New method to assess the water vapour permeance of wound coverings.

A new method for assessing the permeability to water vapour of wound coverings is presented, using the evaporimeter developed by Nilsson. This new method combines the water vapour transmission rate (WVTR) and the vapour pressure difference across a wound covering in one absolute measure: the water vapour permeance (WVP). The WVP of a wound covering is the steady flow (g) of water vapour per unit (m2) area of surface in unit (h) time induced by unit (kPa) vapour pressure difference, g.m-2.h-1.kPa-1. Since the WVP of a wound covering is a more accurate measure for the permeability than the WVTR is, it facilitates the prediction of the water exchange of a wound covering in clinical situations.

Evaluation of a new method for the prevention of neonatal anemia.

Blood sampling for diagnostic purposes is a major cause of neonatal anemia. We propose a new method of blood sampling which preserves the infant's erythrocytes. Upon drawing of 1.5-ml blood samples, the syringe is centrifuged and erythrocytes are injected back into the circulation. Using rabbits as an experimental model, we documented the efficiency of this method in decreasing the need for foreign blood transfusion. Results of a variety of laboratory tests performed with plasma obtained with the new method were identical to those achieved with whole blood. Subsequent testing did not show increased risk of contamination or bleeding phenomena secondary to heparin load. Red blood cells obtained by centrifugation were morphologically normal. This new method may be effective in reducing the need for foreign blood transfusions in neonates.

TMA for all: a new method for the construction of tissue microarrays without recipient paraffin block using custom-built needles.

BACKGROUND: TMAs are becoming a useful tool for research and quality control methods, mostly for immunohistochemistry and in situ hybridization. METHODS: A new technique that allows building TMA blocks with more than 300 tissue cores without using a recipient paraffin block for the tissue cores and without using a commercial TMA builder instrument is described. This technique is based on the construction of TMA needles modifying conventional hypodermic needles to punch tissue cores from donor blocks, which are attached by double-side adhesive tape on a computer-generated paper grid used to align the cores on the block mould, which is filled with liquid paraffin. RESULTS: More than two hundred TMA blocks were constructed using this method, utilized in immunohistochemistry and histochemistry as positive and negative controls and also in research. CONCLUSION: This technique has the following advantages: it is easy to reproduce, affordable, quick and creates uniform blocks with more than 300 cores aligned, adherent and easy to cut, with negligible losses during cutting and immunohistochemistry and in situ hybridization procedures.

New method of treatment of immune infertility.

A history is presented of a couple suffering from long-standing infertility, which on laboratory investigation was found to be immunologic infertility, as expressed in the male partner. This case of male infertility was diagnosed by findings of sperm antibody in both the serum and the seminal plasma of the man, using both the Kibrick and the F-D sperm agglutination test methods. A new effort at treatment, by means of attempted immunosuppression, was proposed and applied, using corticosteroid medication. The special feature of this treatment involved a high dosage (96 mg. per day of methylprednisolone) for a short period (seven days). Laboratory studies revealed that the antibody levels, in both his serum and seminal plasma, did fall considerably. The wife became pregnant in the subsequent cycle. It is proposed that such diagnostic and treatment efforts be tried in other cases of male immunologic infertility.

New method for titration of virus infectivity by immunostaining.

We have developed a new method for titration of viruses utilizing automated microtiter technology. Compared to existing methods such as plaque assay or hemagglutination titration of influenza virus, the new method offers distinct advantages in terms of time and effort. In addition because multiple replicates can easily be employed accuracy can be increased.

A new method for phase identification for electron diffractionists.

An accurate analytical procedure for phase identification for electron diffractionists has been developed. The method opens new frontiers in the identification of solid-state materials, as crystalline samples in the size range 10 microns to 10 A can be accurately characterized. Research with NIST CRYSTAL DATA (a large database with chemical, physical, and crystallographic data on solid-state materials) has proved that a material can be uniquely characterized on the basis of its lattice and chemical composition. To characterize a material, it is sufficient to determine any primitive cell of the lattice and the element types present. Using a modern analytical electron microscope (AEM), the experimentalist can collect the required data on an unknown sample. The lattice information is obtained by rotation of the sample to obtain two or more planes of data. From these planes, a unit cell defining the lattice can be deduced. The chemical data are determined by energy-dispersive spectroscopy (EDS). Once the experimental data are measured, the unknown is identified against the database of knows using lattice/element-type matching techniques. The basic strategy consists of three conceptual steps. First, the unknown lattice is searched against the database to find all lattices that are the same or related; the results are kept in set 1. Second, the unknown is searched against the database to find all materials with the same or similar element types; the results are kept in set 2. Finally, the results in sets 1 and 2 are combined to obtain the answer set. Experience has proved that the procedure is highly selective and reliable.

[Neuroradiologic diagnosis in acute arterial cerebral infarct. Current status of new methods].

At least three questions has to be answered by imaging methods, when new therapies in acute ischemic stroke should be used: 1. Is there a cerebral ischemia? 2. What is the size of the irreversible damaged tissue and what is the size of the safeable tissue? Is there still a vessel occlusion? New MRI-techniques including MR-angiography, diffusion-weighted imaging and perfusion-MRI, have the potential to describe the status of the brain in detail and to answer these questions. However, the value of these techniques for therapeutical decisions (thrombolysis) is unclear and has to be evaluated in clinical studies. Therefore in clinical routine these decisions should still be based on informations from CT and perhaps CT-angiography.

A new method for the isolation of fresh hepatocytes from periportal and pericentral regions of the liver lobule.

A simple method which avoids the use of perfusion with calcium free buffer, hydrolytic enzymes and detergents has been developed to obtain fresh hepatocytes from periportal and pericentral regions of the liver lobule. Cylindrical plugs (200 x 500 microns) of periportal and pericentral areas of the rat liver lobule weighing about 1 mg were collected with a micropunch from fresh or perfused liver. Ninety percent of cells were intact as assessed from trypan blue staining. Glutamine synthetase activity was detected predominantly (ca. 85%) in plugs isolated from pericentral regions indicating that this method allows selective harvesting of pure sublobular zones of the liver lobule. Rates of oxygen uptake measured at 25 degrees C by plugs from livers perfused in the anterograde direction were 56 +/- 5 and 33 +/- 7 mumol/g/h by periportal and pericentral plugs, respectively, values similar to data obtained from the intact organ. This method provides new opportunities to study the regulation of basic metabolic processes in cells from sublobular areas under nearly physiological conditions.

Mesh fixation in laparoscopic repair of ventral hernia: a new method.

During laparoscopic repair of ventral hernia, optimal fixation of the prosthetic mesh to the abdominal wall includes transfascial fixation with sutures in addition to fixation with a stapling, clipping, or tacking device. With the current methods, intracorporeal passage grasping and retrieval of sutures from the abdominal cavity are technically difficult. The reason for this difficulty is the lack of three-dimensional visual feedback during conventional laparoscopy. An easier method is needed. A new method using T-shaped anchors (T-anchors) is described. A T-anchor is a horizontal bar made of rigid titanium that is attached to a vertical limb made of monofilament suture. T-anchors are deployed in pairs, through a needle, and are tied over a musculofascial bridge to achieve transfascial fixation of the mesh to the abdominal wall. This method eliminates the need for intracorporeal grasping and retrieval of the sutures.

A new method of arthroscopic reconstruction of the dislocated acromio-clavicular joint.

BACKGROUND: Symptomatic total acromio-clavicular joint dislocation (Rockwood et al. types III-VI) may be treated by surgical reconstruction. AIM: To describe an arthroscopically assisted technique to reconstruct anatomically the coraco-clavicular ligaments in acute or chronic (> 6 weeks) acromio-clavicular joint dislocation. METHODS: This new technique involves arthroscopic exposure of the coracoid process. Prior to introducing this technique, cadaveric studies were undertaken. RESULTS: Five patients underwent this procedure. All engaged in regular sports or manual-type work. All patients were discharged the same day with the shoulder immobilised for 4 weeks, with no heavy lifting for 3 months. All patients were pain-free at 6 weeks with full function and maximum Constant scores at 3 months. There have been no complications. CONCLUSIONS: A new, safe technique is described which provides a cosmetically acceptable, anatomically solid reconstruction of the coraco-clavicular ligaments.

[A new method of isolation and a new form of transketolase from baker's yeast].

A new procedure for the isolation of homogeneous transketolase from baker's yeast based on the use of enzyme-specific antibodies immobilized on a insoluble matrix has been developed. The enzyme yield is 90% of its total content in the original yeast extract. The eluate from the immunocolumn was found to contain a previously unknown form of transketolase which represents an enzyme-RNA complex.

[New method of canal preparation and new instruments: The Canal Master].

The Canal Master is a new instrument for root canal preparation. Its working portion measures 2 mm and this instrument presents a non aggressive tip. This technique uses mechanical instruments for the straight portion of the canal and manual instruments for the curved portion. Clinically this technique gives good results but is more longer than others.
