Mutations in the 5' nontranslated region of bovine viral diarrhea virus result in altered growth characteristics.

The 5' nontranslated region (NTR) of pestiviruses functions as an internal ribosome entry site (IRES) that mediates cap-independent translation of the viral polyprotein and probably contains additional cis-acting RNA signals involved in crucial processes of the viral life cycle. Computer modeling suggests that the 5'-terminal 75 nucleotides preceding the IRES element form two stable hairpins, Ia and Ib. Spontaneous and engineered mutations located in the genomic region comprising Ia and Ib were characterized by using infectious cDNA clones of bovine viral diarrhea virus. Spontaneous 5' NTR mutations carrying between 9 and 26 A residues within the loop region of Ib had no detectable influence on specific infectivity and virus growth properties. After tissue culture passages, multiple insertions and deletions of A residues occurred rapidly. In contrast, an engineered mutant carrying 5 A residues within the Ib loop was genetically stable during 10 tissue culture passages. This virus was used as starting material to generate a number of additional mutants. The analyses show that (i) deletion of the entire Ib loop region resulted in almost complete loss of infectivity that was rapidly restored during passages in cell culture by insertions of variable numbers of A residues; (ii) mutations within the 5'-terminal 4 nucleotides of the genomic RNA severely impaired virus replication; passaging of the supernatants obtained after transfection resulted in the emergence of efficiently replicating mutants that had regained the conserved 5'-terminal sequence; (iii) provided the conserved sequence motif 5'-GUAU was retained at the 5' end of the genomic RNA, substitutions and deletions of various parts of hairpin Ia or deletion of all of Ia and part of Ib were found to support replication, but to a lower degree than the parent virus. Restriction of specific infectivity and virus growth of the 5' NTR mutants correlated with reduced amounts of accumulated viral RNAs.

Telematics: a new tool for epidemiological surveillance of diarrhoeal diseases in the Aquitaine sentinel network.

A sentinel health information system using telematics and a network of general practitioners was set up in Aquitaine in south western France in 1986. Among the health problems under surveillance was acute diarrhoea. Data for each patient who fulfilled the usual case definition for acute diarrhoea were reported by general practitioners using home terminals (Minitels) connected to a central computer by telephone. Over one year 2234 cases of diarrhoea were reported, the incidence varying from 0.8 to 1.5 cases per doctor per week. Seasonal variations in incidence were observed, with peaks in the winter and in the summer. Only 379 (17%) episodes of diarrhoea were classified as severe, and these patients consulted their general practitioners earlier than patients whose diarrhoea was less severe. Foreign travel was rarely found in the patients' histories, but clusters of cases were found in communities (4.6%) and in families (22.3%). The advantages of this system were easy reporting and immediate feedback, but it was difficult to extrapolate the data, and the system was inadequate for intervening in outbreaks of diarrhoeal disease. Our knowledge of diarrhoeal diseases in south west France improved.

VIDA II: a computerised diagnostic recording system for veterinary investigation centres in Great Britain.

"Veterinary Investigation Diagnosis Analysis" II is a computerised recording system for all the diagnoses made at 33 Government diagnostic laboratories in Great Britain. It was created in 1975 and the records of 150,000 specimens are stored on it each year. The system depends on an agreed list of 393 possible diagnoses (101 cattle, 96 sheep, 72 pigs, 71 birds and 53 for other species). Diagnostic records are sent each month to a computer centre which produces an annual record of all diagnoses made and a monthly record of the number of submissions and financial charges. The computer file can be interrogated from a terminal at the epidemiology unit Weybridge and the diagnostic records are available to research workers.

Can deterministic penalty terms model the effects of synaptic weight noise on network fault-tolerance?

This paper investigates fault tolerance in feedforward neural networks, for a realistic fault model based on analog hardware. In our previous work with synaptic weight noise we showed significant fault tolerance enhancement over standard training algorithms. We proposed that when introduced into training, weight noise distributes the network computation more evenly across the weights and thus enhances fault tolerance. Here we compare those results with an approximation to the mechanisms induced by stochastic weight noise, incorporated into training deterministically via penalty terms. The penalty terms are an approximation to weight saliency and therefore, in addition, we assess a number of other weight saliency measures and perform comparison experiments. The results show that the first term approximation is an incomplete model of weight noise in terms of fault tolerance. Also the error Hessian is shown to be the most accurate measure of weight saliency.

The use of photogrammetry in tissue compensator design. Part I: photogrammetric determination of patient topography.

The surface topography of a patient can be determined by photogrammetry before beginning radiotherapy. The source light of the therapy unit or simulator is used to project a grid pattern onto the patient, and this is then photographed together with control points consisting of miniature light bulbs mounted on a frame suspended from the wedge slot of the therapy machine. When the photograph is projected onto a graphics terminal for data entry into a computer, the three-dimensional topography of the patient's surface can be reconstructed as a two-dimensional matrix of discrete points. A computer algorithm can then design a tissue compensator to fit the individual patient.

Osteosarcoma in adjacent lumbar vertebrae in a dog.

An 8-year-old male Belgian Malinois was referred for evaluation of progressive caudal paresis of 2 to 3 weeks' duration. Radiography revealed a mottled appearance to the body of L4 and misshapen intervertebral foramen at L4-L5. Myelography revealed that the dye column terminated within the body of L4. Computed tomography revealed a soft tissue mass adjacent to or involving the spinal cord and L4, with complete destruction of a portion of the floor of the vertebral foramen. Small circular lesions were also noticed within the body of L3 and L5. A left-sided hemilaminectomy was performed. Histologic examination of a biopsy specimen revealed a high-grade sarcoma. Because of the poor prognosis, the dog was euthanatized. Necropsy examination revealed osteosarcoma, with lesions in L3 to L7, the sacrum, and the lungs. Metastatic lesions in adjacent bones have been termed skip metastases and the primary tumor is typically in long bones. Prognosis associated with skip metastases is similar to or even graver than that associated with pulmonary metastases. In the dog of the present report, the unusual finding of distinct foci of osteosarcoma within 5 adjacent lumbar vertebrae and the sacrum was consistent with skip metastases, potentially spread via the vertebral venous plexus.

[The automated ECG laboratory: equipment and operational problems (author's transl)].

In the light of recent advances in technology, the basic equipment of an automated ECG laboratory is described. The main features of data acquisition terminals, data receiver/controller units, A/D converters, computers, visual displays and systems for storage and retrieval of tracings, are briefly discussed. Three major alternatives are open for computer-aided ECG interpretation today: 1) complete, dedicated system in the hospital; 2) ECG data collection system with offline analysis by hospital business computer; 3) ECG service center outside of the hospital. Advantages and possible limitations of these methods are discussed. At the Ospedale Civile Regionale of Udine we have choosen the first method. An HP 1530 ECG interpretative system and the 12-lead ECG analysis program developed by Caceres-USPHS are used. Analog tracing and interpretative printout are available in the laboratory and/or at the patient location in about one minute. Our system has been working for less than one year. At present, 150-200 ECG are processed daily. Such an ECG processing system has proven to yield considerable savings in time and manpower. Some operational problems related to shifting from manual to computer work have been gradually overcome and will be discussed.

[Identification of the gene for the immunodominant p35 protein from vaccinia virus].

A major immunodominant envelope protein p35 of vaccina virus was purified by means of extraction from virions with detergent NP-40. The protein was cleaved with CNBr, four homogenous peptides were isolated and their N-terminal amino acid sequences were determined. Computer search in a protein sequences data bank revealed that the immunodominant protein p35 of vaccinia virus is encoded by H3 gene in HindIII-H fragment of vaccinia virus genome.

Bootstrapped ultrasound calibration.

This paper introduces an enhanced (bootstrapped) method for tracked ultrasound probe calibration. Prior to calibration, a position sensor is used to track an ultrasound probe in 3D space, while the US image is used to determine calibration target locations within the image. From this information, an estimate of the transformation matrix of the scan plane with respect to the position sensor is computed. While all prior calibration methods terminate at this phase, we use this initial calibration estimate to bootstrap an additional optimization of the transformation matrix on independent data to yield the minimum reconstruction error on calibration targets. The bootstrapped workflow makes use of a closed-form calibration solver and associated sensitivity analysis, allowing for rapid and robust convergence to an optimal calibration matrix. Bootstrapping demonstrates superior reconstruction accuracy.

Structure-activity studies of novel casomorphin analogues: binding profiles towards mu 1-, mu 2- and delta -opioid receptors.

The beta -casomorphin-5 sequence was systematically modified by substitution of the naturally occurring amino acids. The derivatives are compared on the basis of their affinities towards mu 1-, mu 2 and delta -opioid binding sites estimated by means of binding studies with [3H]dihydromorphine and [3H]D-Ala2-Leu5-enkephalin as labels and computer curve fitting. A C-terminal amide group which is known to increase mu-site affinity of beta -CM-4 and beta -CM-5 mainly enhances the mu 2-site affinity. Furthermore, the Pro4-amide structure, which seems to be responsible for the affinity enhancement can be substituted by the pyrrolidide ring structure. Modifications in position 2, 3 and 4 can lead to an increase in mu 1-, mu 2- or delta -site affinity and/or selectivity. The specificity of these effects might be dependent on the respective changes in the charge or hydrophobicity due to the introduction of other amino acids. The results suggest firstly that the alternating proline residues in the beta -CM-5 molecule are not absolutely necessary for its mu-site affinities, and secondly that both opioid receptor subtype affinity and selectivity may be modified by changing charge and/or hydrophobicity in the middle part of the beta -casomorphin molecule.

Amino acid sequence determination of vaccinia virus immunodominant protein p35 and identification of the gene.

A major immunodominant envelope protein of vaccinia virus (protein p35) was purified by extraction from virions with the nonionic detergent Nonidet P-40. The protein was cleaved with cyanogen bromide. Four homogeneous peptides were isolated and their N-terminal amino acid sequences determined. A computer search of a protein-sequence data bank revealed complete identity of the determined sequences with sequences 44-63, 144-149, 154-165, and 224-238 of ORF H3 of the HindIII-H fragment of the vaccinia virus genome (Rosel et al 1986). It has therefore been established that the immunodominant protein p35 of vaccinia virus is encoded by the gene in the HindIII-H fragment of the vaccinia virus genome.

Automated microdensitometry and quantification of lipoproteins by agarose gel electrophoresis.

A major obstacle in the application of quantitative microelectrophoresis has been tedious manipulations and calculations. To overcome these difficulties, we have developed an automatic system for the microdensitometry and calculations as part of a quantitative agarose gel electrophoresis facility. Results are internally standardized by serum cholesterol and/or triglyceride measurements. The hardware consists of a densitometer, an analog to digital converter, a cathode ray tube terminal, a teleprinter, and a small computer. A program in 4K words allows sample coding, electrophoretic scan display, indexing, and systematic identification of each peak. Data are acquired from scans of electrophoretic patterns of serum alone or in combination with the 1.006 gm/ml VLDL top and/or bottom preparative lipoprotein fractions. As many as 30 scans can be stored in 4K words of memory and then sent via high-speed telephone line to a larger computer for remote processing. The analysis corrects for baseline drifts and pre-beta asymmetry and will properly identify and quantify the amount of VLDL, LDL, and HDL with corrections for "sinking pre-beta" and "floating beta" in LDL and VLDL, respectively. Results are given in milligrams per 100 milliliter as well as percentile rank and standard deviation score ranking of each lipoprotein class as compared to an appropriate normal reference population. The latter data are in a form more meaningful to the physician and patient and provide a quantitative dimension to lipoprotein phenotyping.

The Medical Gopher--a microcomputer system to help find, organize and decide about patient data.

We have developed a microcomputer-based medical workstation that does some of physicians' "gopher" work of fetching, organizing, reviewing and recording. For two years physicians have used the first version of this system to order all diagnostic tests in a general medicine clinic. They are about to use a newer version to write prescriptions and office visit notes and to find general medical and patient-specific information. Users can enter data into this system by "pointing" with a "mouse" to menu items displayed on a video terminal. In the course of a computer activity, a physician can obtain information about drugs, tests and different diagnoses, as well as about the patient. The microcomputer workstations are linked to each other and to a central hospital information system through a high-speed network link. Our physicians have accepted the initial order-entry system, and early experience suggests they prefer this faster and more sophisticated system.

A computer-assisted personnel data system for a hospital department of dietetics. I. Development of the data base.

A data base was developed for a computer-assisted personnel data system for a university hospital department of dietetics which would store data on employees' employment, personnel information, attendance records, and termination. Development of the data base required designing computer programs and files, coding directions and forms for card input, and forms and procedures for on-line transmission. A program was written to compute accrued vacation, sick leave, and holiday time, and to generate historical records.

[Component analysis of the reactions of neuronal populations in the human brain during the memorizing of visual stimuli].

The components of the evoked impulse activity of neurons and neuronal populations were studied in different nuclei of the thalamus and strio-pallidar system of the human brain, in patients treated with indwelled electrodes. The impulse activity was recorded during recognition of polygonal shapes with and without semantic meaning, during memorizing of visual stimuli without meaning. Peri-stimulus-time histograms for the cases of presentations of familiar and unfamiliar patterns, for the cases of stimulus presentation in the initial and terminal parts of the memorizing were computed and compared with each other. In some neuronal populations, a group of late components was revealed, altering during learning and depending upon subject evaluation of the meaning of the stimulus.
