Mechanism of the Gibbs Reaction. Part 4.(1) Indophenol Formation via N-Chlorobenzoquinone Imine Radical Anions. The Aza-S(RN)2 Chain Reaction Mechanism. Chain Initiation with 1,4-Benzoquinones and Cyanide Ion.

The mechanism of the Gibbs reaction, a colorimetric phenol assay that applies N-chlorobenzoquinone imines 1 in an aqueous basic medium, was investigated. It is concluded that N-chloroimine radical anion 7 generated in a single electron transfer (SET) from the anion of phenol 4 to N-chloroimine 1 can produce indophenol dye 3 in three distinct routes. For more reactive reagent-substrate pairs, a route is proposed that involves a fast combination of the radical pair in the solvent cage and, consequently, the total rate of which exhibits a pH-independent second-order kinetics, as does the preceding SET itself. For less reactive reagents, a route is proposed in which the N-chloroimine radical anion 7 escapes from the solvent cage to initiate a chain reaction, evidenced by its characteristic kinetics. It has been found in the kinetic experiments that during propagation the chlorine of the chain carrier N-chloroimine radical anion 7 is substituted by the anion of 4 in a bimolecular rate-determining step. Therefore, the mechanism of the chain reaction is termed S(RN)2. In the case when the anion of 4 is less active, a competitive reaction along a third route can proceed in which the N-haloimine radical anion 7 yields benzoquinone imine 6 by the elimination of halogenide and the abstraction of an H-atom from the medium. Compound 6 is also known to give indophenol 3 with a second-order but pH-dependent rate that is considerably faster than the rate in the first route. On the basis of the different kinetic characteristics outlined above a clear distinction can be made among these three pathways. In this paper, evidence is also presented for the initiating SET. Furthermore, it is of high importance that the N-haloimine radical anion 7 can also be generated from reagent 1 using external electron donors and, independently of its origin, it can be spin trapped with 2,2,6,6-tetramethylpiperidine-N-oxyl.

(3R)-Chiral Control of 3-Alkyl-3-hydroxy-beta-lactams via Addition Reaction of Imines to Enolates of 1,3-Dioxolan-4-ones.

A method is described for the chiral construction of (3R)-3-alkyl-3-hydroxy-beta-lactams in a versatile and predictable manner. This protocol follows Seebach's synthetic principle of self-regeneration of stereocenters and has been applied to addition reactions among a selected number of imines and (2S)-chiral enolates of 1,3-dioxolan-4-ones. These reagents are easily available from the acetalization of (S)-alpha-hydroxy acids (lactic, mandelic, isovaleric, malic) and pivalaldehyde or pinacolone. In several cases, the addition of the enolate to the imine, the cyclization, and the removal of the auxiliary center occur in a one-step sequence, affording the corresponding beta-lactams as (3R,4S)-Z and (3R,4R)-E diastereomeric mixtures with high enantiomeric excesses. Four N-unsubstituted (3R,4S)-3-hydroxy-3-methyl-beta-lactams bearing 2-furyl (4e), phenylethenyl (4h), methoxycarbonyl (4i), and 2-thienyl (4l) substituents at C4 were obtained as major diastereomers and were purified by crystallization. The simultaneous presence of these substituents at C3 and C4 make these beta-lactams useful intermediates for the synthesis of new taxoids with interesting structural modifications at the isoserine moiety.

The structures of complexes between polyethylene imine and sodium dodecyl sulfate in D2O: a scattering study.

The association between a highly branched polyelectrolyte with ionizable groups, polyethylene imine (PEI), and an anionic surfactant, sodium dodecyl sulfate (SDS), has been investigated at two pH values, using small-angle neutron and light scattering. The scattering data allow us to obtain a detailed picture of the association structures formed. Small-angle neutron scattering (SANS) measurements in solutions containing highly charged PEI at low pH and low SDS concentrations indicate the presence of disklike aggregates. The aggregates change to a more complex three-dimensional structure with increasing surfactant concentration. One pronounced feature in the scattering curves is the presence of a Bragg-like peak at high q-values observed at a surfactant concentration of 4.2 mM and above. This scattering feature is attributed to the formation of a common well-ordered PEI/SDS structure, in analogue to what has been reported for other polyelectrolyte-surfactant systems. Precipitation occurred at the charge neutralization point, and X-ray diffraction measurements on the precipitate confirmed the existence of an ordered structure within the PEI/SDS aggregates, which was identified as a lamellar internal organization. Polyethylene imine has a low charge density in alkaline solutions. At pH 10.1 and under conditions where the surfactant was contrast matched, the SANS scattering curves showed only small changes with increasing surfactant concentration. This suggests that the polymer acts as a template onto which the surfactant molecules aggregate. Data from both static light scattering and SANS recorded under conditions where SDS and to a lower degree PEI contribute to the scattering were found to be consistent with a structure of stacked elliptic bilayers. These structures increased in size and became more compact as the surfactant concentration was increased up to the charge neutralization point.

The conformation of amine- and amide-terminated poly(propylene imine) dendrimers as investigated by molecular simulation methods.

The parameters that influence the conformation of poly(propylene imine) dendrimers were investigated by molecular simulations using molecular mechanics and simulated annealing methods. Dendrimers with two types of peripheral units able to communicate via hydrogen bonding-amine and amide moieties-were considered in order to study the role that secondary interactions among the end groups have in the spatial organization of the dendritic branches. Radial atomic density profiles and radial atomic probability distributions were used to extract global properties, such as the degree of packing of the branches, the distribution of the monomers throughout the molecular volume, and the extent and characteristics of the surface region. Information was also obtained about the nature, location, and extent of formation of the hydrogen bonds, as well as their evolution with dendrimer generation and their assembly into networks. The analyses were supported by a detailed investigation of the first two generations, with an emphasis on the relationship between hydrogen bonding and the compactness and stability of the molecules; this allowed us to account for the generational evolution of hydrogen bonding that is experimentally observed in several poly(propylene imine) dendrimers.

Highly enantioselective addition of diethylzinc to diphenylphosphinoyl imines under dual amino Alcohol/Halosilane mediation

Arylethylene-derived, enantiomerically pure amino alcohols have been evaluated as ligands for the dual-catalyzed (amino alcohol/halosilane) enantioselective addition of diethylzinc to diphenylphosphinoyl imines. Among them, the conformationally restricted 9-fluorenone-derived ligand 4c provides the highest enantioselectivities so far reported over a range of substrate imines.

In situ oxidation-imine formation-reduction routes from alcohols to amines.

Manganese dioxide is employed as an in situ oxidant for the one-pot conversion of alcohols into imines. In combination with polymer-supported cyanoborohydride (PSCBH), a one-pot oxidation-imine formation-reduction sequence is reported. This procedure enables alcohols to be converted directly into both secondary and tertiary amines.

Novel sulfinyl imine ligands for asymmetric catalysis.

[structure: see text] A novel class of P,N-sulfinyl imine ligands has been prepared that incorporates chirality solely at sulfur. The Pd complex of ligand 14 catalyzes the allylic alkylation reaction with high enantioselectivity (94%), and the first crystal structure of a Pd-bound sulfinyl imine provides insight into binding mode and origins of stereoselectivity.

Palladium-catalyzed coupling of allyl acetates with aldehyde and imine electrophiles in the presence of Bis(pinacolato)diboron.

[reaction: see text] An efficient one-pot procedure was developed for palladium-catalyzed electrophilic substitution of allyl acetates (2a-h) in the presence of bis(pinacolato)diboron (1). These reactions proceed with an excellent regioselectivity and with a remarkably high stereoselectivity. The catalytic transformations take place via palladium-catalyzed formation of allyl boronates, which subsequently react with aldehyde (3) and sulfon-imine (4) electrophiles to afford homoallylic alcohols (5a-h) and amines (6a-d), respectively. A particularly interesting mechanistic feature is that the allylic substitution of the transient allyl boronate with sulfon-imine requires palladium catalysis. This finding indicates that the formation of the homoallylic amine derivatives (6a-d) involves bis-allylpalladium intermediates.

beta-Isocupreidine-catalyzed asymmetric Baylis-Hillman reaction of imines.

[reaction: see text] beta-Isocupreidine (beta-ICD)-catalyzed asymmetric Baylis-Hillman reactions of aromatic imines with 1,1,1,3,3,3-hexafluoroisopropyl acrylate (HFIPA) give (S)-enriched N-protected-alpha-methylene-beta-amino acid esters. In contrast to the corresponding aldehydes, imines show the opposite enantioselectivity. A mechanistic proposal governed by hydrogen bonding is presented.

Catalytic additions of acylsilanes to imines: an acyl anion strategy for the direct synthesis of alpha-amino ketones.

The addition of acylsilanes to imines catalyzed by neutral carbenes (or zwitterions) generated in situ from readily available thiazolium salts is described. The general reaction successfully utilizes acylsilanes as carbonyl anion precursors and is tolerant of a range of structural diversity on the acylsilane or imine electrophile. The overall reaction utilizes easily available precursors and directly accesses protected alpha-amino ketones in the correct oxidation state.

Samarium diiodide-induced asymmetric synthesis of optically pure unsymmetrical vicinal diamines by reductive cross-coupling of nitrones with N-tert-butanesulfinyl imines.

[reaction: see text] An efficient method for the preparation of optically pure unsymmetrical vicinal diamines by the SmI(2)-induced reductive cross-coupling of nitrones with chiral N-tert-butanesulfinyl imines was developed. This is the first successful example of the highly diastereoselective and enantioselective cross-coupling between two different imine species. It provides a straightforward access to enantiopure unsymmetrical vicinal diamines that are widely applicable in asymmetric synthesis.

Enantioselective recognition of 1,2-amino alcohols by reversible formation of imines with resonance-assisted hydrogen bonds.

A chiral aldehyde with three H-bond donating groups (2) has been synthesized. This aldehyde binds a variety of chiral 1,2-amino alcohols in benzene with the same sense of stereoselectivity. Computational and experimental data indicate that one imine bond, one resonance-assisted H-bond to the imine nitrogen, and two H-bonds to the alcoholic oxygen all play an important role in the stereoselective recognition. General approach to the coupling of organoindium reagents with imines via copper catalysis.

[reaction: see text] A copper-catalyzed three-component coupling of organoindium reagents with imines and acid chlorides is described. This mild carbon-carbon bond forming reaction requires only one-third of an equivalent of indium reagent to proceed in high yield, with the sole byproduct being indium trichloride. The reaction demonstrates broad generality, with aryl-, heteroaryl-, vinyl-, and alkylindiums, as well as functionalized imines and acid chlorides, all providing alpha-substituted amides or N-protected amines in a single step.

[reaction: see text] Highly stereoselective nucleophilic monofluoromethylation of (R)-(tert-butanesulfinyl)imines with fluoromethyl phenyl sulfone was achieved to afford alpha-monofluoromethylamines with a nonchelation-controlled stereoselectivity mode. By using the same chemistry, (R)-(tert-butanesulfinyl)imines bearing a terminal tosylate (OTs) group can be converted to alpha-monofluoromethylated cyclic secondary amines with high stereoselectivity.

Direct electrophilic alpha-fluorination of imines: efficient synthesis of mono- and difluoroimines.

[reaction: see text] A mild and efficient procedure to synthesize alpha-fluoro- and alpha,alpha-difluoroimines was developed. Various N-alkylimines derived from acetophenones were successfully monofluorinated using NFSI (N-fluorobenzenesulfonimide) in a mixture of CH(3)CN and DMF at 0 degrees C. Alternatively, the same procedure without DMF gave rise to difluorinated imines when performed at room temperature. The obtained alpha- and alpha,alpha-difluorinated imines were subsequently reduced to give the corresponding beta-fluoro- and beta,beta-difluoroamines in good yield.
