[Study on the method of adsorbing albumin for enwrapping human hard tissue substitute biomaterials].

The aim of this study was to cover up the surface of human hard tissue substitute biomaterials with albumin. The cidex cross-link method was adopted to adsorb albumin so as to form a membrane for wrapping hydroxyapatite(HA), bio-glass ceramics(BGC), and hydroxyl poly-calcium sodium phosphate (HP). The results showed that this membrane of protein could enwrap the biomaterials so firmly that urea solution could not wash it off.

[New technologies in the prevention, detection, diagnosis and treatment of tuberculosis in children].

If the epidemiological situation is tense, new technologies should be developed and put into practice to enhance the efficiency of specific prevention, early detection, diagnosis and treatment of tuberculosis in children. There is evidence for the high efficacy and low reactogenicity of lower antigenicity-loading BCG-M vaccine that causes a 15-fold decrease in infant morbidity, as compared with that among non-vaccinated children, and this vaccine shows a 5-fold reduction in postvaccination complications as compared with BGC vaccine. The 26-year use of tuberculin diagnosis via Mantoux test with 2TE PPD-L during mass vaccination of children and adolescents has proved itself in early identification of tuberculosis and risk groups. A new risk group has been identified. This includes children with increasing tuberculin reactions; three-month intermittent chemoprevention with isoniazid reduces tuberculin sensitivity in the children and prevents tuberculosis in them. The developed short-term (6-9 months) courses of chemotherapy in preschool and school children by using drugs (isoniazid + rifampicin + pyrazinamide) yield the best healing without residual changes of uncomplicated forms of tuberculosis in 83 and 60% of the children with complicated events, respectively. The chemotherapy regimens have been divided into 4 groups of different dosage schedules.

[The antitumor effect of adriamycin conjugated with monoclonal antibody against gastric cancer in vitro and in vivo].

Adriamycin (ADM), an anthracycline cytotoxic agent, was conjugated with monoclonal antibody 3H11 against gastric cancer via the dextran bridge method. The conjugate 3H11-DEX-ADM, with molar ratio of 3H11 to ADM being 1:73, retained antibody activity to 86%. In the cytotoxicity assay, 3H11-DEX-ADM was shown to exhibit increased cytotoxicity against the target cell line BGC 823. Its IC50 was 3.75 fold less than that of free ADM. The antitumor effect of the conjugate was evaluated in tumor-bearing nude mice. The results indicate that the specific antibody conjugate 3H11-DEX-ADM can significantly inhibit the tumor growth. At the dosage level used in the present study (5 micrograms/mouse x 6), 3H11-DEX-ADM showed an inhibition rate of 51.5%, whereas only moderate inhibition rates were observed with free ADM and the control conjugate NIgG-DEX-ADM. In addition, experiment was performed to evaluate the combined cytotoxicity of 3H11-DEX-ADM and the conjugate of mitomycin C (3H11-HSA-MMC) at different ratios. It was shown that the combination has no synergistic effect when their IC50 was compared with that of the two conjugates used alone. The same result was observed on combinations of the two corresponding free drugs.

[Influence of tumor cells on endothelial cells in vitro].

OBJECTIVE: To observe in vitro changes of endothelial cells after confrontation with tumor cells. METHODS: Dynobeads were used to isolate the endothelial cells from the rat lung. Mouse dendritic cell sarcoma cells (DCS), human gastric carcinoma cells (BGC-823) and mouse lung adenocarcinoma cells (LA795) were added to the endothelial cells when the latter was at the confluence phase. Phase contrast microscope, scanning electro-microscope, immunohistochemistry, transwell and fluorescence dye transfer were used to detect morphological and functional changes of the endothelial cells. RESULTS: Endothelial cells may look like cobble stones or long spindle shaped. Direct contact of tumor cells with endothelial cells induced round vascular-like space formation between confluent endothelial cells. Tumor cells were often found at the newly appeared spaces. Tumor cell conditioned medium could support the growth and promote the locomotion of endothelial cells through transwell. It was observed that luciffer yellow was directly transported from tumor cells to endothelial cells. CONCLUSIONS: Tumor cells can directly induce morphological and functional changes in endothelial cells. Direct intercellular communication between tumor cells and endothelial cells is present.

[Production of gamma-interferon by blood mononuclear cells in patients with various types of a tuberculous process].

To elucidate the diagnostic value of gamma-interferon (gamma-IFN) in the human body, the gamma-IFN production that was spontaneous and induced by BCG Mycobacterium bovis in the supernatants of short-term cultures of peripheral mononuclear cell from 31 patients with active tuberculosis was studied. The sandwich method of enzyme immunoassay was used, by employing two monoclonal antibodies in the avidin-biotin system. The studies have established the low spontaneous production of gamma-IFN in patients with the severe progressive course of a tuberculous process (22.5 +/- 2.5 pg/ml in Group 1 versus 41.5 +/- 8.6 pg/ml in patients with the favorable course of a tuberculous process (Group 2) and 34.0 +/- 3.5 pg/ml in healthy individuals (Group 3); p(1-2) < 0.05 and p(1-3) < 0.05)). Moreover, there was a reduction in the induced production of gamma-IFH in response to BGC (37.7 +/- 33.4 pg/ml in Group 1 versus 214.9 +/- 81.9 pg/ml in Group 2 and 206.0 +/- 31.1 pg/ml in healthy individuals; p < 0.05 and p(1-3) < 0.001), which is of diagnostic and prognostic values.

[Study on the acute toxicity experiment of mice and anti-tumor function in vitro of the qinglongyi].

OBJECTIVE: To study the anti-tumor effect and acute toxicity in vitro of the separation compositions from qinglongyi. METHOD: The conventional acute toxicity experiments of mice, the MTT [3-(4,5-dimethylthiazol-2 yl)-2,5-diphenylterazolium bromide, MTT] and the SRB (sulforhodamin B) were used to make preliminary selection to qinglongyi and its separation compositions. RESULT: The half-deadly dose (LD50, the half-lethiferous dose) of the chloroform separation composition in qinglongyi was 575.38 mg x kg(-1) (i.g.), and of the acetic ether separation compositions in qinglongyi was 1303.59 mg x kg(-1) (i.g.). From the other parts of separation compositions in qinglongyi, the LD50 were more than 5 g x kg(-1). When acetic ether separation composition of qinglongyi was at 100 microg x mL(-1) the growth inhibitory rate (GIR) was < 50% to the leukaemia cell HL-60 of human. When chloroform separation composition and the acetic ether separation compositions of qinglongyi was at 100 mg x mL(-1), GIR was 52% to the leukaemia cell HL-60, the gastric carcinoma cell BGC-823 and the cervical carcinoma cell Hela of human. CONCLUSION: The separation composition of the chloroform and the acetic ether from qinglongyi have obvious, anti-tumor effect.

[Antibodies to mycobacterial antigens in patients with pulmonary tuberculosis].

The levels of IgG and IgM antibodies to different mycobacterial antigens were studied in patients with varying pulmonary tuberculosis. The higher levels of IgG and IgM to the used antibodies were simultaneously found in patients with infiltrative tuberculosis as compared with healthy individuals. As compared to patients with fibrocavernous pulmonary tuberculosis, those with infiltrative tuberculosis had significantly lower levels of IgG antibodies to BGC sonicate and protein with a molecular weight of 40-70 kD isolated from the strain H37Rv. A correlation was found between the IgM antibodies to protein isolated from the strain H37RV and to H37RV sonicate and the degree of the activity unique to fibrocavernous pulmonary tuberculosis.

[Tuberculosis in children from the Sverdlovsk Region].

The tuberculosis morbidity rate in BGC-unvaccinated infants is 18 times greater than that in BCG-vaccinated infants and 6 times higher than the rate of complications due to BCG vaccination. If a scar due to BCG vaccination is absent or 1-2 mm, the tuberculosis morbidity in children of 3 years of life is 7 times higher than when a 3-mm or more scar has formed after vaccination. The technological failures are main causes of complications due to BCG vaccination. The type of a vaccine (BCG-1 or BCG-M) does not play a substantial role in the development of complications.

[Mammalian gene-transfer and expression efficiencies of baculovirus bacV-CMV-EGFPA].

It has been reported that baculoviruses could serve as a new gene-transfer vehicle for mammalian cells. We have previously constructed recombinant baculovirus BacV-CMV-EGFPA and have proven that mammalian cells could be effectively infected by the recombinant baculovirus. In this report, we studied the efficiency of baculovirus to deliver exogenous gene into twenty mammalian cells, including twelve human cell lines (WI-38, Hela, HepG2, 293, PLC/PRF/5, 143B, MCF-7, BGC-223, DMS 114, CNE, Raji, LCL-cm), seven murine cell lines (BNL 1ME A.7R.1, CHO-K1, L-929, JC, PT67, NIH3T3, P815) and one monkey cell line (CV1). Results showed that most mammalian cell lines could be transduced by the recombinant baculovirus, the transduction efficiencies of the human and monkey cell lines were markedly higher than that of murine cell lines, and the transduction efficiencies in adherent culture cell lines higher than that of suspend culture cell lines, implying that the infection efficiency of the baculovirus may be correlative with the organism used and the growth properties of the cell lines. The plasmid pcDNA3. 1-EGFP, which contains the CMV promoter and EGFP reporter gene, was next transfected by LipofectAMINE into a number of mammalian cells, especially those cells that were low in the baculovirus transfection. Results showed that the CMV promoter could effectively direct the expression of the reporter gene in these mammalian cells. Therefore the gene-expression efficiencies in different mammalian cell lines by the recombinant baculovirus which contains the same CMV promoter were dictated by the ability of the baculovirus to enter the cell lines. This study suggested that the recombinant baculovirus vector is more suitable for gene expression in primate adherent culture cells than in murine cells and suspend culture cells.

[Anticancer spectrum of pingyangmycin in vitro].

Pingyangmycin (PYM), produced by Streptomyces pingyangensis n. sp., was found to be identical to bleomycin A5. In the present study, a comparative observation was carried out in 10 human cancer cell lines. As determined by a colony-forming assay, the dose-response curves obtained from cells exposed to PYM for 1 h were of one type only: biphasic exponential. The sensitivities of these cells derived from different types of tumors, however, varied with a broad range of ID50 values (0.03-0.82 microgram/ml). A hepatoma cell line (BEL-7402) and three lines derived from squamous carcinomas of the esophagus (Eca109 and CaEs17) or the nasopharynx (CNE) were relatively sensitive (ID50 less than 0.20 microgram/ml) to PYM which is known to have clinical activity against these diseases. Two gastric adenocarcinoma cell lines (MGc80-3 and BGC-823) and a pulmonary adenocarcinoma cell line (SPC-A-1) appeared to be less sensitive to the drug, with ID50 values of 0.21-0.47 microgram/ml. PYM was 7-fold more effective against LTEP-78 cells derived from pulmonary squamous carcinoma as opposed to SPC-A-1 cells, resulting in a low ID50 value of 0.04 microgram/ml. However, PYM as a single agent has not yet received full evaluation in relation to this type of lung cancer. In contrast with other cell lines of squamous cancer origin, HeLa and CC-801 cells derived from uterine cervix carcinomas which have been evaluated as highly responsive to PYM had the highest ID50 values (greater than 0.70 microgram/ml).

Quantitation of HLA antigens on red cells by immunoradiometric assay.

Immunoradiometric assay was employed to quantitate HLA antigens on red cells. Using this technique HLA-B7, HLA-B17 and HLA-A28 were detected on the red cells of all individuals studied irrespective of the serological status of the Bennett-Goodspeed (Bg) antigens. However, HLA antigenic sites for serologically Bg positive red cells were significantly more than that for Bg negative red cells (P less than 0.001). Bga positive red cells possessed maximum number of antigenic sites as compared to Bgb and Bgc positive red cells.

[The role of BCG in the prevention of tuberculosis].

Tuberculosis is still a major international public health problem. The role of vaccination with BCG in the prophylaxis of the tuberculosis has been a source of controversy almost since ist introduction in the thirties, because of the disparity of the results regarding efficacy and protection of the several studies that have been done. Currently, everybody seems to agree that in countries with annual infection risk lower than 1%, a massive vaccination campaign with BGC is not profitable. On the other hand, BCG interferes the lecture of the Mantoux intra-dermo-reaction and, thus, it invalidates the most effective test that we have for the diagnosis of the tuberculosis infection. In our country, the battle against tuberculosis is currently focused on the identification and treatment of the tuberculous patient, chemoprophylaxis with contacts of these patients and avoidance of the development of the tuberculosis infection to tuberculosis disease.

The antisense c-Ha-ras oligodeoxynucleotides inhibit the proliferation of the gastrocarcinoma-DNA-transformed rat 3-3 cells and reduce the p21 expression of the cells.

The Rat 3-3 is a secondary transformant of the rat fibroblast cell line (Rat-1) transfected with total DNA of a gastrocarcinoma cell line BGC-823. The cells over-express the c-Ha-ras oncogene which contains point mutation at the 12th codon. In order to determine how the activated c-Ha-ras oncogene expression governs the cell's transformation, two pentadecadeoxy-nucleotides AS-1 and AS-2 were synthesized. AS-1 was complementary to the single strand of the first three codons and the upstream sequence close to the ribosome binding site of c-Ha-ras mRNA. AS-2 was complementary to the 3' end of the first intron and the 5' end of the second exon of c-Ha-ras unripe RNA enclosed in the nucleus. The oligonucleotides could block either the translation of c-Ha-ras mRNA or the splicing of c-Ha-ras unripe RNA, thus inhibiting the expression of the activated c-Ha-ras oncogene and the proliferation of the transformed cells Rat 3-3. The inhibitory effect increased with a growing concentration of the antisense oligodeoxynucleotide (from 4-10 mumol/L) and reached its peak at 12 h after Rat 3-3 was treated with AS-1 and AS-2. This effect became weaker afterwards. The p21 level, product of c-Ha-ras, in the Rat 3-3, was examined by ELISA method. The result shows that the amount of p21 in the growth-inhibited cells is about 30% of that of the control cells.

[Two cases of familial idiopathic basal ganglia calcifications (BCG) with non-symptomatic family members].

We reported two families each propositus of which exhibited extrapyramidal signs and dementia with bilateral basal ganglia calcifications (BGC), while some of the other non-symptomatic family members showed BCG on brain CT by further examinations. Family 1) A 49-year-old woman was normal until her mid 40s when her memory began to fail. At age 40, dementia, finger-tremor and rigidity were observed and with brain CT and Magnetic Resonance Imaging, BCG and dentate calcifications were found. Her two daughters (20 years old and 26 years old) were free of any neuropsychiatric symptoms, but with CT examinations disclosed BCG. Family 2) A 40-year-old man. His symptoms started at 33 years old. He noticed gradually increasing finger-tremor, rigidity and dysarthria. At 40 years he showed mild dementia and BCG on Brain CT. His mother (64 years old) was non-symptomatic but CT showed that she had BCG. In the two families the calcium, phosphorous and parathyroid hormone levels, and Ellsworth-Howard test were normal. Other specific etiology including infections and somatic abnormalities was not discovered. Familial idiopathic basal ganglia calcification was considered to be rare. But the main purpose of this report is to point out that we must pay attention to the possibility of BCG of non-symptomatic family members if one showed dementia and extrapyramidal signs, and BCG on Brain CT in middle age.
