The Phadiatop test, a new in-vitro test for inhalant allergy, was evaluated in relation to a RAST panel, and in comparison with total IgE. In 248 patients, aged 12-64 years, who had been prescribed pulmonary medication in general practice, the Phadiatop, total IgE (PRIST), and specific IgE (RAST) test were carried out with seven inhalant allergens common in the Netherlands. RAST was considered positive at class greater than or equal to 2 and total IgE elevated at values greater than or equal to 200, greater than or equal to 150, and greater than or equal to 100 kU/l in age groups 12-14, 15-16, and 17-64 years respectively. The result of the Phadiatop test is given as positive or negative. The predictive value of the Phadiatop test for one or more positive RAST results was 100% and its predictive value for all RAST results being negative was 97.1%, while these predictive values for total IgE were 84.7% and 71.2% respectively. It is concluded that the Phadiatop test is a valuable test to confirm or exclude sensitization to common inhalant allergens.
The clinical study for four years running of desensitization treatment to adsorbed pollen calcium phosphate, prepared "at request" by Institut Pasteur, shows that these preparations produced particularly good results. The population of allergic subjects is divided into two categories as from first year of treatment: good responders and non responders. A study on the serums of these patients, in view of a different "immuno-imprint" in these two groups, has not yet been conclusive, whereas we were expecting much from this prognosis test, which would have given us better treatment information.
The biologic role and clinical relevance of IgE (reaginic) antibodies in immediate-hypersensitivity (allergic) reactions are reviewed, and methods useful in the laboratory diagnosis and management of human allergic diseases are discussed. Quantitative immunoassays for total and allergen-specific IgE antibodies are examined within the context of their use in diagnosis. The radioallergosorbent test inhibition assay and specific IgG antibody immunoassays are described as useful methods in the planning and monitoring of immunotherapy.
Paper disc-based solid phase radioimmunoassays are widely used in vitro diagnostic test kits for measuring total IgE (Phadebas PRIST) and specific IgE antibodies (RAST). Recently, these kits have been modified by the substitution of an enzyme-linked immunosorbent assay detection system (Phadezym PRIST/RAST). We studied the performance characteristics of Phadezym PRIST and RAST kits. Phadezym PRIST was sensitive to 0.5 IU/mL IgE. Reproducibility was excellent in the range of 5 to 200 IU/mL IgE and adequate in the range of 5 to 1000 IU/mL using 1:10 serum dilutions (average inter-assay coefficient of variation = 19%). Phadezym RAST was specific, but sensitivity was limited by absorbances in the RAST class 1/0 range indistinguishable from background values. Average inter-assay coefficient of variation was 29% for the semi-quantitative 'Phadezym RAST Unit' (PRU) reporting system. We modified the test kit procedure by disc incubations in microtiter plate wells with rotational agitation and use of ELISA-dedicated spectrophotometer and computer software. These microplate accelerated computerized assays ('MacPRIST' and 'MacRAST') were shown to perform similarly to the conventional Phadezym procedures with advantages in speed, ease, and handling of data.
The measurement of total and specific IgE antibody by in vitro methods offers a significant advance in the diagnosis and management of allergic patients. These tests allow the physician to combine a precise diagnosis with an evaluation of patient sensitivity to provide care in a cost-effective manner.

The Authors report the results obtained with the histamine radioenzymatic test in the evaluation of the histamine content of granulocytes of 91 subjects, suffering from urticaria and urticaria-angioedema syndrome. The laboratory investigation was also integrated, according to the clinical implications, by other in vitro tests such as: kallikrein, RAST, PRIST, secretory IgA, precipitins assays. In urticaria-angioedema syndrome the quantitative and functional evaluation of C1-esterase inhibitor was also performed, to exclude the heredity of these pathologic forms. Basing on the results obtained, the authors expect that the granulocyte histamine radioenzymatic assay is highly reliable from the diagnostic viewpoint in the urticaria and angioedema forms.

Antisera were raised in rabbits to 6 human melanoma cell lines. The cell-surface antigens recognized by these antisera were examined using cell-surface labelling with (125)I, followed by immunoprecipitation of soluble extracts of the cells and polyacrylamide-gel electrophoresis of the immunoprecipitates in the presence of sodium dodecyl sulphate (SDS). Up to 16 cell-surface antigens were recognized by these antisera, and 5 of the melanoma cell lines had a similar profile of cell-surface antigens. Digestion of labelled melanoma cells with neuraminidase before immunoprecipitation revealed that 8 of the larger antigens were sialoglycoproteins. The melanoma antisera produced haemagglutination of human erythrocytes at high dilutions, but the antigens involved could not be detected by iodination. In contrast, absorption of the melanoma sera with lymphocytes and fibroblasts revealed that these cells did contain some cell-surface glycoproteins in common with melanoma cells. The melanoma antisera contained antibodies to foetal calf serum proteins, but the amounts of these proteins on the surface of melanoma-cells were very low. Immunoprecipitation of labelled melanoma cell extracts with monospecific antiserum to β(2)-microglobulin produced 2 bands with mol. wts corresponding to β(2)-microglobulin and the HLA-determinant polypeptide chain. After absorption of melanoma antisera with cross-linked foetal calf serum, erythrocytes, lymphocytes and fibroblasts, antibodies against 10 labelled antigens remained in the absorbed antisera. However, antibodies against 8 of these antigens were still detected after absorption of the melanoma antisera with melanoma cells.
