The common, Near-Eastern origin of Ashkenazi and Sephardi Jews supported by Y-chromosome similarity.

About 80 Sephardim, 80 Ashkenazim and 100 Czechoslovaks were examined for the Y-specific RFLPs revealed by the probes p12f2 and p49a,f on TaqI DNA digests. The aim of the study was to investigate the origin of the Ashkenazi gene pool through the analysis of markers which, having an exclusively holoandric transmission, are useful to estimate paternal gene flow. The comparison of the two groups of Jews with each other and with Czechoslovaks (which have been taken as a representative source of foreign Y-chromosomes for Ashkenazim) shows a great similarity between Sephardim and Ashkenazim who are very different from Czechoslovaks. On the other hand both groups of Jews appear to be closely related to Lebanese. A preliminary evaluation suggests that the contribution of foreign males to the Ashkenazi gene pool has been very low (1% or less per generation).

Blood groups in the Chueta community (Majorcan Jews).

A sample of 443 Chuetas (descendents of Majorcan Jews) was typed for the ABO, Rh, Lewis, Duffy, MNSs, Kell and P blood groups. Significant deviations from the Hardy-Weinberg equilibrium were observed in the MNSs and Duffy systems with a deficiency of heterozygotes. The gene frequencies were compared with those of the Balearic non-Jewish populations and significant differences were found. Genetic distances and cluster analysis demonstrated that the Chuetas resemble more their neighboring non-Jewish populations than other Jewish populations. Discriminant analyses showed that the Chuetas and other Jews considered in this study do not resemble each other but their host peoples with respect to these markers.

A mutation in CYP11B1 (Arg-448----His) associated with steroid 11 beta-hydroxylase deficiency in Jews of Moroccan origin.

Steroid 11 beta-hydroxylase (P450c11) deficiency (failure to convert 11-deoxycortisol to cortisol) causes less than 10% of cases of congenital adrenal hyperplasia in most populations, but it is relatively frequent in Jews of Moroccan origin. P450c11 is encoded by the CYP11B1 gene which is located on chromosome 8q22 along with a homologous gene of unknown function, CYP11B2. To identify mutations in CYP11B1 associated with 11 beta-hydroxylase deficiency in Moroccan Jews, oligonucleotides were used that selectively amplified portions of CYP11B1 in polymerase chain reactions without amplifying CYP11B2. Sequence analysis of amplified fragments from one patient revealed a single base substitution in exon 8, codon 448 from CGC (arginine) to CAC (histidine). This residue is within the "heme binding" peptide that contains a cysteine that is a ligand to the heme group. The equivalent of Arg-448 is found in every known eukaryotic P450, and therefore it seems likely that a mutation of this residue would adversely affect enzymatic activity. 11 of 12 affected alleles from six Moroccan Jewish families carried the mutation in codon 448. This mutation is not normally present in CYP11B2 and thus appears to have arisen in CYP11B1 as a true point mutation rather than a gene conversion.

Differential perceptions of the seriousness of male violence against female intimate partners among Jews and Arabs in Israel.

Crime seriousness studies have consistently shown wide consensus regarding the high perceived seriousness of violent offenses. However, socio-cultural models suggest wide variability between social groups with regard to the seriousness of certain types of violent crime. This is particularly true in the case of male violence against female intimate partners. The present study examines these contradictory propositions in Israel focusing on its two main ethnic groups--Jews and Arabs. In an attitudinal survey, respondents were required to evaluate the seriousness of hypothetical crime scenarios representing a number of criminal offenses including male violence against female intimate partners. As expected, the findings revealed significantly more permissive attitudes to male violence against female intimate partners among Arab respondents than among Jews. The implications of these findings are discussed.

Ashkenazi Jews and breast cancer: the consequences of linking ethnic identity to genetic disease.

We explored the advantages and disadvantages of using ethnic categories in genetic research. With the discovery that certain breast cancer gene mutations appeared to be more prevalent in Ashkenazi Jews, breast cancer researchers moved their focus from high-risk families to ethnicity. The concept of Ashkenazi Jews as genetically unique, a legacy of Tay-Sachs disease research and a particular reading of history, shaped this new approach even as methodological imprecision and new genetic and historical research challenged it. Our findings cast doubt on the accuracy and desirability of linking ethnic groups to genetic disease. Such linkages exaggerate genetic differences among ethnic groups and lead to unequal access to testing and therapy.

[The persecution of Jews during the time of the plague (1349-50) in southern Netherlands].

Research on the persecutions of the Jews at the time of the 'Black Death' in the Southern Low Countries has overemphasized the responsibility of the flagellant movement. In fact, the specific role of the flagellants was confined to spreading the rumor that the Jews were conspiring to kill all Christians by well-poisoning. A closer look at three examples--the persecutions in Hainaut, Brussels and Leuven--reveals that the driving forces behind the pogroms and executions were local and municipal authorities, while the countess of Hainaut and the duke of Brabant were too weak politically to defend the lives of their Jewish subjects.

[The Black Death as a cause of the massacres of Jews: a myth of medical history?].

In the middle of the 14th century, most towns in German-speaking territories and beyond massacred their Jewish communities. Thousands of Jews were burnt, often connected with accusations of well-poisoning. Medical and socio-historical literature usually attributes these massacres to the anxiety created by the Black Death, which was sweeping over Europe during this period. This article argues that there is no direct link between the massacres and the plague. How other researchers showed before, far from acts of plague-terrified, frenzied mobs, the massacres were the carefully planned and executed work of the Christian local governments. In addition, the slaughtering of Jews began long before the Black Death broke out in Europe. No relation can be found between the intensity of the disease and the violence of the murderers, even though there were wide regional differences. Causes of the persecutions other than the effects of plague seem evident, mainly religious fears fueled by the Church, financial profit, and political interests. This article wants to draw the attention to a myth in the history of medicine, the myth of the plague as the main cause of the massacres in the 14th century. It also raises the question, whether the plague as a trigger for the massacres really was a basic requirement.

[Jews and physicians].

The purpose of this paper is to examine the well established yet hardly explored connections between the Jews and the field of medicine. In other words, it is our intention to inquire into the reasons, sometimes historical and social, sometimes cultural and religious - or all of these at the same time - which have led and maybe still lead - such vast numbers of Jews to embark upon a course of study in medicine. Reasons which might be closely linked with their traditional perception of the figure of the father, of the rabbi, maybe even of God.

Soviet Jews in the United States: an analysis of their linguistic and economic adjustment.

"This article reviews the literature and analyzes 1980 Census data to study English language fluency and earnings among Soviet Jews [in the United States]. The literature review reveals: 1) the importance of employment and attaining premigration occupational status for self-esteem; 2) the difficulty of adjusting to the wide range of choices in the United States; 3) the greater difficulty and economic importance of learning English; and 4) the rapid linguistic and economic mobility. The multivariate analysis supports the latter two points. Soviet Jews have a difficult initial adjustment, but after five years in the United States they achieve parity in English fluency and earnings with other European immigrants, ceteris paribus."

Mortality patterns among elderly American Jews.

"Mortality studies of ethnic and religious subgroups within a nation are of interest as they provide indicators of health differentials that may result from differences in life style and risk-factor exposures. The mortality experience of North American Jews has been documented over many years and is of particular interest because of the unusual pattern that has been observed, a crossover from relatively low rates at younger ages to relatively high rates at older ages. This study examines mortality in 1979-81 among more than 100,000 Medicare enrollees who held 22 surnames common among American Jews. The findings substantiate those of a recent mortality study of a Canadian Jewish population which indicated more rapid improvement in life expectancy among elderly Jewish than non-Jewish males, and a lessening of the relative disadvantage of elderly Jewish women."

[Bernard Schapiro--an orthodox Jew as an early andrologist in the 20th century].

The unusual history and professional background of one of the first andrologists is reported. Bernhard Schapiro, born in 1888 in Dvinsk in Latvia, then a city in the Russian Pale of Settlements for Jews, grew up as an orthodox (hassidic) Jew receiving exclusively talmudic lessons until he was 18 years old. During the final years of this period of life he was educated at the famous Slobodka Talmud Academy Kenesset Israel in Kovno where he absorbed the ideals of Musar-doctrines, thus being influenced for the rest of his life. The Rogachover Gaon J. Rozin supported his desire to study medicine. After a brief stay in Frankfurt/Main he acquired by own efforts the necessary general knowledge to matriculate for access to university. Medical studies at Zurich University (1913-1919) were followed by a one-year-internship at a dermatologic department in Breslau/Silesia. The thesis for his doctorate at Zurich University in 1920 was on, Relations between Nodular Erythema and Tuberculosis'. He spent two years training in dermatology at Breslau University under Jadassohn. Back in Berlin, he married and had four children, while he worked at Magnus Hirschfeld's Institute for Sexual Sciences. After initial clinical studies in venerology he more and more turned to andrological problems, including treatment of underdeveloped male genitals, premature ejaculation and impotence in general. In this context he tested the new drug Praehormon and developed the two remedies Testifortan and Praejaculin. He was the first to describe the effect of anterior pituitary lobe hormone on the descent of cryptorchid testicles, thus initiating a treatment modality still in motion today. When Hitler came to power he and his family were spared as Swiss citizens, but he lost his base for working after the Institute was looted. He established an andrologic practice at Zurich. What he had witnessed in Germany caused him to set up a Swiss branch of Mizrachi, the spiritually based center of Zionism in Switzerland. He left Europe in 1940 for New York/USA. After compulsory repetition of medical exams in a new language he managed to establish a new existence as andrologist. His practice flourished, the more as he was trusted to treat Jewish patients according to Jewish Law. The decisive step brought him to Jerusalem, in 1951, where he founded and headed an Endocrinologic Department of the University, pursuing andrologic questions, until his death on December 31, 1966. What actually controlled his life can be read on his tombstone: 'He remained a pupil of Slobodka all the days of his life'.

Crohn's disease and the Jews.

Several epidemiologic studies of Crohn's Disease (CD) from the northern hemisphere show high rates of the disease among Jews. Israel has absorbed many Jews from those countries and the rates of CD in Israel are lower than in their place of birth. Important, too, among immigrants from the middle east Mediterranean countries, the rates appear to be higher than those in their countries of origin, where the disease is rare. CD has a genetic predisposition, but we physicians cannot ignore the environmental factors.

Y-chromosome-specific haplotypes of Jews detected by probes 49f and 49a.

A sample of Ashkenazic and Sephardic Jews has been studied with respect to haplotypes at the 49f-49a Y-specific DNA probes. Only seven haplotypes were found in Jews, three of them (VII, VIII, and XI) being the most widespread. Haplotype distribution in the European non-Jewish population is different.

Infantile spasms: neurological and developmental follow-up--a comparison between two ethnic groups: Israeli Jews and Bedouin in the South of Israel.

BACKGROUND: The aim of this study was to examine all cases of infantile spasms (IS) diagnosed at the Soroka University Medical Center, Ben Gurion University, Israel during 1981--1997. MATERIAL/METHODS: 31 children, 17 (55%) males. 17 (55%) were Jews and 14 (45%) Bedouins. Four (13%) died. Data was gathered from hospital files, neuropediatric unit and Zusman Child Development Center. Demographic and ethnic data, characteristics of the disease type of seizures, EEG pattern imaging studies, type of treatment, psychomotor development, rehabilitation and educational services were analysed. RESULTS: Mean age at diagnosis 7.22 months. Etiology for one third was pre- or perinatal insult, one third postnatal and one third unknown. 26 (84%) were symptomatic and five (16%) cryptogenic. Significant statistical difference was found with more Bedouin children symptomatic with moderate or severe mental retardation, cerebral palsy with severe motor difficulties and recurrence of the disease. Statistically significant correlation existed between poor response to initial treatment and placement in special education, recurrence of disease and cerebral palsy with moderate or severe motor difficulties, the appearence of a different type of epilepsy during follow-up and placement in special education. 18 children (58%) received first treatment with ACTH, 10 (32%) children with IVIG (immunoglobolin iv), 3 (10%) with neither ACTH nor IVIG. Regarding therapy response we found no significant difference between Jews and Bedouins. CONCLUSIONS: No significant statistical difference was found between the two treatments concerning clinical course or developmental follow-up. Due to small numbers multi-center research is needed.

mtDNA polymorphism in two communities of Jews.

Twenty-one types of mtDNA were found in a survey of 39 Israeli Jews, of whom 18 were Sephardic and 21 Ashkenazic. The survey was made with six restriction enzymes that together recognize an average of 600 bp/genome. The differences among the types appear to be due to base substitution at 19 cleavage sites, one deletion, and one conformational mutation. The numbers of differences imply that these modern Jews stem from a minimum of 21 maternal lineages that were already distinct from one another 4,000-5,000 years ago. In three of the four cases where a type was found in more than one person, it occurred in both Ashkenazic and Sephardic populations. The diversity of types in the combined sample of two Jewish populations is lower than both that in a sample from various parts of Africa and that in a sample from various parts of East Asia. Nevertheless, it is as high as that in a sample from diverse parts of New Guinea, an area much larger than that in Israel to which the Jewish population traces back.

Alleles at the PRB3 locus coding for a disulfide-bonded human salivary proline-rich glycoprotein (Gl 8) and a null in an Ashkenazi Jew.

From electrophoretic analysis, we identified in the saliva of an Ashkenazi Jew a disulfide-bonded major glycoprotein variant (Gl 8) that is a product of the proline-rich protein (PRP) locus PRB3. A previous study of this variant protein misidentified it as Pa 2 and as a product of a different PRP locus. The other PRB3 allele in this individual is an apparent null. To identify the mutations, we sequenced the tandemly repetitious exon 3 (the major protein-coding portions) of both alleles. A CGT----TGT (Arg----Cys) mutation was found in one allele (PRB3Scys), which accounts for the disulfide-bonded and peroxidase-modifying properties of Gl 8. A single nucleotide insertion was found in the other allele (PRB3Mnull) that leads to a frameshift with a premature termination codon that causes an apparent lack of gene expression. Null alleles are frequent at PRP loci coding for basic and glycosylated PRPs, and the mechanism described might explain other null phenotypes among PRPs. From nucleotide comparisons, a model of intragenic unequal crossing-over is proposed to explain, in part, the generation of the PRB3Mnull allele. The Gl 8 protein variant is found in Ashkenazi Jews (gene frequency around .008) but not in the general white, black, or Japanese populations. It is interesting that products of different PRP genes, Gl 8 from PRB3 and Pa 1 from PRH1, are both disulfide bonded and probably modify salivary peroxidase (part of an important intraoral antibacterial system) through formation of disulfide-bonded heterodimers.

Segregation analysis of idiopathic torsion dystonia in Ashkenazi Jews suggests autosomal dominant inheritance.

The results of segregation analysis applied to a family study of idiopathic torsion dystonia in Ashkenazi Jews are reported. The study is based on 43 probands (with age at onset prior to 27 years) from 42 nuclear families; pedigrees were extended systematically through all available first- and second-degree relatives, who were directly examined and videotaped. Final diagnoses were based on exam information and blinded videotape review. Segregation analysis demonstrated that the data are consistent with autosomal dominant inheritance with 30% penetrance. Recessive and polygenic inheritance were strongly rejected. There was no evidence for sporadic cases or new mutations. The high incidence and dominant inheritance of early-onset idiopathic torsion dystonia in Ashkenazi Jews suggests genetic homogeneity within this population, making it especially useful for linkage studies of this disorder.

Inheritance of idiopathic torsion dystonia among Ashkenazi Jews.

The mechanism(s) of inheritance of primary dystonia are unclear. An autosomal recessive form among Ashkenazi Jews and an autosomal dominant form among non-Jews have been proposed. However, the patterns of inheritance, particularly among Ashkenazim, are controversial. In this report we have reviewed the literature particularly as it pertains to the mode of inheritance among Ashkenazim. We also report the results of a pilot study of the families of 25 independently ascertained Ashkenazi probands with onset of primary dystonia before age 27 years. A total of 91/98 living first-degree relatives were examined; of these 91, 86 were greater than or equal to 8 years of age at time of examination and were included in our analysis. Overall, 14/86 (16.3%) of first-degree relatives were affected. We found 11.4% (4/35) of parents, 22.2% (8/36) of siblings, and 13.3% (2/15) of offspring were definitely affected. This finding of an approximately equal risk to parents, siblings, and offspring is consistent with autosomal dominant transmission with a minimum penetrance of 32.6%. Our findings do not support autosomal recessive or multifactorial inheritance.
