Hypocholesterolemic activity of Marsilea minuta in gerbils.

Feeding of a Marsilea minuta leaf extract [Fr. I] reduced serum cholesterol and triglycerides by 31 and 63%, respectively, in athero diet fed gerbils. Liver cholesterol and triglycerides were also lowered by 71 and 27%, respectively, in comparison to athero fed controls. Moreover, treatment with Fr. I prevented the accumulation of cholesterol and triglycerides in liver and aorta and was able to dissolve atheromatous plaques of thoracic and abdominal aorta. Fecal excretions of cholesterol and triglycerides were significantly increased in Fr. I fed gerbils.

Internode and petiole elongation of soybean in response to photoperiod and end-of-day light quality.

Elongation of main stem internodes and petioles of soybeans, Glycine max 'Ransom,' was examined in response to various photoperiod/temperature combinations and to end-of-day (EOD) light quality. Photoperiod treatments consisted of 10, 14, and 16 h in combination with day/night temperatures of 18/14, 22/18, 26/22, 30/26, and 34/30 C. The EOD treatments consisted of exposing plants to illumination from either incandescent (high far-red component, FR) or fluorescent (high red component, R) lamps during the final 0.5 h of a 10-h photoperiod. Internode elongation was not significantly promoted by the photoperiod treatments, and, in fact, under the two highest temperature regimes, internode elongation was suppressed under the longer photoperiods. Petiole elongation, however, was enhanced under the longer photoperiods at all temperatures. In the EOD light study, internode and petiole elongation was significantly greater on plants exposed to 0.5 h EOD from incandescent lamps than from fluorescent. Under the incandescent EOD treatment, plants increased dry matter production by 41% and exhibited greater partitioning of assimilates in stem and root portions than under fluorescent EOD.

Prediction of fetal lung maturity in infants of diabetic mothers using the FLM S/A and disaturated phosphatidylcholine tests.

The authors evaluated the performance of the amniotic fluid surfactant to albumin ratio (FLM S/A), and disaturated phosphatidylcholine (DSPC) tests in assessing fetal lung maturity in infants of mothers with insulin-dependent diabetes mellitus antedating pregnancy. The distribution of the study population (n = 180) by class of diabetes was class B (27%); class C (28%); class D (29%); class F, FR and T (8%); and class R patients (8%). The diagnosis of respiratory distress syndrome (RDS) was the standard for evaluating the performance of FLM S/A and DSPC. The mean estimated gestational age was 37.4 weeks. Three infants (1.7%) were diagnosed with RDS. All three were delivered before 36 weeks. FLM S/A at the cut-off for "maturity" of > or = 70 mg/g, had a sensitivity of 66.6%, specificity of 94.9%, positive predictive value (PPV) of 18.2%, and negative predictive value (NPV) of 99.4%. DSPC at the cut-off for "maturity" of 1,000 micrograms/dL, had identical sensitivity and NPV, but lower specificity (89.2%) and PPV (9.5%) than FLM S/A. Both tests mispredicted maturity in the same case of RDS. The false "mature" rate of FLM S/A was 0.6% (95% confidence interval 0.0%-3.2%). The FLM S/A result of > or = 70 mg/g, obtained at or near-term, is a reliable predictor of the absence of RDS in infants of mothers with diabetes mellitus antedating pregnancy.

Screening for asbestos-related pleural disease with digital storage phosphor radiography.

Digital storage phosphor radiography (SR) has a wide dynamic range and unique postprocessing capabilities that may improve the performance of screening studies for asbestos-related pleural disease compared with conventional film radiography (FR). In a group of 32 asbestos-exposed and nine control subjects with established pleural data, we compared the screening performance of FR and SR obtained with a single isoexposure, dual-energy technique (system resolution 0.2 mm, 10 bits). Performance was evaluated for 7320 observations by eight readers using a paired t test (P less than .02 with Bonferroni correction) of averaged receiver operating characteristic curve (ROC) areas (Az +/- standard error). We found that SR alone and SR supplemented by dual-energy soft-tissue and calcium images (SRde) were superior to FR in the overall detection of pleural abnormalities (Az = 0.90 +/- 0.01, 0.90 +/- 0.01, and 0.88 +/- 0.01, respectively). In the specific detection of pleural calcification, SRde was superior to FR (Az = 0.91 +/- 0.01 and 0.87 +/- 0.01, respectively; P less than 0.01). Analysis of variance indicated that SRde most closely reproduced an established pleural score based on the International Labor Organization (ILO) classification of the pneumoconioses (P less than 0.05, Scheffé's multiple comparison test). We conclude that isodose SR performs at least as well as FR in screening for asbestos-related pleural disease. SR supplemented by dual-energy images might improve the specific detection of pleural calcifications compared with FR.

Effect of Vernalization, Photoperiod, and Light Quality on the Flowering Phenotype of Arabidopsis Plants Containing the FRIGIDA Gene.

We have compared the flowering response to vernalization, photoperiod, and far-red (FR) light of the Columbia (Col) and Landsberg erecta (Ler) ecotypes of Arabidopsis into which the flowering-time locus FRIGIDA (FRI) has been introgressed with that of the wild types Col, Ler, and San Feliu-2 (Sf-2). In the early-flowering parental ecotypes, Col and Ler, a large decrease in flowering time in response to vernalization was observed only under short-day conditions. However, Sf-2 and the Ler and Col genotypes containing FRI showed a strong response to vernalization when grown in either long days or short days. Although vernalization reduced the responsiveness to photoperiod, plants vernalized for more than 80 d still showed a slight photoperiod response. The effect of FRI on flowering was eliminated by 30 to 40 d of vernalization; subsequently, the response to vernalization in both long days and short days was the same in Col and Ler with or without FRI. FR-light enrichment accelerated flowering in all ecotypes and introgressed lines. However, the FR-light effect was most conspicuous in the FRI-containing plants. Saturation of the vernalization effect eliminated the effect of FR light on flowering, although vernalization did not eliminate the increase of petiole length in FR light.

Hepatitis B Vaccination of Health Care Workers is Not Yet a Reality.

It is well documented that health care workers (HCW) have a higher prevalence of hepatitis B markers, and a higher risk of acquiring hepatitis than the general population does. In this study, we obtained the prevalence of vaccination against hepatitis B among HCW in a tertiary hospital, evaluated the reasons why hospital personnel did not use the vaccine, and we determined the prevalence of accidents which carried the potential of infection among the professional staff. HCW at possible risk were included in the study. The investigation showed that 39.3% of HCW were completely vaccinated, 12.9% received only partial vaccination, 8.4% were in the process of a vaccination series, 36.5% were never vaccinated, and 2.8% had passive immunization against hepatitis B virus. Lack of opportunity and difficulty in obtaining the vaccine were the main reasons given for non-vaccination. Of the non-vaccinated, 84.5% declared an intention to take the vaccine. Accidents with a potential for infection were observed in 57.9% of the population (sharp object accidents in 48.6%, accidents involving contact with blood or body fluids on mucous membranes in 27.6% and incision related accidents in 5.3%). The accidents were grouped into categories of risk of infection according to profession or medical specialty. Surgeons had the highest number of accidents. Strategies for vaccination campaign plans include an emphasis on the risks of exposure to the virus, discussion about the efficacy and safety of vaccination, and counselling to eliminate resistance to immunization.

Immunolocalization of androgen receptors in testicular cells of prepubertal and pubertal pigs.

In the testis, androgen receptors are known to mediate autocrine and paracrine effects of androgens on Leydig cell function and spermatogenesis. The pig presents some unusual features with regard to the synthesis of testosterone and estrogens in the male gonads. In testes from prepubertal males, testosterone level was lower than in testes from adult boars, while estrogen secretion was relatively high and comparable to that of mature porcine gonad. Immunolocalization of androgen receptors and intensity of immunohistochemical staining was age-dependent. In testis sections from adult boars, androgen receptors were found in nuclei of all somatic cells such as Leydig cells, Sertoli cells, and peritubular-myoid cells, whereas in sections from immature pigs only in the Leydig cell cytoplasm showed positive immunoreaction for androgen receptors. In control tissue sections incubated with omission of the primary antibody, no positive staining was observed. Detection of the androgen receptors in testicular cells of the pig is important for understanding of their central role in mediating androgen action.

Microbial degradation of a single branched isomer of nonylphenol by Sphingomonas TTNP3.

The endocrine disrupting chemical nonylphenol (NP) is a technical product which consists of a complex mixture of nonylphenols with different alkyl side-chain isomers. Since the bio-degradation of each NP isomer may lead to its own range of metabolites, the isolation and identification of transformation products is very difficult. In order to overcome this difficulty, the nonylphenol isomer 4(3',5'-dimethyl-3'-heptyl)-phenol (p353NP) was synthesized, and its degradation by an axenic culture of Sphingomonas TTNP3 was investigated with [ring-U-14C]-labelled and non-labelled p353NP including a time-course study. Radioactive mass balancing resulted in different polar soluble fractions, in insoluble radioactivity associated with biomass, and volatile radioactivity in the form of the mineralization product 14CO2. In the extracellular media, the presence of nonanol corresponding to the nonyl chain of the NP isomer was confirmed and its concentration was determined during the course of fermentation. No other radioactive compounds were detected beside the parent isomer. Radioactive metabolites were only found in the intracellular fraction of S. TTNP3.

A comparison of the effectiveness of soy protein isolate and fish oil for reducing the severity of retinoid-induced hypertriglyceridemia.

The effectiveness of soy protein isolate (SPI) to reduce the severity of retinoid-induced hypertriglyceridemia has been demonstrated in the rat, but not in human subjects. Because fish oil has been demonstrated to be effective at lowering serum triglyceride concentration in human subjects undergoing retinoid therapy, a study was conducted to compare the ability of SPI with that of fish oil to reduce the severity of retinoid-induced hypertriglyceridemia in the rat. Male Fischer 344 rats, n=8/group, were fed one of four isonitrogenous, isoenergetic diets, consisting of a control diet containing 24% casein +20% corn oil (C), and three 13-cis retinoic acid (13cRA)-supplemented diets containing 24% casein +20% corn oil (R), 24% SPI +20% corn oil (SR), and 24% casein +15% fish oil and 5% corn oil (FR). There was no effect of diet on food intake or final body weight. Serum triglyceride concentration for group R was higher (P<0.001) than for groups C, SR, and FR (7.20 vs. 2.50, 2.84, and 2.02 mmol/L, respectively); values for groups SR and FR did not differ for this parameter. The serum concentration of 13cRA for group R did not differ from that for groups SR and FR. Thus, SPI was as effective as fish oil in reducing the severity of retinoid-induced hypertriglyceridemia in an animal model, suggesting that it may be effective for this purpose in human subjects.

Adrenocortical responsiveness to adrenocorticotropic hormone is enhanced in chronically food-restricted rats.

Chronic food restriction (FR) of rats and mice results in moderate hyperadrenocorticism, which may play a role in activating cellular mechanisms that retard aging. Previously, we reported that the FR-induced hyperadrenocorticism is not due to an activated hypothalamo-pituitary unit. Therefore, we investigated in a series of experiments whether adrenal responsiveness to adrenocorticotropic hormone (ACTH), in vitro and in vivo, is enhanced by FR. Three mo-old male Fischer 344 rats were either given free access to food (AL rats) or restricted to 60% of food consumed by AL rats (FR rats) from 6 wk of age. They were killed by decapitation in the morning (AM) and afternoon (PM) when endogenously circulating corticosterone levels are at their nadir and peak, respectively. In vitro, adrenal glands from FR rats (1.5 mo FR) produced more corticosterone per mg at all doses of ACTH than those from AL rats in both the AM and PM (diet main effect, P < 0.001). FR (1.5 to 2.5 mo) also enhanced adrenal responsiveness to physiologic (diet main effect, P < 0.05) and superphysiologic (diet main effect, P < 0.001) levels of ACTH administered in vivo to dexamethasone-treated rats. ACTH-receptor (ACTH-R) mRNA, normalized to adrenal mass or to total RNA, was not influenced by FR (1.5 mo). However, adrenal ACTH-R mRNA, as well as adrenal mass, per unit body weight was greater in FR than in AL rats (diet main effect, P < 0.001). These results indicate that enhanced adrenocortical responsiveness to ACTH plays a major role in the hyperadrenocortical state of chronically FR rats.

Pore disappearance in a cell after electroporation: theoretical simulation and comparison with experiments.

The process of pore disappearance after cell electroporation is analyzed theoretically. On the basis of the kinetic model, in which the formation and annihilation of a metastable hydrophilic pore are considered as random one-step processes, a distribution function of cell resealing times, Fr(t), is derived. Two cases are studied: 1) the rate of pore resealing, k(r), is significantly greater than the rate of pore formation, k(f); and 2) the rate of pore formation, k(f), is comparable with k(r). It is determined that the shape of the distribution function depends on the initial number of pores in a cell, n(i). If in the absence of an external electric field the rate of pore formation, k(f), is significantly less than the rate of pore resealing, k(r) (case 1), pores disappear completely, whereas when k(f) approximately k(r) (case 2), the cell achieves a steady state in which the number of pores is equal to k(f)/k(r). In case 1, when n(i) = 1, the distribution function Fr(t) is exponential. The developed theory is compared with experimental data available in the literature. Increasing the time of incubation at elevated temperature increases the fraction of resealed cells. This indicates that the time necessary for the resealing varies from cell to cell. Although the shape of experimental relationships depends on the electroporation conditions they can be described by theoretical curves quite well. Thus it can be concluded that the disappearance of pores in the cell membrane after electroporation is a random process. It is shown that from the comparison of presented theory with experiments, the following parameters can be estimated: the average number of pores, n(i), that appeared in a cell during an electric pulse; the rate of pore disappearance, k(r); the ratio k(f)/k(r); and the energy barrier to pore disappearance deltaWr(0). Estimated numerical values of the parameters show that increasing the amplitude of an electric pulse increases either the apparent number of pores created during the pulse (the rate of pore resealing remains the same) or the rate of pore resealing (the average number of pores remains the same).

Food restriction enhances endogenous and corticotropin-induced plasma elevations of free but not total corticosterone throughout life in rats.

Chronic food restriction (FR), which retards many aging processes, enhances the endogenous diurnal peak of plasma total corticosterone (B) in young rats. Although the FR-dependent enhancement of total B disappears in aged rats, increased levels of the bioavailable fraction, free B, appear to be maintained. In young rats, we previously found that the FR-induced increase in the diurnal peak of total B is associated with increased adrenal response to corticotropin, also known as adrenocorticotropic hormone (ACTH). Here we show that the FR-enhanced adrenal response of total B to ACTH disappears with age but that the enhanced response of free B is maintained. We measured the endogenous diurnal peak and the response to ACTH of total and free B in 10-, 16-, and 22-month-old ad-libitum fed and FR male Fischer 344 rats in the afternoon, when plasma B peaks. At 10 and 16 months, FR rats showed enhanced total plasma B responses to ACTH relative to ad-libitum fed rats, but not at 22 months. By contrast, the response of free B to ACTH was enhanced by FR at all ages. The effect of FR on patterns of endogenous total and free diurnal B in these three age groups paralleled the ACTH-response data. The enhanced adrenocortical response of FR rats to ACTH does not reflect an increased expression of ACTH-receptor (ACTH-R) mRNA, because ACTH-R mRNA/microg adrenal RNA and ACTH-R mRNA/mg adrenal weight did not differ between ad-libitum fed and FR rats at any age.
