Dracorhodin perchlorate induces apoptosis in HL-60 cells.

Dracorhodin perchlorate, an anthocyanin red pigment, induces human premyelocytic leukemia HL-60 cell death through apoptotic pathway. Caspase -1, -3, -8, -9, and -10 inhibitors partially reversed the cell death induced by dracorhodin perchlorate. Caspase-3 and -8 were activated followed to the degradation of caspase-3 substrates, inhibitor of caspase-activated DNase (ICAD) and poly-(ADP-ribose) polymerase (PARP). Dracorhodin perchlorate up-regulated the expression ratio of mitochondrial proteins, Bax/Bcl-XL. The cell death was accompanied with phosphorylation of ERK, JNK and p38 MAPK and partially reduced by MEK inhibitor (PD98059), JNK MAPK inhibitor (SP600125) and p38 MAPK inhibitor (SB 203580). Taken together, dracorhodin perchlorate-induced apoptosis in HL-60 cells via up-regulation of Bax, activation of caspases and ERK/p38/JNK MAPKs.

[Mechanism of dracorhodin perchlorate-induced Hela cell apoptosis].

AIM: To study the mechanism of dracorhodin perchlorate-induced Hela cell apoptosis. METHODS: Cell viability was measured by MTT method. Morphological changes were observed by phase contrast microscopy and Hoechst 33258 staining. DNA fragmentation was assayed by agarose gel electrophoresis. Protein expression was detected by Western blot analysis. RESULTS: Dracorhodin perchlorate induced Hela cell apoptosis. The apoptosis was partially reversed by caspase-1, -3, -8, -9 and caspase family inhibitors. Treatment of Hela cells with dracorhodin perchlorate for 12 h increased the protein expression ratio of Bax/Bcl-XL; procaspase-3, -8, ICAD and PARP were cleaved to smaller molecules. CONCLUSION: Dracorhodin perchlorate induced Hela cell death via alteration of Bax/Bcl-XL ratio and activation of caspases.

Detection of hexamethonium-perchlorate association complexes using NACE-MS.

Perchlorate (ClO(4) (+)) and other chlorine oxide anions were observed to complex weakly with hexamethonium (1,6-bis-(trimethylammonium)-hexane) in both aqueous and polar nonaqueous solvents. The resultant positively charged complexes were resolved by NACE using 2-propanol/acetone electrolytes prior to mass spectrometric detection using an Agilent(3D)CE system coupled to a Bruker Esquire 3000+ quadrupole IT mass detector. Using electrokinetic injection, the method detection limit for perchlorate in nonaqueous media was 10 microg/L. The isotope patterns due to the presence of (35)Cl and (37)Cl in complex mass spectra allowed for unambiguous identification of perchlorate, chlorate (ClO(3) (+)), chlorite (ClO(2) (+)), and chloride (Cl(+)) in photoreaction samples.

An ion-pairing high-performance liquid chromatographic method for the direct simultaneous determination of nucleotides, deoxynucleotides, nicotinic coenzymes, oxypurines, nucleosides, and bases in perchloric acid cell extracts.

An ion-pairing high-performance liquid chromatographic method for the direct and simultaneous determination of nucleotides, deoxynucleotides, cAMP, nicotinic coenzymes, oxypurines, nucleosides, and bases in perchloric acid cell extracts is presented. By using an Alltima C-18, 250 x 4.6-mm, 5-microns particle size column, a high resolution of 38 acid-soluble compounds, including ATP, GTP, dTTP, CTP, UTP, ADP, GDP, dTDP, CDP, UDP, dATP, dGTP, dCTP, dUTP, dADP, dGDP, dCDP, dUDP, and cAMP, is obtained. Elution is performed with a step gradient from buffer A (consisting of 10 mM tetrabutylammonium hydroxide, 10 mM KH2PO4, 0.25% methanol, pH 7.00) to buffer B (consisting of 2.8 mM tetrabutylammonium hydroxide, 100 mM KH2PO4, 30% methanol, pH 5.50). Perchloric acid extracts of resting and phytohemagglutinin-stimulated human lymphocytes were analyzed. Data indicate that this chromatographic method offers, for the first time to the best of our knowledge, the possibility of simultaneously determining di- and triphosphate nucleosides and their corresponding deoxynucleosides without any chemical manipulation of samples except for perchloric acid deproteinization. Hence, the present HPLC assay minimizes the risks of modification or loss of metabolite concentration and allows one to obtain, with a single chromatographic run, the complete pattern of those metabolites which are known to be involved in energy metabolism and in DNA and RNA synthesis, resulting therefore of great advantage in cell biology studies.

The perchlorate discharge test with and without supplement of potassium iodide.

The purpose of the study was to compare the discharge of radioactive iodine from the thyroid after po administration of perchlorate with and without supplement of 2 mg potassium iodine (Kl) in 42 euthyroid controls, 26 patients with nontoxic goiter, 39 patients with thyrotoxicosis treated with radioactive iodine, and 39 patients with Pendred's syndrome. The discharge test was found to have a satisfactory specificity with supplement of 2 mg Kl, and was more sensitive to detect an organification defect than the standard perchlorate discharge test without Kl. In all groups of patients a significant discharge occurred after addition of Kl in contrast to healthy persons. Only in the group of patients with Pendred's syndrome a discharge of iodine was found in the perchlorate test without Kl.

Treatment of amiodarone associated thyrotoxicosis by simultaneous administration of potassium perchlorate and methimazole.

Amiodarone iodine induced thyrotoxicosis occurs frequently in patients residing in areas of mild iodine deficiency and in patients with preexisting goiter. Drug therapy of the hyperthyroidism is often unsuccessful. Twenty-three patients with amiodarone induced thyrotoxicosis were either not treated, treated with 40 mg methimazole daily or with methimazole and 1 gm potassium perchlorate daily for up to 40 days and then with methimazole alone. Thyrotoxicosis was more likely to spontaneously remit in patients without goiter. Therapy with methimazole alone was unsuccessful in inducing euthyroidism in 5 patients with goiter. However, combined therapy with methimazole and potassium perchlorate rapidly alleviated hyperthyroidism in almost all patients with goiter. This drug combination is successful because perchlorate inhibits the active transport of iodine into the thyroid and methimazole blocks the intrathyroidal synthesis of thyroid hormones.

Differential extractability of creatine phosphate and ATP from cardiac muscle with ethanol and perchloric acid solution.

To compare the extractability of creatine phosphate with that of ATP by alcohol extraction, both compounds were extracted from normal perfused rat heart tissues by using various stepwise concentrations of ethanol and 0.4 M HClO4. Powdered samples (6-15 mg wet wt) from the freeze-clamped tissues were homogenized in 2 ml of the ethanol solutions. After centrifugation, the supernatant was removed; each centrifuged sediment was rehomogenized with 2 ml of 0.4 M HClO4 and centrifuged. The supernatant was neutralized with 0.4 m KHCO3. The same powdered samples were directly homogenized with 2 ml of 0.4 M HClO4 and treated in the same manner. Only a small amount of ATP in the tissues was extracted by an 85% or higher concentration of ethanol. Further, about 13% of the tissue ATP was not extractable by the subsequent perchloric acid extraction. In contrast to ATP, creatine phosphate in the tissues was partially extracted by 95% ethanol and nearly all of the tissue creatine phosphate was extracted by 70% ethanol. The total creatine phosphate obtained by 70% ethanol and by subsequent perchloric acid extraction was significantly higher than that obtained by direct perchloric acid extraction. From these results, it was concluded that the extractability of creatine phosphate in the tissue by alcohol extraction is clearly different from that of ATP. Additionally, the stepwise extraction is recommended as a useful method for the extraction of energy metabolites in perfused rat heart tissue.

Resistance of horse alpha 1-proteinase inhibitor to perchloric acid denaturation and a simplified purification procedure resulting therefrom.

Addition of perchloric acid (6.4% w/v final concentration) to horse alpha 1-proteinase inhibitor or to horse plasma neither precipitated nor inactivated alpha 1-proteinase inhibitor. None of the isoinhibitors of alpha 1-proteinase inhibitor was altered by dilute perchloric acid. This unexpected behavior led to a simplified procedure for the purification of horse alpha 1-proteinase inhibitor, consisting of removal of the bulk of plasma proteins, by perchloric acid precipitation and by gel filtration on Sephadex G-75 and G-200. The resulting preparations of alpha 1-proteinase inhibitor were immunogenically pure. The simplified purification procedure permitted the immunochemical comparison of the isoinhibitors of alpha 1-proteinase inhibitor, which proved to be immunologically identical.

Reversed-phase liquid chromatograhy/electrospray ionization tandem mass spectrometry for analysis of perchlorate in water.

A new reversed-phase liquid chromatograhy/electrospray ionization tandem mass spectrometry method was developed for the analysis of perchlorate in water. The improved separation of perchlorate from common anions along with sample dilution effectively reduced matrix effects, primarily ion suppression caused by common anions. The (18)O-enriched perchlorate used as an internal standard provided further compensation for potential changes associated with instrument sensitivity, retention time shifting, peak broadening, ion suppression, and other matrix effects. The mean recoveries and relative standard deviations were 92-107% and 2.5-9.5% for simulated water matrix spikes at 0.05-1.0 microg/L, and 80-106% and 3.8-13% for real water sample matrix spikes at 2.0 microg/L, respectively. The method detection limits were 0.007 microg/L for reagent water and 0.014 microg/L for the simulated water matrix that contained 100 mg/L of SO(4)(2-), CO(3)(2-), and Cl(-) anions; 2 mg/L of PO(4)(3-) as P and NO(3)(-) as N; and 0.1 mg/L of Br(-), BrO(3)(-), ClO(2)(-), ClO(3)(-), and F(-) anions in reagent water, respectively. When using cartridge pretreatment to remove problematic SO(4)(2-), CO(3)(2-), and Cl(-) anions, the minimum reporting level could be set to 0.05 microg/L or lower. With 10-fold dilution, the minimum reporting level was conservatively set to 0.5 microg/L.

Synthesis, crystal structure, magnetic property and oxidative DNA cleavage activity of an octanuclear copper(II) complex showing water-perchlorate helical network.

A new octanuclear copper(II) complex has been synthesized and structurally characterized by X-ray crystallography: [Cu(8)(HL)(4)(OH)(4)(H(2)O)(2)(ClO(4))(2)].(ClO(4))(2).2H(2)O (1) (H(3)L=2,6-bis(hydroxyethyliminoethyl)-4-methyl phenol). The complex is formed by the linkage of two terminal bimetallic cationic units and a tetranuclear mu(3)-hydroxo bridged dicubane core by a very short intramolecular hydrogen bond (O-H...O, 1.48(3)A and the angle 175 degrees). The coordination sphere of the terminal copper atoms is square pyramidal, the apical positions being occupied by water and a perchlorate ion. Complex 1 self-assembles to form a new type of water-perchlorate helical network [(H(2)O)(2)(ClO(4))](infinity) involving oxygen atoms of coordinated perchlorate ion and the two lattice water molecules through hydrogen-bonding interaction. The variable temperature-dependent susceptibility measurement (2-300K) of 1 reveals a strong antiferromagnetic coupling, J(1)=-220cm(-1) and J(2)=-98cm(-1) (J(1) and J(2) representing the exchange constant within [Cu(2+)](4) and [Cu(2+)](2) units, respectively). The complex binds to double-stranded supercoiled plasmid DNA giving a K(app) value of 1.2x10(7)M(-1) and displays efficient oxidative cleavage of supercoiled DNA in the presence of H(2)O(2) following a hydroxyl radical pathway.

How perchlorate improves excitation-contraction coupling in skeletal muscle fibers.

The effect of the "chaotropic" anion, perchlorate, on the activation of contraction has been studied in voltage clamped frog skeletal muscle fibers. It was found that the voltage dependence of either the contractile force or the intramembrane charge movement was shifted towards more negative membrane potentials. The maximum values of force or charge movement attained with large depolarizing pulses did not change significantly. It is concluded that a specific perchlorate effect on the movement of charged particles can explain the potentiating effect of perchlorate anions on contractile force, strengthening the view that these charged particles serve as voltage sensors regulating Ca2+ release from the sarcoplasmic reticulum.

Abrogation of c-MYC protein degradation in human lymphocyte lysates by prior precipitation with perchloric acid.

Conventional lysis buffers, though containing cocktails of protease inhibitors, did not prevent the degradation of c-MYC recombinant protein added immediately prior to lysis to cell pellets from human mixed lymphocyte cultures. Treatment of the cells with 4.2% perchloric acid, however, prevented protein degradation and facilitated the detection of c-MYC protein by Western blotting even in unstimulated lymphocytes, where previously it had been reported to be undetectable or barely detectable using this technique. PHA stimulation of lymphocytes induced an approximately six fold increase in measured c-MYC protein within 5 h if cell extracts were prepared using perchloric acid precipitation. However, using conventional lysis buffer the proto-oncogene protein was undetectable until 48-72 h after mitogen addition. Pretreatment with perchloric acid may be useful for Western blotting analysis of protein in other systems where it may be desirable to dispense with the use of toxic protease inhibitors or where these may be incompletely effective.

Evidence of molten globule like state(s) of interferon gamma in acidic and sodium perchlorate solutions.

Recombinant porcine interferon gamma (IFN gamma) at neutral pH is characterized by a tryptophan (Trp) fluorescence maximum around 343 nm and a rigid conformation, evidenced from tryptophan polarization values. Guanidine HCl shifts the protein emission spectra further to the red and decreases the fluorescence polarization values, indicating denatured IFN gamma in these solutions. In acidic solutions (3 < pH < 4), the emission spectra show a blue shift and lower tryptophan polarization. The midpoint of transition of these fluorescence properties occurs around pH 3.5-3.6. The protein in NaClO4 solution at neutral pH is similarly characterized by a blue shift in the tryptophan fluorescence maxima and low polarization values. The extent of quenching of tryptophan fluorescence by acrylamide is less in acid and in NaClO4 solutions of IFN gamma compared to its native form. This indicates a lower accessibility of the tryptophan in the altered conformation of the protein. The emission spectra of IFN gamma in NaClO4 solution shows a decrease in the tryptophan fluorescence intensity with simultaneous shift of the emission spectra over time. The presence of two conformational forms of IFN gamma in perchlorate solution is evidenced from an isofluorescent point at 315 nm. The change in the conformational state in perchlorate solution is characterized by first order kinetics. The dye anilinonaphthalene sulfonic acid does not bind either to the native IFN gamma or to its denatured form. However, the dye binds to the acid form of IFN gamma, as well as when the protein is present in NaClO4 solution at neutral pH. These observations, together with the results from literature that IFN gamma retains its secondary structure in acid solution to a considerable degree, would suggest that the protein exists as a molten globule-like state in acidic solution. Similarities of the protein fluorescence and 1-anilino-8-naphthalene-sulfonic-acid (ANS) binding properties of the protein in NaClO4 and acid solutions indicate that IFN gamma also exists in a molten globule-like state in perchlorate solution at neutral pH.

High-performance anion-exchange chromatography of human urine using perchlorate gradient elution systems.

A 100-microliter volume of urine was chromatographed on a 50 X 0.4 cm I.D. column packed with a macroreticular anion-exchange resin. Elution was performed with a concave ammonium perchlorate gradient from 0 to 0.25 M at a flow-rate of 0.75 ml/min and a pressure of 7.5-11 MPa. With this perchlorate gradient, no baseline drift occurred in the detection at 254 nm, and even detection at 200 nm was possible. The effect of the addition of ethanol or acetonitrile to the ammonium perchlorate solution was investigated. For the assignment of peaks, ultraviolet spectra of the peaks were measured with stopped-flow scanning spectrophotometry.

13C cross-polarization magic angle spinning NMR and gauge-independent atomic orbital, coupled Hartree-Fock calculations of buspirone analogues. Part 2. Hydrochlorides and perchlorates of 1-arylpiperazine-4-alkylimides.

13C cross-polarization (CP) magic angle spinning (MAS) solid state NMR spectra of hydrochlorides and perchlorates of buspirone analogues (2-5) were recorded. In the spectra for each compound, one set of signals appeared, in agreement with single crystal X-ray diffraction data indicating the presence of one molecule per crystal unit. The resonances of 2-5 hydrochlorides were assigned by comparison with the solution chemical shifts. For perchlorate 2b and diperchlorate 2c, the reasonable assignment of signals was made with the aid of the theoretical studies. Ab initio calculations of the carbon shieldings were performed by means of the GIAO-CHF method for two model systems: perchlorate and diperchlorate of quinoline-(N-methyl)piperazine. As no remarkable differences between carbon chemical shifts of hydrochlorides 3-5 in solid state and in solution were observed, it was concluded that in solution these compounds adopted the same conformation as in the solid state.

Methods for the stable isotopic analysis of chlorine in chlorate and perchlorate compounds.

Chlorate and perchlorate compounds, used as herbicides, solid fuel propellants, and explosives, are increasingly recognized as pollutants in groundwater. Stable isotope characterization would permit both environmental monitoring of extent of remediation and forensic characterization. Stoichiometric reduction to chloride (greater than 98% yield), by Fe(II) for chlorate and alkaline fusion-decomposition for perchlorate, allows analysis by standard methods to give highly reproducible and accurate delta37Cl results (0.05/1000, 2 x standard error). Analysis of various compounds from different suppliers yielded delta37Cl values for chlorate samples near to +0.2/1000 (SMOC), but one has within-sample heterogeneity of 0.5/1000, possibly due to crystallization processes during manufacture. Results for perchlorate samples also are generally near +0.2/1000, but one is +2.3/1000 (SMOC). The initial results suggest that both forensic and environmental applications might be feasible.

Perchlorate-induced denaturation of ribonuclease A: investigation of possible folding intermediates.

Perchlorate-denatured ribonuclease A (PDR) is known to show a circular dichroism (CD) spectrum suggestive of substantial secondary structure. Thus, PDR may be a molten globule form of ribonuclease A. We find that any secondary structure present in PDR does not provide measurable protection against amide proton exchange, and PDR does not belong to the class of structured molten globules. CD spectra of short peptides show that the perchlorate anion affects these spectra in a way that could be mistaken for secondary structure formation; thus, caution must be used in interpreting CD spectra of peptides and proteins taken in perchlorate solutions.

Perchlorate in milk.

Perchlorate was unambiguously detected by ion chromatography-suppressed conductivity (IC-CD) and/or ion chromatography-electrospray mass spectrometry (IC-MS) in seven of seven supermarket milk samples bought randomly in Lubbock, TX. Quantitation by IC-MS and IC-suppressed conductivity detection in conjunction with a preconcentration-preelution method provided comparable results. With a sample cleanup procedure that involved protein removal by ethanol and sequential passage though activated alumina and C-18 silica, the limit of detection for perchlorate in milk was 0.5 microg/L. The levels found ranged from 1.7 to 6.4 microg/L. An evaporated milk sample contained perchlorate at 1.1 +/- 0.6 microg/L level, while we did not find detectable levels in a reconstituted powdered milk sample.
