Synthesis and pharmacological properties of N-arylpiperazine-N'-alkylindanes.

Ten new compounds, N-aryl substituted piperazinealkylindanes, have been synthesized. The most active one 1-(2-[4-(3-chlorophenyl)-1-piperazinyl]-ethyl)-indane (compound 9), displayed evident central serotoninolytic properties in the 5-hydroxytryptamine (5-HTP) head twitch test in mice, as well as in the tryptamine convulsions test and the quipazine-stimulated hind paw flexor reflex test in rats.

Nonpeptide endothelin receptor antagonists. III. Effect of SB 209670 and BQ123 on acute renal failure in anesthetized dogs.

Endothelin (ET) is a potent vasoconstrictor that has been implicated in the pathogenesis of acute renal failure (ARF). In order to investigate the potential role of ET in ARF in the dog, the effect of a mixed ETA and ETB receptor antagonist, (+/-)-SB 209670, [(1RS-2SR,3RS)-3-(2-carboxymethoxy-4-methoxyphenyl)-5- (prop-1-yloxy) indane-2-carboxylic acid], and a selective ETA antagonist, BQ123, were evaluated in anesthetized uninephrectomized dogs undergoing 60 min of renal occlusion. (+/-)-SB 209670 (1 microgram/kg/min) and BQ123 (10 micrograms/kg/min) were infused directly into the renal artery (intrarenal) for 30 min before renal occlusion, during occlusion and for 60 min after reperfusion at doses that inhibited the renal vasoconstrictor effects of intrarenal renal artery infusions of ET-1. Renal occlusion resulted in a significant reduction in inulin clearance (from 26.2 +/- 1.8 to 3.2 +/- 1.1 ml/min). This response was significantly (P < .05) attenuated by (+/-)-SB 209670 (from 23.5 +/- 2.2 to 7.6 +/- 2.0 ml/min) but not by BQ123 (from 23.5 +/- 1.7 to 4.9 +/- 1.2 ml/min). Endogenous creatinine clearance showed the same pattern. After renal artery occlusion, fractional sodium and fractional potassium excretions were increased significantly. (+/-)-SB 209670, but not BQ123, resulted in a significant reduction in fractional sodium; however, neither compound altered fractional potassium excretion. The data suggest that ET receptor antagonists, possibly by altering tubular sodium reabsorption, may be beneficial in ischemia-induced ARF.

Assessment of the discriminative stimulus effects of the D3 dopamine antagonist PNU-99194A in rats: comparison with psychomotor stimulants.

The present study examined the discriminative stimulus effects of the D3 dopamine receptor antagonist PNU-99194A [5,6-di-methoxy-2-(dipropylamino)indan-hydrochloride] in male Sprague-Dawley rats. Eight rats were trained to discriminate cocaine (10 mg/kg, i.p.) from saline in a two-choice, water-reinforced drug discrimination procedure. In tests of stimulus generalization, PNU-99194A (1.25-40.0 mg/kg, s.c. and i.p.) did not substitute for cocaine. PNU-99194A (5.0-20 mg/kg) also did not significantly block the discrimination of cocaine (10 mg/kg), nor did it potentiate a low dose (1.25 mg/kg) of cocaine. A separate group of eight rats were trained to discriminate PNU-99194A from saline. These subjects met the discrimination criterion within an average of 68 (S.E.M. = 6.5) training sessions; the ED50 for PNU-99194A was 2.6 mg/kg. In stimulus generalization tests, cocaine (1.25-10 mg/kg) did not substitute for PNU-99194A, when administered by either i.p. or by s.c. injection. In addition, neither amphetamine (0.25-1.0 mg/kg) nor caffeine (8.0-64 mg/kg) produced stimulus generalization in these rats. These results indicate that D3 receptors do not play a critical role in the discriminative stimulus effects of cocaine. Furthermore, although PNU-99194A is capable of establishing and maintaining discriminative stimulus control in rats, the effects of this D3-preferring antagonist are dissimilar from those of psychomotor stimulants. Given the unique behavioral profile of D3 receptor antagonists, the potential utility of these agents as adjunctive treatments for psychostimulant abuse is discussed.

Structure-activity relationships in the mutagenicity and cytotoxicity of putative metabolites and related analogs of benzene derived from the valence tautomers benzene oxide and oxepin.

A series of putative metabolites and related analogs of benzene, derived from the valence tautomers benzene oxide and oxepin, was tested for mutagenicity (reversions to histidine prototrophy and forward mutations to resistance to 8-azaguanine) and for cytotoxicity by the Ames Salmonella mutagenicity test. Benzene was not mutagenic in either assay. The benzene oxide-oxepin system and benzene dihydrodiol induced point mutations but not frameshifts. 4,5-sym-Oxepin oxide, which is a putative metabolite of the oxepin valence tautomer; 3,6-diazo-cyclohexane-1,6-3,4-dioxide, a synthetic precursor of sym-oxepin oxide; and transoid-4,11-dioxatricyclo(5.1 0)undeca-1,6-diene, a stable bridge-head diene analog of sym-oxepin oxide, were toxic but not mutagenic in both assays. 4H-Pyran-4-carboxaldehyde, a stable acid catalyzed rearrangement product of sym-oxepin oxide, was not mutagenic and much less cytotoxic than sym-oxepin oxide. Stable analogs of the valence tautomer benzene oxide, namely syn-indan-3a,7a-oxide and syn-2-hydroxyindan-3a,7a-oxide, were mutagenic and induced point mutations. All compounds were cytotoxic to Salmonella. Firstly, the apparent decay times of these chemicals, especially that of sym-oxepin oxide, were surprisingly longer than expected, as judged by quantitative plate diffusion assays. Secondly, it is concluded that if benzene oxide is further metabolized in its oxepin tautomeric form, toxic but not mutagenic products are formed. Thirdly, the relatively weak mutagenicity of benzene oxide may be mainly due to its instability and corresponding low probability to reach intracellular polynucleotide targets, whereas stable analogs of benzene oxide are relatively more potent mutagens.

Hydrogenolysis of benzylic alcohols on rhodium catalysts.

Alumina-supported catalysts from various sources and with different rhodium dispersions predominantly yield the hydrogenation product perhydro-1-indanol in the liquid-phase hydrogenation of 1-indanol, while carbonsupported catalysts mainly give the C-O bond scission - hydrogenation product perhydroindane. Addition of organic or inorganic bases to the reaction mixture suppresses C-O bond scission. To distinguish between the two possible pathways for C-O bond scission of direct hydrogenolysis or dehydration followed by hydrogenation, deuteration studies have been carried out with carbon-supported catalysts. Not only 1-indanol but also indane and indene (the two possible mechanistic intermediates in the C-O bond scission routes) were deuterated. Information about the actual pathway has been obtained by determining the degree of deuteration and the positions at which deuterium is incorporated in the resulting perhydroindane product by means of mass spectrometry and 13C NMR spectroscopy. The results prove that C-O bond scission takes place primarily through the direct hydrogenolysis pathway on the carbon-supported catalysts. Direct hydrogenolysis occurs on the carbon support because of the formation of a better leaving group (OH2+) from the benzylic hydroxy group and its subsequent substitution by spillover hydrogen.

Benzoannelated cis,cis,cis,trans-[5.5.5.6]fenestranes: syntheses, base lability, and flattened molecular structure of strained epimers of the all-cis series.

Tribenzofenestranes possessing the strained cis,cis,cis,trans-[5.5.5.6]-fenestrane skeleton have been synthesized from cis-2,6-diphenylspiro[cyclohexane-1,2'-indane]-1',3'-diols by two-fold cyclodehydration, in striking analogy to the strategy used previously to construct the stereoisomeric all-cis-tribenzo[5.5.5.6]fenestranes from the corresponding trans-diphenylspirodiols. In this manner, both of the parent hydrocarbons, all-cis-tribenzo[5.5.5.6]fenestrane 3 and cis,cis,cis,trans-tribenzo[5.5.5.6]fenestrane 4, have been made accessible from the spirodiketones 5 and 6, respectively. The C6-functionalized derivatives of 4-cis,cis,cis,trans-fenestranol 9 and cis,cis,cis,trans-fenestranone 12-were prepared through cis-diphenylspirotriol 8 and cis-diphenyldispiroacetaldiol 11, by using the same strategy. The cis,cis,cis,trans-[5.5.5.6]fenestrane framework readily epimerizes to the more stable all-cis isomers under basic conditions, but is stable under neutral or acidic conditions. For example, cis,cis,cis,trans-fenestranone 12 yielded all-cis fenestrane 3 under Wolff-Kishner conditions, but cis,cis,cis,trans-isomer 4 under Clemmensen conditions. Epimerization was also circumvented by radical-induced desulfurization of fenestrane dithiolane 15 with nBu3SnH/AIBN, producing 4 in excellent yields. A single-crystal X-ray structure analysis of 4 revealed that, in accordance with force field and semi-empirical MO calculations, the extra strain of the benzoannelated cis,cis,cis,trans-[5.5.5.6]fenestratriene framework [Estrain(4)-Estrain(3)=46 kJmol(-1)] is due both to the almost perfect boat conformation of the six-membered ring and to considerable bond angle widening at the central, non-bridged C4b-C15d-C11b unit (121 degrees). H/D exchange experiments with the cis, cis, cis, trans hydrocarbon 4 under basic conditions demonstrated that the strain-induced epimerization to 3 occurs through direct deprotonation of the "epimeric" benzylic bridgehead C7a-H bond, which was found to be more acidic than the two C-H bonds at the benzhydrylic bridgeheads.

Stereocontrolled synthesis of angularly fused tricyclic ring systems by means of 1-metalla-1,3,5-hexatrienes (M=Cr, W).

An efficient pathway for the stereocontrolled synthesis of functionalized, angularly fused tricyclic ring systems from readily available (1-alkynyl)carbene complexes [(OC)(5)M=C(OEt)C(triple bond)CR] (M=Cr, W; R=Ph, c-C(6)H(9)) is described. The synthesis involves the formation of a 1-metalla-1,3,5-hexatriene from the (1-alkynyl)carbene tungsten complex [(OC)(5)W=C(OEt)C(triple bond)C-c-C(6)H(9)] and a secondary amine, and its thermally induced pi-cyclization to a tetrahydroindene, which undergoes a spontaneous isomerization to another tetrahydroindene. Condensation of these tetrahydroindenes with pyran-2-ylidene complexes derived from (1-alkynyl)carbene complexes [(OC)(5)M=C(OEt)C(triple bond)CPh] (M=Cr, W) proceeds smoothly giving angularly fused tricyclic ring systems, rearrangement of which may generate spiro(cyclopentane-1,1-indanes) as side products. The synthesis is highly versatile and can be applied to the formation of various ring systems, such as steroid-type ring skeletons.

Metalloporphyrin-mediated asymmetric nitrogen-atom transfer to hydrocarbons: aziridination of alkenes and amidation of saturated C-H bonds catalyzed by chiral ruthenium and manganese porphyrins.

Chiral metalloporphyrins [Mn(Por*)(OH)(MeOH)] (1) and [Ru(Por*)(CO)(EtOH)] (2) catalyze asymmetric aziridination of aromatic alkenes and asymmetric amidation of benzylic hydrocarbons to give moderate enantiomeric excesses. The mass balance in these nitrogen-atom-transfer processes has been examined. With PhI=NTs as the nitrogen source, the aziridination of styrenes, trans-stilbene, 2-vinylnaphthalene, indene, and 2,2-dimethylchromene catalyzed by complex 1 or 2 resulted in up to 99 % substrate conversions and up to 94 % aziridine selectivities, whereas the amidation of ethylbenzenes, indan, tetralin, 1-, and 2-ethylnaphthalene catalyzed by complex 2 led to substrate conversions of up to 32 % and amide selectivities of up to 91 %. Complex 1 or 2 can also catalyze the asymmetric amidation of 4-methoxyethylbenzene, tetralin, and 2-ethylnaphthalene with "PhI(OAc)(2) + NH(2)SO(2)Me", affording the N-substituted methanesulfonamides in up to 56 % ee with substrate conversions of up to 34 % and amide selectivities of up to 92 %. Extension of the "complex 1 + PhI=NTs" or "complex 1 + PhI(OAc)(2) + NH(2)R (R=Ts, Ns)" amidation protocol to a steroid resulted in diastereoselective amidation of cholesteryl acetate at the allylic C-H bonds at C-7 with substrate conversions of up to 49 % and amide selectivities of up to 90 % (alpha:beta ratio: up to 4.2:1). An aziridination- and amidation-active chiral bis(tosylimido)ruthenium(VI) porphyrin, [Ru(Por*)(NTs)(2)] (3), and a ruthenium porphyrin aziridine adduct, [Ru(Por*)(CO)(TsAz)] (4, TsAz=N-tosyl-2- (4-chlorophenyl)aziridine), have been isolated from the reaction of 2 with PhI=NTs and N-tosyl-2-(4-chlorophenyl)aziridine, respectively. The imidoruthenium porphyrin 3 could be an active species in the aziridination or amidation catalyzed by complex 2 described above. The second-order rate constants for the reactions of 3 with styrenes, 2-vinylnaphthalene, indene, ethylbenzenes, and 2-ethylnaphthalene range from 3.7-42.5x10(-3) dm(3) mol(-1) s(-1). An X-ray structure determination of complex 4 reveals an O- rather than N-coordination of the aziridine axial ligand. The fact that the N-tosylaziridine in 4 does not adopt an N-coordination mode disfavors a concerted pathway in the aziridination by a tosylimido ruthenium porphyrin active species.

Expanded radialenes with bicyclo[4.3.1]decatriene units: new precursors to cyclo[n]carbons.

A new method for the formation of conjugated polyynes has been developed based on both the rearrangement of vinylidenes to alkynes and the [2+1] cheletropic fragmentation of dialkynylmethylenebicyclo[4.3.1]deca-1,3,5-triene derivatives. A model study of the photolysis of simple dialkynylmethylenebicyclo[4.3.1]deca-1,3,5-trienes resulted in cheletropic fragmentation followed by 1,2-migration to give the corresponding linear polyynes, although undesired isomerization to methylenebicyclo[5.3.0]triene derivatives took place concurrently. Expanded [3]-, [4]-, [5]-, and [6]radialene derivatives with exocyclic bicyclo[4.3.1]decatriene units were prepared by oxidative coupling of the monomeric units as precursors to the corresponding cyclo[n]carbons, monocyclic forms of carbon clusters. The spectroscopic properties of the expanded radialenes were investigated in connection with cross conjugation of the core pi system and with its perturbation by the extraannular bicyclic pi system. In negative-mode laser-desorption time-of-flight (LD-TOF) mass spectra, the expanded radialenes exhibited peaks due to the corresponding cyclo[n]carbon anions (n = 18, 24, 30, and 36) formed by the stepwise loss of the aromatic indane fragments.

Size-selective formation of C78 fullerene from a three-dimensional polyyne precursor.

Multicyclic cagelike cyclophanes 2 a and 2 b containing cyclobutene rings have been prepared as precursors of three-dimensional polyynes C78H18 (1a) and C78H12Cl6 (1b), respectively. Laser irradiation of 2a and 2b induced expulsion of the aromatic fragment, indane, to give the three-dimensional polyyne anions C78H18- and C78H12Cl6-, respectively. Whereas the former anion lost only four hydrogen atoms to form C78H14-, complete loss of all hydrogen and chlorine atoms was observed from the latter anion, to yield a C78- ion that has a fullerene structure which was proven by its characteristic fragmentation pattern.

Solid-supported [2+2+2] cyclotrimerizations.

The transition-metal-catalyzed [2+2+2] cyclotrimerization of a diyne and an alkyne provides a convergent route to highly-substituted aromatic rings. This reaction possesses distinct drawbacks, especially low chemo- and regioselectivities, which hamper its application in combinatorial synthesis. These problems have been solved by the development of solid-supported [2+2+2]-cycloaddition reactions. If conducted on a solid-support, this reaction enables rapid combinatorial access to diverse sets of carbo- and heterocyclic small-molecule arrays. The scope of this methodology has been investigated by examining different immobilization strategies, different diyne precursors, and a variety of functionalized alkyne reaction partners. Overall, isoindoline, phthalan, and indan libraries were assembled in good to excellent yields and with high purities.

Pharmacokinetics of the anticancer agent sulofenur in mice, rats, monkeys, and dogs.

The absorption and pharmacokinetics of sulofenur [N-(indan-5-sulfonyl)-N'-(4-chlorophenyl)urea, LY186641] and its major metabolites were examined in mice, rats, monkeys, and dogs. The compound is a diarylsulfonylurea currently being evaluated as an oncolytic agent in phase I and II trials. In all species, sulofenur was well absorbed after an oral dose, but over a prolonged period, and sulofenur exhibited a fairly long half-life of elimination from plasma. These values ranged from 6 h in rats up to 30, 110, and 200 h in mice, monkeys, and dogs, respectively, at doses (240-1000 mg/m2) within the range of those used in clinical trials. Experiments describing the high degree of binding of sulofenur to plasma proteins (consistently > 99%) help to explain these relatively long half-lives. There is, however, a large difference between these plasma half-lives in the species studied. Sulofenur was previously found to be extensively metabolized to products that are excreted primarily into the urine. In this study, its major metabolites, which are found mainly in the urine, were also minor components of the drug-related material (< 10% of the sulofenur concentrations) in the plasma of rats treated with sulofenur. The absorption, binding characteristics, and elimination of these major metabolites after their administration to rats were also compared with sulofenur.(ABSTRACT TRUNCATED AT 250 WORDS)

Stereoinversions using microbial redox-reactions.

This paper aims to provide a summary of the recent literature on the use of redox enzymes to carry out stereoinversion reactions on chiral secondary alcohols. Emphasis has been placed on biotransformations which result in the deracemization of a racemic substrate to give high value synthetic intermediates in a theoretical 100% yield. Most of the biocatalysts which are competent to carry out such transformations are whole cell systems, which contain the necessary cofactor recycling machinery to facilitate this otherwise entropically disfavoured process. The first section deals with deracemization of compounds such as mandelic acid and pantoyl lactone using two microorganisms which display enantiocomplementary stereospecificity. The deracemization of chiral alcohols such as beta-hydroxyesters, aryl ethanols and terminal 1,2-diols with single microorganisms will then be discussed and the influence of growth and reaction conditions on the selectivity observed will be emphasised. Then the ability of several microorganisms to deracemize by double stereoinversion substrates with two stereocentres such as cyclohexan-1,2-diol, cis and trans indan-1,2-diol and pentan-2,4-diol will be presented and some mechanistic rationale proposed. Lastly enzymes known as epimerases which are important in sugar and deoxysugar biosynthesis will be discussed with reference to some recent work on the mechanism of UDP-glucose epimerase.

Beta 2-adrenoceptor-mediated positive inotropic effect of adrenaline in human ventricular myocardium. Quantitative discrepancies with binding and adenylate cyclase stimulation.

Experiments were designed to unravel the relative contribution of beta 1- and beta 2-adrenoceptors to the positive inotropic effects of adrenaline and noradrenaline in isolated tissues of left ventricular myocardium of man. We also analyzed relationships between the fractions of human left ventricular beta 1- and beta 2-adrenoceptors, estimated from binding assays, and stimulation of adenylate cyclase and contractile force by adrenaline and noradrenaline. Selective blockade of beta 2-adrenoceptors by erythro-(+/-)-(alpha-methyl-indan-4-yloxy)-3-isopropylaminobuta n-2-ol (ICI 118,551) attenuated the increase of contractile force caused by adrenaline but not by noradrenaline, suggesting some involvement of beta 2-adrenoceptors. Selective blockade of beta 2-adrenoceptors without affecting beta 1-adrenoceptors still enabled both adrenaline and noradrenaline to cause maximum possible increases of contractile force through beta 1-adrenoceptors. A direct involvement of beta 2-adrenoceptors became manifest by selectively antagonizing beta 1-adrenoceptors by 1-[2[3-carbamoyl-4-hydroxy)phenoxy)ethylamino]- 3-[4(1-methyl-4-trifluoromethyl-2-imidazolyl)phenoxy]-2-propanol (CGP 20712 A) without affecting beta 2-adrenoceptor. beta 2-adrenoceptors can mediate half of the maximum increase of contractile force elicited by low concentrations of adrenaline and also contribute to the increase of contractile force caused by high concentrations of noradrenaline. beta-adrenoceptors were labelled in membrane particles with 3H-(-)-bupranolol in the absence (beta 1 & beta 2) and presence of 500 nmol/l CGP 20712 A (beta 2). 71% of the beta-adrenoceptors were beta 1 and 29% beta 2. Binding inhibition experiments with CGP 20712 A and ICI 118,551 yielded 74% beta 1 and 26% beta 2.(ABSTRACT TRUNCATED AT 250 WORDS)

Studies on the synthesis, characterisation and antimicrobial activity of new Co(II), Ni(II) and Zn(II) complexes of Schiff base derived from ninhydrin and glycine.

For the first time, Co(II), Ni(II) and Zn(II) complexes have been synthesized involving an intermediate Schiff base, indane-1,3-dione-2-imine-N-acetic acid the condensed product of ninhydrin and glycine. These coloured complexes were characterised by elemental analysis, molar conductivity, thermogravimetric analyses/differential thermal analysis, infrared, magnetic susceptibility, NMR and electronic spectral studies. Mechanisms for their formation have been proposed. The experimental studies reveal that the complexes possess octahedral stereochemistry whereas the Schiff base behaves as a monobasic tridentate ligand. A molecular structure for the metal complexes is also proposed. A comparative study of the antimicrobial activity of ninhydrin and the corresponding metal complexes against Escherichia coli, Proteus mirabilis, Staphylococcus aureus and Streptococcus faecalis has been undertaken and the results are discussed.

A simple, rapid and simultaneous analysis of complex volatile hydrocarbon mixtures in blood using gas chromatography/mass spectrometry with a wide-bore capillary column.

A screening method for detecting volatile hydrocarbons in blood has been developed using gas chromatography/mass spectrometry with a wide-bore capillary column and a headspace method. Toluene-d8 and indan were used as the internal standards for quantitative analysis. Hydrocarbons with retention indices from 600 to 1200 were simultaneously and quantitatively detected in relatively low concentrations (0.01 microgram/ml) in reconstructed ion chromatography. This method could prove useful in forensic cases in which urgent examination of complex hydrocarbon mixtures, e.g. petroleum components, is required.

The Indian picture puzzle test - a developmental test designed and standardised for Indian children.

OBJECTIVE: To construct a test used by community pediatricians and other professionals in the UK as a screening test for verbal and non-verbal development in children aged 2 to 4 years of age. METHODS: A multifaceted developmental test of cognitive skills was constructed, modeled on the Bus Puzde Test (Egan 1984) for its case of administration and appeal. Each stage in the design was piloted in Rajasthan in all socioeconomic groups. Stages include simple ethnic modification of the original test, development of more socioculturally appropriate scenes, a detailed statistical procedure of item analysis and reliability studies. The picture was converted into a wooden insert puzzle, called The Indan Picture Puzzle Test (IPPT) and standardized on a random sample of 616 children to construct the norms. RESULT: The IPPT assesses aspects of early language, picture interpretation, performance skills and conceptual development in children aged 2 to 5 years. Analysis of the standardized data highlighted the need for separate norms for each socioeconomic group. CONCLUSION: Verbal abilities were significantly different between advantaged and disadvantaged (slum and rural) groups though performance skills were comparable.

Labelling of recombinant human and native rat serotonin 5-HT1A receptors by a novel, selective radioligand, [3H]-S 15535: definition of its binding profile using agonists, antagonists and inverse agonists.

The novel benzodioxopiperazine, 5-HT1A receptor weak partial agonist, S 15535 (4-(benzodioxan-5-yl)1-(indan-2-yl)piperazine) bound with high affinity and selectivity to membranes of Chinese Hamster Ovary cells stably expressing the human (h) 5-HT1A receptor (Ki = 0.6 nM versus [3H]-8-hydroxy-dipropylamino-tetralin, [3H]-8-OH-DPAT): its affinity at h5-HT1A receptors was more than 70-fold higher than its affinity at > 50 other binding sites. S 15535 was tritiated to high specific activity (50 Ci/mmol) and its binding profile characterised. At 22 degrees C, [3H]-S 15535 associated and dissociated from h5-HT1A receptors with half-times of 2.9 and 5.0 min, respectively, yielding a Kd estimate of 3.6 nM. In saturation binding experiments, [3H]-S 15535 displayed a Bmax value for h5-HT1A receptors (1630 fmol/mg), higher than that obtained with the agonist [3H]-8-OH-DPAT (1023 pmol/mg). Guanylyl imidodiphosphate (GppNHp, 100 microM) reduced the binding of [3H]-S 15535 by only 25% compared with 79% for [3H]-8-OH-DPAT at h5-HT1A receptors. [3H]-S 15535 also showed high affinity, saturable binding to rat hippocampal membranes (Bmax = 820 fmol/mg versus 647 fmol/mg for [3H]-8-OH-DPAT). For both h5-HT1A and rat 5-HT1A receptors, the Ki values for competition binding of 15 serotonergic ligands with [3H]-S 15535 was highly correlated with that of [3H]-8-OH-DPAT. However, important differences were also observed. The agonist, 5-hydroxytryptamine (5-HT), displayed biphasic competition curves with [3H]-S 15535 but not with [3H]-8-OH-DPAT at h5-HT1A receptors. Similarly, the 'antagonists', spiperone, methiothepin and (+)butaclamol, showed biphasic competition isotherms versus [3H]-S 15535 but not [3H]-8-OH-DPAT. When [3H]-S 15535 competition binding experiments were carried out in the presence of GppNHp (100 microM) the 5-HT and 8-OH-DPAT competition curves shifted to the right, whereas the spiperone and methiothepin competition curves shifted to the left. In contrast, in the presence of GppNHp, the competition isotherms for N-[2-[4-(2-methoxyphenyl)-1-piperazinyl]ethyl]-N-(2-pyridinyl)cyclo-h exanecarboxamide (WAY 100,635) were not altered. Taken together, these data show that (i) [3H]-S 15535 is a highly selective 5-HT1A receptor ligand which labels both G-protein-coupled and uncoupled 5-HT1A receptors, (ii) antagonists, such as WAY 100,635, which yield monophasic isotherms in competition with both [3H]-agonists and [3H]-antagonists, are not sensitive to the G-protein coupling state of the receptor, but (iii) spiperone and methiothepin behaved as inverse agonists, their competition isotherms with [3H]-S 15535 being modulated in an opposite manner to those of agonists.
