The effects of single exposures of Aedes aegypti larvae and pupae to Plagiorchis noblei cercariae in the laboratory.

The mortality of Aedes aegypti pre-imagos harboring metacercariae of Plagiorchis noblei Park, 1936, is governed by the stage of development of the host at the time of infection and the location of the parasite in the insect body. First and second instar larvae generally succumbed to infection, regardless of site. Infections of the head and thorax of third and fourth instar larvae were generally lethal or gave rise to imparied adults. However, older instars frequently survived abdominal infections. Pupae showed greater tolerance to cephalic, thoracic and abdominal infections and generally emerged as adults. Again, many such infected adults were impaired.

A Drosophila melanogaster G protein alpha subunit gene is expressed primarily in embryos and pupae.

A G protein alpha subunit gene has been isolated from a Drosophila melanogaster genomic library using a combination of bovine rod and cone transducin alpha subunit cDNAs as a probe under reduced stringency conditions. The gene, DG alpha 1, encodes a protein with an amino acid sequence 78% identical to bovine Gi alpha 1. However, unlike all reported Gi alpha subunit the DG alpha 1-encoded protein is not expected to be a pertussis toxin substrate, because it lacks a cysteine at the appropriate site. The protein coding region of the gene is split by four introns. The sequence of a head tissue cDNA clone, as well as amino acid similarities to mammalian G proteins, confirms this exon/intron structure. Northern blots of total cellular RNA reveal a major 2.3-kilobase transcript and a less abundant 1.7-kilobase transcript. These transcripts are most abundant in RNA from embryos and pupae. The DG alpha 1 gene is located on band 65C on the left arm of the third chromosome, on the basis of in situ hybridizations to Drosophila salivary gland polytene chromosomes.

An inapparent infection with a probable picornavirus in several stocks of laboratory reared and naturally occurring populations of Dacus oleae Gmel. pupae in Greece.

Pupae from several stocks of wild and laboratory reared olive fruit fly, Dacus oleae, were fractionated by a series of steps designed to identify occluded and nonoccluded viruses. Two different size of particles were isolated, the smaller of which contained a single-stranded RNA molecule of about 2.8 x 10(6). This small RNA virus was found to inapparently infect a Ceratitis capitata continuous cell line.

Some effects of temperature on the adults, eggs and pupae of Stomoxys calcitrans Linnaeus (Diptera: Muscidae).

Adults could only live and reproduce to their full capacity at temperatures between 20 degrees C and 30 degrees C. At 15 degrees C the females laid no eggs, the adult life span was relatively short and the reproductive capacity of females kept at 35 degrees C was low. The thermal histories of the flies had no apparent effect on their later reactions to temperature in any of the parameters tested. The viability rates of S. calcitrans eggs exposed to temperatures between 10 degrees C and 40 degrees C exceeded 84%, but 45 degrees C was lethal. The optimum temperatures for incubation of the eggs was 30 degrees C. Pupae of S. calcitrans seemed to tolerate temperatures between 20 degrees C and 30 degrees C, but their mortalities increased markedly outside this temperature range. Tests showed that pupal mortalities increased linearly with increasing periods of exposure to a temperature of 15 degrees C.

[Optimization of the storage of housefly pupae as a utilizer of organic wastes in a human biological life support system].

The method of experimental orthogonal planning was used to study the combined effect of three factors (ambient temperature, humidity, and storage time) on the yield characteristics of the storage of Musca domestica L. pupae as utilizers of organic wastes in the biological life support system. The application of the type 2(3-1) fractional factorial experiment with the generating ratio x3 = x2x1 and analysis of the resultant regression equation showed that the pupal viability tended to grow as the temperature increased and the time storage and humidity decreased. Step-by-step optimization according to Box-Wilson gave the following optimal conditions for pupal storage (without viability losses); ambient temperature +16 degrees C, relative humidity 37-39%, and storage time 15 days.

Long chain oxoaldehydes and oxoalcohols from esters as major constituents of the surface lipids of Manduca sexta pupae in diapause.

Lipid constitutents of diapausing pupae of the tobacco hornworm, Manduca sexta (L), were identified by thin layer and gas-liquid chromatography, IR spectroscopy, and gas-liquid chromatography-mass spectrometry. Surface wax was a mixture of long chain polar compounds: oxoalcohol esters, oxoaldehydes, primary alcohol esters, and oxoalcohols, as listed in descending order of abundance. The distribution of the alcohols and aldehydes was C28 (75-85%), C27 (5%), and C26 (10-15%). The C26 compounds were largely 11-oxo isomers, but the C28 compounds consisted of similar amounts of 11- and 12-oxo isomers. The identities of the oxoaldehydes were confirmed by selective and complete NaBH4 reductions to yield oxoalcohols and diols, respectively. Mass spectral interpretations were verified with mass spectra of the oxoaldehyde, oxoalcohol, and diol synthesized from 12-hydroxystearic acid. Reduction of the total lipids with NaBH4 and hydrolysis of the product with ethanolic KOH gave oxoalcohols (85%), primary alcohols (8%), and oxoacids (5%); 30-40% of the oxoalcohols were derived from NaBH4-reduced oxoaldehydes, 5-10% were from free oxoalcohols, and 50% were from esters. Primary alcohols only existed as esters. Large quantities of fatty oxoalcohols relative to fatty oxoacids in the saponified mixture suggested the presence of esters other than those composed of long chain acids and alcohols. Oxoacids appeared to be mainly oxidation products of the oxoaldehydes.

Characterization and distribution of the hydrocarbons found in diapausing pupae tissues of the tobacco hornworm, Manduca sexta (L.).

The cuticular hydrocarbons of diapausing pupae of the tobacco hornworm, Manduca sexta (L.), are composed of two types of hydrocarbons: n-alkanes (3%) and unsaturated hydrocarbons (97%). The integument, fat body, muscle, and hemolymph are composed of three types of hydrocarbons: n-alkanes (9, 6, 9, and 2%, respectively), branched alkanes (75, 85, 80, and 56%, respectively), and unsaturated hydrocarbons (16, 9, 11, and 42%, respectively). The absence of branched alkanes on the cuticular surface indicated that hydrocarbons are selectively synthesized or deposited according to the presence or absence of methyl branches in the molecule. The hydrocarbons consisted of homologous series of n-alkanes from 21 to 41 carbon atoms, monomethylalkanes from 23 to 39 carbon atoms, dimethylalkanes from 25 to 43 carbon atoms, and trimethylalkanes from 33 to 37 carbon atoms. All branched components had methyl groups near the center of the molecule, and the di- and trimethyl branched components had three methylene units between the branch points. Straight chain alkenes, alkadienes, and alkatrienes from 23 to 44 carbon atoms were partially characterized. The percentage composition of each homologous series varied from tissue to tissue.

Multisegmental motor activity in the segmentally restricted gin trap behavior in Manduca sexta pupae.

Stimulation of sensory neurons innervating hairs in the gin traps on the abdomen of Manduca sexta pupae evokes a rapid bending of the abdomen that is restricted to one or more of the three articulating posterior segments. However, electrical stimulation of the gin trap sensory nerve in an isolated abdominal nerve cord evokes characteristic motor neuron activity in every abdominal segment. To determine if the segmentally distributed motor activity also occurred in intact animals and how it contributed to the segmentally restricted reflex movement, mechanical stimulation of the sensory hairs in intact animals was used to evoke reflex responses that were recorded as electromyograms synchronized with video recordings of the behavior. Motor activity was monitored during movements to determine if there was activity in many segments when the movement was restricted to one segment. Coordinated muscle activity was evoked throughout the abdomen in response to stimulation of any of the three gin traps, even when movement was restricted to one segment. Differences in the timing of ipsilateral and contralateral motor activity among segments allowed the closing of gin traps to be segmentally restricted. These findings suggest that the neural circuit underlying the gin trap reflex is distributed throughout the abdominal nerve cord. This network generates a complex, yet coordinated, motor pattern with muscular activity in many abdominal segments that produces a localized bending reflex.

Tracheal volume in the pupa of the Saturniid moth Hyalophora cecropia determined with inert gases.

Tracheal volume (VTr) was measured in pupae of the Giant silkworm moth Hyalophora cecropia (Saturniidae, Lepidoptera, Insecta) using inert gas wash-out techniques. The animal was placed in a small vessel that was continuously ventilated (rate, V) by a gas mixture containing 20% O2 in N2; the inflowing (F1) and outflowing gas fractions (FE) of the vessel could be continuously measured by a respiratory mass spectrometer. At the onset of a spiracular constriction period, which was evidenced from the FECO2 trace, the mixture was rapidly replaced by pure Ar. At the subsequent burst, the amount of N2 emerging from the animal, MN2, was calculated from V and the difference (FE--F1)N2. VTr was calculated from MN2 and the N2 concentration in the tracheal system before constriction (assumed to equal that in the ventilating gas before replacement by Ar). Measurements were repeated with N2 and Ar replacing each other. VTr average 48 microliter . g-1 (range 39 to 59) for animals of 5.8 g average body weight (range 3.4 to 9.9), when inert gas solubility in body fluids was accounted for. Both size and stage in pupal development appear to affect VTr. These values show reasonable agreement with literature data, mostly obtained by emptying the tracheal gas space by mechanical compression.

A material (PUPA) presenting both the properties of polyurethanes and the capacity of adsorbing a high quantity of heparin.

Polyurethanes are widely used for biomedical applications, but there is still a constant search for improved blood-compatible materials. We studied a material (PUPA) obtained by the interconnection between a poly(amido-amine), N2LL, capable of forming stable complexes with heparin and a commercial polyurethane, Pellethane 2363-80AE, using hexamethylenediisocyanate as the crosslinking agent. The amount of absorbed heparin (evaluated by biological tests) was generally much higher than that found on the poly(amido-amine) surface-grafted polyurethane.

Action of a cysteine proteinase from pupae of the blowfly Aldrichina grahami on the oxidized B-chain of bovine insulin.

A cysteine proteinase purified from pupae of the blowfly (A. grahami) was tested for its peptide-bond specificity against the oxidized B-chain of insulin. Fifteen peptides were separated on HPLC using both gradient and isocratic elution methods. Analyses of amino acid content and N-terminal amino acids indicated that these were eleven homogeneous peptides produced by digestion and undigested insulin B-chain. Glu13-Ala14 and Tyr26-Thr27 were the major cleavage sites, and Asn3-Gln4, Cys7-Gly8, Tyr16-Leu17, Leu17-Val18 and Cys19-Gly20 were also often cleaved. These findings show the similarity between this enzyme and cathepsin L.

Summer and winter diapause in pupae of the cabbage butterfly, Pieris melete Ménétriés.

The cabbage butterfly, Pieris melete is multivoltine with a pupal summer and winter diapause. Summer and winter diapause are induced principally by relatively long and short daylengths, respectively. The intermediate to relatively short daylengths of autumn permitted some pupae to develop without diapause in the field. A short daylength had a stronger diapause inducing effect than a relatively long one under higher temperatures. The principal sensitive phase for photoperiodic response occurred before the late 3rd larval instar. The critical daylength for wild autumnal populations was between 12h 30min and 12h 40min at an average temperature of 20.5 degrees C. A night interruption by 2h of light averted diapause most effectively when it was placed 10 to 12h after lights-off. High temperatures and long days during summer inhibited the incidence of diapause, suggesting that the occurrence of summer diapause is due to the specific climatic conditions occurring in April and early May, rather than to the high temperatures in summer. This indicates that the butterfly has a cryptic ability to reproduce in summer. High temperatures delayed diapause development, whereas low temperatures enhanced it, indicating that the optimum temperature of diapause development is lower. The diapause regulating mechanisms thus ensure that the species synchronises its development and reproduction with the growth seasons of the host plants and provide the species with a high degree of flexibility in its life cycle.

Studies on the posterior silk gland of the silkworm, Bombyx mori. II. Cytolytic processes in posterior silk gland cells during metamorphosis from larva to pupa.

Cytolytic processes in posterior silk gland cells of the silkworm, Bombyx mori, during metamorphosis from larva to pupa have been studied. During this stage, the wet weight and the amounts of RNA and protein of the gland decrease rapidly and markedly, while the amount of DNA decreases slowly and slightly. The ultrastructural changes observed at the beginning of the prepupal stage consist of the appearance or the increase in the number of autophagosomes containing endoplasmic reticulum (ER), or "early autophagosomes" as we have called them, which seem to be gradually transformed to autolysosomes. A number of usual lysosomes, which frequently contain myelin figures, also appear in the cytoplasm. Sometimes they fuse with each other to form large conglomerates. In the middle of the prepupal stage, a number of smooth membrane-bounded vacuoles appear in cytoplasm. Towards the end of the prepupal stage the partition or sequestration of cytoplasm was observed. Thus large autophagosomes containing cytoplasmic organelles such as rough ER and/or mitochondria are formed. The nucleus is partitioned in a similar way by smooth membranes, and then autophagosomes containing condensed chromatin blocks are formed. These various kinds of autophagosomes, or "late autophagosomes" as we have generally called them, are continuously released into the hemolymph until the gland is completely disintegrated.

Purification of a lectin from the hemolymph of Chinese oak silk moth (Antheraea pernyi) pupae.

A lectin with affinity to galactose was purified to homogeneity from the hemolymph of diapausing pupae of the Chinese oak silk moth, Anteraea pernyi. The molecular mass of this lectin was 380,000 and it formed an oligomeric structure of a subunit with a molecular mass of 38,000. The hemagglutinating activity in the hemolymph was found to increase with time after immunization with E. coli. Studies with antibody against the purified lectin showed that increase in the hemagglutinating activity was due to the same lectin, suggesting that the amount of the lectin increased in response to intrusion of foreign substances. The function of this lectin in the defence mechanism is discussed.

Isolation and characterization of malic enzyme from the pupa of Bombyx mori.

Malic enzyme, which requires NADP+ as a coenzyme, was isolated and purified from pupae of the silkworm, Bombyx mori. The purified enzyme appeared homogeneous and had a molecular weight of 195,000 on polyacrylamide gel electrophoresis. The optimum pH for the oxidative decarboxylation of malate, measured in terms of the increase of NADPH (MH activity) and CO2 (MC activity), was pH 7.5, while that for the decarboxylation of oxaloacetate measured in terms of the increase of CO2 (OC activity) was pH 4.6. Several differences between MH and OC activity were investigated.
