Systems analysis of intestinal hemodynamics and oxygenation.

A systems analysis of intrinsic regulation of intestinal blood flow and oxygenation is presented. The model is based on current concepts of metabolic control of tissue oxygen delivery and incorporates recent data from the literature regarding the influence of oxygen availability-to-demand ratio on intestinal vascular resistance and perfused capillary density. The model was used to evaluate the relative importance of resistance and exchange vessels in preventing cellular hypoxia during reductions in oxygen delivery or increments in oxygen demand. The model predicted that capillary recruitment is of greater quantitative significance than blood flow autoregulation in preventing cellular hypoxia when intestinal perfusion pressure is reduced. However, the combination of capillary recruitment and blood flow autoregulation provides a large margin of safety against tissue hypoxia in the intestine. Simulation results also predict that oxygen extraction plays a greater role than blood flow in providing additional oxygen to the hypermetabolic intestine irrespective of which control system (resistance or exchange vessel) is operating.

A comparison of the TMS-1 and the corneal analysis system for the evaluation of abnormal corneas.

We used two computerized corneal topography instruments (EyeSys Laboratories' Corneal Analysis System and the Topographic Modeling System (TMS-1) from Computed Anatomy, Inc.] to evaluate 18 eyes (ten patients) with abnormal corneas. The diagnoses included keratoconus, corneal scars, and residual postoperative astigmatism following refractive surgery. The patients were randomly selected from a busy corneal practice to include those with significant corneal refractive error whose management, it was believed, would benefit from corneal topography imaging. Images were taken with both systems, and comparisons were made of the ease of operation, the accuracy of corneal readings, and the usefulness of generated data. Three millimeter zone (Corneal Analysis System) and Simulated Keratometry (TMS-1) values were also compared to each other and to standard keratometric readings. We found the two systems to be very similar in their application and results, although certain differences were apparent in terms of operation, patient acceptance, and the ability to obtain useful data from the more distorted corneas. Neither system worked ideally for severely irregular surfaces.

[Automatic ECG analysis system based on TMS320F206].

An automatic ECG analysis system based on DSP is introduced in this paper. It can fulfill the functions of signal regulation, adaptive filtering, QRS waves positioning, and automatically recognize more than 10 kinds of arrhythmia and report alarm. TI DSP product--TMS320F206 is applied in this system to accomplish the adaptive filtering and automatic diagnosis of arrhythmia for the need of real time signal processing.

A versatile video image analysis system for microcirculatory research.

A modular image analysis system is presented consisting of a personal computer equipped with a real time video digitizer, an interactive control unit and a graphic tablet. Together with the corresponding software modules this system can be used for a number of image processing procedures in microcirculatory research including image enhancement, measurement of morphological parameters and image brightness as well as determination of vessel diameter and blood flow velocity.

Rapid diagnostic DNA cytometry with an automatic microscope and a TV image-analysis system.

A TV-based image-analysis system (Leitz TAS plus) combined with a Leitz automatic microscope was used for rapid DNA cytometry for diagnostic purposes. Malignant or suspicious cells found by the cytopathologists in conventionally stained smears were automatically relocated and measured. A program for automatic detection and measurement of nuclear DNA content was developed. DNA data are processed by an algorithm for the diagnosis and grading of malignancy. This diagnostic procedure is performed in real time and yields highly reproducible results. With this equipment, diagnostic DNA cytometry may be introduced in the routine cytology laboratory. Our mode of application does not replace the cytopathologist, but assists him or her in cases of doubtful diagnosis and in the grading of malignant tumors.

Early development of systems analysis in natural resources management from man and nature to the miami conservancy district.

Contemporary approaches to natural resources and environmental decision-making typically draw on a "systems" perspective to assess and improve management strategies. This paper describes the early genesis of the systems analysis approach. It concentrates on a period between the mid-19th to early 20th centuries. During the early part of this period, George Marsh's Man and Nature and related works laid out an approach to problem-solving that recognized the relationship among physically disperse elements in the environment, the need to balance benefits against costs, the potential for using quantitative modeling to understand management options, and the importance of integrating human and natural components into solutions. In the early 20th century, the Miami Conservancy District project brought this approach to fruition with its use of complex simulation and optimization modeling, detailed cost-benefit analysis, and its linking of economics, engineering, science, and law into a far-reaching solution to a complex water resources problem. The objective of this paper is to describe the early development and application of this conceptual approach to problem-solving. An examination of the origins of natural resources systems analysis can broaden one's perspective of the contemporary field to understand its roots as a philosophy for environmental problem-solving.

[Systems analysis as a necessary preliminary to improving psychiatric services].

The systems analysis approach to the work of a regional psychiatric service is described. Standard rehabilitation programs for principal groups of patients have been developed. These programs allow one to choose an optimal structure of the rehabilitation complex based on real requirements of the population. The complex includes a dispensary division, a hospital, a home treatment service, daytime and night wards, and workshops for occupational therapy. The disjunction of purposes and functions has provided complete succession in the work of all those services. The purposes and functions of each service are presented.

Anterior chamber angle measurement with Anterior Eye Segment analysis system Nidek EAS-1000: improving the repeatability.

Previous studies have reported good intraobserver and interobserver repeatability of anterior chamber angle (ACA) assessment using the Anterior Eye Segment analysis system, EAS-1000. This study investigated the influence of fixation pattern, corneal plotting method and camera position on ACA measurements. A Nidek EAS-1000 system was employed for image acquisition and analysis, to obtain measurements of ACA. Twenty-five young healthy subjects participated. The ACAs were different in different quadrants and using different corneal plotting methods (Repeated measures 3-way anova, p < 0.05). However, the difference was <5 degrees and therefore not clinically significant. Alignment along the visual axis or optical axis did not yield any significant difference. Repeatability was improved by asking the subjects to fixate at the flashing light-emitting diode (LED). Temporal and nasal angles also demonstrated better repeatability than vertical angles. In measuring the vertical angles, the camera can be set at either the temporal or nasal side (Paired t-test, p > 0.05). The Anterior Eye Segment analysis system has good clinical repeatability in measuring ACA. It can be further improved with the use of the flashing LED during image acquisition.

Database and analysis system for cDNA clones obtained from full-length enriched cDNA libraries.

We have developed an efficient sequence-analysis system and a database system for clones obtained from full-length enriched cDNA libraries made by using the oligo-capping method. We developed a semi-automatic analysis system for 5'- and 3'-end sequences. It pre-processes raw sequences (vector cut and accurate-sequence region extraction), clusters the sequences, searches for similarities through public databases, annotates completeness of clones and analyzes the ORFs in the sequences. Newly developed or improved programs are used in each step. A new program, ESTiMateFull is used to evaluate and to predict the sequence-fullness based on comparisons with mRNA and EST sequences, respectively. The ATGpr program is used to predict sequence-fullness based on statistical information. The combination of full-length enriched cDNA clones and ATGpr fullness prediction resulted in 70% accuracy in the specificity and the sensitivity of the fullness predictions. For the ORFs predicted by the ATGpr, the signal peptides are predicted and a motif search is performed by our new system. We also developed a program that assembles our sequences with dbEST sequences and developed a system to retrieve clones by the characteristics of the ORFs. As keywords, combination of various results of the analyses can be used for retrieval. And various results such as ORF features and database search results can be shown on the same screen by multiple displays. Full-length clones having interesting functions can thus be retrieved efficiently by using this system.

RNA aptamer evaluation using comparative molecular interaction profile analysis system.

In this paper, we introduce a system named Comparative Molecular Interaction Profile Analysis system (CoMIPA) that utilizes virtual docking study for comprehensive analysis of molecular interactions. Research of RNA aptamer is very useful to creation of functional molecules, because RNA aptamer is excellent in binding selectivity and the stability of structure. By using 3-bases RNA fragment, the system can use computational molecular docking results to help for RNA aptamer screening. We believe that the system has the potential to be a useful for RNA aptamer design.

A simple analysis system for the estimation of recombination efficiency using fluorescence-activated cell sorting.

A simple and accurate analysis system for the estimation of recombination efficiency in vivo using fluorescence-activated cell sorting (FACS) was designed and was subsequently used to compare the efficiency of recombination related to different spacer mutants. F(3) and F(5) mutant sequences were used for Flpe-mediated cassette exchange, and m2 and lox2272 mutant sequences were used for Cre-mediated cassette exchange due to their high incompatibility with wild-type sequences. The incompatibilities with wild-type were almost the same between mutant sequences. However, the recombination efficiencies were different. F(3) and m2 could mediate more efficient recombination than F(5) and lox2272, respectively. These results are consistent with the fact that the sequence of spacer region affects not only the reactivity upon wild-type sequence but also the recombination efficiency. It was also confirmed that the recombination process was mediated in a site-specific manner through PCR analysis using different sizes of exchange cassettes. In this experiment, the feasibility of the FACS analysis system for the estimation of recombination efficiency was verified. This system should be readily applicable for estimating recombination efficiency of other various mutant candidates, which will contribute to more precise and efficient site-specific recombination.

Enzyme-based flow injection analysis system for glutamine and glutamate in mammalian cell culture media.

We present the setup of a flow injection analysis system designed for on-line monitoring of glutamate and glutamine. These amino acids represent a major energy source in mammalian cell culture. A cycling assay consisting of glutamate dehydrogenase and aspartate aminotransferase produces NADH proportional to the glutamate concentration in the sample. NADH is then measured spectrophotometrically. Glutamine is determined by conversion to glutamate which is fed into the cycling assay. The conversion of glutamine to glutamate is catalyzed by asparaginase. Asparaginase was used in place of glutaminase due to its relatively high reactivity with glutamine and a pH optimum similar to that of glutamate dehydrogenase. The enzymes were immobilized covalently to activated controlled pore glass beads and integrated into the flow injection analysis system. The application of the immobilized enzymes and the technical setup are presented in this paper.

Methodology of the modular ECG analysis system MEANS.

The methodology, used in the Modular ECG Analysis System (MEANS) is described. MEANS consists of modules for signal analysis and diagnostic classification. The basic structure of the modular interpretation system remained intact over a period of 20 years, while all modules underwent many changes as a function of experience and insight, and the continuously changing information technology. The article describes the advantages of a modular approach to decision-support systems, the most important ones being easier maintenance of the software package and separate optimization and testing of each module. The overall evaluation of MEANS was done in the CSE study. Evaluation results for modules and for the entire system are presented.

Reliability and validity of the Metrecom Skeletal Analysis System in the assessment of sagittal plane lumbar angles.

Seventeen asymptomatic subjects were examined by the Metrecom Skeletal Analysis System and by lateral lumbar radiography. Measurements of sacral base angle, lumbar curve angle, and lumbosacral disc angle were compared with the values derived from the lumbar radiographs. Repeated readings using the Metrecom showed high intraexaminer reliability in measuring these angles. However, the Metrecom values for the lumbar curve and lumbosacral disc angles showed virtually no agreement with those obtained radiographically, while only a weak agreement existed for the sacral base angle. RELEVANCE: Our results suggest that the Metrecom Skeletal Analysis System (version 2.2) is not a valid technique for the determination of either sacral base angle, lumbar curve angle, or lumbosacral angle. It would appear that the values that it generates are more a reflection of back shape than of osseous positioning.

Evaluation of optical function using a new point spread function analysis system in cataractous and pseudophakic eyes: preliminary results.

PURPOSE: To evaluate optical function in cataractous and pseudophakic eyes using the new point spread function (PSF) analysis system in a clinical setting. METHODS: We applied this new analysis system in the study of two cataractous eyes and one pseudophakic eye of two patients. Using a PSF analyzer, double-pass PSF was measured directly for each subject, and the single-pass modulation transfer function (MTF) and single-pass PSF were calculated. The simulated retinal images of various sizes of Landolt's rings and their contrast characteristics were also calculated by the PSF analyzer. RESULTS: The MTF and the contrast of the simulated retinal images degraded in cataractous eyes were compared with data for normal eyes; the degradation pattern depended on the opacification pattern. The MTF and the contrast of the simulated retinal images in the pseudophakic eye improved significantly compared with the cataractous eyes, although both values were lower in the pseudophakic eye than in young normal eyes. CONCLUSIONS: Our data showed degradation of optical function in cataractous and pseudophakic eyes in comparison with optical function in young normal eyes. If further accumulations of PSF data are made, it may be possible to establish an objective standard by which to measure the progression of cataract, as well as an objective indication for treatment in the future.

Image analysis in immunohistochemistry. Factors with a possible influence on the performance of VIDAS version 2.0, a commercially available true color image analysis system.

This paper describes the evaluation of several factors with a possible influence on the performance of a commercially available image analysis system capable of true color image analysis. The software used by this system is VIDAS Version 2.0. The following factors were evaluated: illumination, power supply, warming up, shading correction, averaging of image intake, hue luminance and saturation images and relation of illumination to quantification of area percentage (area %) of positively staining structures. The first six factors were evaluated by using a macro, with which it is possible to obtain information on variations over the image and over time. The last factor was evaluated by repeated measurement of area % of positive staining in a routinely processed tissue section. In our setting, stability of illumination and warmup time of the camera appeared to be the most important factors with influence on the performance of the image analysis system.

The systems analysis of spinal X-ray films (SASXF) developed by the author solves the problem of an integral and, concurrently, detailed study of the spine of a patient on the basis of routine X-ray study. The examination of the patient gave rise to a patient's systems spinal model that considers the individual features of the vertebral column, which are most significant for a manual therapist's work. The standards of a X-ray study, a protocol form, and examination tools are described. An example of systems spinal model is given. The experience accumulated during studies of more than 2000 patients has shown that this procedure is highly effective in implementing medical manipulations by the methods of manual therapy.
