphyrin IX substituted for heme were compared with those of native cytochrome b5 and the apoenzyme on the oxygenation of substrates in the reconstituted system containing liver microsomal cytochrome P-450, NADPH-cytochrome P-450 reductase, and phosphatidylcholine. Mn-b5, unlike b5, remains essentially fully oxidized in the presence of NADPH and NADPH-cytochrome P-450 reductase under aerobic conditions. The effects of various concentrations of b5 and its derivatives were determined at constant P-450 and reductase concentrations. Cytochrome b5 inhibits benzphetamine demethylation by isozyme 2, the effect increasing up to the highest concentrations tested, and stimulates 7-ethoxycoumarin deethylation by isozyme 2 and acetanilide p-hydroxylation by isozyme 4, the optimal b5:P-450 molar ratio being about 2. In contrast, Mn-b5 inhibits all three reactions and apo-b5 is either inactive or slightly inhibitory. The activities of the three substrates as well as testosterone were determined with P-450 isozymes 2, 3b, 3c, and 4 in the reconstituted system with no additions or with b5 or Mn-b5 present. Cytochrome b5 is stimulatory, inhibitory, or without any effect, the result depending on both the substrate and P-450 isozyme present, whereas Mn-b5 is inhibitory in most instances. Both b5 and its manganese derivative alter the rates of testosterone 6 beta- or 16 alpha-hydroxylation by most of the P-450 cytochromes. The activities are influenced by the molar ratio of reductase to P-450. The Km values of benzphetamine, ethoxycoumarin, and acetanilide are, with one exception, significantly decreased in the presence of b5 or Mn-b5. We conclude that some of the effects of b5 on the oxygenase system are not accounted for by its role as an electron donor to cytochrome P-450.

Effect of 6-MFA, an interferon inducer obtained from fungus Aspergillus ochraceus on hepatic mixed function oxidase system in rats.

Effect of 6-MFA (sixth mycelial fraction of acetone), an interferon inducer obtained from fungus A. ochraceus on hepatic mixed function oxidase system (MFO) of rat has been investigated. Treatment with 6-MFA, 100 mg/kg/day, ip for 1-5 days to adult rats inhibited significantly the different indices of MFO system, viz. hepatic cytochrome P-450, cytochrome b5 content, cytochrome c reductase, aminopyrine-N-demethylase and acetanilide hydroxylase activities. Similar treatment for 3 days in young growing rats significantly inhibited MFO system's components except acetanilide hydroxylase activity which showed marked elevation. These effects seem to be specific as in vitro experiments suggested that 6-MFA does not compete with subsdtrates nor it acts as a sponge reacting with the end product to give false inhibitory effect. It is concluded from the present study that 6-MFA like other interferon inducers depresses MFO system in rats. Its possible clinical implications are discussed.

PHOTOCHEMICAL ACTION SPECTRUM OF THE TERMINAL OXIDASE OF MIXED FUNCTION OXIDASE SYSTEMS.

The reversal of the carbon monoxide inhibition by bands of monochromatic light was determined for the oxidative demethylation of codeine and monomethyl-4-aminopyrine and the hydroxylation of acetanilide by rat liver microsomes and for the hydroxylation of 17-hydroxyprogesterone at carbon-21 by bovine adrenocortical microsomes. Maximum reversal occurred at 450 millimicrons, the light absorption maximum of the CO compound of the CO-binding pigment of microsomes. The agreement between photochemical action spectrum and spectrophotometric difference spectrum supports the conclusion that the CO-binding pigment is the terminal oxidase of mixed function oxidase systems of mammals.

Partial purification and characterization of an esterase from Fusarium sporotrichioides.

Kinetics analysis of the growth of Fusarium sporotrichioides T-424 at 15 degrees C and 25 degrees C in liquid culture for 35 days revealed that production of deacetylated trichothecenes was associated with an increased activity in fungal esterases. High temperature (25 degrees C) favored enzyme production and enhanced esterase activity. Electrophoresis of crude extracts from the mycelia of F. sporotrichioides T-424 with carboxylesterase staining revealed that several esterases were produced by the fungus. Four carboxylesterase isoenzymes (I-IV) were separated on a DEAE-Sephadex anion exchange column. Type (III) esterase, having activities with the substrate 4-nitrophenylacetate and acetanilide, as well as hydrolytic activity for T-2 toxin and acetyl-T-2 toxin, was partially purified with ammonium sulfate precipitation, immunoaffinity column chromatography, and DEAE-Sephadex A-50 chromatography. The esterase (III) had a molecular weight around 68 kDa in SDS-PAGE. For the deacylation of T-2 toxin and acetyl-T-2 toxin, type (III) esterase had a high specificity for the acetyl group at the C-3 and C-4 positions. The Km values for acetyl-T-2 and T-2 toxin were found to be 41.35 microM and 0.38 microM, respectively. The Km value for the acetyl group at C-3 is 110 times greater than for that at C-4.

Analysis of polyanionic macromolecular carrier poly-(N-vinylpyrrolidone-co-maleic acid) and its bioconjugats by capillary electrophoresis.

Anionic carrier poly(N-vinylpyrrolidone-co-maleic acid) and its conjugates, prepared with coupling of 2-cyano-3-hydroxy-5-amino-2-pentenoyc(4-trifluoromethyl anilide) or (6', 7'-dimethyl-l'-quinoxalinyl)-4-(2' amino) acetanilide to the carrier, were analyzed by capillary zone electrophoresis (CZE) and micellar electrokinetic chromatography (MEKC) in the following buffers: 0.25 N triethylammonium phosphate (TEAP); sodium dodecyl sulfate (SDS; 25-150 mM) in TEAP (pH 2.25-6.30); 0.1 N Na-borate buffer (pH range 7-11) and SDS (25-150 mM) in Na-borate buffer (pH range 7-11). The presence of strong carboxyl groups (dissociated even at pH 2.25) on the polymer chain was proved by the CZE method. It was also proved by potentiometric titration that carboxyls with a wide range of acidity were on the polymer chain. CZE was able to differentiate among the analytes possessing carboxyl groups of different acidic strengths at pH 2.25. These components were not distinguished by CZE at high pH values (11.0). Interaction between the analyte and SDS affected the separation at this pH, and hence good resolution was obtained by MEKC. Informative separations were achieved both for the carrier and the conjugates in TEAP buffer at pH 2.25 by the CZE method. Optimal separation was achieved in borate buffer containing 75 mM SDS at pH 11.0 for the carrier and at pH 7.7 for the conjugates in MEKC.

Buccal drug absorption. II: In vitro diffusion across the hamster cheek pouch.

The in vitro diffusion of a series of substituted acetanilides across the hamster cheek pouch was studied. The keratinized epithelial layer of the cheek pouch appeared to provide the major barrier to diffusion of these compounds. Linear relationships were found for plots of log epithelial permeability (Pe) versus the log of the octanol-buffer partition coefficient (PCoct; r = 0.712), and log Pe versus the log of the isooctane-buffer partition coefficient (PCiso; r = 0.869). Comparison of the in vitro data with in vivo data obtained in humans using the buccal absorption test showed good correlation (r = 0.878) between Pe and percent buccal absorption in 5 min. These results suggest that diffusion across the hamster cheek pouch may have some utility in the prediction of human buccal absorption.

1H-NMR assay of papaverine hydrochloride and formulations.

A precise and specific 1H-NMR method has been developed for the assay of papaverine hydrochloride (1) as a bulk drug, as well as in tablet and injection dosage forms. The assay depends upon the integration of the 12 protons of the four methoxy groups of 1 relative to that of the three methyl protons of acetanilide (internal standard). In addition to the accurate quantitative determination of 1, the method provides a specific means of identifying 1 as well as detecting other alkaloids or impurities.

Biomonitoring of aniline and nitrobenzene. Hemoglobin binding in rats and analysis of adducts.

Covalent binding to hemoglobin was studied to further substantiate the proposal that it may be used for biomonitoring N-substituted aryl compounds. (14C)-Labeled acetanilide and nitrobenzene were orally administered to female Wistar rats and binding indices [Binding(mmol/mol Hb)/Dose(mmol/kg)] determined; these were 12 +/- 1 and 73 +/- 10, respectively. After mild acidic or alkaline hydrolysis, 90% of the bound material was released and identified as aniline by radio thin layer chromatography. This supports the hypothesis that nitroso aryl derivatives, common intermediates in the metabolism of N-substituted aryl compounds, react with SH-groups of hemoglobin to yield sulfinic acid amides. Aniline was furthermore identified and quantified by capillary gas chromatography, using hemoglobin from animals treated with unlabeled aniline and nitrobenzene. Binding indices in this case were 30 +/- 3 and 85 +/- 19, respectively. With this method human blood samples may also be analysed. Although nitrobenzene is known to produce less methemoglobin than aniline, hemoglobin binding is higher. This indicates that hemoglobin binding may be a better index of body burden than methemoglobin levels in biomonitoring N-substituted aryl compounds.

The fate of phenylhydroxylamine in human red cells.

Phenylhydroxylamine added to human red cells under aerobic conditions and in the presence of glucose was partly reduced to aniline. About half the hydroxylamine was recovered as amine after a 2-hr incubation. The aniline, after acetylation, was identified as acetanilide by melting point, Rf-value in TCL as well as UV, IR, and NMR spectroscopy. The fate of the remaining phenylhydroxylamine was followed by use of 14C-labeled phenylhydroxylamine. About 30% of the total radioactivity was bound to hemoglobin or other proteins and about 20% was found in highly polar low-molecular substances which were insoluble in organic solvents. The elucidation of the sites at which phenylhydroxylamine was bound to hemoglobin was complicated by the lability of the bonds. When purified human hemoglobin had reacted with radioactive phenylhydroxylamine, large proportions of the radioactivity bound to hemoglobin were removed by treatment with acid or with PMB for separation of alpha- and beta-chains. The radioactive compound liberated from hemoglobin by acid was found to be aniline. After reaction with phenylhydroxylamine the number of SH groups titrable with PMB was found to be diminished. Pretreatment of hemoglobin with N-ethylmaleimide or PMB decreased the amount of phenylhydroxylamine bound to hemoglobin but did not fully prevent the reaction. Tryptic digestion of hemoglobin after reaction with radioactive phenylhydroxylamine yielded tryptic peptides with lower specific activity than that of hemoglobin. Chymotryptic digestion of the tryptic core yielded a core with specific activity much higher than that of hemoglobin. Fingerprinting of the tryptic or chymotryptic hydrolyzates showed the presence of peptides with high and other ones with low or no radioactivity and of radioactive compounds which did not react with ninhydrin. In the covalent binding of phenylhydroxylamine to globin the SH group beta93 plays an important role, but other yet unknown sites are also reactive.

Microsomal mediated metabolism of dialkylaryltriazenes. II. Isolation and identification of metabolites of 3,3-dimethyl-1-phenyltriazene.

After incubating 3,3-dimethyl-1-phenyltriazene with rat liver microsomes, acetanilid and derivatives of aniline and possibly of 3-methyl-1-phenyltriazene were found as metabolites and identified by mass spectrometry. This is the first time that directly formed metabolites (other than formaldehyde) of a dialkyltriazene were identified. The isolation of a derivative of 3-methyl-1-phenyltriazene as 3-acetyl-3-methyl-1-phenyltriazene supports other evidence that the triazenes are enzymically demethylated. Indication for the formation of phenylhydrazine was also obtained. In addition, hydrolysates of the polar fractions of the incubation mixture contained aniline and 4-hydroxy-aniline as a aglycones.
