Impact of tributyltin and triphenyltin on ivory shell (Babylonia japonica) populations.

We histopathologically examined gonads and chemically determined organotin compounds in tissues of the ivory shell, Babylonia japonica. Imposex (a superimposition of male-type genital organs on females) occurred in approximately 80-90% of B. japonica specimens that we examined, with the penis and vas deferens both well developed. No oviduct blockage by vas deferens formation was observed. Ovarian spermatogenesis and suppressed ovarian maturation were observed in the females that exhibited imposex, although no histopathological abnormalities were found in males. Tissue distributions of organotin compounds [tributyltin (TBT), triphenyltin (TPhT), and their metabolites] were different for butyltins and phenyltins; a remarkably high accumulation of TBT was observed in the ctenidium, osphradium, and heart, whereas high concentrations of TPhT were detected in the ovary and digestive gland. More than one-third of TBT accumulated in the digestive glands of both males and females, followed by the testis, ctenidium, muscle, and heart tissues in males and in the muscle, ovary, ctenidium, and head tissues (including the central nervous system ganglia) in females. In both males and females, more than half of total TPhT accumulated in the digestive glands, followed by the gonads. The next highest values were in the muscle, ctenidium, and heart tissues in males and in the muscle, oviduct, and head tissues in females. Both TBT and TPhT concentrations in the gonads were positively correlated with penis length in females. Our findings strongly suggest that reproductive failure in adult females accompanied by imposex, possibly induced by TBT and TPhT from antifouling paints, may have caused the marked decline of B. japonica populations in Japan.

Distribution of tributyltin, dibutyltin and monobutyltin in the liver, brain and fat of rats: two-generation toxicity study of tributyltin chloride.

The distribution of tributyltin (TBT) and its metabolites, dibutyltin (DBT) and monobutyltin (MBT), was examined in the liver, brain and fat tissues in a two-generation reproductive toxicity study of tributyltin chloride (TBTCl) in rats using dietary supplementation at concentrations of 5, 25 and 125 ppm. In the liver, irrespective of TBTCl dietary concentration, gender or generation, the highest concentration of metabolite was consistently MBT, followed by DBT, and then TBT. In contrast, TBT was consistently present at the highest concentration in the brain, nearly always followed by DBT and MBT. In fat tissues, the concentrations of the three butyltin compounds showed similar relationships to those observed in the brain, although the concentrations were much lower. In the liver, the concentration of TBT was higher in females, and those of DBT and MBT were higher in males. Factorial ANOVA also suggested the effect of gender on the concentrations of the three butyltin compounds in the liver. The results of this study suggest tissue-dependent distribution of TBT, DBT and MBT and gender-dependent distribution of the three metabolites in the liver of rats.

[Peculiarities of alkyl tin effects on respiration and oxidative phosphorylation of rat liver mitochondria].

Electron transfer and oxidative phosphorylation were studied as affected by tinalkyls. It is shown that all of them inhibit effectively respiration of the rat liver mitochondria. The action of bis(tributyl tin)oxide and dibutyl diisooctyl thioglycolate tin on the mitochondria in a state of dissociation is localized in the terminal step of the respiration chain. The other alkyls act in the site located before cytocrome c. The effect of bis (tributyl tin) oxide on both the ADP-activated respiration and DNP-activated ATPase of intact mitochondria is similar to that of oligomycin. These effects might be explained by the presence two electrophilic centres in a molecule of this compound.

Simultaneous determination of mono-, di- and tributyltin in environmental samples using isotope dilution gas chromatography mass spectrometry.

The development of a rapid, precise and accurate speciation method for the simultaneous determination of mono-, di- and tributyltin in environmental samples is described. The method is based on using isotope dilution gas chromatography/mass spectrometry (GC/MS) with electron ionization, a widely used technique in routine testing laboratories. A mixed spike containing (119)Sn-enriched monobutyltin (MBT), dibutyltin (DBT) and tributyltin (TBT) was used for the isotope dilution of the samples. Five molecular ions were monitored for each analyte, corresponding to the (116)Sn, (117)Sn, (118)Sn, (119)Sn and (120)Sn isotopes. The detection at masses corresponding to (116)Sn and (117)Sn were used to correct for m + 1 and m + 2 contributions of (13)C from the organic groups attached to the tin atom on the (118)Sn, (119)Sn and (120)Sn masses with simple mathematical equations and the concentrations of the butyltin compounds were calculated based on the corrected (118)Sn/(119)Sn and (120)Sn/(119)Sn isotope ratios. The (119)Sn-enriched multispecies spike was applied with satisfactory results to the simultaneous determination of MBT, DBT and TBT in three certified reference materials: two sediments, PACS-2 and BCR 646, and the mussel tissue CRM 477. The method was compared with a previously published GC/inductively coupled plasma MS isotope dilution procedure, developed in our laboratory, by injecting the same samples into both instruments. Comparable analytical results in terms of precision and accuracy are demonstrated for both atomic and molecular mass spectrometric detectors. Thus, reliable quantitative organotin speciation analysis can be achieved using the more widespread and inexpensive GC/MS instrument.

Organotin-induced apoptosis as observed in vitro is not relevant for induction of thymus atrophy at antiproliferative doses.

The organotin compounds di-n-butyltin dichloride (DBTC) and tri-n-butyltin chloride (TBTC) selectively cause thymus atrophy. Previously, DBTC and TBTC were shown to inhibit proliferation of immature thymocytes, but other studies demonstrated that TBTC but not DBTC increased apoptosis in vitro and also in vivo. In this study, we examined whether apoptosis is increased in vitro by DBTC and TBTC at various concentrations and periods of incubation and whether apoptosis is involved in the induction of thymus atrophy at selective antiproliferative doses. In vitro, DBTC or TBTC at a concentration of 3 microM significantly increased DNA fragmentation in freshly isolated rat thymocytes after preincubation for 10 min followed by a 22-hr culture period. After continuous exposure for 22 hr, apoptosis was observed to be optimum at 1 microM TBTC and 0.3 microM DBTC and lower or absent at higher concentrations. Apparently, apoptosis induced by organotins in vitro depends on both duration and concentration of exposure. Selective antiproliferative doses of DBTC nor TBTC increased apoptosis on day 1 or 2 after single oral exposure. In contrast, the corticosteroid dexamethasone caused a depletion of both small and large thymocytes and a marked increase of apoptosis on both days 1 and 2 after dosing. Thus, although apoptosis is involved in the in vitro cytotoxic effects of both organotin compounds, it seems not involved in thymus atrophy at a dose that selectively inhibits immature thymocyte proliferation.

Investigating the potential of high-performance liquid chromatography with atmospheric pressure chemical ionization-mass spectrometry as an alternative method for the speciation analysis of organotin compounds.

Liquid chromatography with atmospheric pressure chemical ionization-mass spectrometry (LC-APCI-MS) was applied for the determination of butyl- and phenyltin compounds. Chromatography was performed on a 30 x 2 mm, 3 microm C18 column, enabling the separation of mono-, di- and trisubstituted butyl- and phenyltin compounds in less than 10 min using a water/1% trifluoroacetic acid/methanol gradient. While satisfactory retention and resolution is achieved for the di- and trisubstituted butyl- and phenyltin compounds, monobutyltin and monophenyltin cannot be resolved chromatographically. Depending on the parameter values of the interface, APCI-MS detection allows both specific detection of the molecular ion or cluster ion at low to intermediate fragmentor voltages or quasi-element specific detection of the Sn+ ion released from the organotin compounds at high fragmentor voltages. The sensitivity of MS detection is similar for butyl- and phenyltin compounds, but varies largely from mono- to trisubstituted organotin compounds with tributyl- and triphenyltin being the most sensitively detectable compounds. Detection limits are in the 20-65 pg (abs.) range in SIM mode and in the 750-2000 pg (abs.) range in the scan mode for tributyl- and triphenyltin and for dibutyl- and diphenyltin, respectively. Monobutyl- and monophenyltin can be detected with much lower sensitivity which, together with their unfavorable chromatographic behavior, accounts for the fact that they cannot be analyzed at environmentally relevant concentrations. Although LC-APCI-MS is generally less sensitive than comparable GC methods, it is applicable to the analysis of environmental samples as demonstrated by the analysis of the PACS-2 sediment certified reference material. Although the derivatization of the ionic organotin compounds, which particularly in real samples is a potential source of error, is circumvented when LC-APCI-MS is used, the extraction step is still critical and may lead to underestimation when quantitation is not done by the method of standard addition.

The effect of organotin compounds on gender specific androstenedione metabolism in the freshwater ramshorn snail Marisa cornuarietis.

In a recent study, we demonstrated that androstenedione was mainly converted to testosterone (T) and 5alpha-dihydrotestosterone (DHT) by digestive gland/gonad complex microsomal fractions isolated from male Marisa cornuarietis, whereas it was primarily metabolized to 5alpha-dihydroandrostenedione (DHA) by females. In the present work, the sexual dimorphic metabolism of androstenedione was further investigated, and attributed to a higher 17beta-hydroxysteroid dehydrogenase activity in males than in females. Thereafter, the hypothesis was tested that the metabolism of androstenedione might be affected by exposure to tributyltin (TBT) and triphenyltin (TPT), which are known to induce the development of imposex in several gastropod species. The in vitro metabolism of androstenedione, particularly the formation of DHA and DHT, was inhibited by both compounds. However, in vivo experiments showed no significant alteration in the metabolism of androstenedione in males, but a marginal (TBT) and a significant (TPT) inhibition of the formation of DHA in females exposed for 150 days to concentrations that had significantly induced the development of imposex. The ratio DHT+T/DHA, a possible indicator of metabolic androgenization, tended to increase (0.43 versus 0.35, p=0.06) in TPT exposed females. However, this ratio never reached values comparable to those found in males (11+/-1).

Differential sensitivity of three cyanobacterial and five green algal species to organotins and pyrethroids pesticides.

In this work, five organotins and pyrethroids pesticides were tested to examine their effects on the three cyanobacteria Anabaena flos-aquae, Microcystis flos-aquae, Mirocystis aeruginosa and on the five green algae Selenastrum capricornutun, Scenedesmus quadricauda, Scenedesmus obliqnus, Chlorella vulgaris, Chlorella pyrenoidosa through 96 h acute toxicity tests. The results indicated that: (1) the decreasing order of the average acute toxicity to cyanobacteria and green algae of five dissimilar organotins and pyrethroids pesticides was: fentin hydroxide > cyhexatin > azocyclotin > fenbutatin oxide > beta-cyfluthrin. (2) Wide variations occurred in response to the tested pesticides among the eight individual species of cyanobacteria and green algae. The sensitivity of various species of algae exposed to fenbutatin-oxide varied over one order of magnitude, exposed to cyhexatin/fentin-hydroxide/beta-cyfluthrin varied over two orders of magnitude and exposed to azocyclotin varied three orders of magnitude. (3) In contrast with the sensitivity of cyanobacteria and green algae, cyanobacteria were much less sensitive to beta-cyfluthrin than green algae. The pollutants may result in a shift of green algal and cyanobacterial group structure, especially in a shift from dominance by green algae to dominance by cyanobacteria, and may sustain cyanobcterial blooms during the special period. Thus, the decreasing order of the aquatic ecological risk was: beta-cyfluthrin > fentin hydroxide > cyhexatin > azocyclotin > fenbutatin oxide. There was a strong variance between toxicity and ecological risk, i.e. "low toxicity" does not automatically imply "low ecological risk". The toxicity of pyrethroids pesticides was lower than that of organotins pesticides, whereas the aquatic ecological risk of pyrethroids pesticides was higher than that of organotins pesticides.

Specific detection of organotin compounds with a recombinant luminescent bacteria.

Organotin compounds are widely used as biocides in marine and terrestrial environments. Several currently used techniques allow either the measurement of the chemicals or their effects on living organisms. Our current research focuses on the development of a complementary method based on a bacterial bioluminescence-based bioassay for the specific detection of organotin compounds. The performance of the bioassay was assessed. The Escherichia coli bacterial strain used in this study is specific for TBT and DBT (with Cl, Br or I as the halogen group) with the central tin atom important for light production. The assay is conducted after overnight culture of the bacterial strain, followed by 60 min of contact time with the organotin compound for significant light production. The detection limits were found to be 0.08 microM for TBT (26 microgl(-1)) and 0.0001 microM for DBT (0.03 microgl(-1)) with a linear range of one logarithm. The repeatability of the bioassay is 8% and the reproducibility for TBT and DBT was approximately 14%. Lyophilization of the strains did not significantly modify the detection limit as well as the range of detection. Applications of the bioassay to environmental samples are discussed.

Imposex and tributyltin contamination as a consequence of the establishment of a marina, and increasing yachting activities at Phuket Island, Thailand.

The objective was to investigate how tributyltin (TBT) contamination resulting from the establishment of a new marina and increasing yachting activities at Phuket Island, Thailand affects the area. A minimum of 30 specimen of the mangrove dwelling muricid Chicoreus capucinus were collected at five stations 2 months before and 5, 9, 18 and 33 months after Yacht Haven Marina was constructed. Imposex measured as RPLI and VDSI were significantly correlated to duration of time after construction and distance from the marina. All females developed imposex close to the marina after 18 months, whereas no significant increase occurred at stations more than 2 km from the marina. To evaluate the range of effects of the increasing yachting activities the muricid Thais distinguenda was used as an indicator of imposex at two popular mooring sites for yachts at Phi Phi and Raja Island in Phangna Bay, Thailand. The incidence of imposex was 100% at stations close to the mooring sites. A significant correlation existed between the distance from the sites and the incidence of imposex. The increasing incidence of imposex suggest that TBT contamination is worsening, against global trends, because regulations prohibiting the use of TBT-based paints, do not exist in Thailand.

Simultaneous sample preparation and species-specific isotope dilution mass spectrometry analysis of monomethylmercury and tributyltin in a certified oyster tissue.

A rapid, accurate, sensitive, and simple method for simultaneous speciation analysis of mercury and tin in biological samples has been developed. Integrated simultaneous sample preparation for tin and mercury species includes open focused microwave extraction and derivatization via ethylation. Capillary gas chromatography-inductively plasma mass spectrometry (CGC-ICPMS) conditions and parameters affecting the analytical performance were carefully optimized both for species-specific isotope dilution analysis of MMHg and TBT and for conventional analysis of MBT and DBT201Hg-enriched monomethylmercury and 117Sn-enriched tributyltin were used for species-specific isotope dilution mass spectrometry (SIDMS) analysis. As important, accurate isotope dilution analysis requires equilibration between the spike and the analyte to achieve successful analytical procedures. Since the spike stabilization and solubilization are the most critical and time-consuming steps in isotope dilution analysis, different spiking procedures were tested. Simultaneous microwave-assisted spike stabilization and solubilization can be achieved within less than 5 min. This study originally introduces a method for the simultaneous speciation and isotope dilution of mercury and tin in biological tissues. The sample throughput of the procedure was drastically reduced by fastening sample preparation and GC separation steps. The accuracy of the method was tested by both external calibration analysis and species-specific isotope dilution analysis using the first biological reference material certified for multielemental speciation (oyster tissue, CRM 710, IRMM). The results obtained demonstrate that isotope dilution analysis is a powerful method allowing the simultaneous speciation of TBT and MMHg with high precision and excellent accuracy. Analytical problems related to low recovery during sample preparation are thus minimized by SIDMS. In addition, a rapid procedure allows us to establish a performant routine method using CGC-ICPMS technique.

Retrospective monitoring of organotin compounds in marine biota from 1985 to 1999: results from the German Environmental Specimen Bank.

In archived samples from the German Environmental Specimen Bank, organotin compounds including tributyltin (TBT) and triphenyltin (TPT) as well as their degradation products were quantified. Biota samples from North Sea and Baltic Sea areas were analyzed by gas chromatography/atomic emission detection-coupling after extraction and Grignard or ethylborate derivatization. TBT and TPT were detected in nearly all samples. A decrease of TPT contamination was observed in bladder wrack, common mussels, and eelpout muscle tissues in the period 1985-1999. In this period, TPT concentrations in North Sea mussels decreased from 98 to 7 ng/g (as organotin cation concentration in wet tissue). Concentrations of TBT remained relatively constant with 17 +/- 3 ng/g for mussels from a site with nearby marine traffic and 8 +/- 2 ng/g for a more remote area. The results reflect that TBT is still used as a biocide in antifouling paints whereas the use of TPT as a co-toxicant in such preparations had been ceased in the 1980s. The fact that the use of TBT in antifouling paints was banned in 1991 for small boats within the European Community seems not to have resulted in a decrease of TBT levels in marine biota.

Generation of alpha-phosphonovinyl radicals and development of a new route to highly functionalized vinylphosphonates and vinylphosphonate-incorporated carbocyclic or heterocyclic compounds via a radical trapping sequence.

The first direct generation of synthetically useful alpha-phosphonovinyl radicals was achieved by treatment of alpha-phosphonovinyl halides with a tributyltin radical. The alpha-phosphonovinyl radicals 2a-d were trapped with electron-rich olefins and an electron-deficient olefin to produce alpha-functionalized vinylphosphonates 3a-f in 16-55% yields. The alpha-phosphonovinyl radicals 7e-g containing the YCH2CH=CH2 (Y = O, CH2, S) substituent at the beta-position afforded mixtures of 5-exo and 6-endo cyclization products, 5e-g and 6e-g, in good yields. The 5-exo/6-endo product ratios increase in the following order of the beta-substituent: OCH2CH=CH2 > CH2CH2CH=CH2 > SCH2CH=CH2. The effects of the beta-substituents upon the cyclization reaction were discussed. Radical cyclization of alpha-phosphonovinyl radicals bearing functional groups such as geranyloxy, geranylthio, and (2-cyclohexen-1-yl)thio groups at the beta-position afforded 5-exo, 5-exo and 6-endo, and cis-fused-5,6-ring cyclization products incorporating an alpha,beta-unsaturated phosphonate unit within the ring, respectively, in good yields. The alpha-phosphonovinyl radical 20 underwent tandem radical cyclization-radical cyclization to produce a mixture of two isomeric bicyclo[4.3.0]nonenes including a vinylphosphonate moiety in high yield.

Binding of triethyltin to cat haemoglobin and modification of the binding sites by diethyl pyrocarbonate.

Cat haemoglobin binds 2 mol of triethyltin/mol of haemoglobin. Pretreatment of the haemoglobin with diethyl pyrocarbonate at pH6.0 prevents binding to one site only, whereas photo-oxidation with Methylene Blue removes both sites. Pretreatment of rat haemoglobin with diethyl pyrocarbonate also leads to the loss of one binding site. The possibility is discussed that the two binding sites for triethyltin on both cat and rat haemoglobin have a different chemical nature.

Oxidative phosphorylation. Halide-dependent and halide-independent effects of triorganotin and trioganolead compounds on mitochondrial functions.

1. Each of five triorganotin and five triorganolead compounds was shown to perturb mithochondrial functions in three different ways. One is dependent and two are independent of Cl- in the medium. 2. Structure-activity relationships for the three interactions are described, and compounds suitable as tools for the separate study of each process are defined. 3. In a Cl- -containing medium trimethyltin, triethyltin, trimethyl-lead, triethyl-lead and tri-n-propyl-lead all produce the same maximum rate of ATP hydrolysis and O2 uptake; this rate is much less than that produced by uncoupling agents such as 2,4-dinitrophenol. 4. Increase in ATP hydrolysis and O2 uptake are measures on energy ultilization when triogranotin and triorganolead compounds bring about an exchange of external C1- for intramitochondrial OH- ions. Possible rate-limiting steps in this process are discussed. 5. In a C1- -containing medium ATP synthesis linked to the oxidation of beta-hydroxybutyrate or reduced cytochrone c is less inhibited by triethyltin or triethyl-lead than is ATP synthesis linked to the oxidation of succinate, pyruvate or L-glutamate. 6. The inhibition of ATP synthesis linked to the oxidation of both beta-hydroxybutyrate and reduced cytochrome c consists of two processes: one is a limited uncoupling and is C1- -dependent and the other is a C1- -independent inhibition of the energy-conservation system. 7. The different sensitivities to inhibition by triethyltin of mitochondrial functions involving the oxidation of beta-hydroxybutyrate and succinate are compared and discussed.
