2-Acetamido-4-p-tolyl-1,3-thiazole and 2-amino-4-p-tolyl-1,3-thiazolium chloride dihydrate.

The structures of 2-acetamido-4-tolyl-1,3-thiazole, C(12)H(12)N(2)OS, (I), and 2-amino-4-tolyl-1,3-thiazolium chloride dihydrate, C(10)H(11)N(2)S(+).Cl(-).2H(2)O, (II), reveal that both molecules are essentially planar, with the respective dihedral angles between the benzene and thiazole rings being 2.9 (1) and 10.39 (7) degrees . Compound (I) associates via a single N-H...O interaction to form a flat alternate-facing hydrogen-bonded chain [graph-set C(2)(2)(4)]. Compound (II) packs with the hydrogen-bonding associations of the Cl atoms and the water molecules creating a convoluted hydrogen-bonded ribbon made up of five-membered donor-acceptor rings, involving three water O atoms (with associated H atoms) and two Cl atoms. The thiazolium rings form stacked columns, aligned in the same direction as the hydrogen-bonded ribbons, of alternate-facing molecules that are also involved in the hydrogen-bonding network, linking to the Cl atoms and one of the water molecules. Subsequently, each Cl atom is the hydrogen-bond acceptor for five separate O/N-H associations.

[Pathomorphological studies of experimental thiazole-80 poisoning in sheep].

Investigations on the pathomorphological changes occurring in the organisms of 4 sheep poisoned experimentally by varying Thiazole-80 doses applied orally were made. It was established macroscopically that after opening the rumen a smell of sulfuric hydrogen is felt. The abomasum was with rusty brown haemorrhages and liver distrophy as well as enlarged gall bladder were observed. The histological studies of parenchymal organs revealed vascular disturbances, hyperemia, oedema and intervascular haemolysis. A heavy diffuse nephrosis, hyaline and protein cylinders were found in the kidney cortex, while in the ganglial cells of medula oblongata were evident degenerative changes. Histochemically, in sheep poisoned by thiazole-80, a well expressed reaction to iron in the lungs, leptomeniges and distrophic hepatocytes were established. Glycogen quantity in the liver was considerably reduced.

Nephrotoxicity of thioformamide, a proximate toxicant of nephrotoxic thiazoles, in mice depleted of glutathione.

Administration of thioformamide (TFA) (> 2 mmol/kg, sc) in combination with DL-buthionine sulfoximine (BSO) (4 mmol/kg, i.p.), an inhibitor of glutathione (GSH) synthesis, caused kidney injury in male mice. The injury was characterized by tubular necrosis, increases in relative kidney weight and serum urea nitrogen concentration, and a decrease in renal GSH concentration. In contrast to results in male mice, no overt nephrotoxic effects were observed in female mice given TFA in combination with BSO. These features of nephrotoxicity were very similar to those reported for thiabendazole and other nephrotoxic thiazoles in mice depleted of GSH by treatment with BSO; this resemblance supports the suggestion that TFA as a ring cleavage metabolite, or a further metabolite thereof, is responsible for the toxicity of the nephrotoxic thiazoles (Mizutani, T., Yoshida, K., and Kawazoe, S. (1993) Chem. Res. Toxicol. 6, 174-179).

The interactions between the fluorescent dye thiazole orange and DNA.

The interaction of the fluorescent dye thiazole orange (TO) with nucleic acids is characterized. It is found that TO binds with highest affinity to double-stranded (ds) DNA [log (K) approximately 5.5 at 100 mM salt], about 5-10 times weaker to single-stranded polypurines, and further 10-1000 times weaker to single-stranded polypyrimidines. TO binds as a monomer to dsDNAs and poly(dA), both as a monomer and as a dimer to poly(dG) and mainly as a dimer to poly(dC) and poly(dT). The fluorescence quantum yield of TO free in solution is about 2 x 10(-4), and it increases to about 0.1 when bound to dsDNA or to poly(dA), and to about 0.4 when bound to poly(dG). Estimated quantum yields of TO bound to poly(dC) and poly(dT) are about 0.06 and 0.01, respectively. The quantum yield of bound TO depends on temperature and decreases about threefold between 5 and 50 degrees C.

Synthesis of thiazole, imidazole and oxazole containing amino acids for peptide backbone modification.

Novel 5-ring heterocyclic building blocks are synthesized. These can be incorporated into analogs of peptide antibiotics such as microcin B17, which is a potent DNA-gyrase inhibitor that exhibits eight thiazole and oxazole moieties. In particular, the syntheses of imidazole and bisoxazole amino acids as novel peptidomimetics are reported, this includes a new procedure for the oxidative conversion of the intermediates oxazoline, imidazoline as well as oxazole-oxazoline into the corresponding heteroaromatic compounds. A mixture of 1,8-diazabicyclo-[5.4.0.]-undec-7-ene carbon tetrachloride/acetonitrile and pyridine proved to be a very effective and mild agent.

Heterogeneity of peripheral blood reticulocytes: a flow cytometric analysis with monoclonal antibody HAE9 and thiazole orange.

The expression of a human erythroid cell surface antigen recognized by monoclonal antibody (mAB) HAE9 has been studied on peripheral blood reticulocytes by one- and two-color flow cytometry. Total reticulocyte count was determined using Thiazole Orange (TO) and flow cytometry. In normal individuals, 4.56% of reticulocytes were stained by FITC-labeled mAB HAE9. The correlation between reticulocyte percentage by TO and HAE9 staining was 0.828 (P less than 0.0001) in patients with hematocrits less than 0.25. A HAE9-positive reticulocyte percentage of 6-44% was observed when analyzed by two-color flow cytometry with TO and mAB HAE9. These findings, in conjunction with previous studies, suggest that mAB HAE9 recognizes an early, less differentiated population of peripheral blood reticulocytes. Enumeration of immature reticulocytes may be of clinical utility.

New NO-donors with antithrombotic and vasodilating activities, VI: thiazole-2-nitrosimines.

24 new thiazole-2-nitrosimines were prepared and described by means of spectroscopical methods (NMR, IR, MS, UV). At pH 7 in cell free systems as well as in platelet rich plasma the compounds are stable against hydrolysis and do not react with the platelet glutathione. The chemical stability is underlined by the mass spectra: M+. is of high intensity and sometimes even forms the base peak (e.g. 8a). Thermal elimination of N2 is of minor importance. The =N-NO bond in solution is susceptible to cleavage by visible light. The metabolite so formed is able to inhibit the platelet aggregation induced by collagen (Born-test). Five compounds exhibit this activity in concentrations below 10 mumol/L (IC50). This is due to the release of a NO species, as could be demonstrated by the stimulation of soluble guanylate cyclase in a cell free system (e.g. 8a, KM = 72 mumol/L). In vivo the nitrosimines show antithrombotic properties. Two h after a single oral dose of 8g (60 mg/kg) a 57% inhibition of the laser induced thrombus formation in the mesenteric arterioles of rats is observed. After 8 h a 43% inhibition still is seen.

Synthesis and molluscicidal activity of some 1,3,4-triaryl-5-chloropyrazole, pyrano[2,3-c]pyrazole, pyrazolylphthalazine and pyrano[2,3-d]thiazole derivatives.

2-(5-Chloro-1,3-diphenyl-1H-pyrazol-4-ylmethylene)-malononitrile 1a reacts with the arylidenes of malononitrile 2a-d to afford the triaryl-5-chloropyrazoles 3a-d, respectively. 1a reacts with the active methylene pyrazolinones 5a, b and 12a, b to afford different products 8, 9, 10, 11, and 14a, b--depending on the substitution in the pyrazole ring. Compound 1a reacts also with the pyridazinone derivative 15 to afford the phthalazinone 16, and with the thiazolinones 17a-c to afford the pyrano[2,3-d]thiazoles 20a-c, respectively. It reacts also with the malononitrile dimer 21a and with ethyl cyanoacetate dimer 21b to yield the pyrazolyl pyridines 22a, b, respectively. The synthesized compounds showed a moderate molluscicidal activity towards Biomphalaria alexandrina snails.

Bicyclic dioxetanes bearing an inden-2-yl or a benzo(b)thiazol-2-yl moiety as a CIEEL-active chemiluminescent substrate emitting red light.

Bicyclic dioxetanes bearing 5-(t-butyldimethylsiloxy)inden-2-yl, 3, or 5-(t-butyldimethylsiloxy)benzo(b)thiazol-2-yl, 4, were synthesized. On treatment with large excess of tetrabutylammonium fluoride in DMSO, dioxetane, 3, decomposed rapidly with accompanying emission of red (vermilion) light (lambda(max) (CL) = 637 nm). Comparing the chemiluminescent properties for 3 with those for related dioxetane, 1, in which pi-conjugation system is not fixed in plane, both CIEEL-decay rate and chemiluminescent efficiency were found to be improved for 3. Chemiluminescent decomposition of dioxetane, 4, was similarly induced to emit crimson light (lambda(max) (CL) = 725 nm), though the chemiluminescent efficiency was low.

Forced intercalation probes (FIT Probes): thiazole orange as a fluorescent base in peptide nucleic acids for homogeneous single-nucleotide-polymorphism detection.

Fluorescent base analogues in DNA are versatile probes of nucleic acid-nucleic acid and nucleic acid-protein interactions. New peptide nucleic acid (PNA) based probes are described in which the intercalator dye thiazole orange (TO) serves as a base surrogate. The investigation of six TO derivatives revealed that the linker length and the conjugation site decided whether a base surrogate conveys sequence-selective DNA binding and whether fluorescence is increased or decreased upon single-mismatched hybridization. One TO derivative conferred universal PNA-DNA base pairing while maintaining duplex stability and hybridization selectivity. TO fluorescence increased up to 26-fold upon hybridization. In contrast to most other probes, in which fluorescence is invariant once hybridization had occurred, the emission of TO-containing PNA probes is attenuated when forced to intercalate next to a mismatched base pair. The specificity of DNA detection is therefore not limited by the selectivity of probe-target binding and a DNA target can be distinguished from its single-base mutant under nonstringent hybridization conditions. This property should be of advantage for real-time quantitative PCR and nucleic acid detection within living cells.

Synthesis and antifungal activity of 4-thiazol-2-yl-5-aminopyrazoles and 4-aminopyrazolo [3,4-c] isothiazoles.

Starting from 4-thiocarbamoyl-5-aminopyrazoles (I), the AA could prepare two series of products: 4-thiazol-2-yl-5-aminopyrazoles (II) and 4-aminopyrazolo [3,4-c] isothiazoles (III). The screening for in vitro antifungal activity evidenced that compounds (III) are effective in controlling most examined fungi; in particular 1-phenyl-3-methyl-4-aminopyrazolo [3,4-c] isothiazole (III a) at the concentration of 20 p.p.m. completely inhibited the growth of Pythium ultimum and Corticium solani, responsible for the collar rot of the beet.

Thiazole orange: a new dye for Plasmodium species analysis.

A rapid sensitive method for the determination of Plasmodium falciparum in in vitro culture is presented. The technique employs a fluorescent flow cytometer equipped with a 15-mwatt argon laser that emits light at 488 nm and a membrane-permeable fluorochrome thiazole orange (TO) that stains RNA. Parasitized red cells are stained by suspending them in 1 ml of phosphate-buffered saline (PBS) containing 10(-5) M of TO and incubating this mixture for 15 min in the dark at room temperature. The stained cells may be analyzed fresh or after fixation with 1% paraformaldehyde/PBS or 0.25% glutaraldehyde/PBS. Alternatively the cells may be fixed first and then stained. There is excellent correspondence between the number of fluorescent-labeled parasitized red cells and Giemsa-stained cells.

Determination of the percentage of thiazole orange (TO)-positive, "reticulated" platelets using autologous erythrocyte TO fluorescence as an internal standard.

Determination of the percentage of thiazole orange (TO)-positive or "reticulated" platelets by flow cytometry has been advocated as an aid in the diagnosis of thrombocytopenic disorders. However, a reproducible method for determining control fluorescence and setting threshold values on a routine clinical basis has not been described. We used erythrocyte TO fluorescence in whole blood as an internal standard to set threshold markers for TO fluorescence of autologous, purified platelets. Since platelets have approximately a threefold higher TO fluorescence than erythrocytes, multiplying the erythrocyte threshold marker by 3 allowed a determination of the percentage of "reticulated" platelets in 29 normal controls. This value was 3.64% +/- 2.10% (S.D.) in men and 5.79 +/- 2.22% (S.D.) in women; there was a significant (P < 0.05) difference between normal men and women for this measurement. Applying this method to our earlier studies of normals and patients with immune thrombocytopenic purpura confirmed its ability to discriminate between these populations with similar sensitivity and specificity as previously reported. This method controls for fluctuations in TO labeling and fluorescence and does not require daily determination of a new normal control value.
