Studies on the tolerance of silk, nylon, dacron and collagen suture material in the cornea of the rabbit.

With rabbit corneas we did comparative biomicroscopical and histological investigations on material tolerance of the following threads at present used in microsurgery: Barraquer silk (virgin silk), nylon 30 mum (Tübinger nylon), dacron (polyester) 40 mum and plain collagen 7-0. The sutures were implanted into the cornea intra-lamellarly, completely under the microscope. The examinations showed only very little tissue reaction in all these materials.

Chemiluminescent detection of DNA on nylon membranes.

Two types of nonradioactive DNA detection systems were optimized for use with nylon membranes in Southern transfers. A luminol substrate system (consisting of an enhanced chemiluminescent reaction utilizing luminol enzyme substrate) was used with peroxidase-labeled probe DNA, and a dioxetane-based substrate was used with alkaline phosphatase/antibody and digoxigenin-labeled probe DNA. Chemiluminescence was detected by autoradiography. Methods for reprobing the membranes were also optimized for both systems; blots could be reprobed at least ten times. Results showed that excellent sensitivity and low background can be achieved on both amphoteric and positively charged nylon membranes, using either detection system.

Effect of storage conditions on restriction fragment length polymorphism (RFLP) analysis of deoxyribonucleic acid (DNA) bound to positively charged nylon membranes.

The ability to generate an autoradiogram from deoxyribonucleic acid (DNA) immobilized on a positively charged nylon membrane could be compromised by the storage conditions of the membrane. HaeIII-digested human DNA was size fractionated and transferred to two types of positively charged nylon membranes. The membranes were stored at -20 degrees C, 4 degrees C, and ambient temperature and humidity for times ranging from 1 day to 13 weeks, then hybridized to variable number of tandem repeat (VNTR) probes to examine the effect of the storage conditions on the membrane-bound DNAs. It was shown that such membranes could be successfully hybridized and rehybridized if they were stored at -20 or 4 degrees C, but storage under ambient conditions reduced or eliminated the likelihood of successful hybridization.

Nylon cable band reactions in ovariohysterectomized bitches.

Nylon cable bands used as ligatures caused postsurgical complications in 5 ovariohysterectomized bitches. Clinical signs included fever, stiffness, strangury, draining fistulae, vaginal discharge, and tenesmus. Most signs first appeared several years after surgery, and all signs were resolved after removal of the bands. On the basis of our experience, we suggest that nylon cable bands not be used for ovariohysterectomy ligations.

Long-term comparison of Novafil and nylon in corneoscleral sections.

To compare the long-term degradation of Novafil (polybutester, American Cyanamid Company) and nylon suture material, we examined corneoscleral sections of nine rabbits 23 months after the placement of sutures. Only two nylon sutures remained, one in each wound of two rabbits; all the Novafil sutures were still present. However, examination of the Novafil sutures with scanning electron microscopy revealed signs of disintegration.

Nylon suture toxicity after vitrectomy surgery.

A patient underwent an uneventful pars plana vitrectomy with superior sclerotomies closed with 8-0 nylon suture. Four months later, the patient had a suture reaction presumably related to allergy to the suture itself. The signs and symptoms resolved after removal of the suture. Although similar cases have been reported after nylon sutures in cataract wound closure, this is the first reported occurrence of this phenomenon in vitrectomy surgery to our knowledge.

An evaluation of preservative adsorption onto nylon membrane filters.

Pharmaceutical drug products often contain antimicrobial agents as a preservative in their formulation. These excipients are required to destroy or impede the growth of microorganisms that inadvertently enter the product during manufacturing. Unfortunately, these preservatives may also interfere with microbiological assays used to determine product sterility or bioburden levels. The extent of interference by these preservatives can be quite significant, but varies depending on the method used. The most frequently used method for testing parenteral drug products is the membrane filtration technique. Membrane filters are composed of a wide variety of materials such as cellulose, polycarbonate, acrylic polypropylene, Teflon, and nylon. This study evaluated the adsorption characteristics that nylon filters, obtained from five different manufacturers, had on the filtration of solutions of four different antimicrobial compounds (phenol, methylparaben, propylparaben, and benzalkonium chloride). The adsorption properties were determined using both HPLC and microbiological assay techniques. The data revealed that there was a wide range in the amounts of antimicrobial agent (2.3 to 94.1%) bound to the membrane filters when direct product filtration was used without a subsequent rinse step. However, when a rinse step is included, only propylparaben showed any significant "true" adsorption (less than 1 to 33.3%), but showed only marginal bacterial inhibition. Interestingly, the microbiological assays indicated that with a saline rinse step, only benzalkonium chloride was lethal for the two challenge organisms even though the percent adsorbed as measured by HPLC was below 1%. This discrepancy is significant because it demonstrates the analytical limitation when using HPLC to detect minimal concentrations of benzalkonium chloride that may be deleterious to microorganisms.(ABSTRACT TRUNCATED AT 250 WORDS)

Parameters affecting hybridization of nucleic acids blotted onto nylon or nitrocellulose membranes.

Nine different nylon and nitrocellulose membranes were compared utilizing four different methods of attaching the nucleic acid target. Nylon membranes repeatedly demonstrated increased sensitivity as compared to nitrocellulose membranes. Sensitivity could also be enhanced by mildly denaturing the target prior to attachment onto the membrane. This was achieved by either UV cross-linking or baking.

Astigmatism after cataract surgery: nylon versus Mersilene.

To find out if postoperative astigmatism decays less if Mersilene rather than nylon sutures are used, 25 patients had a standard wound closure with 10-0 nylon in one eye and 11-0 Mersilene in the other. There was no difference in the astigmatism decay over a 6-month period.

[Active enhancement of rat kidney grafts by immunization of the recipients with donor-specific nylon-adherent spleen lymphocytes].

At different intervals (2, 4, 8, 12 and 21 days) after preimmunization of BD IX inbred rats with density gradient-separated nylon-wool-adherent spleen lymphocytes from rats of the MHC different inbred strain Sprague-Dawley, (BD IX X SD)F1 kidneys were transplanted into the immunized BD IX rats. The non-immunized control rats died after 11-16 days. Kidneys transplanted 8 or 12 days after preimmunization survived indefinitely without any signs of rejection. In contrast, the rats grafted 2 and 4 days after preimmunization showed elevated levels of blood urea nitrogen (mean: 70-83 mmol/l) on the 7th post-operative day. The transplantation on day 21 resulted in a slight rejection episode on the 7th postoperative day. These results suggest the efficacy of nylon-wool-adherent lymphocytes in inducing an active transplant protection.

In vitro quantitative study of newly made antibacterial braided nylon sutures.

This study was done to examine quantitatively the antibacterial property of a newly made, silver compound coated braided nylon suture and to confirm the previously reported qualitative antibacterial data. Three representative bacterial species were used and they were Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The suture specimens were embedded in a custom built plastic device filled with a fixed concentration of bacteria for predetermined periods of incubation. A direct current ranging from 0.4 to 40.0 microamperes was applied to the suture specimens. The bacterial suspension was periodically removed for a standard plate count in order to determine quantitatively the antibacterial capability of the suture specimens. The amounts of silver ions released to the medium under various direct current levels were also determined by a pH/ion meter. The antibacterial property of the suture was evident in the anode site of the material, and at a fixed current, the degree of bacteriostatic effect depended upon the type of bacterial species. For example, a difference of almost 10(3) in the number of Pseudomonas aeruginosa was observed within a period of six hours. The responses of Staphylococcus aureus and Escherichia coli to the silver compound coated nylon thread, however, were not as drastic as Pseudomonas aeruginosa. These quantitative data were consistent with the previously reported qualitative observation of the width of clear zone in bacterial culture plates. The silver ion concentration in the medium increased with increasing either the current level or time, or both. At the end of six hours, the ion concentrations were 7.3 micrograms per milliliter at 0.4 microampere, 44.3 micrograms per milliliter at 4.0 microamperes and 305.7 micrograms per milliliter at 40.0 microampere.

Hematopoiesis on nylon mesh templates. I. Long-term culture of rat bone marrow cells.

Rodent hematopoietic cells have been perpetuated in long-term bone marrow culture (LTBMC), provided that they were plated onto a pre-established layer of bone marrow stromal cells (fibroblasts, reticular cells, adipocytes, macrophages, etc.). This monolayer-type system supports the self-renewal of murine pluripotent stem cells and produces substantial numbers of progenitors and mature cells of the myeloid lineage. In an effort to increase the growth potential of cells in LTBMC, an adherent matrix of stromal cells was established on a pretreated nylon screen template. Subsequent seeding of hematopoietic cells onto this matrix occurred both on the surface of the adherent layer and in the interstices formed by the developing stromal cells. A three dimensional growth pattern of hematopoietic colonies and clusters was observed. Mature cells and late stage precursors of the myeloid and erythroid series were observed in the non-adherent layer for the duration of the experiment (39 weeks). Cells similar in appearance to small lymphocytes were also seen in both the non-adherent and adherent layers. Cells associated with the adherent layer of this LTBMC system displayed the ability to reconstitute hematopoiesis in the irradiated host. It is suggested that because of its three-dimensional nature the nylon mesh LTBMC system possesses a greater hematologic potential per unit area than monolayer-type LTBMC.

Routine glucose determination in serum by use of an immobilized glucose dehydrogenase nylon-tube reactor.

We report a method for immobilizing glucose dehydrogenase on the inside surface of nylon tubes to produce an immobilized-enzyme nylon-tube reactor. The glucose dehydrogenase reactor is integrated into the flow system of a continuous-flow analyzer to facilitate routine analysis of serum glucose at 50 samples/h. We compared results with those by the reference hexokinase/glucose-6-phosphate dehydrogenase solution method. The coefficient of correlation was r = 0.996. A glucose dehydrogenase reactor made starting with 1 mg (250 U) of enzyme was stable during eight weeks of continuous use, that is, for nearly 3500 tests. This reduced the cost of the assay by at least 50-fold, compared with that for a commercial glucose dehydrogenase test pack method.

Complications of 10-0 nylon sutures.

Three cases are presented which document the finding that 10-0 monofilament nylon suture may cut through corneal tissue after a coup or contrecoup injury to a recently operated eye. The authors speculate that a larger gauge suture does not have the tendency to cut through recently operated corneal tissue as easily as 10-0 monifilament nylon. The authors recommend the use of a larger gauge suture in closing corneal incisions in persons whom the surgeon feels are at a greater postoperative traumatic risk.

Intraocular foreign body from a nylon line grass trimmer.

Nylon line grass trimmers are commonplace in America's lawn armamentarium. As their number has increased, so has the potential for ocular injury from objects thrown from these tools. A patient of ours was injured by an intraocular piece of nylon line thrown from one of these tools. The patient required removal of the foreign body, lensectomy, and anterior vitrectomy to rehabilitate her eye. This report again demonstrates the need for eye protection when using any type of lawn equipment. We think manufacturers of these products should label them to show that eye protection is necessary for the safe use of these tools.

Enzyme-linked immunosorbent assay for detection of antibodies to extractable nuclear antigens in systemic lupus erythematosus, with nylon as solid phase.

In this enzyme-linked immunosorbent assay (ELISA) for detection of antibodies against extractable nuclear antigens (ENA) in sera of patients with systemic lupus erythematosus (SLE), nylon is used as solid phase for antigen binding instead of the commonly used polystyrene surface. Optimal conditions for activation of the nylon beads, antigen coating, and other relevant factors have been investigated. We compared the incidence of anti-ENA antibodies in SLE, using chromogenic and fluorogenic enzyme substrates. Of SLE patients, 54% were positive for anti-ENA antibodies when chromogenic substrate was used as compared with 68% for fluorogenic substrate. Antibody activity against Sm and RNP antigens was distinguished on the basis of ribonuclease sensitivity of the RNP antigen. The method described offers advantages such as decreased background activity, increased surface area, facility for prolonged storage of antigen-coated solid phase, and miniaturization of the assay.

[Immobilization of antibodies against Australia antigen on nylon spheres: use in immunoenzymatic determination of HBsAg].

Antibodies against HBsAg were immobilized by covalent linkage on nylon beads to be used in solid phase enzyme-linked immunoassays. The covalent bond between the protein and the partially hydrolyzed support was obtained via glutaraldehyde or succinylester methods. These particular solid phases were compared, in a conventional enzyme-linked immunosorbent assay for hepatitis B antigen, with polystyrene beads coated by simple adsorption of IgG on plastic surface. The analytical response was more satisfactory when the polystyrene beads were used as immunoextractive support even if more IgG molecules could be immobilized on nylon beads.

Effect of antiarrhythmic drugs on In-111-labeled leukocytes: chemotaxis and adherence to nylon wool.

The influence of lidocaine (L) and procainamide (P) on the chemotactic ability and adherence to nylon wool of In-111-labeled human polymorphonuclear leukocytes (PMNs) was investigated. At the normal therapeutic levels of L (0.022 mM whole blood) or P (0.03 mM whole blood) no change in PMN function was observed. However, at and above five times the aforementioned blood levels of L, significant reduction in the chemotactic ability of PMNs was noted (p less than 0.005). The adverse effects of In-111 radiation appeared insignificant at all L or P concentrations during the 3-hr observation period. The labeled PMNs were resistant to the toxic effects of a higher concentration of P than that of L, and the reduction in PMN chemotaxis and adherence to nylon wool was not apparent until the P concentration reached 1.5 mM.
