Cystic Fibrosis in Ukraine: age, origin and tracing of the delta F508 mutation.

Seven already known CF mutations were searched in 170 unrelated cystic fibrosis patients from different regions of Ukraine. Their frequencies in this sample were: delta F508-50%, 1677delTA-0.3% (10th exon), R553X-0.6%, G551D-0.3% (11th exon), R334W-0.6% (7th exon). 1154insTC (7th exon) and S549I (11th exon) were not found. Heterozygotes for delta F508 were searched in 865 healthy volunteers from different Ukranian regions. Their frequencies ranged from 1:28 to 1:70. We report here unpublished population data from Ukraine in order to discuss the origin, evolution and dispersion of chromosomes bearing the delta F508 mutation. Selection in terms of heterozygote advantage is also discussed.

[The distribution of haptoglobin phenotypes among the population of Ukraine].

The results of investigation of haptoglobin (Hp) types in 596 donor blood samples in some towns of Ukraine (Dnepropetrovsk, Kharkov, Odessa, Kiev, Uzhgorod, Zhitomir) are presented. Three normal Hp types (Hp1-1, Hp2-1 and Hp2-2) have been found. The reliable interpopulation differences in the Hp types frequency were not found. On the whole the Hp types frequency in the type Hp1-1 comprised 12.7%. In the type Hp2-1-48.1% and in the type Hp2-2-36.5%. The frequency of the gene Hp1 is 0.38. The frequency of the Hp types and of the gene Hpl in Ukraine is similar to that in population of Eastern Europe and European Part of Russia.

[Serotypical stratification of HIV-1 in populations of intravenous drug users in the south/southwestern Ukraine].

A total of 101 HIV-1 positive sera from intravenous drug users (IVDU) in Southern and South-Eastern Ukraine were serotyped using V3-mimicking peptides. Serotype B circulates in Nikolaev and type A/C in Odessa and Donetsk. Prevalence of A/C serotype in the population of IVDU is a characteristic feature of the epidemic situation in this region, in contrast to the USA and Western Europe, where subtype B circulates. Serotypical stratification indicates that routes of HIV-1 dissemination in the Ukraine differ from those in the USA and Western Europe, which should be borne in mind when planning the strategy of epidemic control.

Ukrain treatment in a patient with stage IV neuroblastoma. A case report.

A 22-month-old boy with stage IV neuroblastoma underwent laparotomy with tumor removal and nephrectomy, followed by treatment with Ukrain. Two months later, a remaining abdominal tumor, a retroperitoneal tumor (approximately 2 x 1 cm), and lung, brain, pelvis, kidney, and distal femur metastases were found. Growth of the neuroblastoma around the spinal cord and growth into the spinal canal was also found. Ukrain was administered in 3-week therapy series with 3-week pauses between each series. Some tumors disappeared, others were smaller, and growth stopped in the remaining tumors. Various metastases were no longer detectable.

Effect of intermittent three-month treatment with different doses of Ukrain on subregional femoral bone mineral density of sexually mature female rats.

Sexually mature but still growing female Wistar rats received i.p. injections of Ukrain (7, 14 or 28 mg/kg in a volume of 0.5 ml/100g) every other day for 10 days, followed by a 10-day break, and this procedure was performed five times. The control animals received the same volume of injected water. At the end of the experiment the rat right femora were harvested and the bone densitometric parameters of the entire bone, distal metaphyseal and basicervical subregions were assessed using the dual-energy X-ray absorptiometry (DXA) densitometric method. No significant changes were observed in the bone mineral density in experimental groups in comparison with control animals that received the vehicle. A slight decrease in the bone mineral content value was observed in the distal metaphyseal region in animals that were treated with the highest dose of Ukrain.

A study of the influence of a novel drug Ukrain on in vivo effects of low-dose ionizing radiation.

The extrapolation of data obtained with lethal doses of radiation to evaluating low doses, i.e., those not causing immediate death, seems to be unjustified. Thus, models have to be developed that are based on integral parameters (such as the survival of experimental animals), easy to perform, and permit screening procedures to be carried out within 30 days in order to make screening efficient. Two hundred and sixty outbred male white mice were irradiated with a 1 Gy dose at a 0.75 Gy/min. To produce a model of acute infection, the animals were infected by tularemia (2 x LD50) using Gaiskiy-15 strain of the infectious agent. A viral disease was produced by infecting mice by the equine encephalomyelitis (EEM) virus. The animals were infected 24 h after irradiation. The survival of animals was used as an end point. Such conditions are most prevalent in areas contaminated by radioactivity. Irradiation prior to infection was found to decrease the survival rate of experimental animals. Preventive administration of Ukrain increased the survival rate of experimental animals. On the whole, the results suggest that Ukrain deserves the attention of experts in radioprotection.

Induced G2/M arrest and apoptosis in human epidermoid carcinoma cell lines by semisynthetic drug Ukrain.

Exposure of ME180 and A431 carcinoma cells to Ukrain (NSC-631570), a novel semisynthetic drug from Chelidonium majus L, results in cell growth inhibition which is concomitant with reversible G2/M cell cycle arrest and apoptosis at doses as low as 7 microM. In contrast, the same drug concentrations were not affective towards normal human keratinocytes. In order to investigate whether cell cycle control mechanisms are effected in response to Ukrain, we analyzed cell cycle distribution and levels of cyclins and cyclin-dependent kinases in drug treated carcinoma cells. We found alterations in levels of mitotic cyclins A and B1, and cyclin-dependent kinases CDK1 and CDK2, after treatment. We also observed an upregulation of CDK inhibitor p27 in both cancer cell lines which may lead to the G2/M cells accumulation.

Chitotriosidase as a new marker of macrophage stimulation in a tumor model treated with cyclophosphamide and Ukrain.

Ukrain has previously been demonstrated to exert a malignotoxic effect in vivo. This antitumor drug has been effective in the treatment of some malignancies in experimental animals as a result of immunostimulation (macrophage stimulation). In the present study, serum chitotriosidase activity was measured as a biochemical marker of macrophage stimulation in several murine and rat models of macrophage stimulation. It was shown that zymosan, carboxymethylated glucan and Triton WR 1339 administration to CBA mice or Wistar rats was followed by a considerable increase in serum chitotriosidase activity. Murine LS lymphosarcoma development decreased serum chitotriosidase activity. Antitumor treatment by Ukrain or cyclophosphamide did not restore this index to the normal value.

Cystatin C in LS lymphosarcoma and HA-1 hepatoma treated with Ukrain and cyclophosphamide and involvement of apoptosis.

The concentration of cystatin C, a cysteine proteinase inhibitor, was measured during the treatment of murine LS lymphosarcoma with cyclophosphamide and HA-1 murine hepatoma with the antitumor drug Ukrain. It was shown that concentrations of cystatin C were very low in both the tumor tissues studied (HA-1 hepatoma cells and LS lymphosarcoma); increased cystatin C concentrations were found only in Ukrain-treated murine hepatoma, suggesting the mechanism of antitumor effect of this drug. Cyclophosphamide treatment in LS lymphosarcoma did not influence the concentration of cystatin C in tumor cells. At the same time, a marked increase in cathepsin B and cathepsin L activity in LS lymphosarcoma was found, indicating the involvement of apoptosis in the mechanism of antitumor action of cyclophosphamide. While the DNA from untreated LS lymphosarcoma was very homogenous and its molecular weight was high, the DNA from tumors of treated mice broke down, giving rise to the ladder figure characteristically produced by cells dying from apoptosis. Evidence was obtained that cyclophosphamide-induced tumor regression was effected by apoptosis.

Selective inhibition of in vitro cell growth by the anti-tumour drug Ukrain.

The inhibitory effect of Ukrain on malignant cells and on normal cells, in vitro, has been compared. To obtain a 50% inhibition of cell growth, a tenfold concentration had to be used with normal endothelial cells compared to a human osteosarcoma cell line. Hybrids of the two cell types showed nearly the same sensitivity as normal cells. A laser scanning microscope showed a high uptake of Ukrain in malignant cells, while the content in normal cells under the same experimental conditions was substantially lower.

In vitro effects of Chelidonium majus L. alkaloid thiophosphoric acid conjugates (Ukrain) on the phenotype of normal human lymphocytes.

Several plant-derived drugs are used in medical oncology (1-3). The thiophosphoric acid-alkaloid derivative from the plant Chelidonium majus L. (Ukrain) has been produced since 1978 by Nowicky. (Austrian Patent No. 354644, Vienna, 25 January 1980). It has been reported that this drug may exert therapeutic effects in cancer patients by modulating their immune systems (4, 5-8). The aim of this study was to evaluate in vitro the possibility that Ukrain may influence the phenotype of lymphocyte subpopulations isolated from healthy donors.
