A comparative study on the beneficial effects of garlic (Allium sativum Linn), amla (Emblica Officinalis Gaertn)and onion (Allium cepa Linn) on the hyperlipidemia induced by butter fat and beef fat in rats.

Three months feeding of butter fat (BUF) and beef (BF) separately as components of diet at a level of 21% by weight for albino rats, significantly raised their serum and tissue lipids, lipid peroxidation and activities of certain enzymes. BUF was found to be more atherogenic than BF. On incorporation of 5% garlic, amla or onion separately in the above diets, each of them ameliorated the deleterious effects of the animal fats. A higher hyperlipidemic effect of BUF as compared to that of BF may be due to the fact that the ratio of unsaturated to saturated fats is lower for the former (0.56) than for the latter (0.75) and also that the former is richer in cholesterol content than the latter. The order of the curative effects of the vegetables are garlic>amla>onion. The better hypolipidemic effects and correction of elevated levels of certain enzymes shown by garlic and amla may be due to the facts that they contain comparatively better active principles than that found in onions.

[Studies on the stabilities of bioactive selenocompounds in selenium-enriched garlic and onion].

It is reported that selenium (Se) incorporation into garlic increases the bioactivities of garlic. Hence, the chemical changes of selenocompounds during processing and storage will influence the bioactive effectiveness of Se-enriched garlic. The principal selenocompounds in Se-enriched garlic are water-soluble, and several comparative experiments are thus conducted to examine the stabilities of the selenocompounds in water extracts of Se-enriched garlic and Se-enriched onion. The results show that preparing the garlic powder by freeze-drying technique can maintain the chemical properties of the selenium compounds in Se-enriched garlic. Se-methyl-selenocysteine is unstable in water extract of Se-enriched garlic when the extract is prepared and stored at room temperature. Specific alliinase inhibitor hydroxylamine (NHOH.HCl) effectively prevents the loss of Se-methyl-selenocysteine, which suggests that the decomposition of Se-methyl-selenocysteine may be catalyzed by alliinase. Se-methyl-selenocysteine is one of the main bioactive selenocompounds in Se-enriched garlic. The procedures of processing and storage should be carefully chosen to prevent the loss of selenocompounds and the decrease of the bioactivity of Se-enriched garlic. Se-enriched onion also contains alliinase and Se-methyl-selenocysteine, but its Se-methyl-selenocysteine is proved to be stable in the same water extract as that of Se-enriched garlic. The stability differences of Se-methyl-selenocysteine in Se-enriched garlic and onion, the mechanism of selenocompound decomposition and the bio-activities of decomposed compounds in Se-enriched garlic need to be further studied.

Residues of profenofos in spring onion.

Profenofos is one of the commonly used insecticides in the control of Trips tabaci on spring onion in Iran. Residues of profenofos in spring onion were determined in two different fields under the same conditions. In the first field, onion plants were sprayed with profenofos (40EC) at the rate of 1000 g/ha. Spraying was repeated 2 weeks later. In the second field one spraying was preformed at the same rate. Spring onionwere sampled at different time intervals and analyzed for profenofos residues using a GC equipped with NPD detector. In the first field's samples, the residues were 0.097 and 0.025 mg/kg at 2 and 6 days after spraying, respectively. The residues declined to 0.002 mg/kg on the day twelve. Two days after the second spraying the residues was 0.27 mg/kg, which reduced to 0.032 on the day sixth. However the residues were not detectable 32 days after the second spraying. In the second field, residue levels were 0.193 and 0.043 mg/kg at 2 and 6 days after spraying. Residues, which found after 32 days, were less than 0.001 mg/kg. The rate of residue decay in the first field was higher than the second field.

Inhibition of soybean lipoxygenase and mouse skin tumor promotion by onion and garlic components.

Onion and garlic essential oils were previously shown to inhibit mouse skin tumor promotion, as were the enzymes, lipoxygenase, and cyclooxygenase. In the present study, the inhibition of soybean lipoxygenase (EC 1.13.11.12) by onion and garlic components and related compounds was investigated. The IC50 values as well as the kinetic inhibition constants were determined for the most active compounds. Di-(1-propenyl) sulfide, an analog of the substrate moiety required for oxygenase action, was the only irreversible inhibitor observed with Ki = 59 microM and k3 = 0.53/min. Inhibition in the presence of substrate was uncompetitive at 88 and 132 microM linoleic acid with Ki = 129 microM. At 173 microM linoleic acid, however, inhibition was competitive with Ki = 66 microM. Dially trisulfide, allyl methyl trisulfide, and diallyl disulfide were competitive inhibitors, while 1-propenylpropyl sulfide and (E, Z)-4,5,9-trithiadodeca-1,6,11-triene 9-oxide (ajoene) were mixed inhibitors. Nordihydroguaiaretic acid (NDGA), the most potent lipoxygenase inhibitor, was a competitive inhibitor with Ki = 0.29 microM. The results indicate a relative potency of inhibition for structural features in the following order: di(1-propenyl) sulfide greater than an alkenyl trisulfide greater than an alkenyl disulfide. Di(n-propyl) disulfide, a major onion oil component, inhibited neither lipoxygenase nor promotion. Di(1-propenyl) sulfide and ajoene inhibited both. This suggests that the inhibition of lipoxygenase may be involved in antipromotion.

Cloning and characterization of the lectin cDNA clones from onion, shallot and leek.

Characterization of the lectins from onion (Allium cepa), shallot (A. ascalonicum) and leek (A. porrum) has shown that these lectins differ from previously isolated Alliaceae lectins not only in their molecular structure but also in their ability to inhibit retrovirus infection of target cells. cDNA libraries constructed from poly(A)-rich RNA isolated from young shoots of onion, shallot and leek were screened for lectin cDNA clones using colony hybridization. Sequence analysis of the lectin cDNA clones from these three species revealed a high degree of sequence similarity both at the nucleotide and at the amino acid level. Apparently the onion, shallot and leek lectins are translated from mRNAs of ca. 800 nucleotides. The primary translation products are preproproteins (ca. 19 kDa) which are converted into the mature lectin polypeptides (12.5-13 kDa) after post-translational modifications. Southern blot analysis of genomic DNA has shown that the lectins are most probably encoded by a family of closely related genes which is in good agreement with the sequence heterogeneity found between different lectin cDNA clones of one species.

Stimulation of cell growth and proliferation in NIH-3T3 cells by onion and garlic oils.

Onion and garlic oil were seen to shorten the cell-doubling time and stimulate the growth and proliferation of NIH-3T3 cells. Following treatment with either onion or garlic oil, an increase in the growth rate and almost a 2-fold increase in cell number over the control was observed within a 24-hour period. Phorbol myristate acetate when given simultaneously with either oil appeared to nullify both effects. Following exposure to low doses (less than 10 micrograms/ml) of either oil, an increase in cell survival, not seen with the oil control tricaprylin, was observed following a 5-day exposure period. At higher concentrations cell survival decreased proportionately in all cases. The appearance of multinucleated cells, which increased with dose and time, was also observed following treatment with both garlic and onion oil.

[Analysis of a synthetic onion aroma (author's transl)].

A complex mixture of compounds possessing an intensive aroma of onions is obtained, when propionaldehyde, propionaldehyde and acetaldehyde are treated with hydrogen sulfide and ammonia. Dihydrodithiazines, diethyltrithiolane, diethyl-methyl-1,3-dithiine and in small quantities diethyltetrathiane, dimethylthiophene, dipropyldisulfide, dipropenylsulfide, alkylpyridines, and the condensation products of aldehydes can be proved by the GC-MS-technic. The compositions of the synthetic onion aroma corresponds slightly with the composition of the natural onion aroma.

Prevention of allergen-induced bronchial obstruction in sensitized guinea pigs by crude alcoholic onion extract.

Thirteen guinea pigs, sensitized to ovalbumin, were pretreated with either 1 ml alcoholic onion extract (= 12% v/v ethanol) or 1 ml 12% ethanol solution (control) according to a randomized crossover protocol. Thirty minutes later the animals were challenged twice for 30 sec (n = 5), respectively 60 sec (n = 8), by the inhalation of ovalbumin (1 ml 1% ovalbumin solution nebulized in 10 l air). The intensity of bronchial obstruction was measured by whole body plethysmography using as parameter the amount of 'compressed air'. Oral pretreatment of guinea pigs with onion extract markedly reduced the asthmatic response (p less than 0.02). After onion pretreatment near normal values were obtained after 30 sec challenge (0.04 +/- 0.06 ml compared to 0.24 +/- 0.15 ml in the control) and only slightly increased values after 60 sec challenge (0.16 +/- 0.07 ml compared to 0.33 +/- 0.25 ml in the control).

Schwann cell-onion bulb tumor of the trigeminal nerve: hyperplasia, dysplasia or neoplasia?

Onion bulbs are concentric lamellar structures formed by Schwann or perineurial cells, which may be seen in several generalized or localized diseases of the peripheral nerve. There is debate regarding the pathogenesis of localized tumefactions displaying these microscopic structures. We report the fifth case, to our knowledge, of a Schwann cell-onion bulb tumor, which arose in the trigeminal nerve of a child with an unclassifiable, probably distinct, neurocutaneous syndrome; we also provide evidence for a neoplastic or hamartomatous origin. Molecular studies failed to establish an abnormality in the NF1, NF2, PMP22, or Connexin 32 genes. Similar and previously reported cases are discussed, as well as other onion bulb-forming entities.

Characterisation of the welsh onion isolate of Shallot yellow stripe virus from China.

The host range and nucleotide sequence of shallot yellow stripe virus (SYSV) from welsh onion in Shandong province, China is described. Of the plants tested, only shallot and welsh onion became infected but most shallot plants were symptomless. The complete sequence of one isolate (10429 nt) and the 3'-terminal 3540 nts of a second isolate were determined. They had c. 90% nt identity to one another and to published (partial) sequences of SYSV. SYSV was most closely related to onion yellow dwarf virus (OYDV) and resembled it in having a much larger P3 protein than other species in the genus.

Comparative effect of clofibrate, garlic and onion on alimentary hyperlipemia.

The effect of clofibrate on the same subjects in similar test conditions were used as a control to verify the alleged beneficial effects from garlic and onion on alimentary hyperlipemia in normals and in cases with ischemic heart disease. The results showed that clofibrate checked the fat-induced (a) rises of serum triglyceride and plasma fibrinogen, and (b) falls of coagulation time (CT) and blood fibrinolytic activity (BFA). Only garlic had a clofibrate-like effect on CT but both garlic and onion checked the postprandial fall of BFA. Clofibrate, however, increased BFA even above the fasting level. Serum cholesterol and beta-lipoprotein were not appreciably affected by fat with or without any drug. Thus, surprisingly, the so-called beneficial effects of garlic and onion were not seen in subjects who had shown significant changes after clofibrate.

Cytological studies on onion root-tip cells treated with water-soluble extract of tobacco smoke condensate from commercial cigarettes.

The effect of water-soluble extract of tobacco smoke condensate (TSC) from two commercially available cigarettes with dirrerent types of filters was studied on the cytology of root-tip cells of onion (Allium cepa). One of the cigarettes had a 2-cm cellulose acetate filter, and the other had a filter comprised of 1 cm of cellulose acetate and 2 cm of activated charcoal. TSC from these cigarettes induced mitotic abnormalities. To investigate whether these two commercial filters could retain cigarette smoke component (s) responsible for mitotic irregularities, the cigarettes were defiltered, and TSC was prepared and tested on the young roots of onion. Observations revealed that the cytological effect of TSC from defiltered cigarettes was not significantly different from the effect of TSC from cigarettes with filters. Thus, the filters utilized in these cigarettes do not retain compound(s) responsible for mitotic irregularities in the root-tip cells of onion. With increasing concentrations (0.01% ot 0.1%) of TSC from cigarettes with filters and defiltered, precent mitotic abnormalities increased. These abnormalities included scattering, stickiness, lagging, condensation, and breaking of chromosomes during metaphase. Bridging and lagging of chromosomes were observed during anaphase.

Effects of aqueous extracts of onion, garlic and ginger on platelet aggregation and metabolism of arachidonic acid in the blood vascular system: in vitro study.

Aqueous extracts of onion, garlic and ginger were found to inhibit aggregation induced by ADP, epinephrine, collagen and arachidonate in a dose-dependent manner in vitro. In the case of onion and garlic extracts relatively much higher volumes were need to bring about even a modest inhibition (by ca. 13-18%) of thromboxane synthesis in washed platelets from labelled AA. On the other hand a good correlation was found between the amounts of ginger extract needed to inhibit platelet aggregation and those to inhibit platelet thromboxane synthesis. Ginger extract reduced also platelet prostaglandin-endoperoxides. A dose-related inhibition of platelet thromboxane- and prostaglandin (PGF2 alpha, PGE2 and PGD2) synthesis was affected by ginger extract. Extracts of onion, garlic and ginger inhibited biosynthesis of prostacyclin in rat aorta from labelled AA. Ginger extract mildly inhibited the synthesis of prostacyclin from endogenous pool of AA in rat aorta; the other two extracts were without effect.

Changes in non-structural carbohydrate composition during bulbing in sweet and high-solid onions in field experiments.

The composition of non-structural carbohydrate (NSC) content of predominantly long-day onion germplasm has been assessed over several years and in a bi-location trial. It was observed that genetic rather than environmental factors determined the NSC composition of onion bulbs. Glucose was the NSC component which was most closely correlated with genotype. Fructose was the only NSC component that was significantly affected by environment. Sucrose and 1-kestose (DP3 fructans) were not correlated to a large extent to the other NSC components, indicating their transient role in the fructan metabolism. Strong negative correlations were observed between reducing sugars (i.e. fructose and glucose) and dry matter content (DM). Furthermore, it was shown that accessions differed significantly in their fructan accumulation pattern; high DM accessions showed accumulation of fructans over the whole bulbing period, whereas, low DM accessions quickly reached a plateau. Implications for the breeding of high quality onions are discussed.

Organic functionalization and optical properties of carbon onions.

We report, for the first time, the organic functionalization of carbon onions. The functionalization was achieved via 1,3-dipolar cycloaddition of azomethine ylides to a mixture of amorphous carbon and concentric fullerenes. Once functionalized, the onions become soluble in organic solvents and it is possible to purify them from the rest of carbon. Their nonlinear optical properties have been studied in solution for application as broadband optical limiters. It was found that the functionalized onions are an example of soluble material that functions equally well in both the visible and the NIR regions.

The onion-ring structure for Pd-Pt bimetallic clusters.

The onion-ring structure is validated in the Pd-Pt bimetallic clusters of total atom numbers 147 and 309 through the Monte Carlo method by using the second-moment approximation of the tight-binding (TB-SMA) potentials, which is conceived in predicting the possible structures of the bimetallic clusters by He et al. [J. Am. Chem. Soc. 2003, 125, 11034] and Hwang et al. [J. Am. Chem. Soc. 2005, 127, 11140]. In the onion-ring structure, Pd atoms and Pt atoms occupy alternate layers of the clusters. The formation of the onion-ring structure can be associated with the fact that the single Pt impurity is favorable to stay in the subsurface layer and the central part of bimetallic clusters.

Onion phases as biomimetic confined media for silica nanoparticle growth.

Phospholipid onion phases were investigated as biomimetic media for the synthesis of silica in a confined environment. Stable multilamellar nanovesicles incorporating sodium silicate solutions could be obtained. Upon aging, silica condensation occurs in the onion interlayer space while preserving the initial multilamellar organization. The hybrid structure consists of an array of apparently unconnected silica nanoparticles in the 20-30 A size range packed in the vesicular 50 A interlayer space, suggesting that the silica growth was efficiently controlled by its confinement in the onion lamellar organization.
