Synthesis and biological evaluation of new pentaphyrin macrocycles for photodynamic therapy.

This paper describes the synthesis and in-depth characterization of two new porphyrogenic macrocycles 1 and 2, and provides an evaluation of these molecules as photosensitizer agents. By tuning the reaction conditions and starting from readily available 1,9-diformyl-5-phenyldipyrromethane (4) and tripyrrane dicarboxylic acid (3), both the nonaromatic isopentaphyrin 1, composed of a 24 pi-electron macrocycle, and the aromatic pentaphyrin 2, composed of a 22 pi-electron macrocycle, were obtained in good yield and purity. Confocal laser microscopy and cytofluorimetry studies showed that the newly synthesized pentaphyrins penetrate the cell membranes and localize mainly in the cytoplasm. In the absence of light, 1 and 2 exhibit a nonsignificant cytotoxic effect at concentrations up to 3 mug/mL. In contrast, the synthesized pentaphyrins, when delivered to cells at 1.5 or 3 microg/mL and irradiated with white light (8 mW/cm(2)), promoted a strong and dose-dependent phototoxic effect in four different cell lines. FACS and caspase-3/7 activation assays demonstrated that the pentaphyrins cause cell death by apoptosis.

Macrocyclic inhibitors of beta-secretase: functional activity in an animal model.

A macrocyclic inhibitor of beta-secretase was designed by covalently cross-linking the P1 and P3 side chains of an isophthalamide-based inhibitor. Macrocyclization resulted in significantly improved potency and physical properties when compared to the initial lead structures. More importantly, these macrocyclic inhibitors also displayed in vivo amyloid lowering when dosed in a murine model.

Systematic study of the synthesis of macrocyclic dipeptide beta-turn mimics possessing 8-, 9-, and 10- membered rings by ring-closing metathesis.

[structures: see text] A systematic study was performed to establish general synthesis protocols for forming enantiomerically pure macrocyclic dipeptide lactams. Focusing on macrocycles of 8-, 9-, and 10-membered rings, effective syntheses were achieved by a sequence featuring peptide coupling of allyl- and homoallyl-glycine building blocks followed by ring-closing metathesis. The 8-membered lactam-possessing cis-amide and cis-olefin geometry as well as 9-membered [corrected] lactams having trans-amide and cis-olefin [corrected] configurations were effectively prepared by a general strategy employing the respective protected dipeptide, the first generation Grubbs' catalyst, and temporary protection of the central amide as a benzyl derivative. The 10-membered macrocycle was synthesized possessing cis- or trans-olefin geometry by employing similar metathesis conditions in the presence or absence of temporary benzyl amide protection, respectively [corrected]

De novo synthesis of stable tetrahydroporphyrinic macrocycles: bacteriochlorins and a tetradehydrocorrin.

[structures: see text] Bacteriochlorins (tetrahydroporphyrins) are attractive for diverse photochemical applications owing to their strong absorption in the near-infrared spectral region, as exemplified by the bacterial photosynthetic pigment bacteriochlorophyll a, yet often are labile toward dehydrogenation to give the chlorin. Tetradehydrocorrins (ring-contracted tetrahydroporphyrins) are attractive for studies of catalysis analogous to that of vitamin B12. An eight-step synthesis toward such tetrahydroporphyrinic macrocycles begins with p-tolualdehyde and proceeds to a dihydrodipyrrin-acetal (1) bearing a geminal dimethyl group and a p-tolyl substituent. Self-condensation of 1 in CH3CN containing BF3 x OEt2 at room temperature afforded a readily separable mixture of two free base bacteriochlorins and a free base B,D-tetradehydrocorrin. Each bacteriochlorin contains two geminal dimethyl groups to lock-in the bacteriochlorin hydrogenation level, p-tolyl substituents at opposing (2,12) beta-positions, and the absence (H-BC) or presence (MeO-BC) of a methoxy group at the 5- (meso) position. The B,D-tetradehydrocorrin (TDC) lies equidistant between the hydrogenation levels of corrin and corrole, is enantiomeric, and contains two geminal dimethyl groups, 2,12-di-p-tolyl substituents, and an acetal group at the pyrroline-pyrrole junction. Examination of the effect of the concentrations of 1 (2.5-50 mM) and BF3 x OEt2 (10-500 mM) revealed a different response surface for each of H-BC, MeO-BC, and TDC, enabling relatively selective preparation of a given macrocycle. The highest isolated yield of each was 49, 30, and 66%, respectively. The macrocycles are stable to routine handling in light and air. The bacteriochlorins display characteristic spectral features; for example, H-BC exhibits near-IR absorption (lambda(Qy) = 737 nm, epsilon(Qy) = 130,000 M(-1) cm(-1)) and emission (lambda(em) = 744 nm, phi(f) = 0.14). In summary, this simple entry to stable bacteriochlorins and tetradehydrocorrins should facilitate a wide variety of applications.

Is [N+-H...O] hydrogen bonding the most important noncovalent interaction in macrocycle-dibenzylammonium ion complexes?

We report a new host molecule in which one diethylene glycol chain (i.e., a loop possessing only three oxygen atoms) incorporated along with two phenolic aromatic rings is linked by a xylene spacer into a macroring. The design of the molecular structure of this macrocycle "amplifies" any potential [cation...pi], [N+-H...pi], and [N+C-H...pi] interactions between the dibenzylammonium (DBA+) ion and the phenolic rings of the macrocycle; as such, these species display a very strong binding affinity in CD3NO2 (Ka = 15,000 M(-1)). The macroring also coordinates to bipyridinium ions in a [2]pseudorotaxane fashion, which makes it the smallest macrocycle (i.e., a 25-membered ring) known to complex both DBA+ and bipyridinium ions in solution. To confirm unambiguously that these pseudorotaxanes exist in solution, we synthesized their corresponding interlocked molecules, namely rotaxanes and catenanes.

Evaluation of alternative approaches for the synthesis of macrocyclic bisindolylmaleimides.

Approaches for the synthesis of macrocyclic bisindolylmaleimides, in which the indole nitrogens are linked with a tether, are described. Two alternative approaches were investigated: macrocyclisation in either the 'southern' (by adding the tether to the bisindolylmaleimide ring system) or the 'northern' district. With two-, three- and four-atom tethers, both of these approaches were unsuccessful for a wide range of attempted macrocyclisation reactions (palladium-catalysed pi-allyl substitution, ring-closing metathesis, McMurry reaction, iodocyclisation, formation of a silylene derivative, substitution of an alpha,omega-disubstituted electrophile). The failure of all of these reactions was ascribed to the strained nature of the target ring system. However, with longer tethers (six to ten atoms), the macrocycles could prepared using either a ring-closing metathesis reaction or by substitution of an alpha,omega-dibromide). Fourteen successful macrocyclisation reactions are described; deprotection gave eleven macrocyclic bisindolylmaleimides in which an imide substituent had been removed.

The synthesis and conformation of oxygenated trianglimine macrocycles.

The synthesis of series of D(2h) and C(2v) symmetric oxygenated aromatic dicarboxaldehydes, using dilithiation methodology, is described along with their reactivity in the [3+3] cyclocondensation reaction with (1R,2R)-diaminocyclohexane to give oxygenated trianglimine macrocycles. Macrocycles derived from C(2v) symmetric dialdehydes give macrocycles with a stereogenic aromatic plane with complete diastereocontrol, as a mixture of rotamers.

Coordination chemistry of N-aminopropyl pendant arm derivatives of mixed N/S-, and N/S/O-donor macrocycles, and construction of selective fluorimetric chemosensors for heavy metal ions.

The coordination chemistry of the N-aminopropyl pendant arm derivatives (L1c-4c) of the mixed donor macrocyclic ligands [12]aneNS2O, [12]aneNS3, [12]aneN2SO, and [15]aneNS2O2(L1a-4a) towards Cu(II), Zn(II), Cd(II), Hg(II), and Pb(II) in aqueous solution has been investigated. The protonation and stability constants with the aforementioned metal ions were determined potentiometrically and compared, where possible, with those of the unfunctionalised macrocycles. The measured values show that Hg(II) and Cu(II) in water have the highest affinity for all ligands considered, with the N-aminopropyl pendant arm weakly coordinating the metal centres. Crystals suitable for X-ray diffraction analysis were grown for the perchlorate salt (H2L1c)(ClO4)2.dmf, and for the 1 : 1 complexes [Cd(L3a)(NO3)2](1), [Cu(L4a)dmf](ClO4)2(2), [Zn(L1c)(ClO4)]ClO4(3), [Cd(L1c)(NO3)]NO3(4), and [Hg(L2c)](ClO4)2(5). Their structures show the macrocyclic ligands adopting a folded conformation, which for the 12-membered systems can be either [2424] or [3333] depending on the nature of the metal ion. L1c-4c were also functionalised at the primary amino pendant group with different fluorogenic subunits. In particular the N-dansylamidopropyl (Lnd, n= 1-4), and the N-(9-anthracenylmethyl)aminopropyl (Lne, n= 1, 2, 4, ) pendant arm derivatives of L1a-4a were synthesised and their optical responses to the above mentioned metal ions were investigated in MeCN/H2O (4 : 1 v/v) solutions.

Sulfone-linked paracyclophanes via macrocyclic aromatic thioethers: synthetic and structural investigations.

Reaction of 4,4'-sulfonylbis(benzenethiol) with 4,4'-dichlorodiphenylsulfone under pseudo-high-dilution conditions leads to macrocyclic thioethersulfones [-S-Ar-SO2-Ar-]n (Ar = 1,4-phenylene). These include a highly strained [1+1] cyclodimer (n = 2), a cyclotrimer resulting from thioetherexchange reactions, and a [2+2] cyclotetramer which can adopt two entirely different conformations in the crystalline state, one having molecular D2d ("tennis-ball-seam") symmetry. The same type of reaction is successful using 4,4'-thiobis(benzenethiol) instead of 4,4'-sulfonylbis(benzenethiol) and affords macrocycles with a higher ratio of thioether to sulfone linkages. Exhaustive oxidation of macrocyclic thioethersulfones with hydrogen peroxide affords a series of sulfone-linked paracyclophanes, [-Ar-SO2-]4, [-Ar-SO2-]6, [-Ar-SO2-]8 and [-Ar-SO2-]12. Single crystal X-ray analysis reveals [Ar-SO2-]4 to be a near-perfect square box, whilst the cyclic hexamer [-Ar-SO2-]6 adopts a much more irregular conformation. and [-Ar-SO2-]8 displays a "double-box" structure clearly related to that of [Ar-SO2-]4.

Highly functionalized dimeric tetraethynylethenes and expanded radialenes: strong evidence for macrocyclic cross-conjugation.

A selection of dimeric tetraethynylethenes (TEEs) and perethynylated expanded radialenes, containing different donor/acceptor substitution patterns, have been prepared and fully characterized. The first X-ray crystal structure of an expanded [6]radialene, with twelve peripheral 3,5-di(tert-butyl)phenyl substituents, is presented. This macrocycle, the all-carbon core of which is isomeric with fullerene C60, adopts a non-planar, "chair-like" conformation. Also a TEE dimer, carrying N,N-dimethylaniline donor substituents, has been subjected to an X-ray crystallographic analysis. The electronic properties were studied by UV/Vis spectroscopy and electrochemistry, providing fundamental insight into mechanisms of pi-electron delocalization in the acyclic and macrocyclic chromophores. Donor or donor-acceptor-substituted dimeric TEE derivatives show very strong absorptions extending over the entire UV/Vis region; their longest wavelength absorption bands have high charge-transfer character. Macrocyclic cross-conjugation in the expanded radialenes becomes increasingly efficient with increasing donor-acceptor polarization. A dual, strongly solvent-polarity-dependent fluorescence was observed for a tetrakis(N,N-dimethylaniline)-substituted dimeric TEE; this interesting emission behavior is explained by the twisted intramolecular charge-transfer (TICT) state model. Donor-substituted expanded radialenes display huge resonance-enhanced third-order nonlinear optical coefficients.

Crescent oligoamides: from acyclic "macrocycles" to folding nanotubes.

Oligoarylamides whose backbones are rigidified by intramolecular hydrogen bonds (H-bonds) are found to adopt well-defined conformations in both solution and solid state. Depending on the length of their backbones, these folding oligoamides are crescents, broken macrocycles, or helices. A large (approximately 10 A) interior cavity is associated with the "macrocycles" and helices. The folded conformations are independent of chain length and are found in both nonpolar and polar solvents. This strategy is being extended to the construction of folding acyclic "macrocycles" and nanotubes with larger tubular cavities.

Facile preparation of [4.4]metacyclophane- and [5.5]paracyclophane-type macrocycles from arylboronic acids and salicylideneaminoaryl alcohols.

Four different salicylideneaminoaryl alcohols have been treated with arylboronic acids in order to prepare air-stable cyclophane-type macrocyclic systems. In two cases, this objective could be realized with the high-yield formation of [4.4]metacyclophane and [5.5]paracyclophane derivatives. The skeleton in these macrocycles is held together by two chiral boron atoms. In the other two cases, monomeric boronates or polymeric material were obtained. The title structures were characterized by spectroscopic techniques and X-ray crystallography. They show transannular C-H...O hydrogen bonding, but no intramolecular pi-pi interactions. A synthetic strategy for the preparation of further boron macrocycles is presented.

1,1'-Binaphthyl-substituted macrocycles as receptors for saccharide recognition.

The preparation of receptors for saccharide recognition in a natural environment has been an unmet goal for a long time. We present herein the synthesis and binding properties of (R,S)-1,1'-binaphthyl-substituted macrocycles as receptors for saccharide recognition in water/acetonitrile (1:1) and in DMSO. Porphyrin and metalloporphyrin macrocycles with two to four 1,1'-binaphthyl substituents and multiple hydroxy groups generate a binding site for saccharides that incorporates hydrogen-bonding hydroxy groups together with the aromatic hydrophobic pocket. The specificity for di- and trisaccharides is governed by the cavity size. The mechanism of binding has been studied by 1H NMR spectroscopy and the role of H-bonding and CH-pi interactions has been evaluated; the ability to bind saccharides has been demonstrated by the surface plasmon resonance (SPR) technique. The application of these macrocyclic receptors to sensor development is also presented.

Shape-persistent macrocycles: from molecules to materials.

Shape-persistent macrocycles with an interior in the nanometer regime allow the attachment of (functional) side groups at defined positions at the ring. These side groups can have either a fixed orientation relative to the molecular backbone or they can adapt their orientation according to an external stimulus. The properties and applications of the compounds depend strongly on the orientation of these side groups. Macrocycles with intraannular or adaptable long alkyl groups display a new design principle for discotic liquid crystals. Macrocycles with extraannular (oligo)alkyl groups can be used for surface patterning in the nanometer regime and rings with extraannular oligostyryl groups are able to aggregate to supramolecular hollow polymer brushes.

Synthesis and self-assembly of a rigid exotopic bisphenanthroline macrocycle: surface patterning and a supramolecular nanobasket.

The synthesis and characterisation of a rigid nanoscale macrocycle with two exotopic phenanthroline binding sites is reported. Scanning tunnelling microscopy (STM) at the solid-liquid interface reveals the formation of highly ordered monolayers of macrocycles with dimensions that are in good agreement with the calculated structure. Using the HETPHEN concept several bisheteroleptic coordination complexes with other phenanthrolines and a nanoscale basket assembly were prepared in presence of copper(I) ions. NMR spectroscopy, mass spectrometric data and elemental analysis point to three distinct isomers of the basket assembly in solution. A silver basket was prepared and readily converted to its copper analogue. Electrospray ionisation (ESI)-MS and spectrophotometric investigations provided additional mechanistic insight into the assembly process. Hence, the exotopic bisphenanthroline macrocycle in combination with HETPHEN concept proves to be very effective in controlling the compositional aspects of multicomponent self-assembly.

Synthesis, characterization, and properties of subporphyrazines: a new class of nonplanar, aromatic macrocycles with absorption in the green region.

Novel subphthalocyanine analogues that display strong absorption in the green region have been synthesized by using a boron template cyclotrimerization of maleonitrile derivatives. The spectroscopic properties of these macrocycles indicate that, like subphthalocyanines, they have 14 pi electrons and are aromatic compounds with a conical shape. The removal of the three fused benzene rings from the subphthalocyanine skeleton produces a 75-80 nm blue shift of the Q-band and a slight lowering of the absorption coefficients for this band. In addition, the reduction of the pi system from 18 to 14 electrons that accompanies progression from porphyrazines to subporphyrazines causes a hypsochromic shift of the Q-band of around 100 nm. Subporphyrazines that are peripherally functionalized with six thioether chains, and in which the sulfur atoms are attached directly to the pyrrole moieties, exhibit optical features that may be explained in terms of the extension of pi conjugation over the six thiolene groups, as well as strong pi donation from the sulfur lone pairs to the macrocycle. These two effects are quantitatively and qualitatively very similar to those observed for porphyrazines that possess the same type of substitution. In addition, the mesomorphic behavior at low temperatures of a macrocycle that is substituted with six thiododecyl chains was demonstrated by using differential scanning calorimetry and optical polarising microscopy.

Design and synthesis of simple macrocycles active against vancomycin-resistant enterococci (VRE).

16-membered meta,para-cyclophanes mimicking the vancomycin binding pocket (D-O-E ring) were designed and synthesized. The structural key features of these biaryl ether containing macrocycles are (1) the presence of beta-amino-alpha-hydroxy acid or alpha,beta-diamino acid as the C-terminal component of the cyclopeptide and (2) the presence of a hydrophobic chain or lipidated aminoglucose at the appropriate position. Cycloetherification by an intramolecular nucleophilic aromatic substitution reaction (S(N)Ar) is used as the key step for the construction of the macrocycle. The atropselectivity of this ring-closure reaction is found to be sensitive to the peptide backbone and chemoselective cyclization (phenol versus primary amine) is achievable. Glycosylation of phenol was realized with freshly prepared 3,4,6-tri-O-acetyl-2-N-lauroyl-2-amino-2-deoxy-alpha-D-glucopyranosyl bromide under phase-transfer conditions. Minimum inhibitory concentrations for all of the derivatives are measured by using a standard microdilution assay, and potent bioactivities against both sensitive and resistant strains are found for some of these compounds (MIC (minimum inhibitory concentration) = 4 microg mL(-1) against VRE). From these preliminary SAR studies, it was anticipated that both the presence of a hydrophobic substituent and an appropriate structure of the macrocycle were required for this series of compounds to be active against VRE.

Synthesis, structure, and contrasting chiroptical properties of large trianglimine macrocycles.

We have synthesized a number of large trianglimine macrocycles (3a-3f) using our methodology of (3+3) cyclocondensation of (R,R)-trans-1,2-diaminocyclohexane (1) with rigid aromatic dialdehydes (2a-2f), having benzene, biphenyl, terphenyl, styrene, or divinylbenzene spacers. The structures of these chiral macrocycles have been analyzed using computational methods as well as CD spectroscopy. Whereas trianglimines 3a-3d gave uniformly negative exciton Cotton effects, as expected, styrene- and divinylbenzene-based trianglimines 3e and 3f gave opposite-sign exciton Cotton effects. Their contrasting CD behavior was traced by computational methods to subtle differences in the relative orientation of the electric dipole transition moments in the chromophores of the trianglimines. The presence of low-intensity long-wavelength electronic transitions having large rotational strengths in the electronic spectra of 3e and 3f can be accounted for by the symmetry properties of the trichromophoric macrocycles.
