Purification, crystallization and preliminary X-ray analysis of rice BGlu1 beta-glucosidase with and without 2-deoxy-2-fluoro-beta-D-glucoside.

Rice (Oryza sativa) BGlu1 beta-glucosidase was expressed in Escherichia coli with N-terminal thioredoxin and hexahistidine tags and purified by immobilized metal-affinity chromatography (IMAC). After removal of the N-terminal tags, cation-exchange and S-200 gel-filtration chromatography yielded a 50 kDa BGlu1 with >95% purity. The free enzyme and a complex with 2,4-dinitrophenyl-2-deoxy-2-fluoro-beta-D-glucopyranoside inhibitor were crystallized by microbatch and hanging-drop vapour diffusion. Small tetragonal crystals of BGlu1 with and without inhibitor grew in 18%(w/v) PEG 8000 with 0.1 M sodium cacodylate pH 6.5 and 0.2 M zinc acetate. Crystals of BGlu1 with inhibitor were streak-seeded into 23%(w/v) PEG MME 5000, 0.2 M ammonium sulfate, 0.1 M MES pH 6.7 to yield larger crystals. Crystals with and without inhibitor diffracted to 2.15 and 2.75 angstroms resolution, respectively, and had isomorphous orthorhombic unit cells belonging to space group P2(1)2(1)2(1).

Nine members of the myomodulin family of peptide cotransmitters at the B16-ARC neuromuscular junction of Aplysia.

1. Neuromodulation by multiple related peptides with different spectra of physiological effects appears an effective way to integrate complex physiological functions. A good opportunity to examine this issue occurs in the accessory radula closer (ARC) neuromuscular circuit of Aplysia, where, extensive previous work has shown, acetylcholine-induced contractions of the muscle are variously modulated by several families of peptide cotransmitters released under appropriate behavioral circumstances from the muscle's own two motor neurons. 2. In this work we focused on the myomodulins (MMs) released from motor neuron B16. Previous work has characterized MMA (PMSMLRLamide) and MMB (GSYRMMRLamide). We now similarly purified from ARC neuromuscular material and sequenced MMC (GWSMLRLamide), MMD (GLSMLRLamide), MME (GLQMLRLamide), and MMF (SLNMLRLamide). Three additional MMs, MMG (TLSMLRLamide), MMH (GLHMLRLamide), and MMI (SLSMLRLamide), are encoded by a known MM gene. B16 probably synthesizes, and coreleases, all nine MMs. Further MMs have been found in other mollusks. All evidence indicates that the MMs are a major, widely distributed family of molluscan neuropeptides important as neuromuscular modulators and probably also central transmitters or modulators. 3. MM effects on motor neuron B16-elicited ARC muscle contractions were best analyzed as the sum of three distinct actions: potentiation, depression of the amplitude of the contractions, and acceleration of their relaxation rate. We compared the effectiveness of all nine MMs in these respects. We correlated this with their effectiveness in enhancing the L-type Ca current and activating a specific K current in voltage-clamped dissociated ARC muscle fibers, effects we previously proposed to underlie, respectively, the potentiation and the depression of contractions. 4. All nine MMs were similarly effective in enhancing the Ca current and, as far as it was possible to determine, potentiating the amplitude as well as accelerating the relaxation rate of the contractions. 5. In contrast, the MMs' ability to activate the K current and depress the contractions varied greatly. MMB and MMC, in particular, were weak, whereas the other seven MMs were considerably more effective in both respects. 6. Altogether, we were able to explain the potentiating and depressing strengths of the MMs by the magnitude of their modulation of the Ca and K currents, providing further support for our hypothesis that the effects on contraction amplitude are mediated by the effects on the two currents. 7. The net effect on contraction amplitude was determined by the balance between the potentiation and depression. Although most MM concentrations had both potentiating and depressing actions, potentiated contractions predominated at low and depressed contractions (but with accelerated relaxation rate) at high concentrations.(ABSTRACT TRUNCATED AT 400 WORDS)

A refined comparative map between porcine chromosome 13 and human chromosome 3.

We report here the localisation of BAIAP1 (13q24), HTR1F (13q45), PTPRG (13q23) and UBE1C (13q24) by fluorescence in situ hybridisation (FISH), and BAIAP1 (Swr2114; 21 cR; LOD = 11.03), GATA2 (Sw2448; 37 cR; LOD = 8.26), IL5RA (Swr2114; 64 cR; LOD = 3.85), LMCD1 (Sw2450; 61 cR; LOD = 4.73), MME (CP; 50 cR; LOD = 7.75), RYK (Swc22; 12 cR; LOD = 18.62) and SGU003 (Sw1876; 6 cR; LOD = 16.99) by radiation hybrid (RH) mapping to porcine chromosome 13 (SSC13). The mapping of these 10 different loci (all mapped to human chromosome 3; HSA3) not only confirms the extended conservation of synteny between HSA3 and SSC13, but also defines more precisely the regions with conserved linkage. The syntenic region of the centromeric part of SSC13 was determined by isolating porcine bacterial artificial chromosome (BAC) clones (842D4 and 1031H1) using primers amplifying porcine microsatellite markers S0219 and S0076 (mapped to this region). Sequence comparison of the BAC end sequences with the human genome sequence showed that the centromeric part of SSC13 is homologous with HSA3p24.

Casodex treatment induces hypoxia-related gene expression in the LNCaP prostate cancer progression model.

BACKGROUND: The changes in gene expression profile as prostate cancer progresses from an androgen-dependent disease to an androgen-independent disease are still largely unknown. METHODS: We examined the gene expression profile in the LNCaP prostate cancer progression model during chronic treatment with Casodex using cDNA microarrays consisting of 2305 randomly chosen genes. RESULTS: Our studies revealed a representative collection of genes whose expression was differentially regulated in LNCaP cells upon treatment with Casodex. A set of 15 genes were shown to be highly expressed in Casodex-treated LNCaP cells compared to the reference sample. This set of highly expressed genes represents a signature collection unique to prostate cancer since their expression was significantly greater than that of the collective pool of ten cancer cell lines of the reference sample. The highly expressed signature collection included the hypoxia-related genes membrane metallo-endopeptidase (MME), cyclin G2, and Bcl2/adenovirus E1B 19 kDa (BNIP3). Given the roles of these genes in angiogenesis, cell cycle regulation, and apoptosis, we further analyzed their expression and concluded that these genes may be involved in the molecular changes that lead to androgen-independence in prostate cancer. CONCLUSION: Our data indicate that one of the mechanisms of Casodex action in prostate cancer cells is induction of hypoxic gene expression.

Lupus nephritis and renal disease in pregnancy.

Management of pregnant women with renal disease involves awareness of, and allowance for, physiological changes including decreased serum creatinine and increased proteinuria. For women with systemic lupus erythematosus (SLE), pregnancy increases likelihood of flare. These can occur at any stage, and are more difficult to diagnose, as symptoms overlap those of normal pregnancy. Renal involvement is no more common in pregnancy. Worsening proteinuria may be lupus flare but differential includes pre-eclampsia. In women with chronic renal disease, pregnancy may accelerate decline in renal function and worsen hypertension and proteinuria, with increased risk of maternal (eg, pre-eclampsia) and fetal (eg, IUGR, IUD) complications, strongly correlating with degree of renal impairment peri-conception. Pregnancy success rate varies from 20% to 95% depending on base-line creatinine. Best outcome is obtained if disease was quiescent for >6 months pre-conception. Women on dialysis or with renal transplants can achieve successful pregnancy but have higher maternal and fetal complication rates. Acute on chronic renal failure can develop secondary to complications such as HELLP and AFLP. Management needs to be by a multidisciplinary team involving physicians and obstetricians, ideally beginning with pre-pregnancy counselling. Treatment of flares includes corticosteroids, hydroxychloroquine, azothioprine, NSAIDs and MME Blood pressure is controlled with methyldopa, nifedipine or hydralazine.

Calcium channel blockers for reducing cardiac morbidity after noncardiac surgery: a meta-analysis.

Cardiac complications are the leading cause of death after noncardiac surgery. Despite theoretical benefits, calcium channel blockers (CCB) are not widely used in the perioperative setting. This systematic review assessed the efficacy of CCBs during noncardiac surgery. MEDLINE, EMBASE, Science Citation Index, PubMed, and reference lists were searched without language restriction for randomized controlled trials (RCT) evaluating CCBs during noncardiac surgery. Two reviewers independently abstracted data on death, myocardial infarction (MI), ischemia, supraventricular tachyarrhythmia (SVT), and congestive heart failure (CHF). Treatment effects were calculated as relative risks (RR) with 95% confidence intervals (CI). Eleven studies (1007 patients) were included. CCBs significantly reduced ischemia (RR, 0.49; 95% CI, 0.30-0.80; P = 0.004) and SVT (RR, 0.52; 95% CI, 0.37-0.72; P < 0.0001). CCBs were associated with trends towards reduced death and MI. In post hoc analyses, CCBs significantly reduced death/MI (RR, 0.35; 95% CI, 0.15-0.86; P = 0.02) and major morbid events (MME), defined as death, MI, or CHF (RR, 0.39; 95% CI, 0.17-0.89; P = 0.02). In subgroup analyses, diltiazem significantly reduced ischemia, SVT, death/MI, and MMEs. This meta-analysis shows CCBs significantly reduced ischemia, SVT, and combined end-points in the setting of noncardiac surgery. The majority of these benefits are attributable to diltiazem, suggesting the need for further evaluation of this drug in a large RCT.

Molecular pathomechanisms of membranous nephropathy: from Heymann nephritis to alloimmunization.

Membranous nephropathy (MN), the most common cause of idiopathic nephrotic syndrome in white adults, is characterized by an accumulation of immune deposits on the outer aspect of the glomerular basement membrane. In Heymann nephritis, the rat experimental model for MN, megalin--the target antigen of the nephritogenic antibodies--is expressed on the surface of podocytes, where immune complexes are formed, leading to complement activation and nephrotic-range proteinuria. However, megalin cannot be held responsible for human MN because it has not been found in human podocytes or detected in subepithelial immune deposits in patients with MN. Several potential antigens have been identified in so-called secondary forms of MN, but there is no real proof that these antigens are pathogenic. In a subgroup of infants with antenatal MN, neutral endopeptidase (NEP) has been identified as the first protein target on human podocytes of nephritogenic antibodies. The infants' mothers became immunized during pregnancy against NEP expressed on syncytiotrophoblastic cells because they were NEP deficient as a result of truncating mutations in the MME gene. Severity of neonatal renal disease was determined by the mothers' IgG response that led to the formation of the membrane attack complex of complement in the subepithelial deposits. Alloimmunization against NEP is a novel pathomechanism of MN that might also account for some cases of MN after renal or bone marrow transplantation. Other types of alloimmunization should be investigated in MN but also in other renal and nonrenal diseases, particularly those that affect the pediatric age.

Oxytocin and its analogs, methyl-substituted in ortho-, meta- or para-position of aromatic ring of phenylalanine in position 2: NMR study and biological activities.

Complete analyses of NMR data of oxytocin (OT) and 4 analogues, ([o-MePhe(2)]OT, [mMe-Phe(2)]OT, [m-OMePhe(2)]OT and [p-MePhe(2)]OT), are given. The same conformational behavior in solution on one hand and large differences in biological activities on the other hand indicate that the compounds adopt a "biologically active conformation" at the stage of interaction with the receptor when the character of the substituent and its position on the aromatic ring may play a role in hindering attaining the ideal complementarity of both interacting components.

Evaluation of Agnique MMF in man-made ponds for the control of pestiferous chironomid midges (Diptera: Chironomidae).

The efficacy of a monomolecular surface film, Agnique MMF (a liquid isostearyl alcohol ethoxylate containing 2 oxyethylene groups [ISA-2OE]) in suppressing emergence of adult Chironomidae from man-made earthen ponds was studied. Three replicated rates, 0.23, 0.47, and 0.94 ml/m2 (or 0.25, 0.5, and 1.0 gal/acre) of Agnique MMF were applied. Pretreatment and periodic posttreatment samples of adult chironomids were collected by employing submerged metal-cone traps as well as floating traps. Concurrently, water pH, dissolved oxygen, specific conductance, and water temperature were measured. Tanytarsini (>82%), Chironomini (16%), and Tanypodinae (1%) midges were collected during the study period. Midge adult collections in floating traps were not significantly reduced by the 0.23-ml/m2 treatment rate, but were significantly reduced (73-93%) for 1-2 wk posttreatment at 0.47 and 0.94 ml/m2 of Agnique MME The lowest rate significantly reduced midge adult emergence for 1 day posttreatment as measured by metal-cone traps, whereas the 2 higher rates reduced emergence by 78.6-97% for 1 wk posttreatment. Using either trap, the highest rate of Agnique MMF did not produce suppression of adult midges of any greater magnitude or duration than the middle rate. Agnique MMF was not detected by the indicator oil in any treated pond after 7 days posttreatment. Water pH, dissolved oxygen, specific conductance, and water temperature in the ponds were not significantly influenced by Agnique MMF treatments. Analysis of these data supports the possible use of Agnique MMF for chironomid control in areas where the surface film of this material can be maintained for sufficient time to interfere with adult emergence.

NTP Toxicology and Carcinogenesis Studies of Methyl Methacrylate (CAS No. 80-62-6) in F344/N Rats and B6C3F1 Mice (Inhalation Studies).

Toxicology and carcinogenesis studies of methyl methacrylate, a liquid chemical intermediate used in the plastics industry in the manufacture of plexiglass and other acrylic products, were conducted by exposing groups of F344/N rats and B6C3F1 mice by inhalation for 14 weeks and 2 years. In the 14-week studies, groups of 10 male and 10 female rats and mice were exposed to methyl methacrylate at concentrations of up to 5,000 ppm. All male and female rats and eight male and eight female mice exposed at 5,000 ppm died, one male and nine female rats and four male and no female mice exposed at 3,000 ppm died, and one male and three female rats and two male and one female mouse exposed at 2,000 ppm died; all rats and mice exposed at 500 or 1,000 ppm survived. Compared with the controls, the body weights of the exposed male and female rats and mice were lower. Compound-related lesions included inflammation associated with necrosis and loss of olfactory epithelium in the nasal turbinates in both male and female rats; malacia and gliosis in female rats; inflammation of the nasal turbinates and nasal epithelium metaplasia in both male and female mice; and renal cortical necrosis, renal cortical tubular degeneration, renal focal mineralization, and liver necrosis in male mice. Based on these results, 2-year inhalation toxicology and carcinogenesis studies were conducted in which groups of 50 male rats were exposed to methyl methacrylate at 0, 500, or 1,000 ppm; female rats at 0, 250, or 500 ppm; and male and female mice at 0, 500, or 1,000 ppm. In the 2-year studies, the body weights of the low dose and high dose male and female rats were within 10% of those of the controls. There was no difference in survival between the dosed male and female rats and the controls. Incidences of inflammation of the nasal cavity and degeneration of the olfactory sensory epithelium were greater in the dosed male and female rats than in the controls, with lesions seen in virtually all high dose animals. An increased incidence of mononuclear cell leukemia was observed in female rats exposed to methyl methacrylate at 500 ppm compared with the controls (control, 11/50; 250 ppm, 13/50; 500 ppm, 20/50). This increase was not significant by life table tests, the method of analysis most appropriate for this fatal neoplasm. The mean body weights of dosed male and female mice were 5%-8% lower than those of the controls at the end of the 2-year studies. However, during most of the second year of the studies, body weights of dosed male mice and high dose female mice were 10%-18% lower than those of the controls. Survival rates of the dosed and control mice were similar. Incidences of inflammation and epithelial hyperplasia of the nasal cavity and degeneration of the olfactory sensory epithelium were significantly greater in all dosed groups of male and female mice compared with those of the controls. Compound-related neoplastic lesions were not found in the dosed mice. Significant dose-related decreases were observed in the incidences of pituitary gland and preputial gland tumors in male rats, alveolar/bronchiolar adenomas or carcinomas (combined) in male mice, hepatocellular adenomas in both male and female mice, and pituitary gland adenomas or adenocarcinomas (combined) and uterine adenocarcinomas in female mice. Methyl methacrylate was not mutagenic in strains TA100, TA1535, TA97, or TA98 of Salmonella typhimurium in the presence or absence of male rat or hamster liver S9 when assayed by a preincubational protocol but gave a positive response in L5178Y/TK+/- mouse lymphoma cells in the presence or absence of male rat liver S9. In cultured Chinese hamster ovary cells, methyl methacrylate produced a reproducible, dose-related increase in the frequency of sister-chromatid exchanges, both with and without rat liver S9. A slight, dose-related increase in chromosomal aberrations was also induced in cultured Chinese hamster ovary cells in the absence of S9; in the presence of S9, an increase in the frequency of aberrations was seen only at the highest, near-lethalas seen only at the highest, near-lethal dose of 5 mg/ml. An audit of the experimental data was conducted for the 2-year carcinogenesis studies on methyl methacrylate. No data discrepancies were found that influenced the final interpretations. Under the conditions of these 2-year inhalation studies, there was no evidence of carcinogenicity of methyl methacrylate for male F344/N rats exposed at 500 or 1,000 ppm, for female F344/N rats exposed at 250 or 500 ppm, or for male and female B6C3F1 mice exposed at 500 or 1,000 ppm. Inhalation of methyl methacrylate was associated with inflammation of the nasal cavity and degeneration of the olfactory sensory epithelium in male and female rats and mice; epithelial hyperplasia of the nasal cavity was also observed in exposed mice. Synonyms: acrylic acid, 2-methyl, methyl ester; methacrylic acid, methyl ester; methyl a-methylacrylate; methyl methylacrylate; methyl-2-methylpropenoate; methyl-2-methyl-2-propenoate; 2-methyl-2-propenoic acid methyl ester; MME

[The apothicary of the "Maison Royale de Saint-Cyr"].

The "Maison Royale de Saint-Cyr", a royal school for education of poor noble officer's girls, founded by Louis XIV and Mme de Maintenon and working from 1686 to the French Revolution, had an apothicary run by nuns. Its inventory has reached us and its fully and distinct contents well call to mind a 18th century's pharmacy with a large variety of tools for pharmaceutical and chemical preparations, set of furnitures and library. In the garden, a place, carefully cultivated, was reserved to medicinal plants for supply of the apothicary.

[Pharmaceuticals: a strategic national industry].

Asked by Mme Nicole Fontaine, Delegate Minister of Industry, to help the government with its ongoing reflections on pharmaceutical industrial strategy, and the necessary autonomy of our country in the face of major commercial threats, a working group of the French National Academy of Medicine consulted representatives of five French and two foreign major drug companies. Their statements can be classified in four categories:--the first concerns new medications, which must be approved successively by three commissions, whose opinions are often delayed and influenced by economic considerations;--second, public and private research are both insufficient and are sometimes hindered by procedural restrictions,--third, the pharmaceutical industry is unable to deal with frequent and unforseeable political upheavals,--France does not adequately recognize the strategic importance of the pharmaceutical industry in the national economy. The Academy makes several recommendations: the French pharmaceutical industry should be considered as a national priority, the strategic importance of national pharmaceutical companies should be recognized, a multi-annual contract should be signed with manufacturers, clinical trials should be facilitated in France, relationships between the national pharmaceutical industry and public research structures should be reinforced, and an inter-ministerial Council on Pharmaceuticals should be created. This study was supplemented by a survey of veterinary medications, the results and conclusions of which are very similar to those outlined above for human medicines.

[The role of circulating immune complexes and the status of argyrophilic membranes of the vascular walls in the development of brain edema in patients with meningococcal meningoencephalitis].

Based on clinical and immunological examinations of 60 patients with MME and 30 normal persons, the dynamics of the blood CIC content was studied depending on the time and gravity of the disease. The discovered changes in argyrophilic membranes of the vascular walls are determined to a considerable measure by the pathogenic action of CIC on microvessels, which entails the derangement of blood-brain barrier function and contributes to the development of acute purulent meningitis.

[Animal model and multiple trait BLUP applied in poultry genetic evaluation].

Best linear unbiased prediction (BLUP) is a powerful method to estimate genetic values of animals, and is widely applied in many animal species but poultry. Beijing White Leghorn nested data in 1986-1987 with 777 individuals were analysed by animal model and multiple trait BLUP. Two traits (40-week egg production and 36-week egg weight) and two fixed effects (house-pen effect and hatching batch effect) were considered. The way to calculate a large set of mixed model equations in micro-computer was studied. Only non-zero elements of the coefficient matrix of MME were stored on a disk. The iteration process was reduced by block iteration. It also simplified the multiple trait BLUP method, the dimension of equations is only 1/q (q is the number of traits) of regular method. So it saved a lot of compute time and cost, and BLUP became applicable in poultry. The significance of using BLUP in poultry are: (1) Eliminating some fixed effects; (2) Reducing the estimation error for unbalanced data; (3) Estimating breeding values of progeny, so we can shorten the generation interval; (4) It can estimate breeding values of individuals without records from the relatives' information; (5) The breeding value of sires and dams can be estimated from their progeny records, used for family selection; (6) Due to genetic and environmental correlation between traits and all relatives information were considered, it can increase the selection accuracy.

[The French physician and judge confronting the transsexual in 1986. I. The position of French and foreign jurisprudence].

The authors have searched through French and western jurisprudence about transsexualism. Thanks to their experience of how judges and magistrates have reacted they have been able to analyse the evolution of attitudes and the present-day attitudes to the subject and how the condition should be treated and viewed by the law. There are only a relatively few numbers who wish to change sex but these are estimated at 1 in 50,000 to 1 in 100,000 according to different statistics. The proportion of males who want to change is 3 times greater than of females. The fact that transsexualism is a phenomenon much more often found in North America, in North Europe and in the West, and almost not at all in the third world or in socialist countries, or in the Mediterranean basin, makes one think that this is a syndrome connected with a legal attitude that allows people to dispose of their bodies in the way they wish. This in turn derives from the European Convention of the Rights of man as against those found in the legislation of individual nations in the community. The French legal system is justified in being cautious, seeing how hesitant lawyers are and how extremely prudent the French medical corps is. The reserve of jurisprudence in the U.S.A. and Canada (with the exception of two provinces) and a certain hesitation about the indications for medical treatment to convert from one sex to the other voiced by the principal pioneers of such treatment in the U.S.A. together with the absence of specific laws which cover surgical treatment and the change in the legal state (with the exception of Sweden and the Federal Republic of Germany) is to be taken along with the check that Italian law has placed on the operation recently, in 1982. As far as French law is concerned, Mme M.L. Rassat has carried out a recent and very pertinent study which makes it possible to analyse the attitude of magistrates, which until 1975 was basically very restrictive as far as demands for change in civil sex was concerned. Recently, however, they have become laxer but the Appeal Court strictly refuse to recognise the idea of psychological or psychosocial sex.(ABSTRACT TRUNCATED AT 400 WORDS)

Dynamic reciprocity: how do extracellular matrix and hormones direct gene expression?

Using mouse mammary epithelial cells (MME) as a model, we show that extracellular matrix (ECM) plays a fundamental role in the maintenance of tissue-specific function in culture. The ECM affects both the level of mRNA and the rates of synthesis and secretion of milk proteins. Casein gene expression by primary mammary epithelial cells and cell strains is controlled by both ECM and lactogenic hormones (insulin, hydrocortisone and prolactin). In the case of transferrin, the major iron-binding protein of mouse milk, the ECM rather than prolactin, appears to modulate the level of its mRNA. We further show that both ECM and lactogenic hormones influence cell shape and polarity of mammary epithelial cells. The data are consistent with a model of "Dynamic Reciprocity" (Bissell et al. 1982) where the ECM is postulated to exert an influence on gene expression via transmembrane proteins and cytoskeletal components. Cytoskeleton, in turn, is associated with polyribosomes, affecting mRNA stability and rates of protein synthesis, and with the nuclear matrix, affecting mRNA processing and, possibly, rates of transcription. We postulate that hormones and ECM act synergistically to complete the 'reciprocity' loop.

Fine structure and vascular supply of the median eminence (ME) in Acipenser ruthenus (Chondrostei).

The fine structure and vascular supply of the median eminence (ME) was studied in 57 specimens of Acipenser ruthenus (Chondrostei) by means of light microscopy (normal histology, horseradish-peroxidase (HRP), formaldehyde induced fluorescence (FIF), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) as well as by SEM of vascular corrosion casts. Light microscopy revealed that the ME increases in thickness from caudal to rostral with capillaries invading the subependymal layers only at the rostral median eminence (rME). At the middle (mME) and caudal median eminence (cME) capillaries are limited to the ME outer surface. Short term HRP-application (systemically as well as intraventricularly) resulted in reaction product in the intercellular space throughout the ME after a survival time of 2 hours. The ME tanycytes were distinctly marked after 7 days survival. FIF revealed aminergic cerebrospinal fluid (CSF-) contacting neurons; also the apical part of tanycytes showed specific fluorescence. SEM of vascular corrosion casts showed that the arterial supply of the whole ME results from branches of the hypothalamic artery and/or from those arteries supplying the saccus vasculosus. The whole 2-dimensional primary capillary plexus of the ME drains via portal vessels into the adenohypophysis. As demonstrated by SEM, the ME ependymal surface is made up by oligociliated tanycytes, abundant crown cells and intraventricular protrusions of CSF-contacting neurons. Supraependymal cells are lacking, supraependymal fibres are very sparse. TEM revealed the ME tanycytes with long ciliary rootlets. Their basal processes split into numerous delicate branches forming many end-feet, which end at the basal lamina opposite the fenestrated capillaries of the primary plexus. Like tanycytes, also the crown cells possess branching processes which may contribute to the outer glial membrane. CSF-contacting neurons of the type I (with dense core vesicles ranging in diameter from 100-120 nm and of presumed aminergic nature) and type II (with dense core vesicles ranging in diameter from 160-180 nm and of presumed peptidergic nature) were found. Both types are most abundant in the cME. Small astrocytes reveal delicate processes which cover parts of the CSF-contacting neurons and of tanycytes. Another glial cell type is situated near the basal lamina and forms processes parallel the latter. There are few axo-somal and axo-axonal synapses pointing rather to a humoral regulation of the ME with signals from the ME blood vascular bed or from the CSF acting on CSF-contacting neurons.
