[Use of acridine yperite in the selection of Act. nodosus, a producer of amphotericin B].

The effect of acridine-yperite (AY) on Act. nodosus LIA 0861 at various physiological stages of development was studied. It was shown that both the spores and the germs were sensitive to AY. The level of the mutagen effect depended on the exposure time and the physiological state of the Act. nodosus cells during the treatment. The treatment of the spore suspension with AY had an insignificant effect on the strain morphological variation, while the number of the morphologically changed colonies markedly decreased when 5-hour germs were subjected to the mutagen effect. A decrease in the average antibiotic activity of Act. nodosus was observed, when its spores were subjected to the treatment with AY, which was associated with a decrease in the number of the plus variants. The treatment of 5-hour germs increased the average antibiotic activity of the culture which correlated with an increase in the number of the plus variants.

[Late manifestations in former mustard gas workers with special reference to cutaneous findings].

We report on late cutaneous manifestations following exposition to gas (in particular N- and S-LOST) which had not been observed until within the last years. In the years 1945 to 1951, inadequate protective measures during disbandment of the "Heeresmunitionsanstalt St. Georgen" caused contamination and inhalation of these radiomimetics in about 400 persons. Apart from the still today little known pulmonal and cardiac late manifestations, the surviving persons now suffer from multiple skin tumors, such as basal cell carcinomas, Bowen's disease, Bowen's carcinoma and carcinoma spinocellulare even in unexposed parts of the skin. Moreover, there are observed spontaneous flat and partly necrotic, gangrenously imposing, very painful ulcerations with tendency to spread, which keep unaffected by all therapeutic measures. The findings have been documented historically, macroscopically, histologically as well as cytologically. Presentation of these late sequele is of particular importance since at that time containers with mustard gas had been unloaded anywhere in the FRG without any special precautions, so that also contamination of citizens has to be assumed. For this reason, similar findings are to be expected throughout the country, accumulated in certain regions. This additional aspect has also to be considered concerning the application of these radiomimetics therapy of psoriasis ("Russian ointment").

Formation of O6-ethylthioethyldeoxyguanosine from the reaction of chloroethyl ethyl sulfide with deoxyguanosine.

O6-Ethylthioethyldeoxyguanosine has been synthesized from 6-chloro-3',5'-di-O-acetyldeoxyguanosine and characterized by UV, fluorescence, and mass spectrometry. High-pressure liquid chromatography studies have shown that this modified nucleoside is formed when the one-armed sulfur mustard, chloroethyl ethyl sulfide, reacts with deoxyguanosine. This result supports the hypothesis that the mutagenic effects of the sulfur mustards are caused in part by substitution of the O6-position of deoxyguanosine.

[Syntopy of Bowen's disease and Lewisite scar].

Description of a case of Bown's disease in an area of the right lower leg after lewisite contamination 30 years ago. The possible influence of lost derivatives on carcinogenesis is discussed.

[The treatment of ptyalism in children with cerebral lesions (author's transl)].

The cytotoxic and alkylating substance, mustard gas, causes both acute poisoning and also damage to numerous organs following chronic exposure. Especially important is the carcinogenic effect, also confirmed in humans. 34 years after occupational exposure to mustard gas with many intoxication episodes, a former munition-worker died of a glioblastoma; two years before his death a neurofibroma was detected in the thorax and removed by operation. The causal connection between the mustard gas exposure and the development of two neurogenic tumors rests on the statistically significantly raised frequency of malignant tumors and the established psychic changes in former workers with mustard gas and especially on the production of malignant tumors of the central nervous system in experimental animals with alkylating nitrosamines.

[Genesis of malformations of the distal parts of chick embryo extremities (adactylia, hemimelia)].

Two series of experiments were carried out on chick embryos, stages 24 to 28 pairs of somites. (1) Denuded mesenchyme obtained from leg presumptive area previously treated with nitrogen mustard, was grafted into the right flank of embryos, stages 15 to 24 pairs of somites. (2) Conversely, denuded mesenchyme was implanted into the flank of nitrogen mustard-treated embryos. In most of the embryos, a supernumerary abnormal limb appeared; 58% of the legs produced in the first set of experiments had distal parts, whereas only 15% of the legs in the second set of experiments showed toes. Most of these legs had a hemimelic aspect or were reduced to stumps. This study demonstrates that deficiencies localised in the ectodermal apical part of the bud may be responsible for the stumpy limb defect.

[Electrophoretic examination of changes in the protein spectrum of dogs after exposure to beta, beta'-dichlorodiethylsulfide].

In serum proteins, the alpha 3 + 4-globulin subfraction increases with a corresponding decrease of albumins. A statistically significant rise was observed from the 24th-48th hour from application with a progressive tendency reaching its maximum in the terminal stage of intoxication. This elevation reaches the values of 100% of the initial control values in the terminal stage. The drop in albumins is less significant and almost reciprocal with the rise of alpha-globulin content. In the values of beta-globulins, a change was observed in the quotient of beta 1/beta 2 globulins. No changes were ascertained in the values of gamma-globulins. Our results correspond to the data published by Gjessing and Chanutin, who observed a drop in albumins and rise in alpha-globulins after percutaneous implantation of the substance. The results suggest that the latent time between implantation and a statistically significant rise of the alpha-globulin fraction in the serum is 24-48 hours.
