[The proteinase activity and content of trypsin inhibitors in the mammae of female rabbits in physiological states].

We studied the activity of lysosomal cathepsins B and D, neutral trypsin-like proteinase, and the content of trypsin inhibitors in the mammary glands of virgin, pregnant, and lactating rabbit females, as well as during the involution caused by lactostasis. The maximal activity of cathepsin D was found to occur in mammary glands under the condition of lactostasis. The activity of cathepsin B was practically identical in the mammary glands of virgin and pregnant animals and diminished during lactation. The activity of trypsin-like proteinase is low in the mammary glands of virgin animals, increases drastically during pregnancy, and diminishes during lactation. The content of trypsin inhibitors is maintained at a similar level in the mammary glands of virgin and pregnant animals and increases significantly during lactation. The caseinolytic activity of neutral proteinases has not been detected in the mammary glands of virgin rabbit females. At the lactation and involution stages, it is considerably lower than the activity of acidic proteinases and is possibly controlled by endogenous inhibitors.

[The place of galactography in the diagnosis of secreting mamma].

Examinations were carried out in 3,300 women, finding in 210 cases a spontaneous or provoked discharge from one or two breasts. Out of these women 115 patients came for galactographic examination. After the radiological examinations 37 patients were treated surgically including 28 cases with the diagnosis of intraductal hyperplasia, and in this group of patients histopathological examination demonstrated: nine cases of cancer, seven cases of papilloma, seven cases of fibroadenoma, seven cases of benign dysplasia with ductal epithelial hyperplasia, three cases of inflammatory lesions, and one case without pathological changes. The analysis carried out demonstrated that galactographic picture of secreting ducts may suggest a hyperplastic process but determination of its character is possible only in histopathological examination.

[The effect of isoproterenol on protein extrusion from tissue explants of the mouse mamma].

Culture of mouse mammary gland cells was treated with beta-agonist isoproterenol to result in dose dependent increase in protein. Propranolol, beta-antagonist, blocked this effect of isoproterenol. According to electron microscopic analysis, application of isoproterenol leads to disappearance of vesicles from apical zones of the secretory cells, and to increase in casein micelles in alveolar lumen. As follows from the obtained data, catecholamines play part in regulation of mammary gland activity, stimulating process (via beta-adrenoreceptors) of protein extraction from secretory cells of the mouse mammary gland.

[Dose distribution and irradiation technique in the radiotherapy of breast cancer stage I].

A method for homogenous irradiation of the breast by means of opposite standing fields, with the use of wedge filters, is described here. The field is focussed so that the interior thoracic wall under symmetric position of the mamma is still completely situated in the irradiation field, and the lateral area of the ray beam passes 1-2 cm into the pulmonary tissue. On a series of typical breast models, made under consideration of the irradiation position, the isodose distributions were determined by film-dosimetry. The therapeutically most important 80%-isodose corresponds in practically every case with the mamma contour. Peak doses or areas of underdosage were not observed. The thoracic wall is still exposed to about 75% of the maximum dose. In order to reach a more accurate irradiation by the desired maximum dose of, for example, 250 r, the deviation of which does not exceed plus or minus 7%, it is necessary, at the beginning of an irradiation series, to use ionisation chamber measurements with an intensifying hood on the surface of the mamma, 1.5 cm above the border of the field. In the case of lateral irradiation, the measurement need only be made laterally, in medial irradiation it is only necessary on the medial side. The portion of the dose affecting the opposite field on the measuring place can be calculated by the use of depth dose tables. From the values so gained the desired maximum dose within the mamma may be ascertained by multiplication by the factor 1.03.

Normal but not carcinomatous primary rat mammary epithelium: readily transplanted to and maintained in the athymic nude mouse.

Transplantation success rates of primary 7,12-dimethylbenz(a)anthracene [(DMBA) CAS: 57-97-6]-induced rat mammary carcinomas and normal rat mammary glandular epithelium into female athymic mice were compared. The rat mammary carcinomas obtained from female Sprague-Dawley rats were transplanted into host athymic mice (6-8 wk of age) as 1 x 1-cm slices xenografted sc (2 slices/mouse) or as enzymatically dissociated cells inoculated into the gland-free mammary fat pad. Normal rat mammary glands (No. 4 glands and 3- to 5-mo-old virgin rats) were transplanted into host athymic mice as whole, intact mammary glands sc (1 gland/mouse) or as enzymatically dissociated cells inoculated into the gland-free mammary fat pad. All (100%) of the normal rat mammary glands were readily accepted and maintained in the athymic mice when transplanted either sc as whole glands or as dispersed cells inoculated into the gland-free fat pad. In contrast, only 13-14% of the DMBA-induced rat mammary carcinomas were accepted and maintained in the athymic mice (transplanted as slices sc or as dispersed cells inoculated into the gland-free fat pad). Treatment of host athymic nude mice with an intense mammotropic hormonal stimulus (prolactin and/or ovarian steroids) markedly enhanced the developmental growth of the transplanted normal rat mammae (subcutaneous slices and fat-pad inoculates); such a hormonal stimulus did not influence the transplantation success rate of the DMBA-induced rat mammary carcinomas. Thus female athymic nude mice can readily accept and maintain transplants of normal rat mammae but not carcinogen-induced carcinomatous rat mammae; the meager acceptance rate of the carcinomatous rat mammae by the athymic nude mouse was not enhanced by providing the host mice with a potent mammotropic hormonal growth stimulant.

[Staphylococci in skin microbiocenosis of the breasts in healthy women].

The autoflora of different anatomical regions of the mammary glands in 120 healthy nulliparous women aged 18-24 years was studied by P. Williamson and A. M. Kligler's methods of smears and washings. From the nipple, the areola, and the adjacent region of the skin 2,248 strains of anaerobic microorganisms were isolated; of these, 63.83% were staphylococci and micrococci, 6.01% were streptococci, 1.91% were Neisseria, 17.79% were Corynebacterium, 3.87% were bacilli, 2.8% were enterobacteria, and 3.79% were fungi. Coagulase-positive staphylococci occurred in 1.56% of cases. Out of 11 coagulase-negative species of this genus, S. epidermidis occurred most frequently on the skin of the mammary glands. The nipple was found to have the highest bacterial contamination (0.55 X 10(6) +/- 0.7 X 10(5) cells/sq. cm for the right mamma and 0.59 X 10(6) +/- 0.7 X 10(5) cells/sq. cm for the left mamma) and the skin adjacent the areola, the lowest bacterial contamination (0.14 X 10(4) +/- 0.2 X 10(3) cells/sq. cm for the right mamma and 0.25 X 10(4) +/- 0.3 X 10(3) cells/sq. cm for the left mamma). P. Williams and A. M. Kligman's method of washings, more accurate and informative, was found to be preferable for the study of the quantitative characteristics of the dermal microbiocenosis of the mammary glands.

Zeranol (beta-resorcylic acid lactone), a common residous component of natural foodstuffs, stimulates developmental growth of the mouse mammary gland.

ICR mice ovariectomized at 6 weeks of age were divided into 5 groups and injected s.c. daily for 20 days with: (1) 0.9% NaCl solution containing 0.1 mg/ml gum arabic (controls); (2) zeranol (Z) (1.0 microgram/day); (3) progesterone (P) (1.0 mg/day); (4) Z + P or (5) 17 beta-estradiol (1.0 microgram/day) + P (E + P). Mammary gland (no. 4, inguinal) growth was assessed by analysis of mammae wet weight, dry fat free weight, DNA content and mammae whole mount evaluation. Administration of Z, alone or in combination with P, increased all 4 measures of mammary growth (P less than 0.05). The combination of Z + P was highly mammogenic, with mammary wet weight, dry weight and DNA content comparable to that obtained with E + P. Mammae development scores of Z + P treated mice were only slightly less than those from E + P treated mice. The mammary glands of Z treated mice had increased duct number (branching) and thickening. The glands of Z + P treated mice had extensive duct branching with occasional lobule-alveolar development. These data indicate that Z, alone or in combination with P, is an effective mammogen possessing mammogenic properties similar to the potent natural estrogen 17 beta-estradiol.

[New method of MR-guided mammary biopsy].

PURPOSE: With a recently installed new open MR tomograph (Signa SP, General Electric MS) it is possible for the first time to examine patients with obscure lesions of the mamma by MR imaging in a sitting position, similar to mammography. METHOD: The newly developed positioning device with integrated surface coil permits in combination with the vertical gap in the new magnet and imaging of the mamma with a "real time" sequence a biopsy procedure within a short time and in a manner that is acceptable for the patient. By means of an integrated interactive scan plane guiding system ("flash point tracking system") and special MR-compatible needles and wires tissue samples can be taken, or marking by a wire can be carried out within a short time. RESULTS: Phantom experiments and first in vivo experience (10 patients) show that by means of the new mamma fixation and biopsy device a MRI of one breast in a sitting position for detecting an enhancing lesion and a MR-guided biopsy of this lesion is possible. The whole procedure takes about 25-35 minutes, comparable to the conventional stereotactic method. CONCLUSION: Direct patient access and the interactive guided biopsy allow in future both new diagnostic information (histological results after biopsy or after wire-guided open biopsy of a lesion only visible in MR) and the development of minimal invasive therapeutic procedures, e.g. MR-guided and -controlled interstitial thermotherapy.

Soft tissue tumors among beagles injected with 90Sr, 228Ra, OR 228Th.

The occurrence of soft-tissue tumors in beagles given 90Sr (88 dogs), 228Ra (76 dogs), or 228Th (81 dogs) as young adults and followed throughout their lifespans was compared with that of 133 control beagles given no radioactivity. For animals injected with 228Ra, tumors of the eye were more prominent (p < 0.05) than in the controls, and soft-tissue tumors of cavities in the head (excluding the brain, mouth, and eye) were more prominent in dogs given 90Sr than in the controls (p < 0.05). There was some indication that eye tumors in animals given about 0.56 kBq 228Th kg-1 were associated with their radionuclide exposure. For tumors at a few other locations, the relative occurrence was greater (p < 0.05) in the controls. These included malignant tumors of the testis and malignant plus benign tumors of the mammae and vagina in 228Th dogs; both malignant and malignant plus benign tumors of the mouth and testis, and malignant plus benign tumors of the mammae and vagina in 228Ra dogs; and malignant plus benign tumors of the mammae in 90Sr dogs (p > 0.05 by Odds Ratio Chi Square analysis but p < 0.05 by Fisher's Exact Test). Differences in relative occurrence between radioactive dogs and controls of all other tumor types that appeared in any of the animals (notably lymphosarcoma, lymph node tumors, leukemia, mast cell tumors, liver tumors, etc.) were not statistically significant (p > 0.05). Intercurrent mortality, mainly from bone cancer, was higher in the radioactive dogs than in the controls. Mean survival was reduced in the dogs given 90Sr, 228Ra, or 228Th (13.17 +/- 2.64 y in controls, 10.95 +/- 4.06 y in 90Sr dogs, 9.07 +/- 3.61 y in 228Ra dogs, and 9.20 +/- 4.15 y in 228Th dogs). Attenuated lifespans could account, at least in part, for the relative paucity of soft-tissue tumors not induced by radiation among the groups of dogs given radioactivity and occurring near the end of life for control animals.

[Mammogram screening: an on-going debate].

General public mamma-gram screening programs directed at women aged from 50 to 65 years are a common practice in Spain and other European countries. After more than ten years being recognized as early detection programs for breast cancer A debate has opened about the effectiveness of mamma-gram screening to reduce the number of deaths due to this type of tumor. The author includes a wide bibliography and various data bases such as MEDLINE: 1989-2003, CANCERLIT: 1975-2002, CINAHL: 1982-2003, and EMBASE: 1988-2003; and realizes it is not simple to justify mamma-gram screening since there is a great diversity of opinion in medical literature.

Caffeine, theophylline, theobromine, and developmental growth of the mouse mammary gland.

The purpose of this study was to determine the comparative activities of three methylxanthines, i.e., 1,3,7-trimethylxanthine (caffeine), 1,3-dimethylxanthine (theophylline), and 3,7-dimethylxanthine (theobromine) on developmental growth of the mammary gland in ovarian-hormone treated, mature nulliparous female Balb/c mice. When caffeine or theophylline was administered daily (via drinking water, 500 mg/L) for 30 days to 17 beta-estradiol/progesterone-treated intact or ovariectomized mice, a significant (p less than 0.05) enhancement of hormone-induced mammary gland lobulo-alveolar differentiation was observed. Caffeine or theophylline thus accelerated and/or intensified mammae lobulo-alveolar differentiation induced by the ovarian steroids. In contrast, theobromine (500 mg/L drinking water) did not significantly modify this developmental process. The stimulatory effect of caffeine and theophylline on mammae development was comparable quantitatively. In an effort to determine whether or not the stimulatory effect of caffeine or theophylline was directly on the mammary gland, small slow-release Elvax-40P pellets containing these methylxanthines were implanted directly into the mammary gland of mice concurrently treated with estrogen and progesterone. No significant stimulatory effect of caffeine or theophylline (or theobromine) was observed. Furthermore, the addition of methylxanthines (caffeine, 100 microM) to the culture media of whole mouse mammary glands (organ cultures) did not enhance lobulo-alveolar differentiation induced by mammotrophic hormones. Thus, while a consistent significant stimulatory effect of caffeine and theophylline on mammary lobulo/alveolar differentiation was observed when the methylxanthines were consumed orally (drinking water), no direct effect of these methylxanthines, when placed directly into the mammary gland or in culture media, on mammae development was observed. These data demonstrate that certain methylxanthines (e.g., caffeine and theophylline) but not others (e.g., theobromine) can significantly enhance mammotrophic hormone-induced mammary lobulo-alveolar differentiation in female Balb/c mice, an effect that appears not to be manifested via a direct action of the methylxanthines on the mammary gland.

[Malignant fibrous histiocytoma of the breast].

In connection with the mammary tumor of a 41-year-old woman the authors draw attention to the rare occurrence of malignant fibrous histiocytoma in the mamma. The histology of the mammary malignant fibrous histiocytoma, role of immunohistochemical reactions in the diagnosis are described. The literature dealing with the malignant fibrous histiocytoma in the mamma is reviewed. This is the 11th case reported in the literature. The authors discuss the possibilities of the therapy of the malignant fibrous histiocytoma in the mamma.

Correlation of noninvasive surface temperature measurement with rectal temperature in swine.

Skin and rectal temperature measurements were taken on 15 crossbred sows and gilts (225 kg) during the last 5 days of prepartum period and compared with those recorded in the first 8 days after parturition. The correlation of skin and rectal temperature values was not good during either period, although an increased correlation was noted in the postparturient period. The rectal temperature significantly (P less than 0.05) increased in the postparturient period, but the mean surface temperature was unchanged. This would indicate a decreased blood flow to the surface, since an equivalent blood flow to the surface after a rectal temperature increase would result in a concomitant mean surface temperature increase. The decreased flow to the surface of the animal was selective, since a significant (P less than 0.05) decrease in surface temperature was noted at the tailhead, perineum, vulva, mammae-caudal, and eye, whereas the surface temperature of the mammae-cranial and mammae-middle was significantly (P less than 0.05) increased.
