[Studies on protective effect of total flavonoids of Astragalus on liver damage induced by paracetamol].

OBJECTIVE: To study the protective effect of total flavonoids of Astragalus (TFA) on the liver against large doses of paracetamol in mice. METHOD: After oral administration of TFA or Vitamin C 1 h prior to giving large dose of paracetamol in mice, the changes of paracetamol-induced mortality rate, serum enzyme level and liver damage degree were observed. RESULT: Paracetamol produced 80% mortality, within 24 hours of the administration of a dose of 1000 mg.kg-1 to the mice. Pre-treatment of the animals with TFA (100 mg.kg-1) or Vitamin C (1,000 mg.kg-1) reduced the death rate to 20% and 0% respectively. There was also a significant rise in the serum enzyme level of alanine transaminase (P < 0.001) and the area of liver necrosis (P < 0.001), 24 h after paracetamol (400 mg.kg-1) treatment. With pre-treatment with either TFA or Vitamin C, there was an obvious dose-dependent decrease in ALT levels and the area of hepatocellular necrosis. CONCLUSION: TFA has potential protecting effect against the paracetamol-induced hepatic damage.

[Protection against paracetamol-induced hepatic damage using total flavonoids of Astragalus].

OBJECTIVE: To study the mechanism of the protection by total flavonoids of Astragalus protection against paracetamol-induced hepatic damage. METHOD: Analysing paracetamol and its metabolites in mice urine by HPLC and studying the mechanism of anti-damage induced by paracetamol using experiment module of pentobarbital-induced sleeping time. RESULT: Administration of large doses of paracetamol to C57BL/6J mice produced significant hepatic injury with marked elevation in serum ALT activity and severe hepatocellular necrosis. TFA showed a good protective capability against paracetamol-induced hepatic injury. TFA had no marked effect on paracetamol and its metabolites except for the mercapturate-conjugate. The concentration of mercapturate change decreased with increasing TFA dose. TFA had no effect on the pentobarbital metabolites (P > 0.05). However, paracetamol (400 mg.kg-1) prolonged the sleeping time (by 110 min relative to the controls, P < 0.001). The TFA (P < 0.005) caused significant reduction in paracetamol-prolonged pentobarbital-induced sleep. CONCLUSIONS: The mechanism of TFA's protective effect against the paracetamol-induced damage may be related to the inhibition of some metabolism progress of paracetamol and the reduction of the toxicity metabolite such as mercapturate-conjugate.

[Role of Rhizobium in wheat-astragalus mixed cropping system].

Inoculation experiments were carried out in wheat-astragalus mixed cropping system to study the changes of plant biomass, nitrogen content in plants and soil, and enzyme activity of Rhizobium. The results showed that in the mixed cropping system inoculated with Rhizobium, the growth of plants was stimulated, total nitrogen of plants and soil was increased significantly, and the increment of enzyme activity of Rhizobium was also found.

[In vitro selection and characterization of methionine-resistant variant in Astragalus melilotoides Pall].

A variant cell line of Astragalus melilotoides Pall. resistant to 14 mmol/L methionine was selected from calluses via mutagenesis with sodium azide. The plantlets were induced from the variant calluses. The regenerated variant plants were transplanted in soil and had set seeds. After the variant calluses had been cultured for 25 days in the medium containing 15 mmol/L methionine, the relative growth rate was 10.2 folds of the control's. Free amino acids of the aspartate family in the variant cell line increased obviously in comparison with the control. Analyses of SDS-PAGE pattern and isozyme pattern of peroxidase revealed the differences between the variant plants and the control's.

Traditional Chinese medicine Astragalus reverses predominance of Th2 cytokines and their up-stream transcript factors in lung cancer patients.

Th2 cytokine is predominant in tumor patients and was found to be associated with tumor progression. Reversing of Th2 dominant status is thought to be a promising strategy. In the present study, peripheral blood mononuclear cells (PBMNC) of 37 lung cancer patients and 19 healthy subjects were prepared and used for examination of cytokine secretion and gene expression. The positive percentage of mRNA transcripts of Th1 cytokines (8.1% for IFNgamma and 13.5% for IL-2) in patients' PBMNC were lower than those of Th2 cytokines (70.3% for IL-4, 64.9% for IL-6 and 83.8% for IL-10). The gene expression capacity (measured as relative intensity to ratio of beta-actin) of patients for Th1 cytokines was low, but constitutively relatively high for Th2 cytokines. Both positive percentage and relative intensity were lower in transcript factor for Th1 cytokine, T-bet (40.5% and 0.139, respectively) than those for Th2 cytokine, GATA3 (89.2% and 0.364, respectively). Traditional Chinese medicine, Astragalus (AG) was observed to reverse Th2 status of lung cancer. AG enhanced culture supernatant and gene expression levels of Th1 cytokine (IFNgamma and IL-2) and its transcript factor (T-bet), and reduced those of Th2 cytokines in cultured PBMNC of lung cancer patients. These results demonstrated that traditional Chinese medicine AG might reverse the Th2 predominant status in lung cancer patients, which is a probable alternative therapeutic regime in future.

[Inotropic action of Astragalus membranaceus Bge. saponins and its possible mechanism].

Astragalus membranaceus saponins (AMS) 50-200 micrograms/ml had a positive inotropic action on the isolated working heart of rats, but in the case of 30 micrograms/ml the inotropic action turned negative. The contractility recovered after washing out the AMS. Strophanthin K performed in a similar manner as AMS. AMS 500 micrograms/ml decreased the resting potential of cultured rat heart cells by 10 mV. The results suggest that AMS plays its inotropic role through influencing the Na-K-ATPase.

[Effects of Astragalus membranaceus and Tripterygium hypoglancum on natural killer cell activity of peripheral blood mononuclear in systemic lupus erythematosus].

Natural killer (NK) activity of peripheral blood mononuclear cells (PBMC) from 28 patients with systemic lupus erythematosus (SLE) was measured using enzyme-release assay. The SLE patients had significantly decreased NK activity in comparing with normal controls. The levels of NK activity correlated with disease activity. Pre-incubation of PBMC separately with Astragalus membranaceus and Tripterygium hypoglaucum or with their mixture considerably stimulated NK cytotoxicity both in SLE patients and healthy donors. The extent of enhancement was dose-dependent and relevant to pre-incubation periods. The release of a soluble natural killer cytotoxic factor (NKCF) by peripheral blood mononuclear cells was tested by cytotoxicity assay induced in K562 cells. Natural killer cytotoxic factor release was significantly lower in SLE patients than in controls. The levels of natural killer cytotoxic factor were correlated well with NK activities, but correlated negatively with clinical activity. Pre-incubated supernatants from peripheral blood mononuclear cells with above-mentioned agents caused much higher percentage of lysis on K562 targets than that of without pre-incubation.

[Effects of mixture of Astragalus membranaceus, Fructus Ligustri lucidi and Eclipta prostrata on immune function in mice].

A Chinese medicine mixture containing Astragalus membranaceus, Fructus Ligustri Lucidi and Eclipta prostrata(AFE), was fed to mice 9g/kg, 20g/kg, respectively for seven days. Observe the effects of AFE on the immune function in mice. The experiment showed that the two dosages of AFE could obviously raise the conversion percentage of lymphocytes (P < 0.01) and serum IgG level (41%-47%) of the mice. The weights of the thymus and spleensed increased in the two groups compared with those in the normal control. The larger dosage of AFE could also raise activity of Adenosine Deaminase (ADA) obviously (P < 0.01). AFE turned out a resistance to the immunosuppressive effect caused by cyclophosphamide.

[Effect of astragalus injection combined with chemotherapy on quality of life in patients with advanced non-small cell lung cancer].

OBJECTIVE: To observe the effect of Astragalus injection (AI) combined with chemotherapy on quality of life (QOF) in patients with advanced non-small cell lung caner (NSCLC). METHODS: Sixty-NSCLC patients were randomly divided into the treated group (n = 30, treated with AI combined with chemotherapy) and the control group (n = 30, treated with chemotherapy alone). Chemotherapy of MVP protocol was applied to both groups. AI was supplemented to the treated group by intravenous dripping 60 ml per day. Treatment of 21-28 days as one treatment cycle, and 2-3 treatment cycles were applied. RESULTS: The effective rate in the treated group was 40.0% and in the control group was 36.7%, the mean remission rate in the treated and control group was 5.4 months and 3.3 months, the median survival period 11 months and 7 months, and the 1-year survival rate 46.75% and 30.0%, respectively, the differences of these indexes between the two groups were all significant (P < 0.05). Moreover, the clinical improving rate and QOF elevation rate in the treated group was 80.4% and 43.3%, as compared with those in the control group (50.0% and 23.3% respectively), the difference was also significant (P < 0.01). CONCLUSION: AI combined with chemotherapy can significantly improve the QOF in NSCLC patients of advanced stage.

[Study on effects of Astragalus, Angelica and their combination on vascular endothelial cell proliferation in vitro].

OBJECTIVE: To study the effects of Astragalus membranaceus (AM), Angelica sinensis (AS) and their combination on human umbilical vein endothelial cell (HUVEC) proliferation and cells cycle. METHODS: The effects were observed and studied by means of taking the cultured HUVECs as model to determine the cell proliferation with MTT method, cell cycle was analyzed with cytometry, and vascular endothelial growth factor (VEGF) expression with SABC method. The regulatory effects of AM, AS and their combination on the HUVEC proliferation promoting were observed and studied. RESULTS: AM and AS, used singly or in combination, could promote the growth of endothelial cells, increase the cell population in S phase, the effects showed more significant when used in combination (P < 0.05 or P < 0.001). Meanwhile, VEGF expression in all the medicated group was up-regulated, but in the PBS control group, it showed only weak expression (P < 0.05 or P < 0.01). CONCLUSION: AM and AS have effect in promoting vascular endothelial cell proliferation and DNA synthesis, and showed synergistic effect when they were used in combination, suggesting that these two Chinese herbs could have certain effect on the genesis and development of neogenetic vascularization in ischemic myocardium.

[Effect of Astragalus membranaceus on baroreflex sensitivity in spontaneously hypertensive rats].

OBJECTIVE: To observe the chronicity decompression effect of Astragalus Membranaceus(AM) and evaluate the effect on baroreflex sensitivity (BRS). METHOD: Nineteen spontaneously hypertensive rats(SHR) were randomly divided into four groups. The AM groups were intraperitoneally administered with AM parenteral solution 0.9 mL, 1.2 mL and 1.8 mL respectively and the control group was not given AM for eight weeks. Then the change of blood pressure was observed successivly. After eight weeks, BRS were also determined. At last, the difference of blood pressure and BRS among the groups were compared. RESULT: Blood pressure in the control group became higher and higher frome the third week to the eighth week, but the other SHR admistered with AM showed no changein blood pressure level. We also found that the BRS in AM group was higher than that in the control group(P < 0.01). CONCLUSION: AM can promote the BRS in SHR.

[Modulatory effect of Astragalus membranaceus on Th1/Th2 cytokine in patients with herpes simplex keratitis].

OBJECTIVE: To explore the influence of Astragalus membranaceus (AM) on serum cytokines, Th1, including interleukin-2 (IL-2) and gamma-interferon (gamma-IFN), and Th2, including interleukin-4 (IL-4) and interleukin-10 (IL-10), in patients with herpes simplex keratitis (HSK). METHODS: One hundred and six HSK patients were randomly divided into the AM treated group and the ribavirin treated group. Levels of serum IL-2, IL-4, IL-10 and gamma-IFN of all the patients and 62 healthy person, selected from donors for control group, were determined by sandwich enzyme-linked immunosorbent assay (ELISA) technique. RESULTS: Levels of serum IL-4 and IL-10 in HSK patients were significantly higher and those of IL-2 and gamma-IFN were significantly lower than those in the healthy control (all P < 0.01). These parameters were significantly improved in the patients of the AM group after treatment, but with no change in patients of the ribavirin group. CONCLUSION: AM can modulate the imbalance state of Th1/Th2 in HSK patients, improve their immune function disturbance, that shows important significance in treating HSK.

[Preventive and therapeutic effects of astragalus and angelica mixture on renal tubulointerstitial fibrosis after unilateral ureteral obstruction in rats].

OBJECTIVE: To investigate the preventive and therapeutic effects of Astragalus and Angelica mixture A&A on renal tubulointerstitial fibrosis after unilateral ureteral obstruction UUO in rats and their mechanisms. METHODS: UUO rats were randomly divided into Sham, UUO, A&A or ACEI groups. A&A, ACEI or the same amount of water was administered by gavage beginning 24 hours before UUO preparation and continued through ten days after UUO. Sera and the kidney tissues were collected from each group on the tenth day. Scr and BUN were measured. Trichrome staining, measurement of tubulo interstitial damage index and immunohistochemical studies localizing alpha-smooth muscle actin alpha-SMA , TGF-beta1, fibronectin FN , laminin LN were carried out. RESULTS: In UUO rats, the tubular-interstitial damage index, the expressions of alpha-SMA, TGF-beta1, FN and LN were all increased compared with those of Sham group. The tubulo interstitial damage index had positive correlation with expressions of alpha-SMA, TGF-beta1, FN and LN. A&A significantly ameliorated deterioration of renal function, tubulo interstitial damage index and inhibited the over-expressions of alpha-SMA, TGF-beta1, FN and LN in UUO rats. These anti-fibrotic effects were similar to those affected by ACEI. CONCLUSION: In renal interstitial fibrosis-induced UUO rats, A&A retard the progression of renal fibrosis and renal function deterioration by inhibiting myofibroblasts and suppressing TGF-beta1 expression, which may consequently result in a decreased production of extracellular matrix.

Curing of symbiotic plasmid of Mesorhizobium huakuii subsp. rengei isolated from Astragalus sinicus.

Astragalus sinicus (Chinese Milk vetch), a green manure leguminous plant, harbors Mesorhizobium huakuii subsp. rengei strain B3 in the root nodules. The visualization of symbiotic plasmid of strain B3 showed the presence of one sym plasmid of about 425 kbp. Curing of sym plasmid by temperature and acrydine orange was studied. Growing rhizobial cells at high temperature (37 degrees C) or treating the cells with acrydine orange at 50 mg/l eliminated sym plasmid of M. huakuii strain B3, which was confirmed by sym plasmid visualization and plant infection test of cured strains.

Phytochemical analysis of ethyl acetate extract from Astragalus corniculatus Bieb. and brain antihypoxic activity.

Dry ethyl acetate extract containing flavonoids was obtained from above-ground parts of Astragalus corniculatus Bieb. Seven flavonoids were isolated and identified as rutin, hyperoside, isoquercitrin, narcissin, quercetin, kaempferol and isorhamnetin for the first time. The extract was found to be practically non-toxic (acute oral toxicity > 5 g kg(-1) in mice). The extract was investigated for antihypoxic activity in two models of experimental hypoxia--haemic and circulatory. Antihypoxic activity was especially pronounced in the model of circulatory hypoxia. This effect may be attributed, at least in part, to the presence of flavonoids in the extract.

[The protective effect of Rhadiola Astragalus Codonopsis compound on myocardium in early stage of severe burn at plateau in rats].

OBJECTIVE: To investigate the severity of early myocardial injury in rats with 30% full thickness burn at plateau and the protective effects of Rhadiola Astragalus Codonopsis Compound (RACC) on the rat myocardial injury. METHODS: One hundred and four Wistar rats with 30% full thickness burn were randomly divided into RACC application (R, n = 48) and scalding group 1 (S, n = 48), and another 8 healthy Wistar rats as control group 2 (C, n = 8). Four ml of RACC was garaged into the rat stomach in R and 4 ml isotonic saline in S groups respectively, but no treatment in C group. Blood samples from the aorta were harvested in 3, 6, 12, 24, 48 and 72 postburn hours (PBH) for blood gas analysis and for the determination of the changes in myocardial enzymes. Rat heart was harvested for pathomorphological examination. RESULTS: The rat myocardial tissue injury in R and S groups was obvious at 3 PBH and ameliorated gradually thereafter, up to the degree in C group at 72 PBH. The serum levels of myocardial enzymes in R and S groups were significantly higher than those in C group (P < 0.01). Whereas the enzymes in R group were much lower than those in S group (P < 0.01). It was indicated by blood gas analysis that the pH in R and S groups was lower than that in C group (P < 0.05), while that in R group at 12 - 24 PBH was higher than that in S group (P < 0.05). In addition, the base excess in R and S groups was lower than that in C group (P < 0.01), while that in R group at 6 PBH was higher than that in S group (P < 0.05 approximately 0.01). The PaCO2 in R and S groups was evidently lower than that in C group (P < 0.05 approximately 0.01), while that in R group at 48 PBH was no different to that in C group (35.70 +/- 4.23 mmHg vs 37.50 +/- 6.53 mmHg, P > 0.05). The PaO2 in R and S groups at 3 approximately 24 PBH was higher than that in C group and decreased gradually (P > 0.05). There was no difference in SaO2 among 3 groups (P > 0.05). CONCLUSION: RACC exhibited beneficial to the protection of rat heart from myocardial injury at plateau induced by severe burn.

[Intergeneric somatic hybridization between Astragalus adsurgens Pall and Medicago sativa L].

The protoplast fusion was induced between Astragalus adsurgens Pall and Agrobacterium rhizogenes-transformed cell line of Medicago Sativa L. by use of PEG method. Putative somatic hybrid cells were selected based on the observation that only intergeneric hybrid cells continued to divide on the DPD medium without any phytohormone while the cells and clusters derived from parental cells did not survive further. After being cultured, the somatic hybrid calli were obtained. Although both of the parental calli were not capable of regenerating , one of the hydrid cell lines recovered the differentiation ability and produced small shoots. Characterizations of the hybrid calli were carried out by examination of chromsome number, opine synthetase assay, isoenzyme and RAPD analyses.
