The economic effect of estrus synchronization in beef heifers on average weaning weight of calves.

Prostaglandin was used to induce estrus in 129 Hereford x Holstein heifers for artificial insemination. At the start of the breeding season heifers were observed twice daily and inseminated 12 hours after first being observed in estrus. On the morning of the sixth day of the breeding season all heifers not yet observed in estrus or inseminated were treated with prostaglandin (25 mg Lutalyse(R)). Observation was continued and heifers were bred 12 hours after first observed in estrus. Of the 129 heifers 33 were inseminated before prostaglandin treatment, the remainder of the heifers were bred within 4 days of treatment. Two heifers developed cysts and did not express estrus. First service pregnancy rates were 70% before treatment and 79% after treatment. All heifers were inseminated the first time within the first 10 days of the breeding season and 100 pregnancies occurred in the first 10 days of the season. As a result median days to pregnancy was 8. Calves were all weaned and weighed at approximately 150 days of age. These results were then used to model the economic impact of estrus synchronization of beef heifers on the increased weaning weights of their calves. Prostaglandin treatment altered the distribution of pregnancies and resulted in a cluster of conceptions from Day 7 to 10 of the breeding season with no conceptions from Day 11 to 19. In order to study the influence of estrus synchronization on weaning weights a simulation model was constructed. All heifers that conceived on the first service after prostaglandin were assigned an alternate date of conception from Day 7 to 21 of the breeding season with a random number generator. In this way a uniform random distribution of pregnancies over the first 21 days of the breeding season was constructed. An alternate calving date was then estimated using the known gestation length and this date was used to calculate the weaning weight of the calf using the known weight per day of age. The average weaning weight of the uniform random distribution was then compared to the synchronized distribution with Student's t -test. The simulation model was repeated 10 times and each time weaning weights of the uniform random distribution were significantly less than with the synchronization program (p<.01). At a value of $4.00 for prostaglandin and $1.76 for each kilogram of calf the average increase of 3.8 kgs in weaning weight per calf resulted in a return on investment of $1.92 for every dollar invested in prostaglandin.

Effect of service number on estrus duration in dairy goats.

The object of this research was to study the effect of sterile service number on estrus duration in dairy goats. Twenty-four Nubian goats (20 nulliparous and 4 multiparous) were randomly assigned to 1 of 4 treatment groups (n = 6 animals per group). The following Groups were formed: no service (GS-0); 1 service (GS-1); 2 services (GS-2); 3 services (GS-3). Estrus was synchronized by using fluorogestone acetate intravaginal pessaries (40 mg) over a 12-d period plus 400 IU im pregnant mare serum gonadotropin (PMSG) at pessary removal. Estrus was detected by using a vasectomized buck at 6-h intervals over 5 d after pessary removal (at 0600, 1200, 1800 and 2400 h). In the GS-0 group the teaser was outfitted with an apron and was permitted to mount. In the GS-1, GS-2 and GS-3 groups, the teaser was permitted to mount and service 1, 2 and 3 times, respectively, within the first 12 h after initiation of estrus. The duration of estrus for the 4 groups (GS-0, GS-1, GS-2 and GS-3) was (mean +/- SD) 41.0 +/- 5.9, 24.0 +/- 5.4, 22.0 +/- 4.9 and 22.0 +/- 7.2 h, respectively. These results show differences between the serviced groups and the nonserviced group (P<0.01), but they fail to show differences among the serviced groups (P>0.05). It is concluded that sterile service shortens estrus duration and that service number (1, 2 or 3) does not affect estrus duration.

Opposite regulation of XIAP and Smac/DIABLO in the rat endometrium in response to 17beta-estradiol at estrus.

During rat estrous cycle, the endometrium proliferates in response to sex steroids and specific endometrial epithelial cells undergo apoptosis in absence of embryonic factors. The central executioner of apoptosis is a family of aspartic acid-specific cysteine proteases known as caspases. Smac/DIABLO is released from the mitochondria during apoptosis and its stimulation promotes caspases activation by neutralizing members of the inhibitor of apoptosis proteins (IAPs) family, such as X-linked inhibitor of apoptosis protein (XIAP). The aim of this study was to investigate the involvement of Smac/DIABLO and XIAP in the control of caspases activation in endometrium of cycling rats. Polyoestrus female rats were sacrificed at each stage of estrous cycle (diestrus, proestrus, estrus, and metestrus). Endometrial protein extracts were collected to perform Western Blot analysis. Alternatively, uterine horns were sectioned for immunohistochemistry (IHC). We and others showed previously the presence of apoptosis at estrus in rat uterine epithelium. In the present study, cleaved caspase-3, -6, and -7 fragments were detected at estrus. IHC confirmed that caspase-3 was present only in luminal and glandular epithelium at estrus. XIAP was highly expressed at estrus in both epithelial and stromal cells. In contrast, expression of Smac/DIABLO was elevated at diestrus, proestrus and metestrus but was minimal at estrus. Treatment of ovariectomized rats with 17beta-estradiol induced XIAP expression and inhibited Smac/DIABLO protein expression in the endometrium. Cleaved caspase-3, -6, and -7 fragments increased in endometrial protein extracts following 17beta-estradiol treatment. Expression of NF-kappaB and IkappaB proteins, and IkappaB phosphorylation status were detected in the endometrium but were not influenced by the estrous cycle. These findings suggest that Smac/DIABLO and XIAP are regulated differently and may play important roles in the regulation of endometrial cell fate. Moreover, this study confirms a key role for executioner caspases in the control of apoptotic processes at estrus in the rat uterus.

Factors affecting walking activity at estrus during postpartum period and subsequent fertility in dairy cows.

The postpartum period is critical for efficient reproduction, and certain features of the normal estrus cycle during this period have been associated with subsequent reproductive performance. The objectives of the present investigation were to evaluate the effects of environmental, production, and reproductive factors on walking activity at estrus as determined using pedometers during the first 50 days in milk (DIM) (study 1). In a second study, we tried to establish whether pedometer measurements recorded during this period could be used to predict subsequent fertility by considering the number of cows becoming pregnant before 90 DIM. We analyzed data derived from 995 parturitions in a single herd. Detection of estrus was performed using a pedometer system. Variables were screened for associations with walking activity by analysis of variance (ANOVA) through generalized linear model procedures (PROC GLM). Increased milk production and parity, and a mean relative humidity (RH) higher than 95% were associated with lower pedometer measurements. A higher number of animals simultaneously in estrus rendered higher pedometer readings. No significant effects of the year, season, DIM, number of previous estruses, and climatic data other than high mean RH were observed. Relationships between pedometer measurements and other variables recorded during the first 50 days postpartum, and subsequent fertility were assessed by applying logistic regression models. We detected no significant effects of year, milk production, season, and mean activity increase at estrus on high fertility. The likelihood of pregnancy before 90 DIM decreased for each additional lactation and for cows in anestrus between days 0 and 50 postpartum. However, pedometer readings during the first 50 days postpartum were unable to accurately predict subsequent fertility.

Steroid priming shortens prostaglandin-based estrus synchronization program from 14 to 7 days in cattle.

Single injection of estrogen and progesterone before prostaglandin (steroid priming) was used to shorten the prostaglandin-based estrus synchronization program. Sixty-five cyclic Sistani cattle, with parity ranging from 1 to 4 and postpartum period of >80 days were selected at unknown stages of the estrous cycle and assigned to 2 groups according to their age, weight and parity. Females in the control group (n=33; 58.4 +/- 4.3 months; 277 +/- 8 kg LW) received two consecutive injections of prostaglandin F2alpha analogue (500 microg; Cloprostenol, PG) 14 days apart (Day 0 = First PG injection). On Day 7, treated females (n=32; 60 +/- 4.8 months; 292 +/- 9 kg LW) were given an intramuscular injection of 100 mg progesterone and 2 mg estradiol benzoate followed by prostaglandin 7 days later, concurrent with the second PG injection of the control group. Estrus detection was carried out every 6 hours for 7 days, commencing from 24 hours after the last PG injection. Females that allowed to be mounted were identified (standing estrus) and inseminated with frozen semen 12 hours later. Pregnancy was diagnosed on Day 50 after AI through palpation per rectum. Data were analyzed using Chi-squared and t-test. The tightness of estrus synchrony (%), the interval from the end of treatment to estrus (h) and conception rates (%) were similar (P > 0.05) between control (69.6%, 77.7 +/- 5.96 h and 56.5%) and treatment (68.2%, 82.6 +/- 7.64 h and 54.5%) groups. In conclusion, steroid priming is an efficient way to shorten the prostaglandin-based estrus synchronization program from 14 to 7 days without compromising estrous response and fertility.

Comparison of synchronization of ovulation and induction of estrus as therapeutic strategies for bovine ovarian cysts in the dairy cow.

The benefit of using timed-insemination in lactating dairy cows for the treatment of ovarian cysts lies in the fact that cows do not have to be detected in estrus for insemination and achieving pregnancy. We compared the effectiveness of synchronization of ovulation with timed-insemination and induction of estrus with insemination at estrus in the treatment of bovine ovarian cysts in lactating dairy cows. After Day 65 post partum, a total of 368 lactating dairy cows was divided into 3 groups. Cows in Group 1 (n = 209, normal, noncystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and 100 ug, i.m. GnRH on Day 9 and then were time-inseminated 16 h later. Cows in Group 2 (n = 76, abnormal, cystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and 100 ug, i.m. GnRH on Day 9 and time-inseminated 16 h later. Cows in Group 3 (n = 83, abnormal, cystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and inseminated at induced estrus within 7 d after treatment with PGF2 alpha. Day 0 was the day of initiation of the study. Conception and pregnancy rates among groups were compared using logistic regression and adjusted for parity, time of year and days in milk. Conception and pregnancy rates of Group 1 cows (31.5%) were not significantly different from those of Group 2 cows (23.6%). However, the pregnancy rate in normal cows (Group 1) was higher (P < 0.01) than in cystic cows (Groups 2 and 3). Cows in Group 3 had a higher conception rate than cows in Group 2 (51.7% > 23.6%; P < 0.01). However, pregnancy rates for cows in Groups 2 (23.6%) and 3 (18%) were not significantly different. The finding indicated that synchronization of ovulation and timed-insemination resulted in pregnancy rates similar to those of synchronization of estrus and insemination at an induced estrus within 7 d for the treatment of ovarian cysts in lactating dairy cows.

Four years of induction/synchronization of estrus in dairy goats: effect on the evolution of eCG binding rate in relation with the parameters of reproduction.

Ninety-eight Alpine goats of two herds were followed over 4 years in a program of annual artificial insemination after estrus induction/synchronization, including progestagen administration (vaginal sponge) followed by prostaglandin analog and equine chorionic gonadotrophin (eCG) 48 h before sponge removal. Goats were sampled every 4 hours from the 16th to the 56th following sponge removal, for determination of LH surge and tested for estrus by the presence of a buck. Seven days after AI, endoscopic examination of the ovaries was performed to determine the number of corpus lutea. Pregnancy diagnosis was performed at day 21-22 post AI by determination of plasma progesterone and at day 40-45 by ultrasonography. Parturition, number and sex of kids were recorded. All the goats were sampled before and after each treatment, for anti-eCG antibodies screening. Statistical analysis of the results clearly established a significant effect of the treatments on anti-eCG antibodies. Time of estrus and LH surge were significantly different between herd. The antibodies significantly delayed the time of coming out of estrus as well as the time of LH surge. Two antagonistic effects were evidenced: first, the delayed of time of estrus and time of LH surge in relation with the immune reaction to eCG; secondly, the ahead of time of estrus and time of LH surge during the years of treatment, identical to both herd. The antibodies negatively influenced the percentage of ovulating females as well as kidding rate. Finally, no effect of antibodies on prolificacy was found.

Effect of a prostaglandin F2 alpha injection 96 hours after introduction of intact bulls on estrus and calving distribution of beef cows.

Most estrus synchronization schemes facilitate the use of artificial insemination; however, combining estrus synchronization with natural service also offers several benefits to beef producers. This experiment was designed to study the effectiveness of one injection of prostaglandin F2 alpha (PGF2 alpha) 96 h after introduction of intact bulls on estrus synchronization and calving distribution of beef cows and heifers. Three hundred sixty beef cows and 45 beef heifers in seven trials were allotted by age, breed, and prior calving date to one of two treatments. Within each subclass, one group was injected with an appropriate dose of PGF2 alpha and the second group at each location received 5 ml of saline (SAL). Cows were classified as estrual or anestrual at the start of breeding based on serum progesterone levels and were observed for behavioral estrus regularly during the first 25 d of the breeding season in two of the trials (d 0 = day before bull turn-in). Pregnancy data and calving dates were recorded in each of the studies. All cows were palpated per rectum for pregnancy approximately 70 d after the introduction of bulls. During d 5 to 9 after treatment, the percentage of cows exhibiting behavioral estrus (P = .02) and the pregnancy rate (P = .02) obtained was greater for PGF2 alpha-treated cows than for SAL-treated cows. There were 6.5% more (P = .05) calves born during the overall calving period from animals treated with PGF2 alpha compared with those in the SAL group. In summary, treatment of cows and heifers with PGF2 alpha 96 h after bull turn-in was effective in synchronizing behavioral estrus of beef cows and heifers and increasing the calving rate during the subsequent calving season.

Influence of estrus on somatic cell count in dairy goats.

The effect of estrus on the somatic cell count (SCC) of goat's milk was examined by inducing estrus in 24 of 48 seasonally anestrus, lactating dairy goats. Goats were blocked by infection status and ranked on SCC from three preceding herd tests and randomly allocated (within block) to the following three treatment groups: a) "Short," in which an intravaginal progesterone-releasing device was inserted for 12 d plus equine chorionic gonadotropin and dinoprost tromethamine 2 d before device removal (n = 12), b) "Long," in which an intravaginal progesterone-releasing was inserted for 17 d plus equine chorionic gonadotropin on the day of device removal (n = 12), or c) "Control," in which the goats were left as untreated controls (n = 24). Bacteriological status of each gland of each goat was determined before and after synchronization (d -23 and +13) and SCC and milk volumes were determined on d -2, 0, 1, 2, 3, 4, 14, and 25, where d 0 was the day of intravaginal device removal. Goats in the Short group were in estrus before those of the Long group, who were, in turn, in estrus before the Control group. The log10 and log10 absolute SCC (SCC cells/ml x volume) were higher in the Short than in the Control group on d 1, 2, 3, and 4, whereas those of the Long group were higher than those of the Control group on d 2 and 4. These data indicate that estrus resulted in an increase in SCC, and that the increase in SCC was independent of the decline in milk volume at estrus.

Timed artificial insemination with estradiol cypionate or insemination at estrus in high-producing dairy cows.

A total of 799 Holstein cows from 3 herds were randomly assigned at 37 +/- 3 d in milk (DIM) to timed artificial insemination (AI) or insemination at detected estrus. Cows were presynchronized with injections of PGF(2alpha) at 37 and 51 DIM. At 65 DIM, cows received an injection of GnRH, followed 7 d later by PGF(2alpha). Cows in the estrus-detected group were inseminated after being observed in estrus during the 7 d after the last PGF(2alpha). Cows in the timed AI group received an injection of 1 mg of estradiol cypionate (ECP) 24 h after the last PGF(2alpha). If detected in estrus or=1 ng/mL; L = <1 ng/mL), resulting in 8 combinations (LLL, LHL, LLH, LHH, HHH, HHL, HLH, and HLL). Conception rates and pregnancy rates were higher for cows in the timed AI group than in the estrus-detected group at 30, 44, and 58 d (e.g., at 58 d, pregnancy rates were 42.2% for multiparous cows or 34.4% for primiparous cows in the group receiving ECP and timed AI compared with only 20.8 or 18.8% for respective parity subgroups for the treatment group inseminated only at detected estrus). Pregnancy losses were 11.5% from 30 to 58 d and did not differ between treatments. Cyclic cows within both treatments had higher estrous responses, conception rates, and pregnancy rates. Cows that responded to presynchronization and to luteolysis (HHL) had the highest conception and pregnancy rates, followed by cows classified as LHL. Use of 1 mg of ECP to induce ovulation as part of a synchrony regimen improved reproduction at first postpartum insemination in dairy cows.

Stage of estrous cycle, time of insemination, and seasonal effects on estrus and fertility of Holstein heifers after prostaglandin F2 alpha.

Stage of estrous cycle, time of insemination, and season of the year were examined for their influence on estrus and fertility of 223 Holstein heifers after prostaglandin F2 alpha (treatment). Heifers treated during early stages (days 5 to 8) of the estrous cycle had shorter (by 11 h) and less variable intervals to estrus compared with treatment of heifers during late stages (days 14 to 16) of the estrous cycle. A greater proportion of heifers in the early cycle treatment (79/81, 97%) were in synchronous estrus (20-h period) after treatment compared with the late cycle treatment (71/86, 83%). Conception rates were lower when early cycle heifers were inseminated at 80 h after prostaglandin F2 alpha than when inseminated according to estrous detection and also lower as compared with late cycle heifers inseminated at 80 h. No differences in fertility were observed for late cycle heifers inseminated at 80 h compared with those inseminated according to estrous detection. Season of the year had no influence on intervals to estrus or on proportion of heifers expressing estrus after prostaglandin F2 alpha, regardless of stage of estrous cycle. Conception rate during winter, however, was higher in early but not late cycle heifers inseminated after prostaglandin F2 alpha, as compared with fall or spring. Variability of interval to estrus and of fertility after inseminations by appointment at 80 h appears to be due to stage of estrous cycle at time of prostaglandin F2 alpha treatment rather than season of the year.

Mating of captive thirteen-lined ground squirrels and the annual timing of estrus.

Thirteen-lined ground squirrels (Spermophilus tridecemlineatus) have a single annual mating season in Michigan, beginning shortly after their spring emergence from hibernation. Captive females were studied during a 3-year period to determine relations among time of removal from the coldroom, vaginal estrus, and mating behavior. Following a 7-month period females spent in a coldroom, vaginal lavages were taken daily to monitor changes in estrous condition. Females removed from the coldroom about when free-living animals emerge from hibernation were in vaginal estrus within 24-48 hr and had an initial period of persistent estrus (ca. 3 weeks), followed by briefer (less than 1 week) and more sporadic estrous periods. Females left in the coldroom 3 weeks longer than normal had significantly briefer initial periods of vaginal estrus after being removed from the coldroom. Similarly, virgin Yearlings and virgin 2-Year-Olds had significantly briefer initial periods of estrus than nonvirgin Adults (greater than or equal to 2 years old). In 1985, eight of eight females paired with males mated within the first week after removal from the coldroom and subsequently produced litters. Mated females remained in vaginal diestrus from within a few days of mating until after parturition. In contrast, unmated females remained in prolonged vaginal estrus during this period. Females first paired with males 3 weeks after being taken from the coldroom failed to mate. In 1986, five of six females first paired with males 2 weeks after being removed from the coldroom similarly failed to mate. However, five of six females did mate that were removed from the coldroom 10 days after those in the previously described group and paired with a male 4 days after removal. This first report of reliable mating behavior in captive thirteen-lined ground squirrels should facilitate subsequent analysis of reproductive patterns in this species.

Estrus- and steroid-induced changes in circadian rhythms in a diurnal rodent, Octodon degus.

Diurnal Octodon degus exhibited marked alterations in activity and temperature in conjunction with the 3 wk estrous cycle when housed in LD12:12 light cycle. On the day of estrus, mean daily activity increases 109%, mean core temperature rises .4 degree C, activity onset is advanced 2 h, and amplitudes of both rhythms decline compared with the 3 days prior to estrus. On the day following estrus, activity onset was delayed 4.9 h, and mean activity and core temperature fell below that of the preestrus period. Ovariectomy significantly reduced mean temperature (.98 degree C) but did not significantly alter mean activity, and eliminated cyclic effects of estrus. Estrogen replacement led to a nonsignificant elevation in mean activity and core temperature with no change in the phase angle of entrainment. Progesterone replacement significantly reduced mean core temperature and mean activity, while only the phase angle difference between temperature minimum and activity onset was significantly altered. Intact degus maintained in constant darkness displayed only transient fluctuations in activity onset and temperature minimum during and after estrus. Estrogen or progesterone treatment of ovariectomized, free-running degus altered mean temperature and activity levels, but did not influence tau. Changes in phase angle of entrainment during estrus are not the result of hormone effects on the circadian clock but likely reflect increased or decreased levels of activity.

Comparison of three estrus detection systems during summer in a large commercial dairy herd.

The objective of the study was to compare three systems for estrus detection and combinations of these systems on a large commercial dairy (1075 lactating cows) during stress of summer heat. At 37-45 days in milk (DIM), 255 cows were fitted with a HeatWatch device (HW; DDx Inc., Denver, CO), an activity sensor ALPRO (ALPRO; DeLaval Inc., Kansas City, MO), and visually observed (VO) three times daily. Pregnancy status was determined by uterine palpation per rectum 35-49 days following artificial insemination (AI). Effects of DIM, parity, standing events, inseminator, and interval between onset of estrus and AI on conception rates were determined using logistic regression. Efficiencies for detection of estrus, determined by comparing detected periods of estrus with a theoretical total of 570 periods, were 49.3% (VO), 37.2% (ALPRO), 48.0% (HW), and 80.2% for all three systems simultaneously. Conception rates (LSM+/-S.E.) for cows detected by one or more of the three systems were 6.2+/-3.9 for VO, 19.8+/-5.6 for ALPRO, 17.3+/-5.0 for HW, 22.8+/-7.0 for VO+ALPRO, 26.9+/-4.6 for VO+HW, 23.2+/-5.2 for ALPRO+HW, and 18.4+/-4.7 for VO+ALPRO+HW. Inseminations performed during no and mild heat stress (temperature-humidity index; THI< or =76) had greater conception rate (P<0.05; 38.8%) compared to AI performed during moderated heat stress conditions (THI>76; 17.6%). Number of mounts were higher for primiparous versus multiparous cows (P<0.05). Cows over 80 DIM during estrus exhibited fewer (P<0.05) standing events. The highest conception rate occurred with the combination of VO+HW, which confirms the premise that combination of multiple systems enhances both the efficiency and accuracy of estrus detection.

Spine-type densities of hippocampal CA1 neurons vary in proestrus and estrus rats.

Spines-mediated synaptic activity has been associated to learning ability. Dendritic spines from hippocampal CA1 pyramidal neurons of proestrus rats have been reported to be more numerous than in estrus animals, but some behavioral studies have reported a better performance during the estrus stage of the estrous cycle. Because spine shape has been shown to be strongly related to the post-synaptic processing of information, a quantitative morphological study related to the proportional density of each type of spine, was conducted in Golgi material of hippocampal CA1 pyramidal cells of proestrus and estrus rats. After three regular estrous cycles had been asserted, seven Sprague-Dawley female adult rats in proestrus and eight in estrus, were used. Mushroom-shaped spines from hippocampal pyramidal cells predominated in proestrous rats, being 15.1% more numerous in this stage than in estrus; while thin spines were the predominant type of spine in estrous animals, being 15.5% more abundant in estrus than in proestrus. The predominance of the mushroom-shaped or thin spines in the hippocampal CA1 pyramidal neurons during the respective stages of the rat estrous cycle, could be related to the organization of the hippocampal activity-dependent mnemonic information.

1-Iodo-2methylundecane, a putative estrus-specific urinary chemo-signal of female mouse (Mus musculus).

The present study was designed to identify the urinary volatiles across various reproductive stages of female mice in order to detect estrus-specific chemical signal. The urine of adult female mice were extracted with dichloromethane (1:1 ratio, v/v) and analyzed by gas chromatography linked mass-spectrometry (GC-MS). Numerous compounds were identified during estrus cycle of female mice urine. Among these, the compounds, namely, isocroctylhydrazine, 4-methyl-2-heptanone and auzulene were specific to proestrus stage and the compounds, 1-H-cyclopop-e.auzulene, caryophyllene, copanene were specific to estrus stage. However, the compound, 1-iodo-2methyl undecane (1I2MU) was observed both in proestrus and estrus phases and was absent in all other phases. The volatile signal produced at the end of proestrus and the beginning of estrus phase appears to be behaviourally important in the attraction of males. Moreover, the behaviour assay revealed that the compound, 1I2MU, is involved in attracting the male mice. This result concludes that the 1-iodo-2methyl undecane is considered as a putative estrus-specific chemo-signal.

Induction of follicular wave emergence for estrus synchronization and artificial insemination in heifers.

The objective was to synchronize follicular wave emergence among cattle for synchronization of estrus and ovulation, and to determine pregnancy rate after AI at observed estrus. At random stages of the estrous cycle, a controlled internal drug release device (CIDR-B) was inserted intravaginally (Day 0) in 67 cross-bred beef heifers, and they were randomly allocated to receive either no further treatment (Control; n = 18); 5 mg of estradiol-17beta and 100 mg of progesterone im (E/P; n = 16); 100 microg im of GnRH (GnRH; n = 16); or transvaginal ultrasound-guided follicular ablation of all follicles > or = 5 mm (FA; n = 17). All heifers received a luteolytic dose of PGF (repeated 12 h later), and CIDR-B were removed on Days 9, 8, 6 or 5, in Control, E/P, GnRH or FA groups, respectively, so the dominant follicle of the induced wave was exposed to exogenous progesterone for a similar period of time in each group. Mean (+/- SEM) intervals (and range, in days) from treatment to follicular wave emergence in these groups were 3.5 +/- 0.6 (-2 to 8), 3.4 +/- 0.1 (3 to 4), 1.5 +/- 0.3 (-1 to 4), and 1.0 +/- 0.1 (0 to 2), respectively. Although the interval was longest (P<0.01) in the E/P and Control groups, it was least variable (P<0.01) in the E/P and FA groups. Intervals (and range, in days) from CIDR-B removal (and first PGF treatment) to estrus were 2.3 +/- 0.2 (1.5 to 4.5), 2.2 +/- 0.2 (1.5 to 3.0), 2.1 +/- 0.1,(1.5 to 3.5), and 2.5 +/- 0.1 (2.0 to 3.5), and to ovulation were 3.5 +/- 0.2 (2.5 to 5.5), 3.4 +/- 0.1 (3.0 to 4.5), 3.5 +/- 0.1 (2.5 to 4.5), and 3.8 +/- 0.1 (3.0 to 4.5), for Control, E/P, GnRH and FA groups, respectively (ns). The proportion of heifers displaying estrus was higher in the Control than in the FA group (94% versus 65%, P<0.05) and intermediate in EP and GnRH groups (87% and 75%). Heifers were inseminated approximately 12 h prior to ovulation (based on estrous behavior and ultrasound examinations). Pregnancy rates were 78%, 80%, 69% and 65% for Control, E/P, GnRH and FA groups, respectively (P=0.73). Results support the hypothesis that synchronous follicular wave emergence results in synchronous follicle development and, following progesterone removal, synchronous estrus and ovulation with high pregnancy rates to AI. The synchrony of estrus and ovulation in the E/P, GnRH and FA groups suggest that these treatments, in combination with CIDR-B, could be adapted to fixed-time insemination programs.

Lack of an effect of prostaglandin injection at estrus onset on the time of ovulation and on reproductive performance in weaned sows.

We evaluated the effects of a PGF2alpha analogue on time of ovulation and reproductive performance in multiparous Camborough sows (n=47). At onset of first post-weaning estrus, sows received either an intravulval injection of 3.75 mg of prostaglandin analogue (PGF) or, served as a non-injected control (CON). Beginning 24 h after the onset of estrus, transcutaneous ultrasonography was carried out every six h to determine time of ovulation. At 36, 54, and 72 h after the onset of estrus, blood samples were taken for progesterone analysis. Weaning-to-estrus (WEI), duration of estrus, ovulation rate and number of live embryos at d 28 of gestation were recorded. Treatment had no effect (P > 0.05) on any parameters measured. Duration of estrus classified as less or greater than the overall mean also had no effect (P > 0.05) on any of the parameters measured. Results indicate that treatment did not advance ovulation nor did it improve reproductive performance in sows. Overall, a negative correlation of WEI with the ovulation rate (P = 0.0005, r = -0.54) was established.
