[Daphnia magna Straus: a new model for evaluating the antioxidant acton of water-soluble preparations in vivo].

Hydrobionta species of Daphnia magna Straus were used as a test objects for in vivo evaluation of the antioxidant activity of three hydrophilic thiol compounds: reduced gluthatione, unithiol, and cysteine. These compounds exhibited significant differences in activity under oxidative stress conditions, in the dynamics of observed effects, and in the probability of inversion from anti- to pro-oxidant action. The main advantage of the proposed test objects in comparison to the conventional in vitro experiments (where the antioxidant effect is evaluated over a period of time from several minutes to several hours) is that the development of drug activity (pro- and antioxidant effects) can be monitored over a prolonged period of time (up to several days). In comparison to the tests on mammals, the new method is much simpler and allows the entire antioxidant protection (rather than separate systems) to be evaluated. It is recommended to use Daphnia magna Straus species for comparative evaluation of the antioxidant action of water-soluble preparations in vivo.

[Lipid peroxidation and the antioxidant system in the therapeutic effect of hypobaric hypoxia in patients with neurologic manifestations of lumbar osteochondrosis].

The hypobaric therapeutic effect dependence on the initial state of lipid peroxidation (LPO) and on its short-term activation after 3-4 sessions has been shown. The peroxidation action of hypobaric therapy (HBT) was consistent with a substantial increase of the correlation coefficient and regression between diene conjugates (DC) and tocopherol as well as a decrease of ascorbic acid and glutathione peroxidase activity. LPO activation did not take place in patients without changes in the clinical symptoms. Clinical improvement was shown by 94% of patients with the initial content of DC less than 2 o. u/ml. At higher DC values the therapeutic effect was lacking in 50% of cases and in 14% of cases where HBT was employed in combination with unithiol++. The normal level of LPO did not require the use of antioxidants in HBT. Recommendations have been made for differentiated use of HBT and of HBT plus antioxidants. The role of the prooxidant action of HBT in the formation of adaptation mechanisms is discussed.

[Relationship between the kininase- and angiotensin- converting activity under normal conditions and in experimental myocardial infarct].

In rat experiments the depressor and pressor responses to kinins and angiotensins recorded in the carotid artery were compared as to the different methods of their administration. With intravenous injection, the responses to bradykinin and kallidin were lower, and to angiotensin I--higher, than with their intra-aortic administration. The responses to angiotensin II remain identical under these conditions. The administration of all the preparations against the background of 2,3-dimercaptopropanol (unithiol) resulted in levelling of the responses irrespective of the mode of administration. In experimental myocardial infarction induced in rats by ligation of the coronary artery a reduction of the kinin-destructing and an increase of the angiotensin-converting activity was noted in the early postinfarction period. The response of the peripheral vessels to angiotensin, bradykinin and noradrenaline is decreased, while that to isoproterenol remain unchanged. The obtained results support the concept of the existence of the kininase and angiotensin-converting activity connected with a common biochemical factor of pulmonary circulation. Changes in the proportion of these functions under normal conditions and in cases of pathological states are interpreted as a particular regulation mechanism of the vascular tone and arterial pressure.

[The development of a method of nitrosoguanidine inactivation in the mutagenic treatment of bacteria].

A simple method for the inactivation of nitrosoguanidine, a mutagenic agent permitting the simplification and acceleration of the procedure used for obtaining bacterial mutants, is described. This method is based on the addition of unithiol to nitrosoguanidine solution to a final concentration of 0.25-0.5% and the subsequent normalization of the pH of the medium with 0.1 N NaOH solution.

[Criteria of pharmacologic effectiveness of antioxidant and membrane-stabilizing drugs in various disorders of cerebral circulation].

The criteria of the effectiveness of preparations possessing the antioxidative and membrane reparative properties in 135 patients with different clinical forms of disorders of the cerebral circulation were developed. It was found that the effectiveness of antioxidative and membrane stabilizing drugs in relieving consequences of free radical aggression as compared with traditional means varied and manifested itself in such an order: essenciale----alpha-tocopherol----unithiol. The presented methods of therapy and appropriate drugs may be recommended for the introduction in the complex of therapeutic and rehabilitative measures.

[Effect of antioxidants and miscleron on destabilization of cell membranes during nephritis in children].

Effects of alpha-tocopherol, unithiol and mysclerone on lipid peroxidation, activity of phospholipases and the state of a body antioxidant systems were studied in 115 patients with glomerulonephritis. Administration of these drugs caused a decrease in the content of peroxides in blood and urine, a decrease in activity of endogenous phospholipases, an increase in the content of antioxidants as well as in the activity of antioxidant enzymes. Different effects of the preparations on various steps of lipid peroxidation as well as on the activity of phospholipases and antioxidant enzymes were shown.

[A method for determining the strength of the disulphide bonds of glycoproteins from gastric mucus].

The author describes a simple and highly informative method for the assessment of disulfide bonds of gastric mucus glycoproteins, based on studies of the mucus rheology before and after incubation with 5% unithiol for an hour at 37 degrees C. Such an exposure was found to induce in normal subjects just a slight reduction of viscous elastic characteristics of the mucus (under 30% of the initial values), whereas in patients with duodenal ulcer this reduction was marked: in 83 +/- 7% of patients the rheologic characteristics dropped by more than 30%, in 1/3 of the patients the mucous gel was found completely dissolved. Such an effect was observed both in the patients with the initially reduced and with normal rheologic parameters of gastric mucus.

[Acute grey mercury ointment poisoning].

The clinical picture of poisonings with gray mercury ointment in 56 patients after its cutaneous applications is described. The patients developed gastrointestinal disorders, nephro- and hepatopathies, fever, and dermatitis. The concentration of mercury in the blood was as high as 800 micrograms/liter. The principal agent in conservative therapy is unithiol as an antidote. In grave and medium-severe poisonings hemoperfusion is the most effective modality; early administration of this measure helps appreciably alleviate the course of the disease and prevent the complications most hazardous for life (acute renal failure, etc.).

[Acute pancreatitis and biological antioxidants].

The values of lipid free--radical oxidation and the effect of various methods of treatment on these processes were studied in 164 patients with acute pancreatitis. Combined use of oil emulsions with agents possessing antioxidation activity (unithiol, tocopherol) produced the best values. Combined therapy promoted normalization of the main parameters characterizing the activity of lipid peroxidation reactions which was attended with reduced concentrations of pancreatic enzymes and increased concentration of beta-lipoproteins in the blood. This was indicative of stabilization of the pancreatic cell membrane systems.

[Study of the cytogenetic activity of radioprotectors in a human lymphocyte culture. II].

Action of certain radioprotectors: 2-mercaptoethylamine HCl (MEA), cysteine HCl, 2-aminoethylsothiouronium (AET), dithiol 2,3-dimercaptopropansulphate Na (unithiol), aminopropylaminoethylthiophosphoric acid (gammaphos), cystaphos and 5-metoxytriptamine (mexamine) was studied as applied to human chromosome aberrations induced with thio-TEPA. It was shown that with an increase in thioTEPA and protectors concentrations their effect grows.

[The action of sulfur-containing and methemoglobin-forming compounds in renal ischemia].

Rat experiments demonstrated that cystamine showed a dose-dependent effect in renal ischemia. Ischemia was induced by ligation of renal vessels and the ureter for 90 minutes. Unithiol given in a single dose before ischemia produced no protective effect. Preadministration of sodium nitrite in doses causing slight to moderate hemic hypoxia could not diminish reoxygenation ischemic lesions.

[Metabolic components of anesthesiological provision in surgery of suppurative-inflammatory diseases of the abdominal cavity].

The authors analyze the experience gained with the use of multicomponent intravenous anesthesia and epidural anesthesia in combination with a synthetic opioid dalargin, protease inhibitor trasylol, and sulfhydryl group donor unithiol. The studies were carried out in 164 patients operated on for pyoinflammatory diseases of the abdominal and small pelvis organs. Under study were the parameters of central hemodynamics, oxygen-transporting system, and lipid peroxidation. Use of the before-said metabolic components of anesthesia promoted an appreciable reduction of fentanyl and lidocaine doses (almost five- and twofold, respectively) with an adequate antistress defense being the same, hemodynamics and oxygen transport stabilized, and a membrane protective effect observed.

[The action of medichronal (glucose-glycine-formate) on the serotonin blood level in alcoholism patients].

The effects of a novel antialcoholic medication medichronal on the serotonine concentration in blood of patients with second stage alcoholism was investigated. It has been discovered that remission in 75% of the patients was connected with its hyper- and hyposerotoninemic influence, depending on the initial lowered or increased serotonine content. Similarity of medichronal and unithiol effects was observed in the form of palyndromoinversion regulation of serotoninergic activity. In a portion of the patients the increased, compared to healthy persons, serotonine concentration before and after the treatment does not differ from the average values in the whole group.

The use of haemodialysis and 2,3 propanesulphonate (DMPS) to manage acute oral poisoning by lethal dose of arsenic trioxide.

The paper discusses a case of oral acute poisoning by dental paste containing arsenic trioxide. A 33-year-old woman ingested 1/3 of the dental paste package, which corresponds to 1850 mg pure arsenic trioxide. Acute gastroenteritis and oral mucosa burn developed within 3 hours since the poisoning. Considering the amount of the ingested poison, highly exceeding the lethal dose, haemodialysis was undertaken. During the treatment, DMPS (Unithiol) was administered, with the patient's consent. The patient recovered completely, with no complications or side-effects of the therapy.

[Inhalation and peroral mucolytic therapy in mucoviscidosis in children].

The paper summarizes the results of clinical studies of 98 children with mucoviscidosis whose age was 7 to 18 years. There is research evidence for possibility and expediency of using new classes of mucolytic agents in the therapy of mucoviscidosis in children. Analysis of the rheological properties of sputum showed the advantage of inhalation route of mucolytics over their oral administration. There is evidence for the advantage of mucosolvan over carbocysteine and unithiol via all routes of administration, the optimal methods of administration and differentiated indications have been developed.

[Antioxidant system disorder in diabetic patients and the means for its normalization].

The condition was studied on the antioxidant system (AOS)--the activities of catalase, concentration of tocoferoli and reduced glutathione--in 112 patients with type I diabetes mellitus during the course of treatment. It has been established that in decompensation of diabetes mellitus there is a striking deterioration of AOS the degree of which depends upon duration and severity of the illness as well as the presence of angiopathies. Employment in the combined treatment of the donator of sulphhydric groups unithiol and a naturally occurring antioxidant tocopheroli promoted normalization of parameters associated with AOS, diabetic angiopathies included.

[Role of aldose reductase inhibitors in the development of peripheral neuropathy in experimental diabetes].

Streptozotocin-induced diabetes in rats is accompanied by development of diabetic complications such as neuropathies. It was shown that aldose reductase inhibitors (A1-1576, sorbinil and unithiol) administration partially normalized not only polyol pathway. Aldose reductase inhibitors partially restored Na+, K(+)-ATP-ase activity in sciatic nerve of diabetic rats and redox state of nicotinamide adenine dinucleotides. The sorbitol level increases in streptozotocin-diabetic rats as compared to control. Administration of aldose reductase inhibitors to diabetic rats is accompanied by partial reduction of sorbitol level in sciatic nerve. The results obtained confirm the important role of a dose reductase inhibitors, as antidiabetic drugs, in the improvement of diabetic neuropathy.
