Characterization of Chilean, Bolivian, and Argentinian Trypanosoma cruzi populations by restriction endonuclease and isoenzyme analysis.

Ninety-one Chilean, 15 Bolivian, and 9 Argentinian Trypanosoma cruzi stocks, isolated from various hosts and vectors, were characterized by schizodeme analysis with EcoRI and MspI endonucleases. The three major similar pattern groups that emerged from this sample correlated with results of isoenzyme analysis. This result confirms previous work and supports the hypothesis of the clonal structure of natural populations of T. cruzi, fully defined at the level of isoenzyme analysis, quantitative kinetoplast DNA restriction fragment length polymorphism, and kinetoplast DNA hybridization analysis. In Chile, sylvatic and domestic cycles of T. cruzi transmission appear to be mainly independent: genetically different families of natural clones are specific to these cycles. Nevertheless, the possibility of overlap remains unclear. Results described here indicate that natural clones inhabiting Chilean regions appear genetically related to the natural clones identified in neighboring countries. In Chile the more frequently sampled parasite types are natural clone 39 and a genetically closely related clone NP13. In this work an evaluation of T. cruzi natural clone mixtures in T. cruzi stocks from Chile was performed for the first time by schizodeme analysis before and after serial transfer in mouse maintenance. The results indicate that six of nine stocks are composed of two or more natural clones. This observation raises the relevant question of whether specific T. cruzi natural clones generate different clinical features of Chagas' disease.

Distribution of the flux control in convergent metabolic pathways: theory and application to experimental and simulated systems.

1. Metabolic systems involving branched convergent pathways are analyzed under Flux Control Theory, obtaining a relationship between the contribution of every convergent pathway to the total flux and its Flux Control Coefficient. 2. An experimental model system is carried out to demonstrate the physical application of some conclusions of theoretical treatment. 3. Two different types of branched pathways are simulated by computer. 4. In both cases results are in agreement with the theoretical conclusions, showing in addition some new aspects on metabolic control.

Effects of epinephrine on glucose-1,6-bisphosphate and carbohydrate metabolism in skin.

1. Injection of epinephrine induced in skin a decrease in the level of glucose-1,6-bisphosphate (Glc-1,6-P2), which was accompanied by correlated changes in the activities of several enzymes which are modulated by this regulator. 2. These effects were blocked by the alpha adrenergic blocker phentolamine, in contrast to muscle where the hormone increases Glc-1,6-P2, acting through beta receptors. 3. The changes in the enzymes' activities, as well as in glycogen and lactate content induced by epinephrine, reveal that the hormone causes, in skin, a stimulation of glycogenolysis and glycolysis, as well as an acceleration of pentose phosphate pathway. 4. The reduction in glycogen content induced by epinephrine, was blocked by the beta adrenergic blocker propranolol, whereas the hormone's effects on the other processes were mainly mediated through alpha receptors.

A comparative study of Entamoeba histolytica (NIH :200, HK9, etc.), "E. histolytica-like" and other morphologically identical amoebae using isoenzyme electrophoresis.

Cultures of old, well documented strains of Entamoeba histolytica (NIH:200, HK9 etc.) have been compared with "E. histolytica-like" strains and, in turn, with E. moshkovskii, E. invadens and E. chattoni. They have been grouped according to their isoenzyme patterns. The "E. histolytica-like" organisms give patterns similar to E. moshkovskii and both of these are different from E. histolytica Groups I to IV. E. invadens and E. chattoni have easily distinguished characteristic isoenzyme bands. "E. polecki" is indistinguishable from E. histolytica.

Correlation between isoenzyme patterns and biological behaviour of different strains of Trypanosoma cruzi.

Three strains of Trypanosoma cruzi, used previously as prototypes for a classification based on the host-parasite relationships, as well as several stocks isolated from different geographical areas in Brazil, were submitted to isoenzymic analysis. Their isoenzyme patterns revealed a clear correlation with the biological data. The patterns obtained with the enzymes PGM, GPI, ASAT and ALAT permitted discrimination between each of the described types. Only one type was found in each geographical area studied, indicating a possible relationship between regional patterns and clinical presentation of Chagas' disease.

The microdistribution of isoenzymic strains of Trypanosoma cruzi in southern Bolivia; new isoenzyme profiles and further arguments against Mendelian sexuality.

132 Trypanosoma cruzi stocks were collected in southern Bolivia (99 stocks in Tupiza, 33 in Tarija), and were characterized using five enzymes (six loci). From these 132 stocks, a sample of 21 was studied using 10 enzymes (12 loci) to establish the genetic distances between them. Only five different isoenzymic strains were registered among the 132 stocks: the taxonomic status of these strains is discussed. The distribution of the strains indicated that a Founder effect was not a constant fact at the level of the house and of the suburb, but that a Founder effect was more apparent for greater geographical distances. All strains were transmitted sympatrically by the same vector Triatoma infestans. Genotype frequencies demonstrated the lack of Mendelian sexuality among stocks of T. cruzi from southern Bolivia, confirming our previous results.
