Mass discrimination during MC-ICPMS isotopic ratio measurements: investigation by means of synthetic isotopic mixtures (IRMM-007 series) and application to the calibration of natural-like zinc materials (including IRMM-3702 and IRMM-651).

A long known way of anchoring isotope ratio values to the SI system is by means of gravimetrically prepared isotopic mixtures. Thermal ionization mass spectrometry (TIMS) is the traditionally associated measurement technique, but multi-collector double focusing inductively coupled plasma (MC-ICP)-MS now appears to be an attractive alternative. This absolute calibration strategy necessitates that mass discrimination effects remain invariant in time and across the range of isotope ratios measured. It is not the case with MC-ICPMS and the present work illustrates, in the case of Zn isotopic measurements carried out using locally produced synthetic Zn isotope mixtures (IRMM-007 series), how this calibration strategy must be adjusted. First, variation in mass discrimination effects across the measurement sequence is propagated as an uncertainty component. Second, linear proportionality during each individual measurement between normalized mass discrimination and the average mass of the isotope ratios is used to evaluate mass discrimination for the ratios involving low abundance isotopes. Third, linear proportionality between mass discrimination and the logarithm of the isotope ratio values for n(67Zn)/n(64Zn) and n(68Zn)/n(64Zn) in the mixtures is used iteratively to evaluate mass discrimination for the same ratios in the isotopically enriched materials. Fourth, ratios in natural-like materials (including IRMM-3702 and IRMM-651) are calibrated by external bracketing using the isotopic mixtures. The relative expanded uncertainty (k = 2) estimated for n(68Zn)/n(64Zn) and n(67Zn)/n(64Zn) ratio values in the synthetic isotopic mixtures and the natural-like zinc samples was in the range of 0.034 to 0.048%. The uncertainty on the weighing (0.01%, k = 1) was the largest contributor to these budgets. The agreement between these results and those obtained with a single detector TIMS and with another MC-ICPMS further validated this work. The absolute isotope ratio values found for IRMM-3702-material also proposed as "delta 0" for delta-scale isotopic measurements-are n(66Zn)/n(64Zn) = 0.56397 (30), n(67Zn)/n(64Zn) = 0.082166 (35), n(68Zn)/n(64Zn) = 0.37519 (16), and n(70Zn)/n(64Zn) = 0.012418 (23). The derived Zn atomic weight value Ar(Zn) = 65.37777 (22) differs significantly from the current IUPAC value by Chang et al. [1]. Remeasurement, with isotopic mixtures from the IRMM-007 series, of the Zn isotope ratios in the same Chang et al. [1] material have revealed large systematic differences (1.35 (27)% per atomic mass unit) that suggest unrecognized measurement biases in their results.

Recycling of an electric arc furnace flue dust to obtain high grade ZnO.

The production of steel in electric arc furnace (EAF) generates a by-product called EAF dusts. These steelmaking flue dusts are classified in most industrialized countries as hazardous residues because the heavy metals contained in them, tend to leach under slightly acidic rainfall conditions. However, and at the same time they contain zinc species which can be used as a source to obtain valuable by-products. The present investigation shows results on the processing of an EAF flue dust using ammonium carbonate solutions. Once zinc is dissolved: ZnO + 4NH3 + H2O --> Zn(NH3)4(2+) + 2OH- with other impurities (i.e. cadmium and copper), these are eliminated from the zinc solution via cementation with metallic zinc. The purified zinc solution was evaporated (distilled) until precipitation of a zinc carbonate species, which then was calcined to yield a zinc oxide of a high grade. For the unattacked dust residue from the leaching operation, mainly composed of zinc ferrite, several options can be considered: back-recycling to the furnace, further treatment by sodium hydroxide processing or a more safely dumping due to its relatively inertness.

Studies on zinc bioavailability from a representative diet in Chinese urban women of childbearing age using a double label stable isotope technique.

The objectives of the study were to determine dietary zinc bioavailability by using a double label stable isotope technique, and to identify the zinc content and levels of some possible factors which may affect zinc utilization in a typical representative Chinese urban diet. Twenty urban women of childbearing age were fed a diet extrinsically labeled with 67Zn and received intravenous (i.v.) injections of 70Zn. Fecal monitoring, urine monitoring and chemical balance technique were used simultaneously for comparisons. Isotopic enrichment in feces and urine after oral and i.v. administration of stable zinc isotopes was measured to determine the ways of zinc absorption. The mean (+/-SD) zinc intake from the representative Chinese diet was 10.22+/-0.80 mg/day, which is lower than the reference nutrient intake (RNI) level (11.5 mg/day), and mainly came from the subsidiary foods (63%). The mean dietary Fe/Zn weight was 1.1+/-0.1. The phytate/zinc molar ratio in the diet was about 3:1. The phytic acidxCa/zinc molar ratio in the diet was about 45:1. The apparent and true absorption rates of 67Zn were 36.2+/-9.7% and 38.2+/-10.1%, respectively, which were higher than the apparent absorption determined by balance technique (29.7+/-10.6%). The mean fractional absorption (FA) determined in urine was 35.7+/-13.3. In conclusion, the subsidiary food is the main source of dietary zinc in this typical diet. The extrinsic labeling technique by use of stable isotopes was more reliable and accurate to determine zinc absorption. Moreover, the double isotopic tracer ratio (DITR) technique applied to urine samples was a more practical and valuable way to evaluate zinc absorption than other methods. It can be speculated that iron and phytic acid might not be the main inhibitors in the representative Chinese diet.

NMR spectra of some reduced symmetry peripheral fused-ring-substituted phthalocyanines: density functional calculations.

Chemical shifts of some reduced symmetry peripheral fused-ring-substituted phthalocyanines, namely Zn3B1N, Zncis2B2N, Zntrans2B2N, Zn1B3N and Zn3B0N, have been calculated at density functional B3LYP level using the gauge-independent atomic orbital (GIAO) method. The geometries were optimized using the 6-31G(d) basis set and the following NMR calculations were performed using 6-31G(d) and 6-311G(d,p) basis sets, respectively. The calculated NMR shielding tensors and chemical shifts are compared with previous experimental results. The chemical shifts are assigned according to the calculated data and satisfying results are obtained. The NMR shielding tensor simulation of Zn3B0N has been raised as a significant theoretical topic.

Release of zinc from the brain of El (epilepsy) mice during seizure induction.

Brain distribution after i.v. injection of 65ZnCl2 into El mice, an animal model of genetically determined epilepsy, was studied by autoradiography to study the utilization of zinc in the brain. The distribution of 65Zn in the brain of El mice 6 days after injection was almost the same as that of ddY (normal) mice, suggesting that the uptake of zinc by the brain of El mice is normal. To study the movement of zinc in the brain in the course of seizure induction, the concentrations of 65Zn in the brain of seizure-afflicted and untreated control El mice were compared 20 days after 65Zn injection. The concentration of 65Zn in the brain of seized El mice was overall lower than that of control El mice; the concentration of 65Zn was decreased notably in the piriform cortex and amygdaloid nuclei complex during convulsion. These results suggest that the release of zinc from the El mouse brain is enhanced during convulsion. The decrease in actively functioning zinc in the brain may be associated with the increase in susceptibility to seizure in the El mouse.

The use of long-acting oxytetracycline for the treatment of ovine footrot.

The use of long-acting oxytetracycline for the treatment of ovine footrot was investigated under different experimental conditions. In sheep with artificially induced footrot housed under dry conditions, treatment with long-acting oxytetracycline produced a cure in 6/6 affected feet (a cure rate of 100%); foot-bathing in zinc sulphate produced a cure in 2/8 affected feet (a cure rate of 25%). This difference was statistically significant (P < 0.05). In sheep with naturally occurring footrot, housed under dry conditions, treatment with long-acting oxytetracycline produced a cure in 8/9 affected feet (a cure rate of 89%); foot-bathing in zinc sulphate produced a cure in 7/10 affected feet (a cure rate of 70%). This difference was not statistically significant. In sheep with naturally occurring footroot, kept under field conditions, treatment with a combination of long-acting oxytetracycline and foot-bathing in zinc sulphate produced a cure in 49/52 affected feet (a cure rate of 94%); treatment by foot-bathing alone produced a cure in 39/51 affected feet (a cure rate of 77%). This difference was statistically significant (P < 0.05).

Successful medical treatment of severely decompensated Wilson disease.

Delayed response to medical treatment sometimes leads to unnecessary liver transplantation in patients with severely decompensated Wilson disease. We report the course of five patients (mean age 13.4 years, range 11 to 15 years) with severely decompensated Wilson disease who were successfully treated medically. Prothrombin time improved after a minimum of 1 month and returned to normal within 3 months to 1 year or more.

Reversibility of cerebral ventricular enlargement in anorexia nervosa, demonstrated by quantitative magnetic resonance imaging.

OBJECTIVE: To determine the reversibility of the loss of brain parenchyma and ventricular enlargement in patients with anorexia nervosa after refeeding. STUDY DESIGN: Quantitative magnetic resonance imaging was performed on three groups of subjects: (1) 12 female adolescents hospitalized with anorexia nervosa, (2) the same 12 patients after nutritional rehabilitation, a mean of 11.1 months later, and (3) 12 healthy age-matched control subjects. Sixty-four contiguous coronal magnetic resonance images, 3.1 mm thick, were obtained. With a computerized morphometry system, lateral and third ventricular volumes were measured by a single observer unaware of the status of the patient. RESULTS: On admission, patients were malnourished and had lost an average of 11.7 kg (body mass index, 14.3 +/- 2.0 kg/m2). After refeeding, they gained an average of 9.7 kg (body mass index, 17.9 +/- 1.5 kg/m2). Total ventricular volume decreased from 17.1 +/- 5.5 cm3 on admission to 12.4 +/- 3.0 cm3 after refeeding (p < 0.01) and returned to the normal range. The degree of enlargement of the third ventricle was greater than that of the lateral ventricles. There was a significant inverse relationship between body mass index and total ventricular volume (r = -0.63; p < 0.05). CONCLUSION: In patients with anorexia nervosa, cerebral ventricular enlargement correlates with the degree of malnutrition and is reversible with weight gain during long-term follow-up.

Oxalate production by wood-rotting fungi growing in toxic metal-amended medium.

In this report, we have identified oxalic acid as an important metabolite elaborated in the response of wood-rotting fungi to toxic metal stress. The formation of oxalate crystals by white rot fungi (Bjerkandera fumosa, Phlebia radiata and Trametes versicolor) and the brown rot fungus Fomitopsis pinicola, grown on media containing high levels of toxic metal ions has been visualized using scanning electron microscopy (SEM) with energy-dispersive X-ray micro-analysis (EDXA) and HPLC. There were no significant differences between the growth of controls (metal-free) and on the 0.5% CaCO(3), Co(3)(PO(4))(2) or Zn(3)(PO(4))(2)-amended plates. ZnO inhibited the growth of all strains. Crystals were not detected in Zn(3)(PO(4))(2)-amended plates. The four examined strains displayed the formation of crystals on ZnO, Co(3)(PO(4))(2) and CaCO(3)-amended plates.

Kinetics of fluoride release from zinc oxide-based cements.

Considerable attention has been given to the release of the cariostatic fluoride ion from glass-based dental cements (dental silicate and glass ionomer). In these, the total available fluoride content is not precisely known since fluorine is distributed between the cross-linked aqueous salt matrix, partially dissolved glass, and undissolved glass. In analogous cements based on zinc oxide the fluoride is added as highly soluble SnF2. The object of this study is to compare the F- ion release profiles of commercial zinc polycarboxylate and zinc phosphate containing 4.4 and 3.6% SnF2, respectively. Mixed cements were clamped in split ring moulds to produce discs of 10 mm x 1 mm after storage at 37 degrees C for 1 h. Each was weighed and immersed in 10 ml of deionised water. When this changed, at 13 time intervals up to 98 days, the fluoride content was measured using an ion selective electrode. The mean (N = 3) values obtained were expressed cumulatively [F] in micromol F ion/g cement. The total [F] released was 111 for the zinc polycarboxylate and 286 for zinc phosphate compared with total F in the cements of 561 and 464, respectively. When the cumulative [F] was plotted versus t(1/2) close associations were found for both cements. For the polycarboxylate the regression line [F] = 10.6t(1/2) + 9.9 fitted well over the whole 98 days (R = 0.997). For the phosphate a better fit regression line was obtained using results up to 32 days only; [F] = 36.8t(1/2) - 8.4 (R = 0.999). For t > 32 days results increasingly deviated from this line. These results fitted a regression line of the form [F] = 81.7log(e) t - 87.3 (R = 0.9997). Comparisons are made with data from previous authors both for zinc phosphate cement and glass-based cements and with diffusion theory of F ion release. It is concluded that zinc-based cements provide some indications of how glass-based cements may behave over long periods of release and that zinc phosphate is the material of clinical choice for orthodontic cementation if maximal fluoride release is the prime criterion.

Bidirectional transport of cadmium across apical membrane of renal epithelial cell lines via H+-antiporter and inorganic anion exchanger.

The purpose of this study was to investigate whether cadmium (Cd) efflux across the apical membrane of renal epithelial cells is mediated via a H+-antiport system, and to confirm the re-uptake of Cd from the apical membrane via an inorganic anion exchanger. LLC-PK1 and OK cell monolayers cultured on permeable membranes were incubated with 1 microM CdCl2 added to the basolateral medium, and the transport of Cd from the basolateral to apical medium and the accumulation of Cd in the monolayers were measured. Cd transport was increased by lowering the pH of the apical medium and was accompanied by a decrease in Cd accumulation. Coincubation with N'-methylnicotinamide or cisplatin, which act as substrates of the H+-antiport systems, decreased Cd transport and increased Cd accumulation in a concentration-dependent manner. To confirm the re-uptake of Cd, LLC-PK1 cell monolayers were pretreated with 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), a specific inhibitor of an inorganic anion exchanger, before incubation with CdCl2. Pretreatment with DIDS significantly increased Cd transport and decreased Cd accumulation at an apical medium pH of 7.4, but not at pH 5.5. These results suggest bidirectional transport of Cd across the apical membrane of renal epithelial cells via a H+-antiport system (efflux) and an inorganic anion exchanger (influx), depending on the pH of the apical side.

Quantitative analysis of crystalline pharmaceuticals in powders and tablets by a pattern-fitting procedure using X-ray powder diffraction data.

A pattern-fitting procedure for quantitative analysis of crystalline pharmaceuticals in solid dosage forms using X-ray powder diffraction data is described. This method is based on a procedure for pattern-fitting in crystal structure refinement, and observed X-ray scattering intensities were fitted to analytical expressions including some fitting parameters, i.e. scale factor, peak positions, peak widths and degree of preferred orientation of the crystallites. All fitting parameters were optimized by the non-linear least-squares procedure. Then the weight fraction of each component was determined from the optimized scale factors. In the present study, well-crystallized binary systems, zinc oxide-zinc sulfide (ZnO-ZnS) and salicylic acid-benzoic acid (SA-BA), were used as the samples. In analysis of the ZnO-ZnS system, the weight fraction of ZnO or ZnS could be determined quantitatively in the range of 5-95% in the case of both powders and tablets. In analysis of the SA-BA systems, the weight fraction of SA or BA could be determined quantitatively in the range of 20-80% in the case of both powders and tablets. Quantitative analysis applying this pattern-fitting procedure showed better reproducibility than other X-ray methods based on the linear or integral intensities of particular diffraction peaks. Analysis using this pattern-fitting procedure also has the advantage that the preferred orientation of the crystallites in solid dosage forms can be also determined in the course of quantitative analysis.

Formulation of sustained release aqueous Zn-hirudin suspensions.

Sustained release formulations for recombinant hirudin (rHir), an anticoagulant thrombin-specific inhibitor, were developed. Zn-rHir suspensions were formed by precipitation with zinc salts at neutral pH. Characterization of protein precipitation was by UV analysis, capillary electrophoresis (CE), zinc analysis, light and electron microscopy, and particle size analysis. The precipitation of aqueous rHir solution with ZnCl(2) solution at neutral pH resulted in Zn-rHir suspensions. Optimum yields of pelletized Zn-rHir were obtained between pH 7.0 and 7.4. For complete precipitation ( approximately 100%) a molar ratio of zinc to rHir of >28 was necessary. As shown by electron microscopy, the smallest resolvable unit of Zn-rHir suspensions was 20 nm. Agglomerates of up to 200 microm were observed by light microscopy. Zinc salt-induced precipitation phenomena were also investigated using ZnBr(2), ZnI(2), Zn(NO(3))(2) and ZnSO(4) instead of ZnCl(2). ZnSO(4) showed the lowest precipitation efficiency. All other salts behaved similar to ZnCl(2). Upon storage the pelletized protein content of the ZnCl(2) based precipitates was stable ( approximately 95% rHir after 1 year at room temperature), whereas the pelletized protein content of ZnSO(4) based precipitates dropped sharply after precipitation (2% remaining after 13 days at room temperature). This indicates a transition of the ZnSO(4) based precipitates to hexagonal basic zinc sulfate plates and free rHir. The driving force is the lower aqueous solubility of basic zinc sulfate as compared to the higher solubility of basic zinc chloride.

Vital staining with iodine solution in delineating the border of oral dysplastic lesions.

OBJECTIVE: The borders of dysplastic lesions of the oral mucosa are often vaguely delineated. The purpose of this study was to determine if vital staining with an iodine solution can precisely delineate the boundary of superficial spread of dysplastic or malignant epithelium. STUDY DESIGN: Thirty-seven biopsies were obtained from 18 patients with possible malignant lesions of the oral mucosa. Deviations between the clinical boundary obtained with vital staining and the histopathologic boundary determined in hemotoxylin-eosin-and lectin-stained sections were measured with light microscopy. RESULT: The clinical boundaries defined with the vital staining agreed well with the pathologic border lines and were in error by less than 3 mm. CONCLUSION: This result suggests that vital staining with an iodine solution has great potential in determining the extent and precise borders of the dysplastic epithelium. A 5-mm border of normal tissue peripheral to the iodine-positive area would be adequate for the complete removal of the dysplastic epithelium.

Efficiency of the salt flotation technique in the recovery of Ascaris lumbricoides eggs from the soil.

The efficacy of the salt flotation technique using saturated solutions of sodium nitrate (specific gravity 1.30) and zinc sulphate (specific gravity 1.16) for separating Ascaris lumbricoides eggs from clay, loamy and sandy soils has been investigated. Ten samples of each of the egg concentrations of 100, 500, 2500, 5000 and 10,000 eggs/25 g of soil were used for each soil type. Using T-test and ANOVA, the number of eggs recovered from sand was significantly higher than from loam and the number from loam significantly higher than from clay. With sodium nitrate, the maximum egg recovery rate was 25.04% from sandy soil at a concentration of 500 eggs/25 g soil while with zinc sulphate it was 13.88% also from sandy soil and concentration of 500 eggs/25 g soil. While the number of eggs recovered increased with soil egg concentration, the percentage of eggs recovered is inversely proportional to egg concentration. The number of eggs recovered with sodium nitrate was significantly higher than with zinc sulphate solution in the three soil types.

Catalytic asymmetric organozinc additions to carbonyl compounds.

Because of the tremendous effort of a great number of researchers, the catalytic asymmetric dialkylzinc addition to aldehydes has become a mature method. Ligands of diverse structures have been obtained, and high enantioselectivity for all different types of aldehydes have been achieved. Among the representative excellent catalysts are compounds 1, 8, 120, 325, 352, and 360 discussed above. However, compared to the well-developed dialkylzinc addition, the catalytic asymmetric reactions of aryl-, vinyl-, and alkynylzinc reagents with aldehydes are still very much under developed. Although catalysts such as (S)-402 and 210 prepared by Pu and Bolm have shown good enantioselectivity for the reaction of diphenylzinc with certain aromatic and aliphatic aldehydes, the generality of these catalysts for other [formula: see text] arylzinc reagents have not been studied. The vinylzinc additions using ligands 1 and 412 reported by Oppolzer and Wipf were highly enantioselective for certain aromatic aldehydes but not as good for aliphatic aldehydes. Carreira discovered highly enantioselective alkynylzinc additions to aldehydes promoted by the chiral amino alcohol 415, but this process was not catalytic yet. Ishizaki achieved good enantioselectivity for the catalytic alkynylzinc addition to certain aldehydes by using compounds 160, but the enantioselectivity for simple linear aliphatic aldehydes was low. Another much less explored area is the organozinc addition to ketones. Yus and Fu showed very promising results by using ligands 381 and 406 for both dialkylzinc and diphenylzinc additions to ketones, but the scope of these reactions were still very limited. Therefore, more work is needed for the aryl-, vinyl-, and alkynylzinc additions and for the organozinc addition to ketones, although many good catalysts have been obtained for the dialkylzinc addition to aldehydes. Development of these reactions will allow the catalytic asymmetric synthesis of a great variety of functional chiral alcohols that are either the structural units or synthons of many important organic molecules as well as molecules of biological functions. Macromolecular chiral catalysts have become a very attractive research subject in recent years because these materials offer the advantages of simplified product isolation, easy recovery of the generally quite expensive chiral catalysts, and potential use for continuous production. Three types of macromolecules including flexible achiral polymers anchored with chiral catalysts, rigid and sterically regular main chain chiral polymers, and chiral dendrimers have been used for the asymmetric organozinc addition to aldehydes. Among these materials, the binaphthyl-based polymers such as (R)-451 developed by Pu have shown very high and general enantioselectivity. Study of the binaphthyl polymers in the asymmetric organozinc addition has demonstrated that it is possible to systematically modify the structure and function of the rigid and sterically regular polymer for the development of highly enantioselective polymer catalysts. The catalytic properties of highly enantioselective monomer catalysts can also be preserved in the rigid and sterically regular polymer provided the catalytically active species of the monomer catalyst is not its aggregate. The TADDOL-based polymers and dendrimers prepared by Seebach showed very high and stable enantioselectivity for the diethylzinc addition to benzaldehyde even after many cycles. These studies on macromolecular chiral catalysts demonstrate that these materials are potentially very useful for practical applications.

Zn speciation in the organic horizon of a contaminated soil by micro-X-ray fluorescence, micro- and powder-EXAFS spectroscopy, and isotopic dilution.

Soils that have been acutely contaminated by heavy metals show distinct characteristics, such as colonization by metal-tolerant plant species and topsoil enrichment in weakly degraded plant debris, because biodegradation processes are strongly inhibited by contamination. Such an organic topsoil, located downwind of an active zinc smelter and extremely rich in Zn (approximately 2%, dry weight), was investigated by X-ray diffraction, synchrotron-based X-ray microfluorescence, and powder- and micro-extended X-ray absorption fine structure (EXAFS) spectroscopy for Zn speciation and by isotopic dilution for Zn lability. EXAFS spectra recorded on size fractions and on selected spots of thin sections were analyzed by principal component analysis and linear combination fits. Although Zn primary minerals (franklinite, sphalerite, and willemite) are still present (approximately 15% of total Zn) in the bulk soil, Zn was found to be predominantly speciated as Zn-organic matter complexes (approximately 45%), outer-sphere complexes (approximately 20%), Zn-sorbed phosphate (approximately 10%), and Zn-sorbed iron oxyhydroxides (approximately 10%). The bioaccumulated Zn fraction is likely complexed to soil organic matter after the plants' death. The proportion of labile Zn ranges from 54 to 92%, depending on the soil fraction, in agreement with the high proportion of organically bound Zn. Despite its marked lability, Zn seems to be retained in the topsoil thanks to the huge content of organic matter, which confers to this horizon a high sorption capacity. The speciation of Zn in this organic soil horizon is compared with that found in other types of soils.
