Class II genes of miniature swine. IV. Characterization and expression of two allelic class II DQB cDNA clones.

Two cDNA clones coding for allelic miniature swine MHC class II Ag DQB chains have been isolated, characterized, and shown to be expressed after transfection into mouse fibroblasts. The two alleles differ at the nucleotide level by an overwhelming proportion of replacement substitutions, suggesting the influence of selection for polymorphism. Most of the resulting predicted amino acid replacements are in regions commonly polymorphic in mouse Ab and human DQB sequences, corresponding to the predicted Ag recognition site. Nucleotide and amino acid sequence comparisons to homologous mouse and human sequences show more similarity between swine and man than between either swine and mouse or man and mouse. This tendency is most pronounced when comparing the 3' untranslated regions. However, an examination of unique cross-species sharing of amino acid residues suggests a closer relationship between both man and miniature swine and man and mouse than between miniature swine and mouse. The simplest explanation we can envision for these findings is that the mouse DQB gene homologue (Ab) has been subject to a higher substitution rate than either swine or human DQB genes. An additional cytoplasmic exon expressed in mouse Ab gene products and in putative human DQB2 gene products is lacking in both swine and human DQB cDNA clones. Its absence suggests either that the expression of this exon in mouse Ab genes was activated after mammalian speciation or that the expression of this exon was independently inactivated in swine DQB and human DQB1 genes. Alternatively, the mouse Ab gene may be derived from the same primordial gene as human DQB2, whereas the pig DQB gene may be derived from the same primordial gene as the human DQB1 gene.

[Functional morphology of the maxillo-mandibular apparatus in the miniature swine MINI-LEWE. 11. Discussion of the findings of the metric studies and dentition (conclusion)].

The graphs showing the dimensions and angles of the skull and jaw indicate that postnatal development trends are basically uniform. Comparison with the Vietnamese miniature pig shows that the miniature pig MINI-LEWE has a longer skull. Postnatal skull growth is accompanied by a distinct shift in proportions. The sequence of dentition if broadly identical to that of the domestic pig and the Pitmann-Moore miniature pig.

A quality study of 35 mm, miniaturization of full-size radiographic film.

Twenty-one radiologists with 6 months--25 years experience judged the quality and the possibilities for the routine use of miniaturized full-size radiographic films for long time archiving. Twenty-eight examinations on full-size radiographs were photographed on 35 mm. film. After mounting in transparent jackets, the miniaturized films were read with enlarging equipment and diagnoses and quality noted. Comparison was later made with the originals. The consensus was that the quality of the miniaturized film was good enough to permit their use for long-term archiving purposes and subsequent comparison with future full-size radiographs. The use of reading equipment was not considered tedious.

Serum biochemistry of healthy Yucatan miniature pigs.

Sera collected from 24 (12 boars and 12 gilts) healthy, fasted 28-43 week-old Yucatan miniature pigs were analyzed for 21 clinical chemistry parameters. The mean, standard deviation, median, and observed range for each parameter are presented as reference values for the normal blood chemistry for this breed. Comparison with published values reveals that the Yucatan miniature pig has a blood chemistry profile comparable to that of domestic pigs and other breeds of miniature swine.

[Comparison of conventional and miniaturized biplane echoscope: initial clinical results].

A subset of patients experiences substantial discomfort on examination with transesophageal echocardiography using the conventional echoscope, whereby the dimensions of the probe play a decisive role. Miniaturized biplane transducers have recently become available (2 x 32 crystals; dimensions 9.5 x 8.7 mm; circumference approx. 30% less than the conventional echoscope), which allow ultrasound examination at 3.5, 5.0 and 7.0 MHz. A prospective study was carried out in 70 patients to compare difficulties on insertion of the probe, subjective evaluation by the patient during examination, and the 2D-image as well as Doppler and color-coded Doppler quality of the miniaturized biplane echoscope. In 43 patients, intubation of the esophagus proved less difficult with the smaller instrument, it was more difficult in 8 cases. 54 patients reported that the discomfort was definitely easier to bear on use of the narrow instrument. Concomitant parasympathicolytic medication was needed with the smaller probe in 4 cases, and 11 times with the conventional echoscope. The quality of the 2D-image attained by the miniaturized probe was naturally lower. Employing multi-Hertz technology at a frequency of 7 MHz, however, imaging of the anatomy was excellent in the near field of 5-6 cm, and nearly equivalent to that of the conventional probe (5 MHz). The quality of PW-and CW-Doppler as well as color-coded Doppler information was identical. Whenever examination with a conventional echoscope promises to be difficult, or when sedation is contraindicated due to a severe illness or respiratory insufficiency, transesophageal echocardiography should be considered with a smaller biplane probe at higher frequencies.

Nitric oxide inhibition causes an exaggerated pressor response in Yucatan miniature swine.

The involvement to nitric oxide (NO) in cardiovascular and renal function was evaluated in 12 anesthetized Yucatan miniature swine. The effect of NO blockade on blood pressure was measured in six additional conscious swine. In the anesthetized swine, mean arterial pressure (MAP), heart rate, glomerular filtration rate (GFR), and urinary excretion of water, sodium, and potassium were measured after systemic inhibition of NO synthesis by NG-nitro-L-arginine methyl ester (L-NAME), and were compared with values for a control period. After NO synthesis blockade, MAP increased by 63 +/- 5 mm Hg, a far greater increase than those observed in rats, dogs, domestic swine, or humans. The changes in GFR, urine flow rate (UFR), and sodium excretion (UNaV) were time-dependent. The GFR decreased to 50 +/- 6% of control values immediately after L-NAME administration, but returned to control values within 1 h. Significant increases in UFR and UNaV were observed only during the third experimental period, 40 to 60 min after drug infusion. In the conscious swine, L-NAME administration increased MAP by 24 +/- 4 mm Hg. Administration of the sympatholytic hexamethonium bromide fully reversed the increase of MAP in anesthetized and conscious swine. These findings indicate that NO has an important role in the maintenance of cardiovascular and renal function in Yucatan miniature swine. The exaggerated pressor response to NO blockade in miniature swine appears to involve the sympathetic nervous system.

[Histopathological study of miniature pig temporomandibular joint after missing teeth of one side].

This study evaluated the histologic response of the temporomandibular joint (TMJ) following one side posterior teeth extraction in miniature pig. Right posterior teeth were removed in 12 miniature pigs. Six of them were killed after 1 month and the other six after 3 monthes. 6 miniature pigs were taken as control. We found that obvious degenerative changes were common in all TMJs of the experimental pigs. Hyaline thrombus found in the condyles of posterior teeth losing side suggested that local microcirculation was disturbed. Missing one side posterior teeth had various influence on two side TMJs. Both TMJs changes were not identical, and the changes were more marked in teeth lost side than in control side. The results comfirm that occlusal disturbance can cause degenerative change of the TMJ.

Ion/molecule reactions in a miniature RIT mass spectrometer.

Ion/molecule reactions were explored in a newly developed miniature mass spectrometer fitted with a rectilinear ion trap (RIT) mass analyzer. The tandem mass spectrometry performance of this instrument is demonstrated using collision induced dissociation (CID) and ion/molecule reactions. The latter includes Eberlin transacetalization reactions and electrophilic additions. Selective detection of the chemical warfare simulant dimethyl methyl phosphonate (DMMP) was achieved through selective Eberlin reactions of its characteristic phosphonium fragment ion CH3OP(+)(O)CH3 (m/z 93), with 1,4-dioxane or 1,3-dioxolane. Efficient adduct formation as a result of electrophilic attack by the phosphonium ion on various nucleophilic reagents, including 1,1,3,3-tetramethyl urea, methanesulfonic acid methyl ester, dimethyl sulfoxide and methyl salicylate, was also observed using the RIT device. The product ions of these reactions were analyzed using CID and the characteristic fragmentation patterns of the ionic addition products were recorded using multiple-stage experiments in the miniature RIT instrument. This study clearly demonstrates that a small, home-built, miniature RIT mass spectrometer can be used to perform analytically useful ion/molecule reactions and also that instruments like this have the potential to provide a portable platform for in situ detection of organophosphorus esters and related compounds with high specificity using tandem mass spectrometry.

Postsynaptic structure may account for variations in miniature endplate current shapes along frog neuromuscular junctions.

Intracellular recordings were made in the presence of 6-9 microM neostigmine with an electrode placed halfway between the distal end and the proximal region of a long simple branch of a frog neuromuscular junction. During the intracellular recording period, an extracellular electrode was placed alternatively at a distal and at a proximal site. The temporal correspondence between the extracellular miniature endplate current and intracellular miniature endplate potential permitted us to identify two miniature endplate potential (MEPP) populations originating, respectively, from the distal and proximal regions. The frequency and intracellular amplitude of MEPPs were higher in the proximal than in the distal region. The rise time and the decay time of the extracellularly recorded MEPC were longer in the proximal region. A significant correlation was observed between the amplitude (in mV) and surface (in mV.ms) of MEPCs. The slope of this regression line was steeper for the proximal region. Furthermore, the correlation coefficient was lower for that region owing to variations of MEPC shape. Our observations suggest that in the presence of anticholinesterase, ACh rebinds more often to acetylcholine receptors in the proximal region because of the presence of longer postjunctional folds.

Plasma lipid changes in PRCD-affected and normal miniature poodles given oral supplements of linseed oil. Indications for the involvement of n-3 fatty acids in inherited retinal degenerations.

It has previously been shown that miniature poodles with progressive rod-cone degeneration (PRCD) have lower plasma levels of docosahexaenoic acid (22:6n-3) than normal poodles and it has been suggested that affected animals have a defect in the metabolism of 22:6n-3. To test this hypothesis in vivo, PRCD-affected and normal miniature poodles were given daily oral supplements of linseed oil (enriched in 18:3n-3). Blood was drawn from food-deprived animals at predetermined times before, during and after supplementation, and plasma lipid fatty acids were analysed. There were no differences in the levels of 18:3n-3, 20:5n-3, and 22:5n-3 between affected and normal dogs. Therefore, there appears to be no abnormality in the elongation and desaturation system that takes 18:3n-3 to 22:5n-3. Surprisingly, the plasma level of 22:6n-3 was reduced in both groups following supplementation, but to a significantly greater extent in affected dogs. This resulted in a significantly higher 22:5n-3/22:6n-3 ratio in affected animals. These results support the earlier suggestion of an abnormality in 22:6n-3 metabolism in PRCD-affected miniature poodles. To determine the effect of n-3 supplementation on polyunsaturated fatty acid metabolism in dogs (not as a function of disease), results from both groups of dogs were pooled and compared at times before and near the end of supplementation. Dietary 18:3n-3 led to predictable increases in 18:3n-3, 20:5n-3, and 22:5n-3, but to a decrease in 22:6n-3.(ABSTRACT TRUNCATED AT 250 WORDS)

Properties of miniature postsynaptic currents during depolarization-induced release at a cholinergic neuroneuronal synapse.

1. Miniature postsynaptic currents were analyzed at an inhibitory cholinergic neuroneuronal synapse in the buccal ganglion of Aplysia. Under double voltage-clamp, it was possible to induce postsynaptic currents by long-duration depolarizations of the presynaptic neuron and to analyze these as the linear summation of individual miniature postsynaptic currents (MPSCs). The amplitude of these miniature currents (imin) was calculated from the ratio of the variance of the noise (E2) to the mean of the postsynaptic current (Im), according to Campbell's theorem, with imin = 2E2/Im. Their decay time (tau min) was obtained from the cutoff frequencies of the power spectra obtained from the noise. 2. Neither the conductance nor the decay time of MPSCs was voltage dependent. However, imin appeared to decrease when the quantal content of the response increased. Meanwhile, tau min increased slightly with Imin. 3. Carbamylcholine was injected into the neuropile and this led to a decrease in imin and a slight increase in tau min. 4. Power spectra obtained after the application of inhibitors of acetylcholinesterase (AChE), with or without curare, suggested that acetylcholine (ACh) does not accumulate during large depolarizations. 5. The possible origin of the nonlinear relationship between the variance and the mean of the postsynaptic currents is discussed.

Optical coherence tomography of the Ex-PRESS miniature glaucoma implant.

PURPOSE: To describe the localisation of the Ex-PRESS miniature glaucoma implant with an experimental setup for optical coherence tomography (OCT) of the anterior segment of the eye. METHODS: An OCT scanner, central wavelength 1,280 nm, bandwidth 60 nm, resolution of 12 microm, was built onto a slitlamp to scan the anterior segment of the eye. Five ex-vivo porcine eyes received an Ex-PRESS miniature glaucoma implant and were used as a model to visualise the position of the implant in the anterior segment. RESULTS: In the ex-vivo porcine eyes, the OCT images showed the anatomy of the anterior segment in great detail. The anterior segment OCT was able to visualise the whole outline and position of the implant. The abrupt change in reflectivity going from tissue to the implant resulted in a clear border along the circumference of the whole device. CONCLUSION: In this paper, we have shown that we were able to outline the Ex-PRESS miniature glaucoma implant in the anterior segment of the ex-vivo porcine eye by using an experimental OCT setup built onto a slitlamp. The acquisition time of 0.8 s is short enough to allow for the scanning of patients, and anterior segment OCT is expected to aid in providing answers to the question regarding which parameters will determine the success or failure of such a device.

Plasma lipid abnormalities in the miniature poodle with progressive rod-cone degeneration.

The miniature poodle with progressive rod-cone degeneration (prcd) is a model for human retinitis pigmentosa (RP). Since previous studies from several laboratories have shown abnormalities in plasma lipids in human RP, we examined the plasma lipids of prcd-affected animals. Fasting blood was drawn on three separate occasions from affected and control miniature poodles and on one occasion from normal Irish setters and those affected with a different inherited retinal degeneration (rod-cone dysplasia). Plasma phospholipids from prcd-affected animals had significantly lower levels of docosahexaenoic acid (22:6 omega 3) and cholesterol, compared to control miniature poodles. No differences were observed in plasma levels of phospholipids, vitamin E, or vitamin A, and no lipid differences were found between control and affected Irish setters. The ratios of 22:5 omega 3 to 22:6 omega 3 and of 22:4 omega 6 to 22:5 omega 6 were significantly elevated in prcd-affected poodles compared to controls. Since the conversion of 22:5 omega 3 to 22:6 omega 3 and of 22:4 omega 6 to 22:5 omega 6 is catalysed by a delta 4-desaturase, these results are consistent with a defect in desaturase activity in the prcd-affected poodle.

A practical miniature long-term bone marrow culture system for investigating early myelodysplasia.

A method was devised to grow haemopoietic cells in long-term bone marrow culture (LTBMC) which requires only 1 x 10(6) cells/culture. Such miniature cultures were used to study growth patterns of marrow from patients with myelodysplastic syndromes (MDS). Consistent differences in LTBMC cellularity and cellular composition were noted between MDS and normal marrow. These differences were accentuated by rGM-CSF. The criteria which distinguished between and MDS marrows were: cell count at weeks 1 and 4, % neutrophils and % blasts. In 10 patients with unexplained macrocytosis or pancytopenia miniature LTBMC results clearly segregated into either 'normal' or 'MDS' growth patterns. Miniature LTBMC with rGM-CSF may therefore be a useful diagnostic test for early MDS.

The formation of glutamatergic synapses in cultured central neurons: selective increase in miniature synaptic currents.

The formation of synapses between cultured rat thalamic neurons was studied with electrophysiological and immunocytochemical methods. Thalamic neurons in culture form predominantly glutamatergic synapses. Already after 3 days in vitro glutamatergic miniature EPSCs occurred spontaneously and their frequency was strongly increased after K+ depolarization, while GABAergic mIPSCs were found after K+ depolarization at lower frequency. This demonstrates that both, excitatory glutamatergic and inhibitory GABAergic synapses were functional in close succession to initial neurite outgrowth. Synapses formed independent of spontaneous electrical activity, which was absent during the first week in culture. Spontaneous action potentials appeared during the second week and chronic action potential blockade by addition of tetrodotoxin reduced neuronal survival and the number of glutamatergic synapses per neuron. During in vitro differentiation the number of synapsin I immunoreactive presynaptic terminals and the frequency of spontaneous glutamatergic miniature EPSCs increased closely correlated, while the frequency of GABAergic mIPSCs after K+ depolarization did not increase. Thus, the continous formation of presynaptic terminals, including possible maturation of transmitter release, appeared to underlie the increase in mEPSC frequency. Analysis of miniature EPSC amplitudes at different stages in vitro revealed an increase in amplitudes, suggesting synaptic differentiation after initial establishment of functional transmission in glutamatergic synapses. This process was synapse specific as amplitudes of GABAergic mIPSCs were invariant.

Miniature postsynaptic currents recorded from identified rat spinal dorsal horn projection neurons in thin-slice preparations.

Whole-cell voltage-clamp recordings were made from spinothalamic and spinomesencephalic tract neurons in thin-slice preparations of rat spinal cord. In the presence of tetrodotoxin, spontaneous inward and outward postsynaptic currents were observed near the resting membrane potential. These currents were divided into miniature excitatory postsynaptic currents (mEPSCs) mediated by glutamate, and miniature inhibitory postsynaptic currents (mIPSCs) mediated by glycine or gamma-aminobutyric acid (GABA). Glutamatergic mEPSCs had two components mediated by NMDA and non-NMDA receptors. Analyzing these miniature synaptic currents, valuable information concerning the pre- and postsynaptic mechanisms underlying modulation of synaptic transmission in the spinal dorsal horn could be obtained.

Effects of divalent cations on the time course of post-tetanic decay of miniature endplate potential frequency in frogs.

Relatively high external Mg2+ specifically induces a shoulder in the post-tetanic decay of miniature endplate potential frequency at frog neuromuscular junctions. This effect is antagonized by Ca2+, but not by Sr2+ and Mn2+. This shoulder formation is not caused by Mn2+ or dinitrophenol in the medium. It is suggested that Mg2+ enters nerve terminals and displaces Ca2+ from internal stores; Mg2+ subsequently interferes with Ca2+ removal until the Mg2+ itself has been removed. The dinitrophenol result suggests that ATP-dependent Ca2+ extrusion is not permanently slowed. An equally likely and simpler explanation is that Mg2+ enters nerve terminals and the Mg2+ itself causes an increase in miniature endplate potential frequency. External Ca2+ competes with Mg2+ for channel entry and prevents this effect. The time course of the decay of miniature endplate potential frequency reflects the processes involved in Mg2+ extrusion and/or uptake.

Rapid miniaturized chromatography for 111In labeled monoclonal antibodies: comparison to size exclusion high performance liquid chromatography.

Our laboratory investigated the use of a rapid miniaturized chromatography system, ITLC-SG with 0.9% NaCl, to assess the radiochemical purity of 111In labeled monoclonal antibodies (MoAbs). Radiochemical analysis was performed on numerous 111In labeled antibody preparations with labeling efficiencies ranging from 40 to greater than 95% and the results compared to those obtained with size exclusion high performance liquid chromatography (HPLC). The chromatographic procedure involved challenging radiolabeled antibodies with 0.05 M DTPA to chelate unbound and/or non-specific bound 111In, spotting on miniaturized instant thin layer-silica gel chromatography strips, developing in 0.9% NaCl, and counting appropriate segments for radioactivity. Results of the study demonstrated that the miniaturized chromatography procedure was rapid, taking less than 4 min to complete, and accurate in assessing the amount of unbound or non-specific bound 111In in 111In labeled monoclonal antibodies, when compared to size exclusion HPLC.
