Synthesis and aqueous solution properties of novel sugar methacrylate-based homopolymers and block copolymers.

We report the facile preparation of a range of novel, well-defined cyclic sugar methacrylate-based polymers without recourse to protecting group chemistry. 2-Gluconamidoethyl methacrylate (GAMA) and 2-lactobionamidoethyl methacrylate (LAMA) were prepared directly by reacting 2-aminoethyl methacrylate with D-gluconolactone and lactobionolactone, respectively. Homopolymerization of GAMA and LAMA by atom transfer radical polymerization (ATRP) gave reasonably low polydispersities as judged by aqueous gel permeation chromatography. A wide range of sugar-based block copolymers were prepared using near-monodisperse macroinitiators based on poly(ethylene oxide) [PEO], poly(propylene oxide) [PPO], or poly(e-caprolactone) [PCL] and/or by sequential monomer addition of other methacrylic monomers such as 2-(diethylamino)ethyl methacrylate [DEA], 2-(diisopropylaminoethyl methacrylate [DPA], or glycerol monomethacrylate [GMA]. The reversible micellar self-assembly of selected sugar-based block copolymers [PEO23-GAMA50-DEA100, PEO23-LAMA30-DEA50, PPO33-GAMA50, and PPO33-LAMA50] was studied in aqueous solution as a function of pH and temperature using dynamic light scattering, transmission electron microscopy, surface tensiometry, and 1H NMR spectroscopy.

Environmentally responsive "hairy" nanoparticles: mixed homopolymer brushes on silica nanoparticles synthesized by living radical polymerization techniques.

This article reports on the preparation of environmentally responsive "hairy" nanoparticles by growth of mixed poly(tert-butyl acrylate) (PtBA)/polystyrene (PS) brushes from silica particles using living radical polymerization techniques and subsequent hydrolysis of PtBA to produce amphiphilic mixed poly(acrylic acid) (PAA)/PS brushes. Silica particles were synthesized by the Stober process and were functionalized with an asymmetric difunctional initiator-terminated monolayer. Surface-initiated atom transfer radical polymerization of tBA was carried out in the presence of a free initiator. Kinetics study showed that the polymerization was well controlled. By cleaving PtBA off the particles, the molecular weights of the grafted and free polymers were found to be essentially identical. Mixed PtBA/PS brushes were obtained by the nitroxide-mediated radical polymerization of styrene from PtBA particles. The M(n) of the grafted PS was found to be the same as that of the free PS formed in the solution from the free initiator. Amphiphilic mixed PAA/PS brush-coated nanoparticles were synthesized from mixed PtBA/PS particles by hydrolysis of PtBA with iodotrimethylsilane. Tyndall scattering experiments and (1)H NMR study showed that the mixed PAA/PS particles can be dispersed and form a stable suspension in CHCl(3), a selective solvent for PS, and also in CH(3)OH, a selective solvent for PAA, demonstrating the capability of these hairy nanoparticles to undergo chain reorganization in response to environmental changes.

Amphiphilic homopolymer as a reaction medium in water: product selectivity within polymeric nanopockets.

A styrene-based water-soluble polymer has been explored for its use as a host for lipophilic substrates in aqueous medium. Unimolecular reactions, namely, photo-Fries rearrangement of naphthyl esters, alpha-cleavage reaction of 1-phenyl-3-p-tolyl-propan-2-one, and Norrish type I and type II reactions of benzoin alkyl ethers were examined. We find that the hydrophobic domains generated by the polymer not only restrict the mobility of the radicals but also modestly incarcerate the substrate, intermediates, and products during the time scale of the reactions. Comparative studies of the same photoreactions in micelles formed from small molecule surfactants and an amphiphilic diblock copolymer demonstrate that the styrene-based water-soluble polymer aggregates in aqueous medium offer better selectivity.

Binding of sodium dodecyl sulfate to linear and star homopolymers of the nonionic poly(methoxyhexa(ethylene glycol) methacrylate) and the polycation poly(2-(dimethylamino)ethyl methacrylate): electromotive force, isothermal titration calorimetry, surface tension, and small-angle neutron scattering measurements.

We investigated the binding of sodium dodecyl sulfate (SDS) to various linear and star polymers of the nonionic methoxyhexa(ethylene glycol) methacrylate (PMHEGMA) and the ionic 2-(dimethylamino)ethyl methacrylate (PDMAEMA), the latter being a polycation at low pH. The dodecyl sulfate ion selective electrode (EMF), isothermal titration calorimetry (ITC), and surface tension (ST) were applied to gain detailed information about interactions. In all cases there is evidence of significant binding of SDS over an extensive SDS concentration range spanning from ca. 10(-6) to 0.1 mol dm(-3). At pH 3, the polymer PDMAEMA is a strong polycation and here the binding is dominated by electrostatic 1:1 charge neutralization with the anionic surfactant. At their natural pH of 8.6, PMHEGMA and PDMAEMA polymers are essentially nonionic and bind SDS in the form of polymer-bound aggregates in the concentration range of ca. 1 x 10(-3) to 3 x 10(-2) mol dm(-3). All the polymers also bind SDS to a lesser extent at concentrations below 1 x 10(-3) mol dm(-3) reaching as low as 10(-7) mol dm(-3). This low concentration binding process involves the polymer and nonassociated SDS monomers. As far as we are aware, this is the first example that such a low concentration noncooperative binding process could be observed in SDS/neutral polymer systems by EMF and ST. We also showed that the nonionic surfactant hexa(ethylene glycol) mono-n-dodecyl ether (C12EO6) and the cationic cetyltrimethylammonium bromide (C16TAB) interact with star PDMAEMA. We believe that the interaction of C12EO6 and CTAB is of similar noncooperative type as the first SDS binding process in the range from ca. 10(-5) to 0.3 x 10(-3) mol dm(-3). At the high concentration binding limit Csat of SDS, the above polymers become fully saturated with bound SDS micelles. We applied small angle neutron scattering (SANS) to determine the structure and aggregation numbers of the star polymer/bound SDS micelles and calculated the stoichiometry of such supramolecular complexes. The SANS data on PDMAEMA star polymers in the presence of C12EO6 showed only a limited monomer binding in contrast to linear PDMAEMA, which showed monomer C12EO6 binding at low concentrations but micellar aggregates at 6 x 10(-3) mol dm(-3).

Synthesis of homopolymer containing diphenyl end-capped oligothiophene co-oligomer unit in the side chain.

[reaction: see text] A new polymer with the diphenyl end-capped oligothiophene co-oligomer unit in the side chain was obtained by the ROMP method. The polymer showed good photophysical characteristics, thermal stability, and film-forming properties. A photovoltaic cell fabricated from this polymer showed relatively large open-circuit voltage (V(OC) = 0.7 V), moderate short-circuit current (I(SC) = 0.7 microA/cm(2)), and excellent device stability under ambient conditions.

Oxidation of DNA bases, deoxyribonucleosides and homopolymers by peroxyl radicals.

DNA base oxidation is considered to be a key event associated with disease initiation and progression in humans. Peroxyl radicals (ROO. ) are important oxidants found in cells whose ability to react with the DNA bases has not been characterized extensively. In this paper, the products resulting from ROO. oxidation of the DNA bases are determined by gas chromatography/MS in comparison with authentic standards. ROO. radicals oxidize adenine and guanine to their 8-hydroxy derivatives, which are considered biomarkers of hydroxyl radical (HO.) oxidations in cells. ROO. radicals also oxidize adenine to its hydroxylamine, a previously unidentified product. ROO. radicals oxidize cytosine and thymine to the monohydroxy and dihydroxy derivatives that are formed by oxidative damage in cells. Identical ROO. oxidation profiles are observed for each base when exposed as deoxyribonucleosides, monohomopolymers and base-paired dihomopolymers. These results have significance for the development, utilization and interpretation of DNA base-derived biomarkers of oxidative damage associated with disease initiation and propagation, and support the idea that the mutagenic potential of N-oxidized bases, when generated in cellular DNA, will require careful evaluation. Adenine hydroxylamine is proposed as a specific molecular probe for the activity of ROO. in cellular systems.

Confinement free energy and chain conformations of homopolymers confined between two repulsive walls.

Lattice Monte Carlo simulations of polymer solutions confined between two parallel plates were performed. The confinement free energy Deltamicro(conf) per chain and the radius of gyrations of the chains parallel and perpendicular to the plates were obtained. When the concentration of the confined solution is above the overlap concentration, Deltamicro(conf) is found to scale with Na/D in a power law, betaDeltamicro(conf) approximately (Na/D)(m), with an exponent m=1.10+/-0.02 for athermal walls where N is the number of monomers in a chain, D is the slit width, and a is the lattice spacing. The presence of a weak attractive polymer/wall interaction epsilon(w) does not change the scaling variable, but the exponent m increases slightly. Extrapolating the results to melt would suggest that the predictions made by de Gennes [C. R. Acad. Sci. Paris II 305, 1181 (1987)] about the confinement free energy cost per chain in polymer melt is correct as far as the scaling variable is concerned, but is incorrect about the exponent m observed. The implication of this result on the predicted force between plates immersed in polymer melt is discussed. The parallel dimensions of the confined chain is expanded when the slit width D is narrow, however, the expansion is reduced at high concentration. It is conceivable that in melt the chain is not expanded when confined in a repulsive slit.

Composition fluctuations in a homopolymer-diblock copolymer mixture covering the three-dimensional Ising, isotropic Lifshitz, and Brasovskiĭ classes of critical universality.

The phase behavior of a three-component polymer blend consisting of a critical mixture of polybutadiene and polystyrene (PB/PS) with varying amount of a symmetric PB-PS diblock copolymer was explored with small-angle neutron scattering. Our focus were thermal composition fluctuations which we discuss in terms of mean field, three-dimensional Ising, isotropic Lifshitz, and Brasovskiĭ classes of critical universality. Particular attention is spent to the observation of a narrow reentrant two-phase regime and double critical point in the Lifshitz critical regime as well as the Lifshitz line. Critical exponents of the isotropic Lifshitz case are proposed in spite of the demonstrated nonexistence of the isotropic Lifshitz critical point. The Ginzburg number (Gi) and Flory-Huggins parameter were determined over the whole diblock concentration range; Gi changes by three orders of magnitude, two orders of magnitude of that change over a 0.03 diblock concentration interval within the isotropic Lifshitz regime.

A computer simulation study of the segregation of amphiphiles in binary immiscible matrices: short asymmetric copolymers in short homopolymers.

The bulk and interfacial properties of ternary mixtures with asymmetric amphiphiles (A2B8) in A2 and B2 matrices and in A2 and B10 matrices are investigated by the dissipative particle dynamics type of molecular-dynamics simulations. The monomer concentrations of A2B8(phiA2B8) studied are below the critical micelle concentration (phiA2B8(cmc)) for the formation of micelles in the presence of an adsorbed amphiphilic monolayer at the interface. Macrophase separation from the mixed phase to the segregated state with A-rich and B-rich coexisting phases and the segregation of A2B8 at the interface are thermodynamically gradual but are accompanied by a pronounced stretching and orientation of the constituent chains. The segregation of A2B8 at the interface broadens the interfacial region and reduces the interfacial tension. The chain conformation of the asymmetric amphiphilic molecules and the interfacial properties are dominated by the majority block in the amphiphilic chain and dependent on the composition of the matrix in contact with the majority block. In the A2 and B2 matrices, the B8 blocks in A2B8 chains at the interface resemble a wet brush swollen by short B2 chains. Swelling is responsible for the pronounced stretching and orienting of the amphiphilic chains and the reduced interfacial amphiphile enrichment. At the same interfacial amphiphile excess, however, swollen amphiphiles are more efficient in reducing the interfacial tension than nonswollen amphiphiles.

Glassy correlations and microstructures in randomly cross-linked homopolymer blends.

We consider a microscopic model of a polymer blend that is prone to phase separation. Permanent cross-links are introduced between randomly chosen pairs of monomers, drawn from the Deam-Edwards distribution. Thereby, not only density but also concentration fluctuations of the melt are quenched-in in the gel state, which emerge upon sufficient cross-linking. We derive a Landau expansion in terms of the order parameters for gelation and phase separation, and analyze it on the mean-field level, including Gaussian fluctuations. The mixed gel is characterized by thermal as well as time-persistent (glassy) concentration fluctuations. Whereas the former are independent of the preparation state, the latter reflect the concentration fluctuations at the instant of cross-linking, provided the mesh size is smaller than the correlation length of phase separation. The mixed gel becomes unstable to microphase separation upon lowering the temperature in the gel phase. Whereas the length scale of microphase separation is given by the mesh size, at least close to the transition, the emergent microstructure depends on the composition and compressibility of the melt. Hexagonal structures, as well as lamellas or random structures with a unique wavelength, can be energetically favorable.

Density functional theory of homopolymer mixtures confined in a slit.

A density functional theory (DFT) is developed for polymer mixtures with shorted-ranged attractive interparticle interactions confined in a slit. Different weighting functions are used separately for the repulsive part and the attractive part of the excess free energy functional by applying the weighted density approximation. The predicted results by DFT are in good agreement with the corresponding simulation data indicating the reliability of the theory. Furthermore, the center-of-mass profiles and the end-to-end distance distributions are obtained by the single chain simulation; the predictions also agree well with simulation data. The results reveal that both the attraction of the slit wall and the temperature has stronger effect on longer chains than on shorter ones because the intrasegment correlation of chains increases with increasing chain length.

A long purine-pyrimidine homopolymer acts as a transcriptional diode.

Polypurine-polypyrimidine (R.Y) sequences have the unusual ability to form DNA triple helices. Such tracts are overrepresented upstream of eukaryotic genes, although a function there has not been clear. We report that transcription in vitro into one such upstream R.Y tract in the direction that makes a predominantly purine RNA is effectively blocked by formation of an intramolecular triple helix. The triplex is triggered by transcription and stabilized by the binding of nascent purine RNA to the template. Transcription in the opposite direction is not restricted. Polypurine-polypyrimidine DNA may provide a dynamic and selective block to transcription without the aid of accessory proteins.
