The dominant intestinal helminths in the Iranian villages of Kargar Malleh (hookworms), Bandar Abbas (hookworms) and Jazin (Ascaris lumbricoides and Trichuris trichiura), were recorded with their frequency distributions, in 80, 40 and 111 villagers, respectively in 1977-78. Anthelminthics were then used to remove all intestinal stages which were identified and counted. Analyses showed clumped or over-dispersed patterns in all cases. Each community contained 'wormy persons', 1 to 3% of the individuals carried 11, 16, 30, 38 or 84% of all the worms collected. 'Wormy persons' could only be identified for one parasite, thus those with the highest infestations of one species were not prone to higher infestation with another. In Jazin, 111 persons with measured initial worm burdens of A. lumbricoides, T. trichiura and Trichostrongylus spp. were followed without further therapy or change in habits for up to 12 months during re-infection. There was no significant correlation between those persons with high initial worm burdens and those with high burdens after 12 months. The public health significance of these findings is discussed.

The effect of single oral doses of albendazole 600 mg and mebendazole 1 g given to 56 and 60 men, respectively, with T. trichiura and/or N. americanus infestation has been studied. Both albendazole and mebendazole cured more than 90% of T. trichiura infestations, but only albendazole (95%) and not mebendazole (21%) had a high cure rate for N. americanus infestations. Thus, albendazole is the preferred benzimidazole derivate for the mass treatment of subjects with T. trichiura and/or N. americanus infestations.

Solid-state fermentation (SSF) has recently been proposed as an alternative to submerged fermentation for the production of poly(hydroxyalkanoates). In the present work, X-ray diffraction, differential scanning calorimetry, nuclear magnetic resonance and infrared spectroscopy were employed to investigate the chemical structure, as well as the thermal properties and the crystalline morphology of poly(3-hydroxybutyrate) samples produced by SSF, using as raw material either soy cake or soy cake supplemented with 2.5% (m/m) sugarcane molasses. The results obtained showed that the biopolymer obtained by SSF presented the same properties as commercial PHB, except for the higher molar mass and the lower degree of crystallinity that were observed. Thus, the present data indicate that solid-state fermentation is an interesting alternative for the production of PHB, allowing the production of biopolymers with adequate properties from low-cost, renewable resources.

A method for rapid extraction of both double-stranded RNA (dsRNA) and DNA from an obligate biotrophic phytopathogenic fungus is described. Lyophilised fungal material is incubated in a commercial guanidium thiocyanate reagent. Proteins and cell debris are centrifuged by chloroform precipitation. After precipitation in isopropanol and washing in 75% ethanol, nucleic acids are resuspended in water (10 microl/mg fungal dry weight). DsRNA is directly visualised by agarose gel electrophoresis. DNA contained in 10-fold dilutions of the samples proved to be suitable for PCR-based experiments.

Grapevine is subject to a number of diseases that affect yield and wine quality. To limit the excessive use of phytochemicals in the vineyard, alternative strategies have to be developed. Plant treatment with signaling molecules like elicitors stimulates their natural defense mechanisms. To improve grapevine tolerance against fungal pathogens, Vitis vinifera plants were treated with a natural exogenous elicitor, methyl jasmonate (MeJA). MeJA-treated leaves (Cabernet Sauvignon foliar cuttings) reacted by increasing transcript levels coding pathogenesis-related proteins (acidic class IV chitinase, serine protease inhibitor, polygalacturonase-inhibiting protein, and beta-1,3-glucanase) and coding enzymes involved in phytoalexin biosynthesis (one phenylalanine ammonia lyase and one stilbene synthase). This was correlated with the accumulation of stilbenes (antimicrobial compounds). The eliciting activity of MeJA was confirmed by enhanced tolerance of grapevine foliar cuttings and vineyard against powdery mildew (75% and 73%, respectively). On the basis of these original results, MeJA can therefore act as an efficient elicitor in an alternative strategy of grapevine protection.
We investigated environmental factors influencing the distribution of hookworm infection in KwaZulu-Natal, South Africa. Prevalence data were sourced from previous studies and additional surveys carried out to supplement the database. When geo-referenced the data revealed that higher prevalences are limited to areas below 150 m above sea level, and low prevalences to areas above this altitude. Using univariate analysis we investigated the differences in environmental factors in the two areas. The relationship between hookworm prevalence, altitude and climate-derived variables was assessed using Pearson correlation coefficient, and that of soil type using the t-test. Multivariate analysis was used to determine environmental factors that combine best to provide favourable conditions for hookworm distribution. The results revealed that areas 150 m above sea level, i.e. inland, supported low mean hookworm prevalences (x = 6, n = 21), and were characterized by soils with a clay content of more than 45%, variable temperatures and moderate rainfall. Hookworm prevalence also decreased southwards as temperatures became slightly cooler, rainfall remained more-or-less constant and the coastal plain narrowed. In the multivariate model prevalence was most significantly correlated with the mean daily minimum temperature for January followed by the mean number of rainy days for January. This indicates the importance of summer conditions in the transmission of hookworm infection in KwaZulu-Natal and suggests that transmission may be seasonal.
