[Effects of opium peptide system on the temperature regulation of mice under dry-hot environment with different temperature].

The Kunming mice were selected as subjects. Animals were allowed to move and drink freely. The Naloxone(0.02 mg/g) was injected by abdominal cavity. The ability to body temperature stable by mice under the environment with high temperature (41 degrees C, 43 degrees C) was observed. The results showed that after obstructing opium receptor by Naloxone, the ability mentioned above was reduced obviously.

[The disturbances of sensitivity in women with opium addiction].

52 women with opium addiction in remission (deprivation as a result of imprisonment) were examined. The disorders of appreciation pain were found in 50 cases (96 +/- 3%). There were clinical peculiarities of their topography (composed distal, maculary-mosaic and perioral-axial versions). These peculiarities were shown in alcoholism, neuroses, neurosis-like nonpsychogenic conditions of infection and traumatic genesis. The supposed mechanisms of described disorders are also presented.

[Study on the pathogen and its biological characteristics of opium poppy downy mildew].

OBJECTIVE: To study the pathogen of opium poppy downy mildew and its biological characteristic for further research on the disease. METHOD: Development of the disease was observed systematically in the field. Germination rate of sporangium in different temperature, pH and nutrition was examined with suspending-drop method. Slide-germination method was used to observe its germination in different humidity maintained by different concentration of H2SO4. RESULT AND CONCLUSION: The disease manifests itself in two forms: severely infected plants (systematic infection) and leaf spots (nonsystematic infection). Sporangia of the pathogen are oval or globular, thin walled, smooth, hyaline, with 7.74-16.34 microns diameter in base 1 and 8.34-15.05 microns in base 2.0 ospores are light yellow with 33.87-70.54 microns x 19.34-62.64 microns in base 1 and 36.85-49.68 microns x 42.08-55.76 microns in base 2. Conidiophores are stout, erect, whose branching times and length are different between those in base 1 and those in base 2. Sporangia sprot directly in two hours. Film of water is necessary for sporangium to sprot. The optimum temperature range of sporangium sprot is 12-21 degrees C, the best being 16 degrees C, the pH range is 4.53-9.18 the best optimum at pH 7.38, and the extract of leaf of 1:5 is good for its germination.

[Mycotic lesions in the brain in chronic opium-ephedrone addiction].

Histological examination of the brain of a opium-ephedrone addict has detected large spherical cells resembling Coccidioides or Paracoccidioides spherules. Roundish and oval structures were found in subependymal space of the fourth ventricle. These structures may be spores of many fungi. Areas of normal structure and these with productive inflammation were observed in the surrounding nervous tissue.

[The development of the Japanese pharmaceutical industry (Part 8) - the change of opium alkaloid opioid analgesics in Japanese pharmaceutical companies.].

After the Japan-England Friendship and Commercial Treaty of 1858 (Ansei 5), narcotic drugs have been strictly regulated by laws. The production of opium alkaloid analgesics, mainly morphine, became active in Japan after World War I (1918, Taisho 7). Having released the "Cancer Pain Relief" guideline by the WHO in 1986 (Showa 61) and the morphine sulfate controlled-release tablet "MS Contin" in 1989 (Heisei 1) in Japan, the demand for morphine in Japan expanded remarkably. Today, several more kinds of strong opioid analgesics are available for cancer pain treatment in Japan. An increase in the use of strong opioid analgesics is expected in the future.

[Psychopathological heterogeneity in opium drug addicts].

The aim of the study was to assess the presence and nature of psychiatric disorders in opium drug addicts. One hundred and six subjects receiving treatment at the CMAS in Cagliari were included in the study. Hathawai and McKinley's MMPI test was preventively carried out on all subjects; each drug addict was then interviewed three times in the space of three weeks in order to formulate a diagnosis in line with DSM III R criteria. The results obtained show a high incidence of psychopathological disorders which are not included among those caused by drug abuse, and a high degree of diagnostic heterogeneity on both axis I and axis II. The comparative assessment of three subsamples undergoing different phases of treatment reveals both qualitative and quantitative differences.

Environmental effects on morphine content in opium poppy (Papaver somniferum L.).

Ten cultivars of opium poppy were tested for morphine content during the past five years at the Indian Agricultural Research Institute (IARI), New Delhi. Techniques employed to determine the adaptive potential (Finlay and Wilkinson, 1963; Eberhart and Russel, 1966), relative stability and environmental interaction showed the significant varietal differences when the cultivars' mean square was tested over pooled deviations as the error term. Significant V- X -E (linear) component suggests the existence of genetically conditional differences in the stability of cultivars. Highest morphine content was observed in cultivar H and least in R. Four cultivars exceeded the grand mean. Within the five-year study period three were observed to have given the highest morphine content.

Differential and tissue-specific expression of a gene family for tyrosine/dopa decarboxylase in opium poppy.

Two early and potential rate-limiting steps in the biosynthesis of isoquinoline alkaloids, such as morphine and codeine, in opium poppy (Papaver somniferum) involve decarboxylation of L-tyrosine and L-dihydroxyphenylalanine (L-dopa) to yield tyramine and dopamine, respectively. A DNA fragment was amplified by polymerase chain reaction (PCR) using degenerate primers designed to two highly conserved domains found in other aromatic amino acid decarboxylases. A poppy seedling cDNA library was screened with this PCR product and a cDNA (cTYDC1) for tyrosine/dopa decarboxylase (TYDC/DODC) was isolated. Two other independent cDNAs (cTYDC2 and cTYDC3) encoding TYDC/DODC were isolated by heterologous screening with a plant tryptophan decarboxylase (TDC) cDNA as probe. A poppy genomic library was screened with cTYDC1 and two intronless genomic clones (gTYDC1 and gTYDC4) were isolated. The deduced amino acid sequences of all poppy clones share extensive identity with other reported pyridoxal phosphate-dependent decarboxylases from both plants and animals. Based on sequence homology, members of the gene family were divided into two subsets (cTYDC1 and gTYDC4; cTYDC2 and cTYDC3) of proteins with predicted M(r) = 56,983 and 59,323, respectively. Within each subset the clones exhibit greater than 90% identity, whereas clones between subsets share less than 75% identity. Expression of gTYDC1 and cTYDC2 as beta-galactosidase fusion proteins in Escherichia coli resulted in catalytically active enzymes immunodetectable with TDC-specific polyclonal antibodies. Each enzyme showed marginally higher substrate specificity for L-dopa over L-tyrosine, but did not accept L-tryptophan and L-phenylalanine as substrates. Genomic DNA blot-hybridization analysis revealed 6 to 8 genes homologous to cTYDC1 and 4 to 6 genes homologous to cTYDC2 in the tetraploid poppy genome. A premature translation stop codon was found in the gTYDC4 clone suggesting that it may not encode a functional protein. RNA blot-hybridization with probes specific to the gTYDC1- or cTYDC2-like subsets showed that members of the TYDC gene family are differentially expressed in various plant tissues.
