The Interfacial Polarization-Induced Electrorheological Effect.

The Wagner theory, which describes the interfacial polarization in heterogeneous systems, was employed to model the electrorheological (ER) effect under the presumption that the shear stress increment is induced by the interfacial polarization. The currently observed experimental facts, such as the yield stress of some ER fluids, decreases with the applied field frequency increasing or the environment temperature decreasing, while that of other fluids increases with the frequency decreasing or temperature increasing; the strongest ER effect is usually observed in the suspension with the dispersed particle conductivity around 10(-7) S/m; the particle dielectric loss tangent of a good ER fluid usually is above 0.10 at 1000 Hz; and the fluid with a high conductive particle usually has a short response time, can be satisfactorily understood with the extended Wagner model. The Wagner-polarization-induced maximum yield stress of a heterogeneous-type ER fluid is estimated around 7 kPa under the presumption that the dielectric constants of the solid particle and the liquid medium are 10 and 2, respectively, the particle volume fraction is 35%, and the applied electric field strength is 3 kV/mm. It is concluded that the ER effect may substantially correlate with the Wagner polarization, which would help in understanding the mechanism of the ER effect and would provide a strategy for designing high performance ER fluids. Monte Carlo Simulation of Particle Aggregation and Simultaneous Restructuring.

Ultrafine ("nano"-) particles produced from highly supersaturated vapors or liquids often undergo rapid coagulation and slow interspherule coalescence. Resulting "aggregates" typically contain hundreds of small spherules bound together in tenuous structures characterized by mass fractal dimensions much less than 3. Such aggregates have large and relatively accessible initial surface area but are metastable with respect to more compact configurations, especially in high temperature environments (e.g., flames). Subject to deliberately idealized "uncoupled" rate laws for coagulation and coalescence, we illustrate the power of Monte Carlo simulation methods to obtain the self-preserving joint distribution function (with respect to both particle size and surface area) of populations of coagulating fractal aggregates in the continuum regime, simultaneously undergoing finite-rate restructuring (e.g., via surface-energy-driven viscous flow). Unconditional distributions with respect to either particle volume or area are also obtained from the Monte Carlo simulations. These are conveniently quantified by fitting them to log-normal distributions and we report the sensitivity of the associated spreads to characteristic fusion/coagulation time ratio, chi, and particle fractal dimension, Df, here prespecified. We also calculate and report selected "mixed" moments of the joint pdf with respect to particle volume and surface area needed for engineering calculations of deposition or diffusion-controlled vapor scavenging, as well as the important ratio of actual mean area to that area corresponding to the mean particle volume in the aerosol population. This work sets the stage for tractable simulations of particle dynamics in more complex coagulating systems requiring multi-internal (state-) variables for their more realistic and self-consistent description. Effect of ultraviolet light on the dielectric behavior of bone at microwave frequencies.

The dielectric constant of bone and its two components (collagen and apatite) were determined at 9.2 GHz in a room temperature environment by the cavity perturbation technique. After samples were exposed to ultraviolet light, repeat measurements under identical conditions showed decreases in both the real and the imaginary parts of the dielectric constant in all cases. The present work describes the technique and a possible mechanism of ultraviolet interaction with bone.

pH homeostasis and ATP synthesis: studies of two processes that necessitate inward proton translocation in extremely alkaliphilic Bacillus species.

Alkaliphilic Bacillus species that are isolated from nonmarine, moderate salt, and moderate temperature environments offer the opportunity to explore strategies that have developed for solving the energetic challenges of aerobic growth at pH values between 10 and 11. Such bacteria share many structural, metabolic, genomic, and regulatory features with nonextremophilic species such as Bacillus subtilis. Comparative studies can therefore illuminate the specific features of gene organization and special features of gene products that are homologs of those found in non-extremophiles, and potentially identify novel gene products of importance in alkaliphily. We have focused our studies on the facultative alkaliphile Bacillus firmus OF4, which is routinely grown on malate-containing medium at either pH 7.5 or 10.5. Current work is directed toward clarification of the characteristics and energetics of membrane-associated proteins that must catalyze inward proton movements. One group of such proteins are the Na+/H+ antiporters that enable cells to adapt to a sudden upward shift in pH and to maintain a cytoplasmic pH that is 2-2.3 units below the external pH in the most alkaline range of pH for growth. Another is the proton-translocating ATP synthase that catalyzes robust production of ATP under conditions in which the external proton concentration and the bulk chemiosmotic driving force are low. Three gene loci that are candidates for Na+/H+ antiporter encoding genes with roles in Na(+)-dependent pH homeostasis have been identified. All of them have homologs in B. subtilis, in which pH homeostasis can be carried out with either K+ or Na+. The physiological importance of one of the B. firmus OF4 loci, nhaC, has been studied by targeted gene disruption, and the same approach is being extended to the others. The atp genes that encode the alkaliphile's F1F0-ATP synthase are found to have interesting motifs in areas of putative importance for proton translocation. As an initial step in studies that will probe the importance and possible roles of these motifs, the entire atp operon from B. firmus OF4 has been cloned and functionally expressed in an Escherichia coli mutant that has a full deletion of its atp genes. The transformant does not exhibit growth on succinate, but shows reproducible, modest increases in the aerobic growth yields on glucose as well as membrane ATPase activity that exhibits characteristics of the alkaliphile enzyme.

Measuring polymerization shrinkage of photo-activated restorative materials by a water-filled dilatometer.

A water-filled dilatometer specifically designed for determining the polymerization shrinkage of photo-activated composite restorative materials was used to measure the polymerization shrinkage of three visible light-activated composites. Polymerization shrinkage values ranged from 1.82% for P-50 to 2.15% and 2.19% for Herculite XRV and Prisma APH, respectively. Shrinkage data obtained in this investigation were compared with the published data, and the factors which affect shrinkage measurements were reviewed. It was concluded that maintaining a constant temperature environment (+ or - 0.02 degrees C) for the dilatometer during the shrinkage test was the most critical factor for successful application of the dilatometer.

Structural and kinetic properties of chymotrypsin from Atlantic cod (Gadus morhua). Comparison with bovine chymotrypsin.

1. Two chymotrypsins with isoelectric points pI 6.2 and 5.8 were purified from the pyloric caeca of Atlantic cod using a phenyl-Sepharose column and chromatofocusing chromatography. The apparent molecular weight was 26,000 as judged by SDS-polyacrylamide gel electrophoresis and gel filtration. 2. The cod enzymes differed from bovine chymotrypsin in having a slightly higher molecular weight and more acidic pI points. N-terminal amino acid sequence analysis of cod chymotrypsin B showed considerable similarity with bovine chymotrypsin. 3. Heat stability and stability towards acidic pH were reduced in the cod enzymes. Generally, the cod and bovine chymotrypsins responded similarly to various protease inhibitors. However, the cod chymotrypsins were less sensitive to aprotinin inhibition but more sensitive towards soybean trypsin inhibitor and cysteine. 4. Kinetic properties were examined and the cod enzymes found to be more active towards both ester (N-benzoyl-tyrosine ethyl ester) and amide (N-benzoyl-tyrosine-p-nitroanilide) substrates. The observed differences in kinetic properties are indicative of an adaptive response towards the low temperature environment in which the cod lives.

Regulation of ischemic cell death by the lipoic acid-palladium complex, Poly MVA, in gerbils.

Modulation of ischemic cell death can be accomplished via a multitude of mechanisms, such as quenching radical species, providing alternative energy sources, or altering glutamate excitation. Transient cerebral ischemia will induce apoptotic cell death selectively to hippocampal cornus ammon's field 1 of the hippocampus (CA1) pyramidal cells, while neighboring CA3 and dentate neurons are spared. Poly MVA is a dietary supplement based on the nontoxic chemotherapeutic lipoic acid-palladium complex (LAPd). LAPd is a liquid crystal that works in cancer cells by transferring excess electrons from membrane fatty acids to DNA via the mitochondria. Therefore, by its structural nature and action as a redox shuttle, it can both quench radicals as well as provide energy to the mitochondria. To understand the role of LAPd in regulating ischemic cell death, we studied Poly MVA. Male Mongolian gerbils were subjected to 5 min of bilateral carotid artery occlusion under a controlled temperature environment (37.0-38.0 degrees C). Animals were injected with physiological saline or either 30, 50, or 70 mg/kg of Poly MVA every 24 h beginning immediately after the occlusion until being sacrificed on experimental day 4. Damage was evaluated by analyzing nesting behavior and conducting blinded measures of viable CA1 lengths. All Poly MVA treatment dosages significantly (p < 0.05) reduced hippocampal CA1 damage by 72 h. Nesting scores were significantly improved after 30 and 50 mg/kg treatment but not 70 mg/kg. While nesting is usually a very accurate indicator of morphological damage, the 70 mg/kg-treated animals demonstrated excessive energy, thus ignoring the nesting material. While numerous routes offer varying degrees of CA1 neuronal survival after transient global ischemia, only the LAPd complex, which quenches radicals and provides energy to stabilize the mitochondria, offers such significant protection. Thus, the administration of Poly MVA may be a potent neuroprotective agent for victims of transient ischemic attack (TIA), cardiac arrest, anesthetic accidents, or drowning.

SEM observations and differential scanning calorimetric studies of new and sterilized nickel-titanium rotary endodontic instruments.

Scanning electron microscopy (SEM) and differential scanning calorimetric (DSC) studies were utilized to investigate surface and microstructure of two brands of rotary nickel-titanium (NiTi) endodontic instruments, in the as-received condition and after subjection to 1, 6, and 11 sterilization cycles. A total of 66 ProFile (n = 33) and Flexmaster (n = 33) files were examined. SEM observations indicated the presence of surface imperfections and adherent material in all new and sterilized instruments and an increase in surface roughness of the instruments that underwent multiple sterilizations. DSC measurements showed that the specimens of both brands, in the as-received condition and after 11 sterilizations, were completely austenite in the oral environment temperature, suggesting that they are capable of superelastic behavior in appropriate clinical conditions.

Study of 1-3-2 type piezoelectric composite transducer array.

Based on a new structure of precise location and decoupling between the transducer elements, high frequency underwater transmission transducer arrays with 4 elements and 8 elements serried uniform linear array were studied, using novel 1-3-2 type piezoelectric composite as the sensing material. There are ceramic framework supports in the transverse and longitudinal directions in 1-3-2 type piezoelectric composite structure; the transducer is free from the influence of outside mechanical impact and the environment temperature change. Transducer and array samples have been designed, fabricated and measured. The resonant frequency is 150 kHz, resonant transmission response greater than 160 dB, and bandwidth is from 140 kHz to 160 kHz. The results indicate that, the transducer array has wide bandwidth, high sensitivity, stabile performance, and good coherence.

Male breast cancer.

Occurrence of male breast cancer, a rare disease, peaks at age 71 years. Familial cases usually have BRCA2 rather than BRCA1 mutations. Occupational risks include high temperature environments and exhaust fumes, but electromagnetic fields have not been implicated. Hyperoestrogenisation resulting from Klinefelter's, gonadal dysfunction, obesity, or excess alcohol, all increase risk as does exposure to radiation, whereas gynaecomastia does not. Presentation is usually a lump or nipple inversion, but is often late, with more than 40% of individuals having stage III or IV disease. Most tumours are ductal and 10% are ductal carcinoma in situ. Surgery is usually mastectomy with axillary clearance or sentinel node biopsy. Indications for radiotherapy, by stage, are similar to female breast cancer. Because 90% of tumours are oestrogen-receptor-positive, tamoxifen is standard adjuvant therapy, but some individuals could also benefit from chemotherapy. Hormonal therapy is the main treatment for metastatic disease, but chemotherapy can also provide palliation. National initiatives are increasingly needed to improve information and support for male breast cancer patients.

Heat-labile bacterial alkaline phosphatase from a marine Vibrio sp.

Psychrophilic organisms have successfully adapted to various low-temperature environments such as cold ocean waters. Catalysts with increased catalytic efficiencies are produced, generally at the expense of thermal stability due to fewer non-covalent stabilizing interactions. A marine bacterial strain producing a particularly heat-labile alkaline phosphatase was selected from a total of 232 strains isolated from North-Atlantic coastal waters. From partial 16S rRNA sequences the strain was characterized as a Vibrio sp. An alkaline phosphatase was purified 151-fold with 54% yield from the culture medium using a single step affinity chromatography procedure on agarose-linked L-histidyldiazobenzylphosphonic acid. The active enzyme was a 55 +/- 6 kDa monomer. The enzyme had optimal activity at pH 10 and was strikingly heat-labile with a half-life of 6 min at 40 degrees C and 30 min at 32 degrees C. This enzyme from Vibrio sp. had a higher turnover number (k(cat)) and higher apparent Michaelis-Menten factor (K(m)) than the enzyme from Escherichia coli, a clear-indication of cold-adaptation. Inorganic phosphate was a competitive inhibitor with a relatively high K(i) value of 1.7 mM. Low affinity for phosphate may contribute to higher turnover rates due to more facile release of product.

Videocapillaroscopy and Diabetes mellitus: area of transverse segment in nailfold capillar loops reflects vascular reactivity.

UNLABELLED: The value of measuring projected area of transverse segment of hand nailfold capillary loops using computerized videophotometry was studied in 17 healthy individuals (10 women and 7 men) group A and 17 patients with diabetes type 2 (10 women and 7 men) group B with comparable ages (49.94+/-8.62 x 49.11+/-14.63 years old P=0.49). Videocapillaroscopy (VC) was performed under controlled temperature environment (24-26 degrees C). The test of post-occlusive reactive hyperemia was performed using a sphygmomanometer attached to the fourth left hand finger, 20 mmHg above maximum arterial pressure during 1 min. Images were captured each 2 s during 1 min. Time averages to reach the maximum post-ischemia area were determined as well as the area's increment percent in both groups. Both averages were compared by the Mann-Whitney test. There was no differences between area increments percent averages among controls and patients with diabetes (52.49+/-22.6 x 67.99+/-47.48% P=0.67), but the time to reach it during reperfusion was significantly increased among diabetics (5.52+/-2.96 x 18.58+/-8.08 s P<0.0001). CONCLUSIONS: changes of capillary response to ischemia may be observed among patients with diabetes through bi-dimensional measures of projected area of capillary loops transverse segment in VC using a computerized system of image analysis. Patients with Diabetes mellitus do not have significantly different maximum area increments than controls, but spend more time to reach it during reperfusion.

Lack of contact transmission of recombinant Marek's disease virus type 1 expressing the fusion protein of Newcastle disease virus.

To clarify the level of excretion of a recombinant Marek's disease virus type 1 (rMDV1) that confers good protection in chickens against both Marek's and Newcastle diseases, even in the presence of maternal antibodies, contact transmission tests were conducted. Naïve chickens kept in the same cage or room with chickens inoculated with rMDV1 did not produce antibodies against MDV1 or the fusion protein of Newcastle disease virus. Moreover, the rMDV1 was not isolated from the dander of chickens inoculated with rMDV1. Even under the stressful conditions of forced molting and a high temperature environment, rMDV1 was not isolated from the dander of inoculated birds. The viral DNA, however, was detected from the dander of chickens inoculated with rMDV1 as well as a commercial vaccine. These findings indicate that dander from chickens inoculated with rMDV1 includes viral DNA, but does not contain infectious virus.

Comparison and evaluation of the synthetic biopolymer poly-L-aspartic acid and the synthetic "plastic" polymer poly-acrylic acid for use in metal ion-exchange systems.

Poly-L-aspartic acid (PLAsp), a biopolymer, and a similar synthetic polymer, poly-acrylic acid (PAA), each consisting of approximately 50 repeating Asp and acrylic acid monomers, respectively, were immobilized onto controlled pore glass (CPG) and evaluated for use as metal ion-exchange materials. Both polymers achieve metal complexation primarily through their repeating carboxylate side groups resulting in a similar binding trend for the metals tested (Ca(2+), Cd(2+), Co(2+), Cu(2+), Mg(2+), Mn(2+), Na(+), Ni(2+), Pb(2+)), with metal binding capacities ranging from <0.1 to 12 micromol metal/g column and <0.1 to 32 micromol metal/g column for PLAsp and PAA respectively. Cu(2+) and Pb(2+) exhibited strong binding to both materials, while the other metals demonstrated only weak or minimal binding. Both columns allowed for quantitative release of bound metals through acid stripping and experienced increased overall metal binding with increasing pH. Both systems also maintained similar structural and chemical stability when continuously exposed to neutral buffered, highly acidic, oxidizing, large molecule rich, and elevated temperature environments. The main differences between the two systems are the material cost and system biodegradability.

In vitro manipulation and propagation of medicinal plants.

Well developed techniques are currently available to help growers meet the demand of the pharmaceutical industry in the next century. These protocols are designed to provide optimal levels of carbohydrates, organic compounds (vitamins), mineral nutrients, environmental factors (e.g. light, gaseous environment, temperature, and humidity) and growth regulators required to obtain high regeneration rates of many plant species in vitro and thereby facilitate commercially viable micropropagation. Well-defined cell culture methods have also been developed for the production of several important secondary products. An overview of the regeneration of medicinal plants by direct and indirect organogenesis and by somatic embryogenesis from various types of explants is presented, and the use of these techniques combined with other biotechnological approaches to improve medicinal plants through somaclonal variation and genetic transformation is reviewed.

The value of microbial diversity.

In the past few years, due to the use of molecular methods, our knowledge of microbial diversity has increased dramatically, not only from a phylogenetic and taxonomic perspective but also from an ecological basis. We now know that microorganisms exist in every conceivable place on Earth, even in extreme environments. Temperature may be the only limitation as to where they can and cannot exist and/or function. As more small subunit rDNA sequence information becomes available there is a real need to start turning the information into knowledge that can be applied to better elucidate and understand structure-function relationships within ecosystems, develop new culturing methods, and discover new products and processes. It has been stated on numerous occasions that the 21(st) century is the century for biology. Within that context, we must address the real value of microbial diversity.

Effect of polyamines on histone-induced DNA compaction of hyperthermophilic archaea.

The effect of polyamines on histone-mediated DNA compaction was examined in vitro with archaeal histone HpkA from Pyrococcus kodakaraensis KOD1. An agarose gel mobility-shift experiment indicated that histone-bound DNA (compacted DNA) was further compacted by addition of a polyamine (putrescine, spermidine, or spermine) or its acetylated form (N-acetylputrescine, N1-acetylspermidine, N8-acetylspermidine, or N1-acetylspermine) when the mixture was incubated at above 75 degrees C. Spermine was most effective in compaction enhancement among all the polyamines tested. A high concentration of potassium ion (1.0 M) did not stabilize the compacted form of DNA even though double-stranded DNA was stably maintained against thermal denaturation at elevated temperatures under this condition. It appears likely that multivalent polyamines have a nucleosome maintenance function in hyperthermophilic archaea in high-temperature environments.

Structural and thermodynamic studies on a salt-bridge triad in the NADP-binding domain of glutamate dehydrogenase from Thermotoga maritima: cooperativity and electrostatic contribution to stability.

Cooperative interactions within ion-pair networks of hyperthermostable proteins are thought to be a major determinant for extreme protein stability. While the favorable thermodynamic contributions of optimized electrostatics in general as well as those of pairwise interactions have been documented, cooperativity between pairwise interactions has not yet been studied thermodynamically in proteins from hyperthermophiles. In this study we use the isolated cofactor binding domain of glutamate dehydrogenase from the hyperthermophilic bacterium Thermotoga maritima to analyze pairwise and cooperative interactions within the salt-bridge triad Arg190-Glu231-Lys193. The X-ray structure of the domain was solved at 1.43 A and reveals the salt-bridge network with surrounding solvent molecules in detail. All three participating charges in the network were mutated to alanine in all combinations. The X-ray structure of the variant lacking all three charges reveals that the removal of the side chains has no effect on the overall conformation of the protein. Using solvent denaturation and thermodynamic cycles, the interaction energies between each pair of residues in the network were determined in the presence and in the absence of the third residue. Both the Arg190-Glu231 ion pair and the Lys193-Glu231 salt bridge in the absence of the third residue, contribute favorably to the free energy for unfolding of the domain in urea. Using guanidinium chloride as denaturant reveals a strong cooperativity between the two ion-pair interactions, the presence of the second ion pair converts the first interaction from destabilizing into stabilizing by as much as 1.09 kcal/mol. The different energetics of the salt-bridge triad in urea and GdmCl are discussed with reference to the observed anion binding in the crystal structure at high ionic strength and their possible role in a highly charged, high-temperature environment such as the cytoplasm of hyperthermophiles.
