[Effect of tuftsin and hydroxymethacil on tyrosine hydrolase of macrophages in chronic stress].

It has been established that rat peritoneal macrophages possess tyrosine hydroxylase activity which has been characterized by KM4, 2 microM, Vmax 30 nmole/min per mg of protein. After 15 days of electronociseptive stimulation almost all tyrosine hydroxylase activity has been established in the form which has tyrosine KM47.0 M and Vmax 18.6 nmole/min. per mg of protein. Immunostimulator hydroxymethacil++, at its intraperitoneal injections to stressed rats during 7 days induced the appearance of low-affinity form of tyrosine hydroxylase with KM270 microM and Vmax 27.8 nmole/min.per mg of protein. The same low affinity form of the enzyme has been established after injections of tuftsin which possesses immunostimulating properties.

[The effect of tuftsin on various aspects of metabolism in the sensorimotor system of the animal brain].

Rates of synthesis and degradation of monoamines, acetyl choline and protein were distinctly altered in cortex-subcortex structures of the brain locomotory system after a single administration of tetrapeptide tuftsin Thr-Lys-Pro-Arg during the period 15-30 min of its pronounced effect on the animal motor activity. Correlation between the functional and morphological alterations in cellular and subcellular brain structures, caused by the peptide administration, is discussed.

The analgesic activity of some kyotorphin-tuftsin analogs.

The analgesic potency of the C-terminal dipeptide of tuftsin (Pro-Arg) was found to be much stronger than that of the C-terminal of bradykinin dipeptide Phe-Arg. It was also found that the tripeptide fragment of bradykinin (Pro-Phe-Arg) and its D-Pro analog (D-Pro-Phe-Arg) are deprived of analgesic activity. The attempt to synthesize kyotorphin analogs containing fluorescence label (DMAPhe-Arg and D-DMAPhe-Arg; DMAPhe: 4'-dimethylamino-phenylalanine) resulted in inactive compounds.

[Effects of heptapeptide of the tuftsin group with nootropic components of action and piracetam on formation of escape reaction in normal conditions and in conflict situation].

In this work we have compared the effect of Tuftsin Group synthetic heptapeptide (TP-1) and Piracetam on learning of avoidance response and its restoring after disturbance provoked by unexpected changes of previously established cause-effect relations in experimental environment. We have pointed out that both of the drugs improve learning but the dynamics is different, namely: Piracetam causes the greatest improvement on the 1st day, meanwhile the Heptapeptide--on the 4th day. Both of them preclude from disturbing avoidance response and this protective action of peptide is more effective.

[Effects of femto- and picomolar concentrations of thyroliberin and tuftsin on the contractile activity of lymphatic vessels of the rat mesentery].

Thyrotropin-releasing-hormone (TRH) in concentration from I g/ml to 1.10(-10) micrograms/ml and tuftsin (T) (10 micrograms/ml to 1.10(-6) micrograms/ml) biomicroscopy were used in rats to show the ability to activate the contraction of lymphatic microvascular walls. TRH in used concentrations stimulated spontaneously contractility of lymphatic vessel's walls or evoked its contractility when it was absent. T stopped the contraction of lymphatic microvascular walls and caused the reaction of vasodilatation.

Tuftsin a natural peptide with antiviral activity--structural basis of its action.

The influence of tuftsin--the natural phagocytosis stimulating tetrapeptide and its elongated analogues--on the mortality of encephalomyocarditis virus-infected NMRI mice was investigated. It seems that amino acids elongation in the parent peptide chain does not result in significant increase of antiviral activity of the compounds tested.

[Temporal characteristics of the behavioral effects of tuftsin].

Administration of endogenous immunostimulator, tuftsin, (0.3 mg/kg, i/p) in rats produced biphase behavioral response. The first phase lasted for about 30 min and was characterized by the enhancement of orientation behavior and suppression of startle reaction. During the second phase the orientation behavior was depressed and passive avoidance reaction intensified. The second phase lasted for at least 24 hours.

Migration enhancement by tuftsin of human mononuclear cells and its effect on the migration inhibition factor test with tumor antigens.

Tuftsin, a physiological tetrapeptide which stimulates the phagocytic activity of polymorphonuclear neutrophils and macrophages, is found to enhance the migration of normal and sensitized human mononuclear cells. Furthermore, it abrogates the migration inhibition effect of human malignant melanoma antigen. This finding may represent a possible explanation for the unexpected migration enhancement often observed when the migration inhibition test is employed.

Biological aspects of tuftsin activity.

Radioisotopic methods for determination of the biological activity of tuftsin was applied to study the stimulatory effect of this compound as well as of its components. This effect appeared to be dependent on the presence of two amino acids - Proline and Arginine. Administered jointly, they exert much stronger effect than separately.

[Tuftsin-like peptides from a C-reactive protein molecule as regulators of lymphocyte proliferation].

Influence of synthetic peptides from human C-reactive protein (CRP) on proliferation of intact and mitogen stimulated peripheral blood lymphocytes was studied. Effect of the tetra-peptides from CRP was similar to that of tuftsin Thr-Lys-Pro-Arg. All the peptides studied did not increase the 3H-thymidine incorporation into the resting lymphocytes but accelerated it after mitogen stimulation. The peptides efficiency depended on T- or B-specificity of mitogens.

[The anxiolytic action of an analog of the endogenous peptide tuftsin on inbred mice with different phenotypes of the emotional stress reaction].

The anxiolytic effect of the synthetic analogue of the endogenous peptide tuftsin (Thr-Lys-Pro-Arg-Pro-Gly-Pro) was studied in inbred mice Balb/c (C) and C57B1/6 (B6) with different heritable types of emotional-stress reaction. In C strain with genetically high level of anxiety and "passive" type of behaviour under conditions of avoidable and unavoidable stress (open field, elevated plus-maze, light-dark chamber) the heptapeptide prevented manifestations of anxiety being administered intraperitoneally in a wide range of active doses (200-3000 mcg/kg). The drug did not change the behaviour of B6 mice with low level of anxiety and active type of emotional-stress reaction. The described effect is comparable with tranquilizing-activatory action of low doses of benzodiazepine tranquillizers, however, in contrast to the latter, the heptapeptide has no side inhibitory behavioural effects even in high doses. The heptapeptide may be considered as a promising new anxioselective drug without side inhibitory behavioural effects which are typical for benzodiazepine tranquillizers.

[The characteristics of the effect of tuftsin on the behavior and on the level of biogenic amines in the brain of rats with differing resistance to acoustic stress].

The influence of tetrapeptide tuftsin (Tyr-Lys-Pro-Arg) on learning, exploratory activity, emotional behavior, and hypothalamic monoamine content was studied in Wistar rats with different resistance to stress induced by acoustic stimuli. Positive effects of taftsin were more pronounced in low-resistant rats. Administration of taftsin induced in these animals a significant increase in reactivity to stimuli of different modalities, the open-field exploratory activity, rate of alimentary conditioning and its modification in emotionally negative situation. Biochemical examinations showed that in rats with high resistance to stress taftsin administration led to a decrease in hypothalamic noradrenaline level and increase in dopamine and serotonin levels. On the contrary, in low-resistant animals taftsin increased the level of noradrenaline and decreased that of dopamine, serotonin, and 5-hydroxyindoleacetic acid. It is suggested that different behavioral effects of taftsin in stress-resistant and nonresistant rats are caused by its different influence on hypothalamic biogenic amines.
