Occupational asthma due to inhalation of chloramine-T. II. Demonstration of specific IgE antibodies.

In the sera of patients who developed asthmatic symptoms after exposure to chloramine-T, specific IgE antibodies are demonstrated by a polystyrene tube radioimmunoassay. These antibodies are directed against human serum albumin treated with chloramine-T. Specific antibodies of subclasses IgG1, IgG3 and IgG4 were not found. Histamine release in response to chloramine-T-treated human serum albumin was demonstrated using the peripheral blood leukocytes of one of the patients. To define the antigenic determinant to which the specific IgE antibodies are directed, radioimmunoassay and radioimmunoassay-inhibition studies were performed using several autologous and heterologous chloramine-T-treated proteins. Moreover, the inhibition capacities of chloramine-T and structurally related compounds were also determined. The results of these experiments indicate that the antigenic determinant is formed, at least in part, by the p-toluenesulfonyl group of the chloramine-T molecule.

Crystal structure of 2,5-anhydro-1-O-(p-tolylsulfonyl)-D-mannitol.

2,5-Anhydro-1-O-(p-tolylsulfonyl)-D-mannitol, C13H18SO7, Mr = 318.4, monoclinic, C2, a = 26.370(6), b = 7.9741(11), c = 6.6801(6) A, beta = 91.401(11) degrees, V = 1404.3(6) A3, Z = 4, DX = 1.506 g/cm3, CuK alpha, lambda = 1.54184 A, mu = 23.03 cm-1, F(000) = 672, T = 296(1) K, R = 0.042 for 2832 observations with I > 3 sigma (I) (of 2864 unique data). On the esterified side of the molecule, three bond lengths and three bond angles show small changes compared to the unesterified side, which is similar to the symmetrical parent compound, 2,5-anhydro-D-mannitol. The conformation of the five-membered ring is E5 with P = 49.3 degrees and tau m = 38.1 degrees. The hydroxymethyl groups adopt g+ and g- dispositions similar to the parent molecule. The three hydroxyl groups are involved in a network of intermolecular hydrogen bounds both as donors and acceptors.

Trichomes of tobacco excrete zinc as zinc-substituted calcium carbonate and other zinc-containing compounds.

Tobacco (Nicotiana tabacum L. cv Xanthi) plants were exposed to toxic levels of zinc (Zn). Zn exposure resulted in toxicity signs in plants, and these damages were partly reduced by a calcium (Ca) supplement. Confocal imaging of intracellular Zn using Zinquin showed that Zn was preferentially accumulated in trichomes. Exposure to Zn and Zn + Ca increased the trichome density and induced the production of Ca/Zn mineral grains on the head cells of trichomes. These grains were aggregates of submicrometer-sized crystals and poorly crystalline material and contained Ca as major element, along with subordinate amounts of Zn, manganese, potassium, chlorine, phosphorus, silicon, and magnesium. Micro x-ray diffraction revealed that the large majority of the grains were composed essentially of metal-substituted calcite (CaCO3). CaCO3 polymorphs (aragonite and vaterite) and CaC2O4 (Ca oxalate) mono- and dihydrate also were identified, either as an admixture to calcite or in separate grains. Some grains did not diffract, although they contained Ca, suggesting the presence of amorphous form of Ca. The presence of Zn-substituted calcite was confirmed by Zn K-edge micro-extended x-ray absorption fine structure spectroscopy. Zn bound to organic compounds and Zn-containing silica and phosphate were also identified by this technique. The proportion of Zn-substituted calcite relative to the other species increased with Ca exposure. The production of Zn-containing biogenic calcite and other Zn compounds through the trichomes is a novel mechanism involved in Zn detoxification. This study illustrates the potential of laterally resolved x-ray synchrotron radiation techniques to study biomineralization and metal homeostasis processes in plants.

Responses of isolated human internal anal sphincter to drugs and electrical field stimulation.

The effects of drugs and electrical field stimulation on muscle strips from the human internal anal sphincter have been examined to provide information about the receptors and nerves that might be involved in the relaxation of the muscle in vivo. Acetylcholine and bethanechol usually relaxed muscle strips; this effect was abolished by hyoscine and antagonized to a varying degree by tetrodotoxin. Hexamethonium in concentrations sufficient to block relaxations to 1,1-dimethyl-4-phenylpiperazinium iodide or nicotine had no effect on relaxations due to acetylcholine, thereby indicating that acetylcholine was acting on muscarinic receptors. The nerves stimulated by acetylcholine released an unknown transmitter. Both 1,1-dimethyl-4-phenylpiperazinium iodide and nicotine relaxed muscle strips, possibly by releasing an adrenergic neurotransmitter which, because the responses to nicotinic receptor-stimulation were blocked by propranolol, stimulated beta-adrenergic inhibitory receptors. Sphincter muscle was also relaxed by electrical field stimulation of intrinsic nerves; this response was blocked by tetrodotoxin but unaffected by hexamethonium, hyposcine, or propranolol. The nerves responding to electrical field stimulation were therefore post-ganglionic, noncholinergic, and nonadrenergic. Compounds discounted as possible neurotransmitters of the noncholinergic, nonadrenergic inhibitory nerves were prostaglandin E2 and F2 alpha, histamine, 5-hydroxytryptamine, and dopamine. Some evidence allows vasoactive intestinal peptide and adenosine triphosphate to be considered as possible neurotransmitters; this could not be confirmed because selective antagonists are not yet available.

Dual antagonists of the bradykinin B1 and B2 receptors based on a postulated common pharmacophore from existing non-peptide antagonists.

We have recently drawn attention to the fact that most non-peptide antagonists of the kinin B1 receptor reported so far are structurally related, possessing the core motif phenyl-SO2-NR-(spacer(2-4))-CO-NRR. This is found in compound A (N-[2-[4-(4,5-dihydro-1H-imidazol-2- yl)phenyl]ethyl] - 2- [(2R)-1-(2-napthylsulfonyl)-3-oxo-1,2,3,4-tetrahydroquinoxalin-2-yl]acetamide), a very potent and selective B1 receptor antagonist. A subset of specific bradykinin B2 receptor antagonists (LF16-0687, bradyzide and derivatives) possesses a similar 'scaffold' (phenyl-SO2-NR-CRR-CO-NRR). We investigated whether simple molecules mimicking the postulated pharmacophores could be identified in two public chemical databases. Receptor binding to B1 and B2 receptors expressed by rabbit cultured smooth-muscle cells was confirmed for some of these newly identified agents, with a loss of receptor subtype selectivity. For instance, compound 3[2-(3-oxo-1-(toluene-4-sulfonyl)-1,2,3,4-4H-quinoxalin-2-yl)-N-phenyl-acetamide] exhibits IC50 values of 2.13 and 126 microM in the radioligand competition assays for B1 and B2 receptors, respectively, and a pA2 of 6.27 at the rabbit B1 receptor in a functional test (Lys-des-Arg9-bradykinin-induced contractility of the isolated aorta). Compound 5 (a close analog of compound 3) is a more balanced dual antagonist of low potency (IC50 values of 30 and 117 microM, respectively). As predicted, compounds modeled on a postulated pharmacophore common to some non-peptide B1 or B2 receptor antagonists exhibit measurable binding with decreased receptor subtype selectivity. Dual B1/B(2) receptor antagonists are of possible therapeutic interest and should be developed.

Inactivation modifiers of Na+ currents and the gating of rat brain Na+ channels in planar lipid membranes.

Rat brain Na+ channels whose inactivation process had been removed either by batrachotoxin (BTX) or veratridine (VT) were reconstituted into planar lipid membranes. The voltage dependence of the open probability (Po) of the channel, of the opening and closing rate constants, and the conductance and relative permeability for Na+ and K+ were studied in voltage-clamp conditions in the presence of agents known to modify the inactivation of Na+ currents. In relation to alkaloids (BTX, VT, and aconitine), it was found that once a Na+ channel was modified by BTX or VT, the addition of another alkaloid did not change further the gating and permeation properties of the channel over a period of about 1 h. Once the inactivation process of the channels is removed by BTX, the addition of a proteolytic enzyme (trypsin) or an halogenated compound (chloramine-T, CT) induced profound and specific modifications on the opening and closing events of Na+ channels: (1) the voltage dependence of the channel Po shifted to more hyperpolarized potentials; (2) this voltage shift can be explained by equal hyperpolarizing voltage shifts of the opening and closing rate constants of the channel; (3) although the gating properties of the channel were modified by these compounds, the permeation properties of the channel, as evaluated by the conductance and the selectivity to Na+ and K+ ions, were unaltered; (4) trypsin and CT were active only in the intracellular side of the channel and were irreversible within the time course of the experiments, suggesting covalent modifications of the channel. Inactivation modifiers also affected the gating of toxin-activated single Na+ channels.(ABSTRACT TRUNCATED AT 250 WORDS)

Chiral nonsteroidal affinity ligands for the androgen receptor. 1. Bicalutamide analogues bearing electrophilic groups in the B aromatic ring.

A series of chiral analogues of bicalutamide bearing electrophilic groups (isothiocyanate, N-chloroacetyl, and N-bromoacetyl) on aromatic ring B of the parent molecule were synthesized. These compounds were designed as affinity ligands for the androgen receptor (AR). We prepared the (R)- and (S)-optical isomers of these compounds as pure enantiomers. The AR binding affinities of these compounds were measured in a competitive binding assay with the radiolabeled high-affinity AR ligand, [(3)H]mibolerone. In accordance with our previous results for the enantiomers of bicalutamide, we found that all (R)-isomers demonstrated much higher binding affinity to the AR as compared to their corresponding (S)-isomers. The para-substituted affinity ligands in ring B bound the AR with higher affinities than the corresponding meta-substituted analogues. Oxidation of thioester affinity ligands to their sulfonyl analogues for the para-substituted compounds decreased AR binding affinities and similar modification increased binding affinities for corresponding meta-analogues. The least potent para-substituted sulfonyl compounds had higher AR binding affinities than the most potent meta-substituted sulfonyl compounds. Overall, the para-substituted unoxidized molecules demonstrated the highest AR binding affinity. Subsequent research using AR exchange assays and Scatchard analyses showed that the isothiocyanate affinity ligands (R)-7, (R)-9, and (R)-10 reported herein are the first specific chemoaffinity ligands for the AR.

Syntheses of functionalized 1,4-disubstituted gamma-carbolines.

We report the complete synthesis of a variety of 1,4-disubstituted gamma-carbolines. These compounds are of particular interest for their involvement in many biological processes and are believed to possess various medicinal activities. A large number of N-tosylaldimines were condensed with indoles affording an array of 3-aminomethyl indoles. Subsequent additions, followed by intramolecular cyclization, afforded an array of 1,2-dihydro-3H-gamma-carbolones in good yield. Upon subsequent aromatization, the corresponding fully aromatic functionalized 1-aryl-4-hydroxy-gamma-carbolines resulted.

Mineralization of alkylbenzenesulfonates in water by means of contact glow discharge electrolysis.

Mineralization of aqueous alkylbenzenesulfonates (ABS) was investigated by means of contact glow discharge electrolysis (CGDE). Toluenesulfonic and dodecylbenzenesulfonic acids in a neutral phosphate buffer solution were smoothly degraded and eventually converted to inorganic carbon and sulfate ion when CGDE was conducted under the applied DC voltage of 500 V and current of ca. 90 mA. As the intermediate products, some phenolic compounds were detected as well as carboxylic acids such as oxalate, formate and so on. It was demonstrated that the decay of ABS followed the first-order rate law. Based on the detailed analysis of the hydroxylation products and kinetic consideration, it was assumed that hydroxyl radicals would play a crucial role in the oxidative degradation of aqueous ABS. The features of CGDE as a tool for the removal of organic pollutants in water were discussed in comparison with other physicochemical methods.

Synthesis and conformational studies on methyl 4-O-acetyl-3-azido-2,3,6-trideoxy-hex-5-enopyranosides of the L series.

Methyl 3-azido-2,3-dideoxy-alpha-D-xylo-, -alpha-D-lyxo-, and -beta-D-xylo-hexopyranosides were converted into 4-O-acetyl-3-azido-6-iodo-2,3,6-trideoxy analogues via 6-O-p-tolylsulfonyl compounds. The elimination of hydrogen iodide from 6-iodo glycosides yielded methyl 4-O-acetyl-3-azido-2,3,6-trideoxy-beta-L-erythro-, -alpha-L-threo-, and -beta-L-threo-hex-5-enopyranosides. The configuration and conformation of all products are evaluated in depth on the basis of (1)H and (13)C NMR data. Factors determining conformational energy in 4-O-protected-3-azido-2,3,6,-trideoxy-hex-5-enopyranosides are discussed.

Promotion of agonist activity of antiandrogens by the androgen receptor coactivator, ARA70, in human prostate cancer DU145 cells.

Although hormone therapy with antiandrogens has been widely used for the treatment of prostate cancer, some antiandrogens may act as androgen receptor (AR) agonists that may result in antiandrogen withdrawal syndrome. The molecular mechanism of this agonist response, however, remains unclear. Using mammalian two-hybrid assay, we report that antiandrogens, hydroxyflutamide, bicalutamide (casodex), cyproterone acetate, and RU58841, and other compounds such as genistein and RU486, can promote the interaction between AR and its coactivator, ARA70, in a dose-dependent manner. The chloramphenicol acetyltransferase assay further demonstrates that these antiandrogens and related compounds significantly enhance the AR transcriptional activity by cotransfection of AR and ARA70 in a 1:3 ratio into human prostate cancer DU145 cells. Our results suggest that the agonist activity of antiandrogens might occur with the proper interaction of AR and ARA70 in DU145 cells. These findings may provide a good model to develop better antiandrogens without agonist activity.

Experiences using chloramine-T and 1, 3, 4, 6-tetrachloro-3 alpha, 6 alpha-diphenylglycoluril (Iodogen) for radioiodination of materials for radioimmunoassay.

A comparison of labelling compounds with chloramine-T and with 1, 3, 4, 6-tetrachloro-3 alpha, 6 alpha-diphenylglycoluril (Iodogen) has been carried out. For human transferrin, human calcitonin, 1-84 bovine parathyrin, fibrinopeptide-A, human thyrotropin and F-CB3, a cyanogen bromide cleavage peptide of human fibrinogen, the quality of tracer produced by the Iodogen method was better. For rat lutropin, human growth hormone and human prolactin, labelling with Iodogen produced a tracer of unsatisfactory quality. For a further 13 peptides, the results from both methods were comparable. Optimal reaction times using Iodogen were of the magnitude of two to three times longer than when using chloramine-T. Reduction of the volume of radioactive waste by up to 90% could be achieved when the Iodogen method was coupled with a short cation-exchange column to separate unreacted iodide from the labelled compound. Data is presented on the quality of tracer, expressed in terms of elution profiles and radioimmunoassay standard curves. A novel "combi-method" of labelling proteins without tyrosine or histidine moieties is presented where N-succinimidyl-3-(4-hydroxyphenyl)-propionate is labelled at pH 7.5 using Iodogen to give "Bolton-Hunter" reagent, which is then transferred to a vessel containing the peptide to be labelled at ph 8.6.

Synthesis and evaluation of 2-tosylamino and 2-tosyliminopyrimidine derivatives as inhibitors of some leukocyte functions.

We have studied the potential anti-inflammatory effects of 20 2-tosylamino and 2-tosyliminopyrimidine new derivatives in human neutrophils. We have evaluated their interference with some leukocyte functions and 5-lipoxygenase activity. All the compounds reduced neutrophil degranulation process at concentrations in the microM range. Besides, compounds with a phenolic substitution inhibited leukotriene B(4) biosynthesis in neutrophils and decreased the cell-free 5-lipoxygenase activity. This study demonstrates that 2-tosylamino and 2-tosyliminopyrimidine derivatives can reduce the activation of neutrophil cells which may have relevance for the modulation of the inflammatory response.

(4aR,8R,SR)-8-(p-tolylsulfinyl)-2,3,4,4a,5,6,7,8-octahydro-4a-quinolinol.

The title compound, C16H21NO2S, isolated from the reaction of lithiated racemic 2,3,4,4a,5,6,7,8-octahydroquinoline with 0.5 equivalents of (-)-(1R,2S,5R)-menthyl (SS)-p-toluenesulfinate, possesses the unusual conformation in which the OH and bulky p-toluenesulfinyl groups are in axial positions. The S=O bond distance of 1.519 (3) A is longer than expected because of intramolecular hydrogen bonding between the hydroxyl group and the sulfinyl O atom.

Determination of glyphosate and its metabolite aminomethylphosphonic acid in fruit juices using supported-liquid membrane preconcentration method with high-performance liquid chromatography and UV detection after derivatization with p-toluenesulphonyl chloride.

The application of supported-liquid membrane (SLM) technique for effective extraction of N-(phosphonomethyl)glycine (glyphosate) and its primary metabolite aminomethylphosphonic acid (AMPA) from juices (orange, grapefruit, apple and blackcurrant) in combination with HPLC-UV detection after derivatization with p-toluenesulphonyl chloride (TsCl) is presented. The influence of various parameters such as the composition of acceptor phase, flow-rate, concentration of analytes, on the performance of extraction procedure, was studied. It was shown that by appropriate manipulation of SLM parameters the level of detection could be significantly improved. The influence of SLM conditions on extraction efficiency of studied compounds was also discussed. Selection of the optimal conditions enable detection of glyphosate and AMPA in juices at concentrations as low as 0.025 mg/l. The calculated recoveries for glyphosate were-71.1, 72.1, 93.6, and 102.7% and for AMPA-64.1, 64.6, 81.7, and 89.2%, for orange, grapefruit, apple and blackcurrant juices, respectively. The results suggest that the application of SLM extraction as a method for glyphosate and AMPA enrichment from complicated liquid matrices may be useful mean of routine analysis.

Cytotoxicity in pig hepatocytes induced by 8-quinolinol, chloramine-T and natamycin.

The potential cytotoxic effects of the compounds 8-quinolinol, chloramine-T and natamycin have been studied in isolated pig hepatocytes. The relative cytotoxicity of these compounds was evaluated on the basis of the leakage of cytosolic lactate dehydrogenase (LDH), 3-(4,5 dimethyl)thiazol-2-yl,-2,5-diphenyl tetrazolium bromide (MTT) reduction by mitochondrial dehydrogenases, uptake of neutral red (NR) by cytosolic lysosomes, glutathion (GSH) depletion and oxidized glutathion (GSSG) efflux after 24 h exposure. Evaluation of the 20%, 50% and 80% reduced absorbance data obtained from the parameters NR20, NR50, and NR80, and MTT20, MTT50 and MTT80 enabled us to rank these compounds in decreasing order of cytotoxicity: 8-quinolinol > natamycin > chloramine-T. Also for the parameters LDH and GSH, chloramine-T appears to be less cytotoxic than natamycin and 8-quinolinol. Our study demonstrated that pig hepatocytes may be a useful model for examining cytotoxic events of drugs to be used in pigs, therefore avoiding possible extrapolation problems due to species differences.

The effect of arylsulfonate esters on cholesterol--aggravated atherosclerosis in White Carneau Pigeons.

The arylsulfonate esters of linoleyl, stearyl, and decyl alcohols were found to reduce significantly the accumulation of cholesterol in the plasma and livers of White Carneau pigeons subjected to a diet of Purina pigeon pellets coated with 0.25% cholesterol and 10% lard when fed for periods ranging from 9--12 months; no effects were observed in normocholesterolemic pigeons. These compounds produced no toxic side effects and were found to significantly attenuate the development of aortic atherosclerosis. The effect on aortic atherosclerosis was most likely the result of the lowering of plasma cholesterol concentrations. Linoleyl p-toluenesulfonate appeared to be the most effective of the three arylsulfonates tested, both with respect to the reduction of plasma and liver cholesterol accumulation and attenuation of the atherosclerotic process.

Metabolism and enantioselective pharmacokinetics of Casodex in man.

1. Five healthy male volunteers received a single oral dose (50 mg; 42 microCi) of 14C-Casodex, a racemic compound, which has its antiandrogen activity predominantly in R-Casodex, the (-)-enantiomer, with little activity in S-Casodex, the (+)-enantiomer. 2. Plasma concentrations of R-Casodex increased slowly in all subjects to reach a peak of 559-970 ng/ml between 15 and 48 h after dosing and, thereafter, declined monoexponentially with a mean half-life of 4.2 days. Plasma concentrations of S-Casodex rose rapidly to reach a peak of 32-66 ng/ml within the first 2-5 h, and then declined monoexponentially with a mean half-life of 19 h. Plasma concentrations of the racemate were in very good agreement with the sum of the enantiomer concentrations throughout the study and were very similar to concentrations of total radioactivity over the first 4 days. 3. About 80% of the radioactive dose was recovered in urine (35.8 +/- 1.7%; mean +/- SEM) and faeces (42.6 +/- 2.9%) during a total collection over 9 days; this incomplete recovery was consistent with the slow elimination of R-Casodex. 4. T.l.c. of urine extracts indicated extensive metabolism of Casodex to two polar metabolites identified as the glucuronide conjugates of Casodex and hydroxy-Casodex; almost no parent compound was observed. Virtually all of the Casodex glucuronide excreted in urine during the first 2 days was derived from S-Casodex, consistent with the relatively low plasma concentrations and rapid elimination of this enantiomer. 5. T.l.c. of faecal extracts showed the presence of both Casodex and hydroxy-Casodex; these may have been eliminated in bile as the glucuronide conjugates, with subsequent hydrolysis in the intestinal tract.
