Many-body hydrodynamic interactions in charge-stabilized suspensions.

In this joint experimental-theoretical work we study hydrodynamic interaction effects in dense suspensions of charged colloidal spheres. Using x-ray photon correlation spectroscopy we have determined the hydrodynamic function H(q), for a varying range of electrosteric repulsion. We show that H(q) can be quantitatively described by means of a novel Stokesian dynamics simulation method for charged Brownian spheres, and by a modification of a many-body theory developed originally by Beenakker and Mazur. Very importantly, we can explain the behavior of H(q) for strongly correlated particles without resorting to the controversial concept of hydrodynamic screening, as was attempted in earlier work by Riese [Phys. Rev. Lett. 85, 5460 (2000)].

Giant amplification of interfacially driven transport by hydrodynamic slip: diffusio-osmosis and beyond.

We demonstrate that "moderate" departures from the no-slip hydrodynamic boundary condition (hydrodynamic slip lengths in the nanometer range) can result in a very large enhancement--up to 2 orders of magnitude--of most interfacially driven transport phenomena. We study analytically and numerically the case of neutral solute diffusio-osmosis in a slab geometry to account for nontrivial couplings between interfacial structure and hydrodynamic slip. Possible outcomes are fast transport of particles in externally applied or self-generated gradient, and flow enhancement in nano- or microfluidic geometries.

Quasicrystalline and rational approximant wave patterns in hydrodynamic and quantum nested wells.

The eigenfunctions of nested wells with an incommensurate boundary geometry, in both the hydrodynamic shallow water regime and quantum cases, are systematically and exhaustively studied in this Letter. The boundary arrangement of the nested wells consists of polygonal ones, square or hexagonal, with a concentric immersed, similar but rotated, well or plateau. A rich taxonomy of wave patterns, such as quasicrystalline states, their crystalline rational approximants, and some other exotic but well known tilings, is found in these mimicked experiments. To the best of our knowledge, these hydrodynamic rational approximants are presented here for the first time in a hydrodynamic-quantum framework. The corresponding statistical nature of the energy level spacing distribution reflects this taxonomy by changing the spectral types.

Physical mechanisms governing the hydrodynamic response of an oscillating ultrasonic file.

Ultrasonically driven vibrating files are known to enhance the efficiency of root canal debridement. This paper presents a phenomenological view of the hydrodynamic response of an oscillating ultrasonic file and the relationship between the file response and various physical factors such as file size and curvature, file surface properties, file velocity amplitude, root canal geometry, and the type of irrigant. Relevant hydrodynamic properties include the propensity of a file to produce stable and transient cavitation, steady streaming, and cavitation microstreaming. These relationships were explored by experiment. Sonoluminescence was employed as an indicator of transient cavitation activity and photographic analysis was utilized as a means for detecting steady streaming, microstreaming, and stable cavitation. Measurements failed to indicate any strong correlation between registered driving power and the propensity to produce transient cavitation. Files that were pitted or possessed salient edges were very effective at generating transient cavitation. When observed, transient cavitation activity generally occurred near the tip of the straight file, provided the wall-loading did not inhibit file motion. In all cases studied, steady streaming and stable cavitation were observed to varying degrees, depending on the amount of file to wall contact. Stable cavitation was probably enhanced by the addition of moderate amounts of dissolved gas into the irrigant. Although the imposition of file-wall contact served to inhibit the production of transient cavitation, this action had relatively little effect on the ability of a file to produce a nominal level of streaming, microstreaming, and stable cavitation. The relationship between these hydrodynamic properties and the process of root canal debridement is addressed. Observations suggest that it is not prudent to ascribe enhanced cleaning effects to any one phenomenon, for it is likely that several factors are involved to varying degrees depending on the local conditions of application.

Use of high-performance size-exclusion chromatography to measure protein molecular weight and hydrodynamic radius. An investigation of the properties of the TSK 3000 SW column.

We have conducted a study of the TSK 3000 SW high-performance size-exclusion column to define under what conditions proteins would migrate most consistently with their known hydrodynamic properties. Our findings include the following: 1) the residual negative charge of the column does cause charge-exclusion or charge-retention effects at low ionic strengths; with elution in deionized water several anionic proteins elute approximately in the void volume; 2) at mu greater than or equal to 0.5, protein migration is not only independent of ionic strength, but consistent with protein molecular weight and hydrodynamic volume; 3) small hydrophobic peptides are retarded by the column; and 4) very asymmetric proteins and other hydrodynamic particles are likely to be retarded by an "end-on insertion" mechanisms.

The two coiled coils in the isolated rod domain of the intermediate filament protein desmin are staggered. A hydrodynamic analysis of tetramers and dimers.

Desmin protofilaments and the proteolytically derived alpha-helical rod domain have been characterized by high-resolution gel permeation chromatography (GPC) using columns calibrated for the determination of viscosity radii. Additional characterization by chemical cross-linking and the determination of sedimentation values allowed the calculation of the molecular dimensions of the molecular species isolated. In dilute buffers GPC separated desmin rod preparations into two complexes: a dimer species (single coiled coil) with a length of 50 +/- 5 nm and a tetramer species (two coiled coils) with a length of 65 +/- 5 nm. Thus the two coiled coils in the tetramer are staggered by approximately 15 nm. The hydrodynamically derived lengths of the rod dimer and tetramer are supported by electron microscopy after metal shadowing. The hydrodynamic properties of desmin protofilaments follow that of the rod tetramer. The data on the hydrodynamic analysis of the rod tetramer of desmin in solution are in full agreement with the structural information recently deduced from paracrystals of the rod of glial fibrillary acid protein [Stewart, M., Quinlan, R.A. & Moir, R.D. (1989) J. Cell Biol. 109, 225-234]. Our results explain the inhomogeneity of molecules encountered in previous electron microscopical analyses.

Conditions affecting hydrodynamics-based gene delivery into mouse liver in vivo.

1. It has been demonstrated that the hydrodynamics-based procedure has high efficiency to deliver foreign genes into the liver. The widespread use of this procedure in gene function studies and as a treatment option for liver and other organ diseases puts considerable importance on the investigation of various conditions that affect hydrodynamics-based gene delivery into mouse liver in vivo. 2. Various conditions, including the volume, speed and solution of the injection and the state, gender and strain of the animal were manipulated to evaluate their effect on the expression levels in mice of human factor IX (hFIX) 8 h after tail vein injection of the plasmid pCMV-hFIX. 3. It was found that an injection volume of 2-2.5 mL and an injection speed of 5-7 s were very effective in delivering DNA into the mouse liver. Using Ringer's solution as an injection fluid increased the efficiency of hFIX expression. 4. Anaesthetized mice expressed higher hFIX than conscious mice. Males expressed higher hFIX than females. The ICR mouse strain demonstrated higher expression of the foreign gene than did the C57 strain. 5. The effects of these specific factors on hFIX expression may be caused by variations in hydrostatic pressure, the degree of liver damage and liver size. 6. It can be concluded that there are optimal conditions for hFIX expression in the liver. This information may be helpful for the application of hydrodynamics-based procedures in the investigation of gene expression and gene therapy.

Relationship between mechanical and hydrodynamic properties of bioprosthesis produced from canine aortic valve.

Static tensile, stress relaxation, and hydrodynamic tests were carried out to investigate the relationships between the mechanical deformation or stiffness of heart valve leaflets and the opening behavior of bioprosthetic valves. The specimens used were fresh and glutaraldehyde (GA)-treated canine aortic valves. The tensile strength depended on the fiber orientation in the leaflet. The deformability of fresh and 0.05% GA-treated tissues was significantly larger than that induced by 0.1-5.0% GA concentrations according to the stress-strain curves. The stress relaxation function, which expresses the viscoelastic property, did not show significant differences in the 0.05-5.0% range of GA concentrations. In the hydrodynamic tests, the opening resistance of fresh and 0.05% GA-treated valves was less than that of 0.1-5.0% GA-treated valves. Thus, it was shown that the hydrodynamic valve functions were closely related to the material properties of aortic valve leaflets.

Concept for a new hydrodynamic blood bearing for miniature blood pumps.

The most crucial element of a long-term implantable rotary blood pump is the rotor bearing. Because of heat generation and power loss resulting from friction, seals within the devices have to be avoided. Actively controlled magnetic bearings, although maintenance-free, increase the degree of complexity. Hydrodynamic bearings for magnetically coupled rotors may offer an alternative solution to this problem. Additionally, for miniature pumps, the load capacity of hydrodynamic bearings scales slower than that of, for example, magnetic bearings because of the cube-square-law. A special kind of hydrodynamic bearing is a spiral groove bearing (SGB), which features an excellent load capacity. Mock-loop tests showed that SGBs do not influence the hydraulic performance of the tested pumps. Although, as of now, the power consumption of the SBG is higher than for a mechanical pivot bearing, it is absolutely contact-free and has an unlimited lifetime. The liftoff of the rotor occurs already at 10% of design speed. Further tests and flow visualization studies on scaled-up models must demonstrate its overall blood compatibility.

CSF hydrodynamics after subarachnoid hemorrhage.

Recent reports have shown that despite an apparently satisfactory recovery from previous subarachnoid hemorrhage (SAH), many patients still have minor sequelae when specifically looked for. The cause of this so-called post-SAH-encephalopathy is uncertain. This prospective study comprises 54 patients who underwent aneurysmal surgery after SAH between September 1978 and March 1985. One patient died, and 6 patients were non-biased drop-outs. CSF hydrodynamics, determined by infusion test and isotope cisternography, were evaluated on the remaining 47 patients in the recovery stage. Five patients (11%) were found to have typical clinical, CSF dynamic and radiological manifestations of normal pressure hydrocephalus (NPH), and all were shunted with good results. Twelve (26%) had abnormal results consistent with disturbed CSF-hydrodynamics, although there were no clinical or radiological findings supporting the diagnosis of shunt-demanding NPH. Disturbed CSF-hydrodynamics as one of the possible etiological factors of post-SAH-encephalopathy is discussed.

A residence-time distribution analysis of the hydrodynamics within the intestine in man during a regional single-pass perfusion with Loc-I-Gut: in-vivo permeability estimation.

The goal of this study was to determine the most appropriate hydrodynamic model for the Loc-I-Gut in-vivo perfusion system. The general mixing-tank-in-series model, which can approximate single mixing tank and laminar and plug-flow hydrodynamics, was fitted to the observed experimental residence-time distribution curves for the non-absorbable marker [14C]PEG 4000. The residence-time distribution analysis shows that the hydrodynamics of the perfusion solution within the jejunal segment in man is well approximately by a model containing on average between 1-2 mixing tanks in series. The solution is well mixed when using perfusion rates of 2.0, 3.0 and 6.0 mL min-1. The average mean residence time estimates from the fitted residence-time distribution were 12 +/- 7.6, 15 +/- 4.2 and 7.7 +/- 4.6 min, respectively, at these three perfusion rates. The mean volumes of the segment (Vs) were 25 +/- 15, 45 +/- 12 and 46 +/- 27 mL, respectively. There were no statistical differences between 2.0, 3.0 and 6.0 mL min-1 in respect of the number of mixing tanks (n) and mean residence times. This residence-time distribution analysis indicates that the luminal fluid in the Loc-I-Gut perfusion system is well-mixed, and that permeability calculations based on the well-mixed assumption most closely approximate the actual local (average) membrane permeability within the perfused segment.

A self-organized vortex array of hydrodynamically entrained sperm cells.

Many patterns in biological systems depend on the exchange of chemical signals between cells. We report a spatiotemporal pattern mediated by hydrodynamic interactions. At planar surfaces, spermatozoa self-organized into dynamic vortices resembling quantized rotating waves. These vortices formed an array with local hexagonal order. Introducing an order parameter that quantifies cooperativity, we found that the array appeared only above a critical sperm density. Using a model, we estimated the hydrodynamic interaction force between spermatozoa to be approximately 0.03 piconewtons. Thus, large-scale coordination of cells can be regulated hydrodynamically, and chemical signals are not required.

Regional cerebral blood flow, white matter abnormalities, and cerebrospinal fluid hydrodynamics in patients with idiopathic adult hydrocephalus syndrome.

OBJECTIVES--(1) to evaluate regional cerebral blood flow (rCBF) with single photon emission computed tomography and 99mTc-hexamethylpropyleneamine oxime in patients with the idiopathic adult hydrocephalus syndrome (IAHS); (2) to examine regional cerebral blood flow (rCBF), gait, and psychometric functions before and after CSF removal (CSF tap test); (3) to assess abnormalities in subcortical white matter by MRI. METHODS--Thirty one patients fulfilling the criteria for IAHS (according to history and clinical and neuroradiological examination) were studied. Quantified gait measurements, psychometric testing, and rCBF before and after removal of CSF were obtained. Pressure of CSF and CSF outflow conductance were investigated with a constant pressure infusion method. Brain MRI was used to quantify the severity of white matter lesions and periventricular hyperintensities. In IAHS a widespread rCBF hypoperfusion pattern was depicted, with a caudal frontal and temporal grey matter and subcortical white matter reduction of rCBF as the dominant feature. Removal of CSF was not accompanied by a concomitant increase in rCBF. Significant white matter lesions were detected only in a minority of patients by MRI. An altered CSF hydrodynamic state with a higher CSF pressure and lower conductance was confirmed. IAHS is characterised by an abnormal CSF hydrodynamic state, associated with a widespread rCBF reduction with preference for subcortical white matter and frontal-temporal cortical regions. Furthermore in most patients MRI did not show white matter changes suggestive of a coexistent subcortical arteriosclerotic encephalopathy. At least in the idiopathic group of patients with AHS, measurements of rCBF before and after temporary relief of the CSF hydrodynamic disturbance will not provide additional information that would be helpful in the preoperative evaluation but is suggestive of a preserved autoregulation of rCBF.

Intracerebral microdialysis and CSF hydrodynamics in idiopathic adult hydrocephalus syndrome.

BACKGROUND: In idiopathic adult hydrocephalus syndrome (IAHS), a pathophysiological model of "chronic ischaemia" caused by an arteriosclerotic process in association with a CSF hydrodynamic disturbance has been proposed. OBJECTIVE: To investigate whether CSF hydrodynamic manipulation has an impact on biochemical markers related to ischaemia, brain tissue oxygen tension (PtiO(2)), and intracranial pressure. METHODS: A microdialysis catheter, a PtiO(2) probe, and an intracerebral pressure catheter were inserted into the periventricular white matter 0-7 mm from the right frontal horn in 10 patients with IAHS. A subcutaneous microdialysis probe was used as reference. Intracranial pressure and intracerebral PtiO(2) were recorded continuously. Samples were collected for analysis between 2 and 4 pm on day 1 (baseline) and at the same time on day 2, two to four hours after a lumbar CSF hydrodynamic manipulation. The concentrations of glucose, lactate, pyruvate, and glutamate on day 1 and 2 were compared. RESULTS: After CSF drainage, there was a significant rise in the intracerebral concentration of lactate and pyruvate. The lactate to pyruvate ratio was increased and remained unchanged after drainage. There was a trend towards a lowering of glucose and glutamate. Mean intracerebral PtiO(2) was higher on day 2 than on day 1 in six of eight patients. CONCLUSIONS: There is increased glucose metabolism after CSF drainage, as expected in a situation of postischaemic recovery. These new invasive techniques are promising tools in the future study of the pathophysiological processes in IAHS.

Hydrodynamic interaction of AFM cantilevers with solid walls: an investigation based on AFM noise analysis.

The noise power spectrum of the thermally activated motion of an AFM cantilever has been analyzed with respect to viscoelastic and hydrodynamic coupling between the cantilever and a substrate surface. Spheres with radii between 5 and 25 microm were glued to the cantilever to provide a well-defined geometry. The cantilever is modeled as a harmonic resonator with a frequency-dependent complex drag coefficient xi(omega). The variation of the drag coefficient xi(omega) with the tip-sample distance, D, and the sphere radius, R, can be expressed as a function of the single dimensionless parameter D/ R. However, this scaling breaks down close to the surface. There are two sources of a frequency dependence of xi(omega), which are viscoelastic memory and hydrodynamics. Viscoelastic relaxation is observed when the surface is covered with a soft polymer layer. In the absence of such a soft layer one still finds a frequency dependence of xi(omega) which is caused by hydrodynamics. At large substrate-cantilever distances, the drag coefficient increases with frequency because of inertial effects. At small distances, on the other hand, the drag coefficient decreases with increasing frequency, which is explained by the reflection of shear waves from the substrate surface. In liquids, inertial effects can be important when performing dynamic AFM experiments.

Synchronization of rotating helices by hydrodynamic interactions.

Some types of bacteria use rotating helical flagella to swim. The motion of such organisms takes place in the regime of low Reynolds numbers where viscous effects dominate and where the dynamics is governed by hydrodynamic interactions. Typically, rotating flagella form bundles, which means that their rotation is synchronized. The aim of this study is to investigate whether hydrodynamic interactions can be at the origin of such a bundling and synchronization. We consider two stiff helices that are modelled by rigidly connected beads, neglecting any elastic deformations. They are driven by constant and equal torques, and they are fixed in space by anchoring their terminal beads in harmonic traps. We observe that, for finite trap strength, hydrodynamic interactions do indeed synchronize the helix rotations. The speed of phase synchronization decreases with increasing trap stiffness. In the limit of infinite trap stiffness, the speed is zero and the helices do not synchronize.

Light enhances hydrodynamic signaling in the multimodal caudal photoreceptor interneurons of the crayfish.

1. The caudal photoreceptor (CPR) interneurons in the sixth abdominal ganglion of the crayfish are complex, multi-modal interneurons. These cells respond directly to light with tonic spike discharges, and they integrate synaptic input from an array of fili-form mechanoreceptors on the tailfan. They also provide input to rostral command centers, inducing backward walking at high firing frequencies, and thus directly influence behavior. 2. We recorded CPR activity in response to weak hydrodynamic stimulation of the tailfan mechanoreceptors while under varying intensities of light shined on the sixth ganglion. Spike trains were characterized according to the mean discharge rate (MDR) and the power spectrum from which the signal-to-noise ratio (SNR) was calculated. 3. Illumination of the CPR enhances the efficiency of transmitting mechanosensory signals. It does so by increasing the SNR of mechanosensory input received from tailfan receptors. A sevenfold, nonlinear increase in the SNRs was observed with increasing light intensity, an effect especially pronounced for weak hydrodynamic stimuli. In comparison with the dark, illumination of the ganglion lowered the hydrodynamic threshold and heightened the response to suprathreshold stimulation. Unlike the SNR, the MDR is little affected by mechanosensory input. 4. These results are compared with simulated electronic activity from an analogue threshold model and are discussed with respect to the mechanism of stochastic resonance.

Sensitivity of central units in the goldfish, Carassius auratus, to transient hydrodynamic stimuli.

This study describes the discharges of central units in the medulla of the goldfish, Carassius auratus, to hydrodynamic stimuli received by the lateral line. We stimulated the animal with a small object moving in the water and recorded activity of 85 medullary lateral line units in response to different motion directions and to various object distances, velocities, accelerations and sizes. All but one unit increased discharge rate when the moving object passed the fish laterally. Five response types were distinguished based on temporal patterns of unit responses. Ten units were recorded which encoded motion direction by different temporal discharge patterns. In general, discharge rates decreased when object distance was increased and when object speed was decreased. When object size was decreased, discharge rates decreased systematically in one group of units, but they were comparable for all but the smallest object tested in a second group of units. Units responded about equally well whether an object was moved at a constant velocity or was accelerated when it passed the fish. The data indicate that medullary lateral line units in the goldfish can encode motion direction but are not tuned to other aspects of an object moving in the water. The functional properties of units in the medulla of goldfish are similar to those reported for medullary units in the catfish Ancistrus sp., suggesting that the central mechanisms for processing complex hydrodynamic stimuli may be quite similar in fish species that occupy habitats with different hydrodynamic conditions.
