Evidence that Bacillus subtilis bacteriophage SP02 is temperate and heteroimmune to bacteriophage phi-105.

Some characteristics of Bacillus subtilis phage SPO2 which show that it is a temperate phage are presented. Wild-type SPO2 forms turbid plaques, similar to those of other temperate phages. SPO2 lysogenic strains which are resistant to SPO2 can be isolated; these strains remain stable lysogens despite the fact that they can no longer adsorb SPO2. SPO2 lysogenic strains can be grown for many generations in SPO2 antiserum and remain lysogenic. Phage SPO2 plates on phi105 lysogens and phage phi105 plates on SPO2 lysogens; this indicates that SPO2 and phi105 are heteroimmune. Phage phi105 plates on an SPO2-resistant strain; this indicates that SPO2 and phi105 adsorb to different receptor sites on the bacterial surface.

Distinct slime polysaccharide depolymerases of bacteriophage-infected Pseudomonas aeruginosa: evidence of close association with the structured bacteriophage particle.

Five new polysaccharide depolymerases were isolated from cultures of Pseudomonas aeruginosa infected with phages 6, 7, 8, 9, and 10. The production of enzyme paralleled the release of phage. Depolymerase associated with phage 8 was active on slime polysaccharide A, whereas depolymerases associated with phages 6, 7, 9, and 10, like pseudomonas phage 2, hydrolyzed slime polysaccharide B. None of the depolymerases was active on slime polysaccharide C. Despite exhaustive purification, depolymerase activity was found to band with the phage particles at a density of 1.49 to 1.51 g/ml in a density gradient composed to cesium chloride. These results suggest that the depolymerases are firmly bound to the phage particles.

Initiation by bacteriophage T1 of DNA packaging at a site between the P and Q genes of bacteriophage lambda.

The growth of phage T1 on cells tandemly lysogenic for heteroimmune lambdoid prophages leads to a nonrandom packaging of lambda DNA by T1. A site, called esp-lambda, is located between the P and Q genes of lambda and results in increased packaging to the left by T1. When cloned into pBR322, the esp-lambda site causes a significant increase in transduction of the plasmid by T1. The nin5 deletion inactivates esp-lambda.

Lack of nucleotide sequence relationship among the temperate bacteriophage SPO2, Bacillus subtilis, and virulent bacteriophages beta 3 and beta 22.

Radiolabeled phage SPO2 fragments were tested for the abiity to form interspecies deoxyribonucleic acid (DNA) duplexes with DNA from Bacillus subtilis and from phages beta3 and beta22. No reassociation, above control values, occurred between the DNA of phage SPO2 and that of its host or either of the virulent B. subtilis phages. It appears that extensive nucleotide sequence similarities between portions of host and phage DNA species are not necessary for the formation of the lysogenic state. The thermal elution profile of reassociated SPO2 DNA exhibited a normal distribution, indicating no heterogeneity in base composition. This differs from temperate enterophages whose DNA molecules contain regions of differing base composition.

Gamma-T4 hybrid bacteriophage carrying the thymidine kinase gene of bacteriophage T4.

Among 32 lambda-T4 recombinant phages selected for growth on a thymidylate synthetase-deficient (thyA) host, 2 were shown to carry the T4 thymidine kinase (tk) gene. The lambda-T4tk phages contain two T4 HindIII DNA fragments (2.0 and 1.5 kilobases) that hybridize to restriction fragments of T4 DNA, encompassing the tk locus at 60 kilobases on the T4 map. The T4tk insert compensates for the simultaneous host deficiencies of thymidine kinase and thymidylate synthetase in a thymidine kinase-deficient (tdk) host growing in the presence of fluorodeoxyuridine when provided with thymidine and uridine. The lambda-T4tk hybrid phages specified five polypeptides with Mrs of 22,000 (22K), 21K, 14K, 11K, and 9K.

Low-frequency rescue of a genetic marker in deoxyribonucleic acid from Bacillus bacteriophage phi 105 by superinfecting bacteriophage.

Markers in gene L, which maps at the right end of the vegetative and prophage maps, are rescued at a strongly reduced frequency from mature phi105 deoxyribonucleic acid (DNA) by superinfecting phage but at high frequency from vegetative and prophage DNA. It is suggested that the ends of mature DNA are degraded when DNA is taken up by competent cells.
