Special role and role expectation of black administrators of neighborhood mental health programs.

This paper attempts to portray the role conflict and ambiguity of black administrators in community mental health programs. The paper is not about mental health programs themselves but rather focuses on the problems of blacks in leadership positions in such programs. The implication of this paper is that blacks cannot escape being black despite their professional position or training. They are regarded by their superiors, colleagues, staff, and the community as performing what Hughes called a "racial function." This function presents an interesting dilemma. On the one hand, the black administrators are recruited and employed out of the needs expressed by the black community. On the other hand, their white superiors determine whether they remain in those positions. To offend either group risks the loss of their position. The black community and white employer sometimes make conflicting demands on these black administrators. The result is that blacks find themselves unable to please either. Blacks have reacted by withdrawal from the black community or resignation from the organization. This paper attempts to explain these conflicts. Although the problems of black administrators are called special cases it does not mean that other ethnic groups have not experienced any or all of these problems. It is called a special case because the black community is undergoing a special and unique kind of socioeconomical transition. Black administrators in other types of public organizations will recognize some of the conflicts described in this paper.

New roles for health care consumers: a study of role transformation.

The increased emphasis on citizen participation in health planning has caused changes in the traditional roles of health professionals and their clients. Interaction patterns between health care providers and health care consumers as members of planning boards are examined in two health-related organizations: a comprehensive health planning council and a low-income neighborhood health center. Eighteen months of observation, supplemented with an analysis of official records and interviews, provide the basis for the discussion.

Perceptions of sex-role stereotypes, self-concept, and nursing role ideal in Chinese nursing students.

AIM: This study examined the relationship between sex-role stereotypes, self-concept and the requisite personality characteristics of an ideal nurse in a cohort of Hong Kong nursing students. METHODS: To rate these concepts a measure of eight comprehensive dimensions of personality perception was administered to 177 nursing students, studying on preregistration and postregistration programs at a Hong Kong tertiary institution. Both male and female nursing students perceived an ideal nurse to possess a profile of traits including being high on the dimensions of emotional stability, application, intellect, helpfulness and restraint. RESULTS: No significant difference between the self-ratings of the male and female students was found, indicating that male students had undergone a highly self-selective process when choosing nursing education under the influence of Chinese cultural stereotypical attitudes towards nursing. A typical Chinese nurse was rated as similar to the typical female in Chinese society by both male and female nursing students. A typical Chinese nurse was rated relatively low on the masculine dimensions of openness, extroversion and assertiveness. The self-ratings of male nursing students more closely approximated the ideal nurse than did the self-ratings of female nursing students. CONCLUSION: The conclusions highlight implications for the recruitment and education of both male and female nursing students in Hong Kong society.

Gender roles, medical practice roles, and ob-gyn career choice: a longitudinal study.

This paper discusses follow-up data from physicians who were studied ten years earlier when they were medical students. Seventeen physicians were practicing ob-gyn, and 57 of the physicians studied had been interested in an ob-gyn career when they were medical students. At Time 1, women were more likely than men to be strongly interested in ob-gyn, but they were no more likely than men to be ob-gyn physicians at Time 2. The desire to have a surgical specialty was much more important to men than to women practitioners. Men ob-gyn practitioners were significantly more traditional in their sex-role outlook as medical students than were either women practitioners or women who had been strongly interested in the field but did not enter it. The data suggest that women ob-gyn physicians are more likely than their male peers to be egalitarian in their relationships with female patients.

From heroin to methadone-social role changes and reinforcement differentials in relation to outcome on methadone. Part II. The study of social role changes.

The life-styles of addicts who are Successful, Marginally Successful, and Failures in a methadone maintenance program are examined by in-depth interviews in five time periods-early family life, the preheroin period, late heroin, premethadone, and methadone maintenance. The hypothesis is confirmed that the more the methadone patient has managed to develop a new and more conventional set of peer group, familial, and occupational roles, the greater the likelihood of his success on methadone maintenance. The in-depth view of the groups over time suggests that the pattern of alternation of positive reinforcements and punishing situations may be associated with outcome.

A greater role of emotional than physical or sexual abuse in predicting disordered eating attitudes: the role of mediating variables.

OBJECTIVE: Previous research on the role of trauma in eating psychopathology has generally focused on reported childhood sexual abuse. There has been relatively little research addressing the full range of abusive experiences, and none considering their long-term impact on eating. This study investigated the relationships between four forms of reported childhood abuse (physical abuse, sexual abuse, emotional abuse, neglect) and unhealthy eating attitudes in adult life. Within this relationship, depression, anxiety, and dissociation were considered potential mediators, and age of onset of abuse was considered a potential moderator. METHOD: A nonclinical sample of 236 women completed self-report measures of abuse, eating psychopathology, and psychological function. Multiple regression analyses were used to test for associations as well as for mediating and moderating influences. RESULTS: When the intercorrelations of the different forms of reported abuse were controlled for, emotional abuse was the only form of childhood trauma that predicted unhealthy adult eating attitudes. That relationship was perfectly mediated by the women's levels of anxiety and dissociation. Age at onset of emotional abuse did not moderate these relationships. DISCUSSION: Although these results require extension to a clinical sample, the findings underscore the need to consider a history of emotional trauma as a potentially central factor in any abusive history. Treatment may depend on addressing the psychological consequences of such trauma.

Expression of the Celsr/flamingo homologue, c-fmi1, in the early avian embryo indicates a conserved role in neural tube closure and additional roles in asymmetry and somitogenesis.

Flamingo is one of a core group of proteins that regulate planar cell polarity of epithelial structures within the Drosophila embryo while their vertebrate counterparts have been implicated in the coordination of convergent extension movements during gastrulation and in neural tube closure, suggesting that planar polarity mechanisms also function in these processes. Failure of neural tube closure is one of the most common human birth defects, and a murine flamingo (fmi) homologue, Celsr1/fmi-1, was identified as the defective gene in two mouse mutants exhibiting failure of closure 1 of the neural tube. This failure resulted in craniorachischisis in which the neural tube is open from the midbrain posteriorly. The avian embryo provides a tractable system to study neural tube closure. We have identified a chick Celsr1/fmi-1 orthologue, c-fmi1 and provide the first study of expression of an avian flamingo gene. We show that expression is highly dynamic in the early embryo and that c-fmi1 transcripts become enriched within the avian neural epithelium at the initiation of neural tube closure, suggesting a conserved function for Flamingo proteins in this process. Our data also suggest a role for c-fmi1 in myotome development.

The role of charge microheterogeneity of basic protein in the formation and maintenance of the multilayered structure of myelin: a possible role in multiple sclerosis.

Isolation of several of the charge isomers from both normal and multiple sclerosis (MS) myelin basic protein (MBP) was achieved on CM-52 columns at pH 10.6. In liquid x-ray diffraction and aggregation experiments, corresponding charge isomers were equally effective in the formation of multilayers, demonstrating the dominant role of overall net positive charge. These studies demonstrated that the change in overall charge of MBP of one net positive charge was sufficient to produce large changes in aggregation and in multilayer formation. The x-ray diffraction experiments showed that component 1 was twice as effective as component 2 although they differed in charge by a single positive charge. Component 3 was less effective than component 2 and component "8" was not effective at all. Vesicle aggregation also showed a dependence on net positive charge. In order of decreasing effectiveness, component 1 greater than component 2 greater than component 3 greater than component "8". Since overall charge on MBP is determined by contributions from the various charge isomers, the relative proportions of these charge isomers favoring the less cationic components could explain the observation that MBP from MS victims was less effective than MBP from normal brain in vesicle aggregation and multilayer formation. The isolation of myelin-containing white matter fractions from both normal and MS tissue in which the loss of some of the most cationic charge isomers was correlated with presence of less compact myelin supports this hypothesis.

Role of protein kinase C isozymes in activation of human immunodeficiency virus type 1 in chronically infected promonocytic cells: evidence against a role of PKC beta 1.

Protein kinase C (PKC) plays an important role in activation of human immunodeficiency virus type 1 (HIV-1). Because of a molecular and biochemical heterogeneity of PKC, we have studied the effects of PKC isozymes in HIV-1 activation in a latently infected promonocytic cell line, U1, using various PKC isozyme agonists. 12-Deoxyphorbol 13-phenylacetate (dPP), an agonist of both Ca(++)-dependent and Ca(++)-independent isozymes, and thymeleatoxin (TT), an agonist of Ca(++)-dependent PKC isozymes, induced HIV-1 production at 10 nM with increase in a concentration dependent manner, whereas 12-deoxyphorbol 13-phenylacetate 20-acetate (dPPA), an PKC beta I isozyme agonist, did not induce viral production at 100 nM. We verified that dPPA induced translocation of PKC beta isozyme with the isozyme-specific monoclonal antibody using flow cytometry. This study demonstrates that activation of PKC isozymes leads to an induction of latent HIV-1 in U1 cells whereas PKC beta I isozyme may not be important.

Hemispheric cooperation in visuospatial rotations: evidence for a manipulation role for the left hemisphere and a reference role for the right hemisphere.

A survey of mental rotation strategies in 210 normal subjects showed a strong tendency for right-handers to prefer rotating an object on the right and vice versa for left-handers. The differential functioning of the cerebral hemispheres during mental rotation was then assessed in 42 subjects by means of tachistoscopic presentation of two geometrical figures separately to the left and right visual fields--one of which was gravitationally stable and the other unstable. Performance was better when the unstable object was presented to the right visual field and the stable object to the left. This finding is interpreted as indicating more efficient hemispheric cooperation when the active manipulation of a mental image is performed by the left hemisphere, while the reference role is carried out by the right hemisphere.

Specific regulatory role of phosphorylation of calf thymus DNA-topoisomerase I smaller forms on the relaxational activity expression. Phosphorylation role on Topo I smaller forms activity.

Calf thymus Topo I is found to be associated with three active breakdown products, resolved from intact enzyme, which do not appear to be unique to one extraction procedure. They are phosphoproteins, whose enzymatic activity can be modulated through changes in phosphorylation, and which can be phosphorylated 'in vitro', by N II protein kinase, in the same five sites as the intact enzyme. Different amounts of 32P incorporated are observed however, in the corresponding sites. We conclude: 1. proteolysis is probably an 'in vivo' phenomenon, as the Topo I smaller species are observed, during isolation from the earlier crude fractions, and as a minimum of them is always present, even if precautions are taken to minimize proteolysis; 2. a specific regulatory role in the DNA relaxational activity might be played by N II protein kinase phosphorylation, indeed, in the smaller species; 3. the different degrees of 32P incorporation, in analogous phosphorylation sites, might represent a different signal for modulating the gene expression.

The in vivo role of alpha-mannosidase IIx and its role in processing of N-glycans in spermatogenesis.

The surfaces of mammalian cells are covered by a variety of carbohydrates linked to proteins and lipids. N-glycans are commonly found carbohydrates in plasma membrane proteins. The structure and biosynthetic pathway of N-glycans have been analyzed extensively. However, functional analysis of cell surface N-glycans is just under way with recent studies of targeted disruption of genes involved in N-glycan synthesis. This review briefly introduces the potential role of processing alpha-mannosidases in N-glycan biosynthesis and recent findings derived from the alpha-mannosidase IIx (MX) gene knockout mouse, which shows male infertility. Thus, the MX gene knockout experiment unveiled a novel function of specific N-glycan, which is N-acetylglucosamine-terminated and fucosylated triantennary structure, in the adhesion between germ cells and Sertoli cells. Analysis of the MX gene knockout mouse is a good example of a multidisciplinary approach leading to a novel discovery in the emerging field of glycobiology.

Role of mitochondria in the immune response to cancer: a central role for Ca2+.

This study demonstrates that Ca2+ stimulates mitochondrial energy metabolism during spleen lymphocyte activation in response to the ascitic Walker 256 tumor in rats. Intracellular Ca2+ concentrations, phosphorylated protein kinase C (pPKC) levels, Bcl-2 protein contents, interleukin-2 (IL-2) levels, mitochondrial uncoupling protein-2 (UCP-2) contents and reactive oxygen species (ROS) were significantly elevated in these activated lymphocytes. Mitochondria of activated lymphocytes exhibited high free Ca2+ concentrations in the matrix and enhanced oligomycin-sensitive oxygen consumption, indicating an increased rate of oxidative phosphorylation. The production of ROS was largely decreased by diphenylene iodinium in the activated lymphocytes, suggesting that NADPH oxidase is the prevalent source of these species. Accumulation of UCP-2 and the anti-apoptotic protein Bcl-2 is probably important to prevent mitochondrial dysfunction and cell death elicited by the sustained high levels of intracellular Ca2+ and ROS and may explain the observed higher resistance from activated lymphocytes against the opening of the mitochondrial membrane permeability pore (MPT). All these changes were blocked by pretreatment of the rats with verapamil, an L-type Ca2+ channel antagonist. These data demonstrate a central role of Ca2+ in the control of mitochondrial bioenergetics in spleen lymphocytes during the immune response to cancer.

Role of dopamine D1 and D2 receptors in the nucleus accumbens in jaw movements of rats: a critical role of the shell.

Given the differences in the dopamine neurotransmission between the shell and the core of the nucleus accumbens, as well as the differential involvement of these two domains in oral behaviour of rats, it was decided to determine whether or not dopamine D1 and/or dopamine D2 receptors differentially direct oral behaviour in these two domains in rats. Intra-accumbens injections of the dopamine D1 receptor agonist (+/-)-6-chloro-7,8-dihydroxy-3-allyl-1-phenyl-2,3,4,5-tetrahydro-1H-3- benzazepine (SKF 82958: 5 micrograms/0.2 microliters), the dopamine D2 receptor agonist quinpirole (10 micrograms/0.2 microliters) and their combination were used to assess the role of these accumbens domains in jaw movements of rats. The present study shows that the combined administration of SKF 82958 and quinpirole into the shell, but not the core, of the nucleus accumbens produced a highly significant increase in jaw movements, when doses which per se were nearly ineffective, were injected. This effect was fully inhibited by prior administration of either the dopamine D1 receptor antagonist R-(+)-7-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine (SCH 23390: 0.5 microgram/0.2 micrograms) or the dopamine D2 receptor antagonist (-)-sulpiride (25 ng/0.5 microliter) into the same region. It is concluded that dopamine D1 and D2 receptors in the shell, but not the core, of the nucleus accumbens are involved in jaw movements of the rat, providing the first piece of evidence that dopamine D1 and D2 receptors in the shell of the nucleus accumbens mediate a particular behaviour.

Criteria for establishing a physiological role for brain peptides. A case in point: the role of vasopressin in thermoregulation during fever and antipyresis.

This paper has attempted to present and discuss the criteria necessary for the evaluation of a specific physiological role for a peptide in the CNS. These criteria are based on many experimental approaches to the problem and conclusions must be supported by the weight of the evidence. These criteria were illustrated by examining the hypothesis that AVP is an antipyretic neurotransmitter involved in regulating febrile increases in Tb by release and action in the VSA of the brain. The weight of the evidence in this case implies that this hypothesis is essentially correct. The only serious conflicting evidence comes from the work with Brattleboro rats. It is hoped that further research will resolve these discrepancies or result in a suitably modified hypothesis.

The critical role of the nucleolus in cell differentiation and stem cell development - an extension of the concept with a look at the mast cell and its possible role in zinc metabolism.

From the assumptions governing the behaviour of primitive cells, a set of corollaries is established and the corollaries are used in coming to an understanding of the growth and differentiation of the pancreas. A case is made out to show that there is a constant replacement of old acini by new throughout the life of the pancreas, and that new acini are derived from primitive cells (fixed reticulum cells) present in the stroma which envelopes the fine ducts. The part played by the mast cells in the process is discussed, and view put forward that circulating thymocytes are mast cell precursors, and that the function of the mast cell is to provide the primitive cells of the pancreas (and the primitive cells of certain other tissues) with Zn in an assimilatable form. The hypothesis is made that Zn can only be assimilated by primitive cells, and, that for organs other than the thymus, it is assimilated in the form of the histamine-Zn complex liberated by the mast cells. The fate of the mast cell is analysed, and the suggestion made that, following degranulation, the mast cell nucleus is transformed into a cell of the eosinophil series. The role of the basophil is also analysed, and the conclusion reached that basophils are mast cells whose development has been modified by plasma.

Modulatory role of the adventitia on noradrenaline and angiotensin II responses role of endothelium and AT2 receptors.

OBJECTIVE: We have studied the modulatory role of the adventitia on vascular tone and nitric oxide (NO) availability in response to noradrenaline (NA) and angiotensin II (Ang II). METHODS: Changes in isometric tension were determined in carotid arteries from 3-month-old Sprague-Dawley rats denuded from adventitia (-A) and compared to intact rings (+A). NO availability was assessed by the fluorescent NO indicator, 4,5-diaminofluorescein diacetate (DAF-2). RESULTS: Responses to NA (10(-10) to 10(-6) M) were: (i) significantly lower in -A compared to +A rings; (ii) equally enhanced in +A and -A rings without endothelium; and (iii) reduced in +A and -A rings incubated with superoxide dismutase (SOD; 15 U/ml). Responses to Ang II (10(-10) to 10(-7) M) were: (i) similar between +A and -A segments; (ii) equally reduced in both groups by SOD; and (iii) increased by endothelial denudation in both +A and -A arteries. Blockade of AT2 receptors with PD 123,319 (10(-7) M) significantly increased Ang II-induced contractions in +A rings. In segments preincubated with losartan (10(-5) M) and precontracted with NA (10(-7) M), Ang II elicited a relaxation that was abolished by l-NAME (10(-4) M), PD 123,319 (10(-7) M), and endothelium or adventitial removal. NO availability was increased in carotid rings stimulated with Ang II, but not with NA. This NO release was blocked by PD 123,319 (10(-7) M) and endothelium denudation. CONCLUSIONS: These results suggest that the adventitia differently modulates responses to vasoconstrictors and that it is a key layer in Ang II-induced contractions, mediating NO release from the endothelium via AT2 receptors. This increase in NO counterbalances basal superoxide release.
