Ultra-high performance, solid-state, autoradiographic image digitization and analysis system.

We developed a Macintosh II-based, charge-coupled device (CCD), image digitization and analysis system for high-speed, high-resolution quantification of autoradiographic image data. A linear CCD array with 3,500 elements was attached to a precision drive assembly and mounted behind a high-uniformity lens. The drive assembly was used to sweep the array perpendicularly to its axis so that an entire 20 x 25-cm autoradiographic image-containing film could be digitized into 256 gray levels at 50-microns resolution in less than 30 sec. The scanner was interfaced to a Macintosh II computer through a specially constructed NuBus circuit board and software was developed for autoradiographic data analysis. The system was evaluated by scanning individual films multiple times, then measuring the variability of the digital data between the different scans. Image data were found to be virtually noise free. The coefficient of variation averaged less than 1%, a value significantly exceeding the accuracy of both high-speed, low-resolution, video camera (VC) systems and low-speed, high-resolution, rotating drum densitometers (RDD). Thus, the CCD scanner-Macintosh computer analysis system offers the advantage over VC systems of the ability to digitize entire films containing many autoradiograms, but with much greater speed and accuracy than achievable with RDD scanners.

Evaluation of a content-analysis system for therapeutic interviews.

The Zimmer and Cowles system for content analysis was tried out on psychotherapeutic sessions and on interviews with disturbed patients. It was found that the system indeed did seem to pick up improvement in psychotherapy, and to a lesser degree, differences in speech behavior between patients with radically differing pathologies. It also was concluded that the computer program for the content analysis system is not easily portable, but a SNOBOL equivalent is made available.

The clinical evaluation of peripheral nerve regeneration using turns/amplitude analysis system in EMG.

OBJECTIVE: The aim of this study was to explore the value of turns/amplitude analysis system in EMG in evaluating peripheral nerve regeneration. METHODS: Twenty-eight patients were investigated using turns/amplitude analysis system and conventional method in EMG after nerve repair. The patients were grouped according to their muscle power and their parameters in EMG were recorded and compared among groups as well as between the two methods. RESULTS: Statistical differences of recovery percentage were shown among the groups except turns between M3:M4 using turns/amplitude system. However, none of the differences were present among them using conventional method. Among the parameters, amplitude/s advanced the fastest although it was the lowest at early stage and it also got the highest correlation coefficient (r = 0.848). CONCLUSION: Turns/amplitude system was a reliable and sensitive method for evaluating nerve regeneration during nerve repair, which is more precise than the conventional method, and the amplitude was closely related to muscle function recovery.

[Research and development of the surface EMG detection and analysis system].

To meet the requirement of developing myoelectric controller for a man-machine systems, a surface electromyography (EMG) detection and analysis system is discussed in this paper. This system consists of two parts--data acquisition hardware and signal processing software. In the surface EMG acquisition system both wire and wireless TELEMG are available. In the surface EMG processing system various methods are integrated such as the methods in time domain, the methods for frequency analysis, and also including some newly developed methods, e.g., wavelet transform and fractal analysis. This system provides a useful platform for research and development of the myoelectric controller that is widely used in rehabilitation engineering products.

Laboratory test of an automated cell analysis system for cervical screening.

An automated cell analysis system (Autoplan-MIAC) for the early detection of precancerous lesions of the cervix was tested under semi-routine conditions in a clinical cytology laboratory. A set of 1500 specimens, highly enriched with abnormal cases, was analysed. Cervical scrapings were collected in suspension and processed by cytocentrifugation for microscopy. Two slides were prepared from each sample: one for staining according to Papanicolaou for the visual reference diagnosis and one for Feulgen staining for automated analysis. The specimens were evaluated in two ways: the first one, which is referred to as the automated machine classification system (AMC), classifies the specimens according to the number and ratio of selected objects (alarms) and is a fully automated system. The second system classifies the specimens after visual evaluation of the stored alarms as they are displayed on a TV monitor, and is designated the interactive machine classification system (IMC). The AMC results showed a false positive rate of 16.5% when the cut-off threshold was selected so that all 117 positively diagnosed specimens were classified 'positive' by the system. In that case 87.4% of the CIN I and 96.9% of the CIN II cases were AMC-positive. The IMC results showed a false positive rate of 2.5%, when 86.3% of the CIN I cases, 96.9% of the CIN II cases and all CIN III and invasive carcinoma cases were positively classified.

The motion analysis system and the fingertip motion area. Normal values in young adults.

The reliability of the motion analysis system and the normal value of the fingertip motion area have been studied in young adults. The results indicate that the motion analysis system is a reliable tool for the evaluation of fingertip motion. It was found that the fingertip motion area in young adults has a linear correlation with the square of the finger length. Therefore, the normal value of the fingertip motion area can be calculated from the finger length.

A computerized analysis system in chest radiography: evaluation of interstitial lung abnormalities.

We evaluated the usefulness of a computerized analysis system in the detection of interstitial lung abnormalities in digitized chest radiography. This system uses the processes of four-directional Laplacian-Gaussian filtering, linear opacity judgment, and linear opacity subtraction. For qualitative analysis, we employed a combined radiographic index, which was calculated from two normalized radiographic indices obtained by linear opacity judgment and subtraction of linear opacities. We selected 50 regions of interest (ROIs) in patients with mild interstitial lung abnormalities, 50 ROIs in patients with severe interstitial lung abnormalities, and 50 ROIs in individuals with normal lung parenchyma. High-resolution computed radiography (HRCT) findings were used as the standard of reference for this study. These ROIs were processed by our computerized analysis system, and radiographic indices were obtained from each ROI. The area under the receiver operating characteristic curve (Az) was used as the measure of performance. The combined radiographic index provided better results in the mild interstitial lung abnormality group (Az = 0.94 +/- 0.02), but it also yielded good results in the severe interstitial lung abnormality group (Az = 0.98 +/- 0.01). These results indicate that this system of combining radiographic indices has improved the detection performance over that with our previous system.

HESAS: HERVs Expression and Structure Analysis System.

SUMMARY: HESAS (HERVs Expression and Structure Analysis System) database was developed to understand the human endogenous retroviruses (HERVs) that have an effect on the expression of human functional genes. The database products are generated by the exon-based expressed sequence tag clustering and reconstructing of partial HERV structures that result from various mutations during primate evolution. The expression types were classified according to the existence of splicing, transcriptional start and polyadenylation signal sites. The database currently contains HERV information on 26,981 human genes of exon-intron structure. The HERV elements were inserted into 17,317 of these genes and linked to expression with 898 genes. Development of a fully integrated analysis system for ions based on ion-selective optodes and centrifugal microfluidics.

A fully integrated, miniaturized analysis system for ions based on a centrifugal microfluidics platform and ion-selective optode membranes is described. The microfluidic architecture is composed of channels, five solution reservoirs, a measuring chamber, and a waste reservoir manufactured onto a disk-shaped substrate of poly(methyl methacrylate). Ion-selective optode membranes, composed of plasticized poly(vinyl chloride) impregnated with an ionophore, a proton chromoionophore, and a lipophilic anionic additive, were cast, with a spin-on device, onto a support layer and then immobilized on the disk. Fluid propulsion is achieved by the centrifugal force that results from spinning the disk, while a system of valves is built onto the disk to control flow. These valves operate based on fluid properties and fluid/substrate interactions and are controlled by the angular frequency of rotation. With this system, we have been able to deliver calibrant solutions, washing buffers, or "test" solutions to the measuring chamber where the optode membrane is located. An analysis system based on a potassium-selective optode has been characterized. Results indicate that optodes immobilized on the platform demonstrate theoretical responses in an absorbance mode of measurement. Samples of unknown concentration can be quantified to within 3% error by fitting the response function for a given optode membrane using an acid (for measuring the signal for a fully protonated chromoionophore), a base (for fully deprotonated chromoionophore), and two standard solutions. Further, the ability to measure ion concentrations by employing one standard solution in conjunction with acid and base and with two standards alone were studied to delineate whether the current architecture could be simplified. Finally, the efficacy of incorporating washing steps into the calibration protocol was investigated.

Development of a support vector machine-based image analysis system for assessing the thyroid nodule malignancy risk on ultrasound.

An SVM-based image analysis system was developed for assessing the malignancy risk of thyroid nodules. Ultrasound images of 120 cytology confirmed thyroid nodules (78 low-risk and 42 high-risk of malignancy) were manually segmented by a physician using a custom developed software in C++. From each nodule, 40 textural features were automatically calculated and were used with the SVM algorithm in the design of the image analysis system. Highest classification accuracy was 96.7%, misdiagnosing two high-risk and two low-risk thyroid nodules. The proposed system may be of value to physicians as a second opinion tool for avoiding unnecessary invasive procedures.

Nonlinear systems analysis: comparison of white noise and sum of sinusoids in a biological system.

The Gaussian white noise and the sum-of-sinusoids methods of systems analysis provide equivalent descriptions of nearly linear and strongly nonlinear transductions in the cat retina. Smoothness in the frequency domain is a common characteristic of biological transductions. This permits a substantial improvement in the signal-to-noise ratio by using the sum-of-sinusoids method, as is demonstrated for the transductions of the cat retina.

[An APC composite analysis system for analyzing the cause of perinatal death].

As the cause of perinatal death is multi-factorial and complex, its analysis should be a composite one. The method stated here includes the following: The APC analysis system. A is autopsy, P is placental pathological examination and C is the clinical data. The analysis is based mainly on the diagnosis of autopsy, with the findings of placenta as a non-negligible component part of the autopsy. As for the clinical data, they are both as the start point of the autopsy and the final check for the conclusion drawn from the pathological examination. The analysis procedure of a complex case is given in detail for example. Strictly applying this system, the causes of perinatal death were clarified in 99.3% of 137 cases, and in 98.6% (66/67) of stillbirths. The underlying factors of primary perinatal death are usually neglected, which could be found out as well with this system. For example, the underlying factors inducing intrauterine anoxia of 92.3% of cases (36/39) were determined by using the APC system. The role played by the placental pathological examination in this analysis system is clearly shown and stressed, especially in the stillbirths. The quality of causal analysis of perinatal death is greatly improved by applying the APC analysis system.

Development of a micro total analysis system incorporating chemiluminescence detection and application to detection of cancer markers.

We developed a micro total analysis system (mu-TAS) incorporating chemiluminescence detection, in which the chemiluminescence reaction of isoluminol isothiocyanato (ILITC) (as a chemiluminescence reagent for labeling)-microperoxidase (as a catalyst)-hydrogen peroxide (as an oxidant) was adopted. The analysis system performed the following three processes on a microchip: immune reaction for high selectivity, electrophoresis for formation and transportation of the sample plug, and chemiluminescence detection for high sensitivity. The three processes were compactly integrated onto the microchip to give the mu-TAS. The microchip contained two microchannels that crossed at an intersection, while the ends of the microchannels accessed four reservoirs. As the first process, the immune reaction was performed using an antibody-immobilized glass bead. The glass bead was placed in one of the reservoirs along with antigen (analyte) and a known amount of ILITC-labeled antigen to set up a competitive immune reaction. For electrophoresis, as the second process, the reactant after the immune reaction was fed electrophoretically into the intersection resulting in a sample plug. The sample plug was then moved into another reservoir containing hydrogen peroxide solution. At this point, chemiluminescence detection was performed as the third process: the labeled antigen mixed with the hydrogen peroxide and the catalyst included in the migration buffer to produce chemiluminescence. Chemiluminescence was detected by a photomultiplier tube located under the reservoir. The mu-TAS described here was capable of determining, with high selectivity and sensitivity, human serum albumin or immunosuppressive acidic protein as a cancer marker in human serum.

Development and application of the 'Sleep Ukiha' automated sleep analysis system.

We attempted to develop an automated sleep analysis system that uses a personal computer as an aid to the entire sleep research process. Analysis is based on electroencephalogram, chin muscle electromyogram and electrooculography, while other physiological phenomena can be handled arbitrarily. Major characteristics of the system include: (i) simultaneous gathering of physiological phenomena from up to three patients; (ii) high-speed waveform analysis; (iii) user-friendly operating environment through the use of a graphical user interface; and (iv) versatile utilization of analytical data in research.

A multicentre evaluation of the Boehringer Mannheim/Hitachi 704 analysis system.

The selective multitest Boehringer Mannheim/Hitachi 704 analysis system was examined according to the ECCLS guidelines in a multicentre evaluation involving four laboratories. Ten routine parameters, covering most of the application settings of the instrument, were measured in the respective laboratory at temperatures 25, 30 or 37 degrees C. The trial lasted four months and gave more than 40,000 data. It yielded the following results: 1. Within the four laboratories the mean coefficients of variation for three control sera at different concentrations were found to be equal to or better than 1.6% for the within-run imprecision and 2.8% or better for the between-day imprecision. 2. No drift was observed during eight hours. 3. Because of the high linear measuring range a re-run analysis was seldom necessary. 4. Sample-related carry-over was not seen. Reagent-dependent carry-over was measured from cholesterol to uric acid and from triacylglycerols to lipase. Through modification of the cholesterol and triacylglycerol reagents, the carry-over effect was practically eliminated. 5. The recovery of the assigned values of control sera showed average values between 99 and 104%. For bilirubin, creatinine, creatine kinase and alanine aminotransferase some control sera showed deviations greater than 10%. 6. In all cases, regression analysis of the results obtained in comparisons of the present instrument with the Hitachi 705 or 737 yielded slopes close to unity with extreme values of 0.95 and 1.06. 7. During the entire evaluation period there was no malfunction or breakdown of the instruments. The evaluators came to the conclusion that the analytical performance as well as the reliability and practicability of the Hitachi 704 can be rated as excellent.

Radionuclide imaging of miniaturized chemical analysis systems.

We propose radionuclide imaging as a valuable tool for the study of molecular interactions in miniaturized systems for chemical analysis. Sensitive and quantitative imaging can be performed with compounds labeled with short-lived positron-emitting radionuclides, such as (11)C and (68)Ga, within selected parts of the system. Radionuclide imaging is not restricted to transparent materials since the relatively energetic positrons can penetrate high optical density materials. Experimentally, a radiotracer is introduced into the object of study, which is subsequently placed on a phosphor storage plate. After exposure, the plate is scanned with a laser and a digital, quantitative image can be reconstituted. To demonstrate the concept, three types of microstructures suited for integration in chemical analysis systems were imaged with (11)C- and (68)Ga-labeled tracers. The influence of factors such as geometry of the object and type of radionuclide on resolution and sensitivity was investigated. The resolution ranged from 0.9 to 2.7 mm (fwhm). Measuring low amounts of radioactivity in the three structures, 2-20 Bq could be detected, which corresponded to 2.3-500 amol or 2.4-110 pM tracer. The imaging approach was applied to study analyte concentration and sample dilution effects on the performance of a capillary extraction column integrated in an automated LC-ESI-MS system. The utility of the technique was further illustrated by imaging of microchannels in a zeonor plastic compact disk and in a poly(dimethylsiloxane) material for the study of nonspecific peptide adsorption.

Results obtained by using a computerized image analysis system designed as an aid to diagnosis of cutaneous melanoma.

Results obtained using a computerized image analysis system as an aid to clinical diagnosis of melanoma are reported. The system comprises a colour television camera connected through a digitizing board to a 386 personal computer. By means of original algorithms able to measure the shape, the colours and texture of a pigmented lesion of the skin, the system provides eight on/off indicators that are matched with the histological diagnosis to identify benign and malignant pigmented lesions. The chances that a given lesion is malignant increase with the increasing number of positive indicators. The training field of the system was constituted of images and data of 169 cutaneous lesions in 165 patients. Taking two positive indicators as the threshold between pigmented benign and malignant lesions, the efficiency of the system is 0.98, the positive predictive value is 0.45 and the negative predictive value is 0.95. These values were confirmed in a series of 44 pigmented lesions, 10 of which were melanoma, that constitute the present test series. The authors conclude that this computerized image analysis system should be regarded as a useful aid to diagnosis for a non-expert clinician. The system limit is transformation within a naevus.

Environmental planning vs. systems analysis: four prescriptive principles vs. four descriptive indicators.

Four principles for a systems approach to sustainability, which have been suggested as prescriptive, normative and desirable, are analysed. Limitations in the principles are observed. The four principles are developed into descriptive indicators, metrics or tools that can be used in sustainability systems analysis to study important properties of all complex systems.
